The Influence of DNA Methylation on Bone Cells.

DNA methylation in eukaryotes invokes heritable alterations of the of the cytosine base in DNA without changing the underlying genomic DNA sequence. DNA methylation may be modified by environmental exposures as well as gene polymorphisms and may be a mechanistic link between environmental risk factors and the development of disease. In this review, we consider the role of DNA methylation in bone cells (osteoclasts/osteoblasts/osteocytes) and their progenitors with special focus on in vitro and ex vivo analyses. The number of studies on DNA methylation in bone cells is still somewhat limited, n…

[Orthodontic treatment and mechanical stress].

In orthodontic treatment, the role of periodontal ligament is essential for the teeth to move within the alveolar process against the mechan … ch results in constant reconstruction of the alveolar process in 3 dimension. Among the bone cells, osteocytes act as a major role in bone remodelling during orthodontic tooth movement against mechanical stress. Moreover, the side effect during tooth movement is known to be the pain and external apical root resorption.

Mitochondrial Transport in Bone Metabolism Homeostasis: Molecular Mechanisms and Targeted Therapeutic Strategies.

The homeostasis of bone metabolism relies on the precise synergistic regulation among osteoblasts, osteoclasts, and osteocytes. Disruption of this regulatory network underlies the pathogenesis of osteoporosis, osteonecrosis, osteoarthritis, and other metabolic bone diseases. As an emerging mode of intercellular communication, mitochondrial transport delivers functional mitochondria to impaired cells, thereby reshaping cellular metabolism, alleviating oxidative stress, and restoring cell function. It thus plays an irreplaceable role in maintaining bone metabolic homeostasis. However, studies fo…

Integration of Bone-Targeted Delivery and Crosstalk Modulation of Liver-Bone Axis for Improved Osteoporosis Therapy.

Nanomedicines hold great promise for the treatment of osteoporosis, while their nonspecific accumulation in the liver typically reduces the drug delivery efficacy. Herein, we report bone-targeted liposomes encapsulated with arginine and metformin for the treatment of osteoporosis. These liposomes are functionalized with alendronate to enhance the bone-targeting capability. The delivered agents directly modulate osteoblasts, osteoclasts, and osteocytes, promoting bone formation and inhibiting bone resorption to counteract osteoporotic bone loss. In addition to targeted bone delivery, the inevit…

Tackling myeloma bone disease: From pathophysiology to cutting-edge therapies.

Bone involvement in multiple myeloma (MM) is marked by osteolysis, driven by excessive bone resorption and a profound suppression of bone formation. Interactions between MM cells and the bone microenvironment-mediated by integrins, chemokines, and bone marrow stromal cells-play a critical role in the development of myeloma bone disease (MBD). Key players include osteoclasts, osteoblasts, and osteocytes. Osteoclast activation is mainly driven by RANK/RANKL and other pro-osteoclastogenic factors that disrupt bone remodeling, while impaired bone formation involves Wnt signaling inhibition and Runx…

Marrow Fat-Secreted Factors as Biomarkers for Osteoporosis.

PURPOSE OF REVIEW: The age-related accumulation of bone marrow adipose tissue (BMAT) negatively impacts bone metabolism and hematopoiesis. T … BMAT disturbs bone metabolism primarily through palmitate-mediated toxic effects on osteoblasts and osteocytes, including dysregulated autophagy and apoptosis. BMAT-secreted factors are important modulators of bone metabolism. [Calcium and bone metabolism across women's life stages. Bone metabolism and estrogen/androgen balance in women.].

Androgen as well as estrogen has physical, psychological and sexual roles in women. The action of androgen on bone health in women is important. The androgen receptor is expressed in osteoblasts and osteocytes but the mechanism has not been clarified in women. It has been reported that endogenous testosterone level was positively correlated with bone mineral density and higher testosterone level might be associated with decrease in bone fractures. Also, it has been reported that bone mineral density in women who received testosterone with estrogen was higher than that in women who received est…

An unusual breast lump: osseous metaplasia.

We present a rare case of osseous metaplasia in the breast with no other associated breast pathology. A 46-year-old HIV-positive lady presen … psy of the breast lesion, with the final histology demonstrating bone matrix deposition with viable osteocytes within lacunae and associated osteoclasts with spindle cells, consistent with osseous metaplasia. A core biopsy of the axillary lymph nodes was normal. The patient was therefore reassured and discharged from the clinic.

Potential role for therapies targeting DKK1, LRP5, and serotonin in the treatment of osteoporosis.

Osteoporosis is a common disorder in which diminished bone mass leads to progressive microarchitectural skeletal deterioration and increased fracture risk. Our understanding of both normal and pathologic bone biology continues to evolve, and with it our grasp of the highly coordinated relationships between primary bone cells (osteoblasts, osteoclasts, and osteocytes) and the complex molecular signals bone cells use to integrate signals derived from other organ systems, including the immune, hematopoietic, gastrointestinal, and central nervous systems. It is now clear that the Wnt signaling pat…

Osteonecrosis of the femoral head.

New cases of osteonecrosis of the femoral head in the United States number between 10,000 and 20,000 per year. This disease usually affects patients in their late 30s and early 40s. Although a number of authors have related specific risk factors to this disease, its etiology, pathogenesis, and treatment remain a source of considerable controversy. This disorder has been associated with corticosteroid use, substance abuse, and various systemic medical conditions. Either direct damage to osteocytes (e.g., by toxin production) or indirect damage (e.g., due to disorders in fat metabolism or hypoxi…

Circular RNAs in osteoporosis: expression, functions and roles.

Osteoporosis, which is caused by an imbalance in osteoblasts and osteoclasts, is a global age-related metabolic disease. Osteoblasts induce osteocyte and bone matrix formation, while osteoclasts play an important role in bone resorption. Maintaining a balance between osteoblast formation and osteoclastic absorption is crucial for bone remodeling. Circular RNAs (circRNAs), which are characterized by closed-loop structures, are a class of novel endogenous transcripts with limited protein-coding abilities. Accumulating evidence indicates that circRNAs play important roles in various bone diseases

Advances in osteoimmunology: pathophysiologic concepts and treatment opportunities.

Osteoimmunology is an emerging research area that deals with the mutual interactions between bone and the immune system. Osteoclasts have long been the center of attention in osteoimmunological research due to their hematopoietic origin and strong activation through cytokines. However, also the osteoclast's opponent - the osteoblast - has recently sought the spotlight, and novel functions of its descendant - the osteocyte - have been unraveled. A considerable number of investigations carried out over the past decade have identified critical proteins with osteoimmune functions including the pro

Glucocorticoid-induced osteoporosis: pathophysiological data and recent treatments.

Long-term glucocorticoid therapy promptly induces osteoporosis, whose severity depends on the dose and duration of the treatment. Recent data suggest that there is no safety threshold for adverse effects on bone. Glucocorticoid therapy impairs calcium intestinal absorption, dramatically suppresses osteoblastic formation, and stimulates osteocyte apoptosis. In contrast, the contribution of secondary hyperparathyroidism and increased bone resorption, although frequently mentioned, is now a focus of controversy. Beneficial effects on bone have been obtained with calcium and vitamin D supplementat

Tissue material properties, whole-bone morphology and mechanical behavior in the Fbn1 C1041G/+ mouse model of Marfan syndrome.

Marfan syndrome (MFS) is a connective tissue disorder caused by pathogenic mutations in FBN1. In bone, the protein fibrillin-1 is found in the extracellular matrix where it provides structural support of elastic fiber formation, stability for basement membrane, and regulates the bioavailability of growth factors. Individuals with MFS exhibit a range of skeletal complications including low bone mineral density and long bone overgrowth. However, it remains unknown if the bone phenotype is caused by alteration of fibrillin-1's structural function or distortion of its interactions with bone cells.
