Effects of maternal exposure to ammonium perchlorate on thyroid function and the expression of thyroid-responsive genes in Japanese quail embryos.

Perchlorate, a known thyroid disruptor, is deposited in eggs of exposed female birds, raising concerns that the embryos from these eggs may become hypothyroid, which may in turn affect the development and function of thyroid-dependent organs. We hypothesized that exposure to ammonium perchlorate (AP) would decrease hen and embryonic thyroid function and affect the expression of thyroid-responsive genes in embryonic brain and liver. Laying Japanese quail hens were treated with 2000 mg/l or 4000 mg/l AP in drinking water. Thyroid status and expression of thyroid-responsive genes were examined in

Fetal exposure to alcohol as evidenced by fatty acid ethyl esters in meconium in the absence of maternal drinking history in pregnancy.

The detection of fatty acid ethyl esters (FAEE) in neonatal meconium has been proposed as a novel screening method for intrauterine exposure to alcohol. We investigated the potential use of meconium FAEE screening in a high-risk neonatal population in the absence of maternal drinking history. One hundred forty-two meconium samples of neonates suspected of intrauterine illicit substance exposure and referred to the Motherisk Laboratory were analyzed for the existence of drugs by enzyme-linked immunosorbent assay (ELISA) and FAEE by gas chromatography-flame ionization detection (GC-FID). Effects of HCFC-123 exposure to maternal and infant rhesus monkeys on hepatic biochemistry, lactational parameters and postnatal growth.

Peroxisome proliferators are a class of nongenotoxic rodent hepatocarcinogens that cause peroxisome proliferation and liver tumors when administered to rats and mice; but other species, including guinea pigs, dogs, and primates are less sensitive or refractory to the induction of peroxisome proliferation. Therefore, rodent peroxisome proliferators are not believed to pose a hepatocarcinogenic hazard to humans. Some peroxisome proliferators produce developmental toxicity in rats that is expressed as suppressed postnatal growth. To evaluate the relevance of the rat developmental effect to primat

Effects of in utero ethanol exposure and maternal treatment with a 5-HT(1A) agonist on S100B-containing glial cells.

This laboratory previously showed that in utero ethanol exposure severely impairs the development of the cell bodies and projections of serotonin (5-HT) neurons, and that maternal treatment with a 5-HT(1A) agonist prevents many of these abnormalities. Others demonstrated that stimulation of fetal astroglial 5-HT(1A) receptors increases production and release of S100B, a glial trophic factor that is essential for the development of 5-HT neurons. The present study investigated a potential mechanism by which ethanol hinders development of 5-HT neurons, and by which maternal 5-HT(1A) agonist treat

Maternal exposure to atrazine during lactation suppresses suckling-induced prolactin release and results in prostatitis in the adult offspring.

The availability of prolactin (PRL) to the neonatal brain is known to affect the development of the tuberoinfundibular (TIDA) neurons and, as a consequence, lead to alterations in subsequent PRL regulation. Without early lactational exposure to PRL (derived from the dam's milk), TIDA neuronal growth is impaired and elevated PRL levels are present in the prepubertal male. These observations, combined with the finding that alterations in PRL secretion (i.e., hyperprolactinemia) in the adult male rat have been implicated in the development of prostatitis, led us to hypothesize that early lactatio

Perinatal maternal ethanol effects on pregnant mice and on offspring viability and growth: influences of exposure time and weaning diet.

C57BL/6 mice were maintained on liquid diets containing 27.5% ethanol-derived calories beginning on gestational day 12. For one group, the liquid diet was replaced with laboratory chow on the morning of gestational day 18, approximately 24 hr before delivery. For a second group, the diet was continued for 7 days postnatally. Two additional groups were pair-fed isocaloric sucrose diets, and a final group was maintained on normal laboratory chow. The mice consumed sufficient amounts of diet to provide near 30 g ethanol/kg/day during pregnancy and slightly greater amounts during the first postpar

Gestational Exposure to Tri-n-butyl Phosphate Induces Maternal Insulin Resistance and Glucose Intolerance by Antagonizing Estrogen Receptor Alpha.

Epidemiological evidence links tri-n-butyl phosphate (TNBP) exposure to gestational diabetes mellitus (GDM), but mechanistic insights remain unclear. This study demonstrated that gestational TNBP exposure induced a GDM-like phenotype in pregnant mice, characterized by glucose intolerance, but not in nonpregnant female mice. This pregnancy-specific susceptibility might be attributed to significantly higher internal levels of TNBP and its metabolites dibutyl phosphate (DNBP) and dibutyl-3-hydroxybutyl phosphate (3-OH-TNBP) than the nonpregnant mice, subsequently exacerbating insulin resistance i

Maternal deprivation and adolescent alcohol exposure induce sex-dependent alterations in stress-related behavior and lipid signaling in rats.

BACKGROUND: Early-life stress constitutes a major risk factor for the development of neuropsychiatric and substance use disorders, exerting enduring effects on stress responsivity and emotional behavior. Maternal deprivation (MD) is a well-established model of early adversity that induces persistent neurobiological alterations. During adolescence, alcohol exposure presents an additional challenge that may interact with early-life stress, potentially influencing long-term adaptations in a sex-dependent manner. Among the systems involved, lipid signaling pathways, including the endocannabinoid s

Associations between prenatal exposure to ambient air pollutants and preterm birth in the Atlanta African American maternal-child cohort.

BACKGROUND: Substantial differences in preterm birth (PTB) persist across populations, with African Americans facing the highest rate. While air pollution contributes to PTB, its role in these uneven health burdens remains poorly understood. Studies of PTB subtypes might uncover racial variation in etiology, yet the impact of air pollution is largely understudied. We assessed the associations of prenatal exposures to ambient fine particulate matters (PM2.5), carbon monoxide (CO), and nitrogen oxides (NOx) with PTB and its subtypes using data from the Atlanta African American Maternal-Child Coh

Fetal vascular malperfusion and other placental lesions in relation to maternal SARS-CoV-2 exposure: a case-control study.

BACKGROUND: The morphologic characteristics of placentas associated with coronavirus disease-19 (COVID-19) have been described but with varying findings. Studies conducted at the onset of the pandemic reported frequency of Fetal Vascular Malperfusion (FVM), an umbrella term that describes occlusive vasculopathies that affect fetal circulation and their associated sequelae. Morphologic manifestations include thrombosis, avascular villi and vascular stromal karyorrhexis. Associated outcomes include fetal growth restriction, thromboembolic events such as neonatal stroke and fetal demise. Placenta

Associations of ambient air pollution exposure during pregnancy with maternal and neonatal thyroid function: Systematic review and meta-analysis.

Thyroid function in both pregnant women and newborns is vital for ensuring healthy child growth and development. Existing evidence suggests that exposure to air pollutants during pregnancy may disrupt maternal and neonatal thyroid function; however, findings across studies remain inconsistent. This systematic review and meta-analysis included 24 studies (55,814 pregnant women and 84,645 neonates) published until April 30, 2025. Results indicated that each 10 μg/m3 increase in PM2.5 was associated with a significant decrease in maternal FT4 levels (-0.42%; 95% CI: -0.83%, -0.01%) and neonatal F

Embryo-Fetal Development and Maternal-Fetal Exposure of the Von Willebrand Factor Binding Aptamer Rondaptivon Pegol (BT200) in Rabbits.

Rondaptivon pegol (BT200) is a PEGylated RNA aptamer that prolongs von Willebrand factor (VWF) and factor VIII half-lives in patients with hemophilia A and von Willebrand disease (VWD) type 2B. Embryo-fetal safety is particularly relevant given the disproportionate disease burden in women with VWD. Data on placental transfer and developmental safety of PEGylated aptamers remain limited. An embryo-fetal development study was conducted in pregnant rabbits. Animals received rondaptivon pegol subcutaneously (0, 1.5, 5, or 15 mg/kg/day) from gestational days (GDs) 7-19. Maternal toxicokinetic sampl

Maternal Thyroid Supplementation Prevents Autistic-relevant Social Behavior and Hypothalamic Oxytocin Depletion Produced by Developmental Exposure to Environmental Toxicants.

Environmental toxicants that target the developing brain are suspected of contributing to autism spectrum disorder risk but causative evidence is lacking. We and others have shown that the indoor flame retardants, polybrominated diphenyl ethers (PBDEs), reduce prosocial behavior, however, few studies have assessed the central targets and underlying mechanisms. PBDEs are well established endocrine disruptors of the expanded thyroid system, which also regulates the prosocial neuropeptides oxytocin (OXT) and vasopressin (AVP) and their hypothalamic signaling. The potential role of PBDE-induced th

Exploratory MIA study: multimodal magnetic resonance imaging characteristics of rat offspring induced by maternal Poly(I:C) exposure during pregnancy.

BACKGROUND: Maternal infection or inflammatory stimulation during pregnancy can disrupt fetal brain development through immune activation, increasing the risk of psychiatric disorders in offspring, including schizophrenia. Using a maternal immune activation (MIA) model induced by polyinosinic-polycytidylic acid (Poly(I:C)) during pregnancy, this study employed multimodal magnetic resonance imaging (MRI)-including T2 structural imaging, diffusion tensor imaging (DTI), arterial spin labeling (ASL), and magnetic resonance spectroscopy (MRS)-to comprehensively assess offspring brain alterations ac

Developmental programming: gestational exposure to excess testosterone disrupts maternal steroid homeostasis and perturbs the steroid-lipid relationship in sheep.

Gestational hyperandrogenism may disrupt the steroid and lipid metabolism homeostatic balance that is important for pregnancy progression. We hypothesized that excess gestational testosterone would disrupt the maternal steroid profile and the steroid-lipid relationship. Using sheep, we investigated maternal delta-4 (Δ4) and delta-5 (Δ5) steroids and the steroid-lipidome relationship in gestational testosterone excess (n = 12, 100 mg T-propionate days 30-90 of gestation twice-weekly intramuscularly) and control (n = 5, vehicle) Suffolk sheep. Steroids were measured using liquid chromatography-t

Ductus Arteriosus Variability Linked to Maternal Drug Exposure: Assessment of the USFDA Adverse Event Reporting System using Disproportionality Analysis.

INTRODUCTION: The ductus arteriosus is a vital fetal vessel connecting the pulmonary artery to the aorta, facilitating blood flow away from the non-functional fetal lungs. Drug exposure during pregnancy may affect DA physiology, leading to conditions like premature DA closure, DA stenosis, or Patent Ductus Arteriosus (PDA). To identify drugs with maternal exposure that may be linked to alterations in the fetal and neonatal Ductus Arteriosus (DA). This study examines associations between various drugs and alterations in DA using data from the USFDA Adverse Event Reporting System (AERS) through …

Modifying effects of prenatal exposure to rare earth elements on birth outcomes through maternal thyroid function in early pregnancy.

The potential health effects of exposure to rare earth elements (REEs) remain largely unexplored. This prospective cohort study aimed to elucidate the association between early pregnancy REE exposure and maternal thyroid function, as well as neonatal birth outcomes, in a cohort of pregnant women in Beijing, China. Additionally, the study explored the mediating role of thyroid homeostasis in the effects of REE exposure. Serum concentrations of fifteen REEs, along with Free Thyroxine (FT4), Thyroid Stimulating Hormone (TSH), and Thyroid Peroxidase Antibodies (TPOAb), were measured in 195 pregnan
