Paternal ribosome biogenesis dysfunction caused by cyclophosphamide-busulfan chemotherapy contributes to early embryonic developmental impairment in mice.

BACKGROUND: Combined cyclophosphamide (CTX) and busulfan (BUS) chemotherapy, while effective against malignancies, poses significant risks to male reproductive health. This study systematically investigates the proteomic basis underlying CTX + BUS-induced paternal reproductive toxicity and its impact on early embryonic development in mice. METHODS: We utilized a mouse model of acute CTX + BUS exposure. Systemic germ cell apoptosis, sperm parameters (concentration, motility, morphology), and fertilization rates were assessed. Testicular and sperm proteomic profiling was performed. Embryonic dev

Haplotype-based association between paternal DNMT1 variants and nonsyndromic cleft lip or without cleft palate in Chile.

BACKGROUND: DNA methyltransferase 1 (DNMT1) is responsible for epigenetic remodeling of the genome during spermatogenesis and maintenance of DNA methylation. The current study aimed to assess the possible association between paternal polymorphic variants of the gene encoding DNMT1 enzyme and the risk of nonsyndromic cleft lip with or without cleft palate (NSCL/P) expression in offspring. METHODS: Nine DNMT1 polymorphic single nucleotide polymorphism (SNP) variants were analyzed in 101 fathers of NSCL/P Chilean cases and 187 fathers of controls. Single marker association for additive, dominant

Therapy to teratology: chronic paternal antioxidant supplementation alters offspring placental architecture and craniofacial morphogenesis in a mouse model.

Oxidative stress is an important regulator of the mammalian epigenome, with redox imbalances triggering stress-responsive epigenetic modifications linked to various diseases. Accordingly, antioxidant therapies are commonly used to reduce oxidative damage and are widely employed in cases of male infertility. Interestingly, in ostensibly healthy males, recent research suggests that antioxidants may have a negative influence on sperm DNA methylation, indicating a potential epigenetic liability. However, whether male antioxidant treatment can induce paternal effects on offspring growth and develop

Effects of paternal age and interaction with smoking, alcohol consumption and maternal age on implantation failure in women undergoing ART.

It remains unclear whether there is an independent association of paternal age with implantation failure (IF) in couples undergoing Assisted Reproductive Technology (ART) treatment. Little is known about the interactive effects of paternal age with maternal age, paternal smoking and alcohol consumption on IF. The Anhui Maternal-Child Health Cohort Study (AMCHS) interviewed 1910 infertile couples (paternal age ranged 22-57 years) to look for risk factors associated with IF. Implantation outcome was assessed using serum β-HCG between 0 and 25 IU/L 14 days post-embryo transfer. Odds ratios (ORs)

Unraveling sex differences in maternal and paternal care impacts on social behaviors and neurobiological responses to early-life adversity.

Early-life stress (ELS) affects the development of prosocial behaviors and social-cognitive function, often leading to structural brain changes and increased psychosocial disorders. Recent studies suggest that mother- and father-child relationships independently influence social development in a sex-specific manner, but the effects of impaired father-child relationships are often overlooked. This review examines preclinical rodent studies to explore how parental neglect impacts neuroplasticity and social behaviors in offspring. We highlight that disruptions in maternal interactions may affect

Association of Paternal Age Alone and Combined with Maternal Age with Perinatal Outcomes: A Prospective Multicenter Cohort Study in China.

Maternal and paternal age at birth is increasing globally. Maternal age may affect perinatal outcomes, but the effect of paternal age and its joint effect with maternal age are not well established. This prospective, multicenter, cohort analysis used data from the University Hospital Advanced Age Pregnant Cohort Study in China from 2016 to 2021, to investigate the separate association of paternal age and joint association of paternal and maternal age with adverse perinatal outcomes. Of 16,114 singleton deliveries, mean paternal and maternal age (± SD) was 38.0 ± 5.3 years and 36.0 ± 4.1 years.

Longitudinal links among paternal and maternal harsh parenting, adolescent emotional dysregulation and short-form video addiction.

BACKGROUND: Due to its diversifying and appealing content, short-form videos have developed very rapidly since its emergence, with more and more users enjoying the transient pleasures from such videos, which have triggered widespread concern over the detrimental influence of short-form video addiction on adolescent health. OBJECTIVE: The present study intended to examine the longitudinal relations among paternal/maternal harsh parenting, adolescents' emotional dysregulation and their short-form video addiction using a cross-lagged longitudinal design. PARTICIPANTS AND SETTING: We recruited a

Paternal and maternal psychiatric history and risk of preterm and early term birth: A nationwide study using Swedish registers.

BACKGROUND: Women with psychiatric diagnoses are at increased risk of preterm birth (PTB), with potential life-long impact on offspring health. Less is known about the risk of PTB in offspring of fathers with psychiatric diagnoses, and for couples where both parents were diagnosed. In a nationwide birth cohort, we examined the association between psychiatric history in fathers, mothers, and both parents and gestational age. METHODS AND FINDINGS: We included all infants live-born to Nordic parents in 1997 to 2016 in Sweden. Psychiatric diagnoses were obtained from the National Patient Register

Case report: Paternal uniparental disomy on chromosome 7 and homozygous SUGCT mutation in a fetus with overweight after birth.

Background: Paternal uniparental disomy (UPD) of chromosome 7 is extremely rare, and only a few postnatal cases have been reported. The effects on growth were discordant in these cases, and the relevance of paternal UPD(7) to growth caused by imprinting remains questionable. Case presentation: Here, we report a prenatal case that underwent invasive prenatal diagnosis due to the high risk of Down's syndrome and failed noninvasive prenatal screening. The fetus had a normal karyotype and no apparent copy number variation. Homozygous copy-neutral regions on chromosome 7 were identified using a sin

A Cross-Sectional Analysis of Paternal Intimacy Problems, Stress Levels, and Satisfaction from Families with Children Born with Mucoviscidosis.

There is an increasing interest in father-child interactions and their effects. Due to the rising number of working mothers, marital interruptions, divorces, and child custody arrangements, paternal duties and the relevance of fathering continue to be re-evaluated. As there are rising expectations for men to undertake more childcare and household responsibilities, it was hypothesized that the presence of a disabled or chronically ill child would have a significant impact on the couple's future family situation, marital conduct due to paternal dissatisfaction, and increased stress levels. There

CDKN1C hyperexpression in two patients with severe growth failure and microdeletions affecting the paternally inherited KCNQ1OT1:TSS-DMR.

BACKGROUND: Two imprinting control centres, H19/IGF2:IG-differentialy methylated region (DMR) and KCNQ1OT1:TSS-DMR, reside on chromosome 11p15.5. Paternal deletions involving the KCNQ1OT1:TSS-DMR result in variable phenotypes, namely, normal phenotype, Silver-Russel syndrome (SRS) and fetal demise. However, expression analyses for CDKN1C in these patients are very limited. CASES: Patient 1 (adult woman) and patient 2 (boy in early childhood) showed prenatal and postnatal growth failure and clinical suspicion of SRS. MOLECULAR ANALYSES: Both patients showed hypermethylation of the KCNQ1OT1:TS

The effect of kangaroo care practice after caesarean section on paternal-newborn interaction: A mixed-methods study in Turkey.

OBJECTIVES: The first meeting of the mother with the newborn after the caesarean section is usually delayed due to some reasons in Turkey. Although there are many benefits of applying kangaroo care (KC) between mother and newborn, there are a limited number of studies on the application of KC between newborn and father after caesarean section in international literature. This study was conducted to determine the effect of kangaroo care between father and newborn after caesarean section. METHOD: A mixed-methods study was used. Data were collected through semi-structured interviews and video re

Paternal High-Fat Diet Altered Sperm 5'tsRNA-Gly-GCC Is Associated With Enhanced Gluconeogenesis in the Offspring.

Background: Paternal lifestyle, stress and environmental exposures play a crucial role in the health of offspring and are associated with non-genetic inheritance of acquired traits, however the underlying mechanisms are unclear. In this study, we aimed to find out how the sperm tsRNA involved in paternal high-fat diet induced abnormal gluconeogenesis of F1 offspring, and explore the underlying molecular mechanism of its regulation. Method: We generated a paternal high fat diet (42% kcal fat) model to investigate the mechanism by which paternal diet affects offspring metabolism. Four-week-old C

Paternal high protein diet modulates body composition, insulin sensitivity, epigenetics, and gut microbiota intergenerationally in rats.

Mounting evidence demonstrates that paternal diet programs offspring metabolism. However, the contribution of a pre-conception paternal high protein (HP) diet to offspring metabolism, gut microbiota, and epigenetic changes remains unclear. Here we show that paternal HP intake in Sprague Dawley rats programs protective metabolic outcomes in offspring. Compared to paternal high fat/sucrose (HF/S), HP diet improved body composition and insulin sensitivity and improved circulating satiety hormones and cecal short-chain fatty acids compared to HF/S and control diet (P < .05). Further, using 16S rRN

Partial trisomy 16q and partial monosomy 7p of a fetus derivated from paternal balanced translocation: A case report.

INTRODUCTION: Subchromosomal deletions and duplications could currently be detected by noninvasive preliminary screening (NIPS). However, NIPS is a screening test that requires further diagnosis. Here we report a fetus with an autosomal abnormality revealed by NIPS and conventional karyotype combined with copy number variations sequencing (CNV-seq) confirmed the fetus with an unbalanced translocation. PATIENT CONCERN: This was the fourth pregnancy of a 30-year-old woman who underwent 2 spontaneous abortions and gave birth to a child with a normal phenotype. The woman and her husband were heal

Early life adoption shows rearing environment supersedes transgenerational effects of paternal stress on aggressive temperament in the offspring.

Prenatal experience and transgenerational influences are increasingly recognized as critical for defining the socio-emotional system, through the development of social competences and of their underlying neural circuitries. Here, we used an established rat model of social stress resulting from male partner aggression induced by peripubertal (P28-42) exposure to unpredictable fearful experiences. Using this model, we aimed to first, characterize adult emotionality in terms of the breadth of the socio-emotional symptoms and second, to determine the relative impact of prenatal vs postnatal influe

[Paternal Risk and Protective Factors for Endangerment of Child Well-Being and their Consideration in the Risk Inventories Used Throughout Germany].

Paternal Risk and Protective Factors for Endangerment of Child Well-Being and their Consideration in the Risk Inventories Used Throughout Germany In view of the current family and role models, paternal risk and protective factors can have a significant impact on the risk of violence within a family and should therefore - in addition to the factors on the part of the mother, the child and the closer or other family systems - be taken into consideration when estimating the endangerment of child well-being. In this article we explore which empirical findings on paternal risk and protective factor
