A Physiological Microfluidic Blood-Brain-Barrier Model for In Vitro Study of Nanoparticle Trafficking and Accumulation.

Although the blood-brain barrier (BBB) restricts passage of most molecules, various naturally occurring and synthetic nanoparticles (NPs) are nonetheless found within the brain parenchyma. To study the mechanisms underlying this phenomenon, we developed a microfluidic BBB model (mBBB) using human cerebral microvascular endothelial cells (HCMECs) in direct contact with primary human astrocytes and pericytes within a physiologically relevant extracellular matrix. The horizontal architecture enables high-resolution imaging across the full barrier interface and allows direct assessment of nanopart…

Innovations in preclinical biology: ex vivo engineering of a human kidney tissue microperfusion system.

Kidney disease is a public health problem that affects more than 20 million people in the US adult population, yet little is understood about the impact of kidney disease on drug disposition. Consequently there is a critical need to be able to model the human kidney and other organ systems, to improve our understanding of drug efficacy, safety, and toxicity, especially during drug development. The kidneys in general, and the proximal tubule specifically, play a central role in the elimination of xenobiotics. With recent advances in molecular investigation, considerable information has been gat

Blood flow patterns in mice are regulated by interpericyte tunneling nanotubes connecting functionally-opposite neuronal areas.

Functioning of the central nervous system is critically dependent on precise blood delivery achieved through the formation of blood flow patterns or differences in flux between vessels. Nevertheless, how these blood flow patterns are generated is a fundamental question that is incompletely understood. Here, we show, in live imaging experiments on mice, that specialized pericytes connected via tunneling nanotubes regulate flux patterns by modulating the response of different capillaries in opposing ways, homogenizing blood flow, which is vital to supply functionally-opposite neuronal areas of t…

The heterogeneity of dermal mesenchymal cells reproduced in skin equivalents regulates barrier function and elasticity.

The heterogeneity of dermal mesenchymal cells, including perivascular mesenchymal cells and papillary and reticular fibroblasts, plays critical roles in skin homeostasis. Herein, we present human skin equivalents (HSEs), in which pericytes, papillary fibroblasts, and reticular fibroblasts are spatially organized through autonomous three-cell interactions among epidermal keratinocytes, dermal fibroblasts, and vascular endothelial cells. The replication of dermal mesenchymal cell heterogeneity enhances skin functions, including epithelialization, epidermal barrier formation, and dermal elasticit…

Molecular Mechanisms Governing Vascular Function in Heart Failure.

Heart failure is a leading cause of morbidity and mortality worldwide. Emerging evidence points to vascular dysfunction as a major contributor to the development and progression of this grievous syndrome. Functional interplay between the myocardium and vasculature involves endothelial cells, vascular smooth muscle cells, and pericytes governing tissue perfusion, vascular tone, inflammation, and metabolism. Vascular cells play a central role in maintaining cardiovascular homeostasis by sensing hemodynamic and inflammatory stimuli and releasing vasoactive and paracrine factors. Dysregulation of …

Leptomeningeal fibroblasts promote glioblastoma progression by regulating cerebrospinal fluid dynamics.

Cerebrospinal fluid contributes to homeostasis in the central nervous system, but how its dynamics are altered in glioblastoma is unclear. We find that glioblastoma drives leptomeningeal perivascular fibrosis that is associated with impaired fluid transport and clearance. Lineage tracing and single-cell analyses in male tumor-bearing mice identify leptomeningeal fibroblasts as the principal source of this fibrotic response, with limited contribution from pericytes. Fibrosis is linked to activation of nuclear factor kappa B signaling in collagen-producing fibroblasts, extracellular matrix depos…

Emerging role of mesenchymal cells in cardiac and cerebrovascular diseases: Physiology, pathology, and therapeutic implications.

In recent years, the therapeutic utility of mesenchymal stem cells (MSCs) has received substantial attention from investigators, owing to their pleiotropic properties. The emerging insights from the developments in tissue engineering provide perspectives for the repair of damaged tissue and the replacement of failing organs. Perivascular cells including MSC-like pericytes, vascular smooth muscles, and other cells located around blood vessels, have been acknowledged to contribute to in situ angiogenesis and repair process. MSCs offer a wide array of therapeutic applications in different patholo…

Retroperitoneal Hemangiopericytoma in a young woman. Case report and literature review.

UNLABELLED: Hemangiopericytoma (HPC) is a primary tumor with mesenchymal vascular origin that represents 1% of all vascular neoplasm. HPC develops from the Zimmerman's pericytes around capillaries venules and it is possible to observe it frequently in the extremities, pelvis, retroperitoneum, head, neck and meninges. The only definitive parameter of malignancy is the development of recurrence or distant metastases. This report describes a case of symptomatic retroperitoneal HPC in a young female patient treated by surgical complete removal of the mass, and literature review. Despite the relati…

Electron microscopy of renal and ocular changes in virus-induced diabetes mellitus in mice.

The eyes, kidneys and pancreas of mice (SJL/J) infected with encephalomyocarditis virus were examined by light and electron microscopy. Diabetic mice with the longest duration (6 months) of diabetes showed marked renal and ocular alterations. Fasting blood glucose levels were 17.8-21.9 mmol/l and glycosuria was present. Clinically, based on ophthalmoscopy and fluorescein angiography, retinal vessels were normal. Histologically, moderately decreased numbers of pericytes were noted following trypsin digestion. The basement membrane of inner retinal vessels showed significant thickening in diabet…

Light and electron microscopic study of early lesions in angiomatosis retinae.

Multiple early untreated lesions of angiomatosis retinae in the peripheral retina and optic disk were discovered after death in both eyes of a patient with von Hippel-Lindau disease. Each of the four peripheral angiomas was studied by a different technique: routine light microscopy of serial histologic sections, electron microscopy, flat mount and trypsin digestion, and frozen section with special stains for lipid. The large capillaries comprising the core of each angioma displayed normal endothelium, basement membrane, and pericytes. Thus, capillary hemangioma of the retina is a more correct …

Chemopreventive agents targeting tumor microenvironment.

Recent studies have shown that tumor development and progression depend not only on the perturbed genes that govern cell proliferation, but is also highly determined by the non-tumor cells of the stromal compartment surrounding the tumor called tumor microenvironment (TME). These findings highlight the importance of targeting the microenvironment in combination with therapies aimed at tumor cells as a valuable approach. The innate and adaptive immune cells in the TME interact among themselves and also with the endothelial cells, pericytes and mast cells of the stromal compartment through vario…

Dysfunction of the neurovascular unit as a temporal driver in Alzheimer's pathogenesis.

Extensive studies have shown that cerebrovascular dysfunction is a critical factor in the onset and progression of Alzheimer's disease (AD). Neurovascular unit (NVU) is impaired in AD brains, including damage of tight junction between endothelial cells, degeneration of pericytes, activation of astrocytes and microglia, and apoptosis of neurons. As decreased cerebral blood flow is observed before amyloid-beta (Aβ) generation, it is supposed that NVU dysfunction may precede and exacerbate the pathological state of AD neural system, and that the events of NVU dysfunction and Aβ deposition synergi…

Brain remodeling in stroke patients: A comprehensive review of mechanistic and neuroimaging studies.

Stroke-induced brain remodeling involves a complex interplay of neurovascular components, including endothelial cells, microglia, astrocytes, and pericytes, which collectively contribute to the restoration of brain function. These processes are crucial for repairing the blood-brain barrier, regulating inflammation, and promoting neurogenesis. This review examines the mechanisms underlying brain remodeling and the role of advanced neuroimaging techniques-such as functional MRI (fMRI), positron emission tomography (PET), functional near-infrared spectroscopy (fNIRS), and functional ultrasound (f…

Hybrid Management of Giant Solitary Intracranial Fibrous Tumors: A Report of Two Cases.

Solitary fibrous tumors (SFTs) are rare tumors of vascular origin, derived from pericytes surrounding blood vessels. Their frequency of presentation is mainly in adults, and they can be in various parts of the body, such as the extremities, head, neck, retroperitoneum, and abdomen. These tumors can be either benign or malignant, and their behavior and clinical course are variable, with a significant risk of recurrence and metastasis. We present a two-case report: the first involving a 25-year-old man with right hemiparesis and a history of two prior surgeries at another institution for a diagn…

Müller cells and retinal angiogenesis: critical regulators in health and disease.

Müller cells are the most abundant glial cells in the mammalian retina. Their morphology and metabolism enable them to be in close contact and interact biochemically and physically with almost all retinal cell types, including neurons, pericytes, endothelial cells, and other glial cells, influencing their physiology by releasing bioactive molecules. Studies indicate that Müller glial cells are the primary source of angiogenic growth factor secretion in the neuroretina. Because of this, over the past decade, it has been postulated that Müller glial cells play a significant role in maintaining r…
