TENT5A-associated osteogenesis imperfecta: long-term follow-up and molecular insights.

OI is a genetically diverse disorder characterized by bone fragility and deformities, with most cases attributed to dominant mutations in collagen I-related genes. However, mutations in TENT5A, encoding a poly(A) polymerase, have recently been implicated in severe, recessive forms of OI. We reported 3 individuals from a consanguineous family with a novel homozygous TENT5A variant (c.672G>T, p.Arg224Ser). Our patients presented with severe bone fragility, generalized osteopenia, skeletal deformities, short stature, and early wheelchair dependence. Patient-derived fibroblasts demonstrated impair

Smart "Thrombus": Self-Localizing UCST-Type Microcage.

Embolization is often used to block blood supply for controlling the growth of fibroids and malignant tumors, but limited by embolic agents lacking spontaneous targeting and post-treatment removal. So we first adopted nonionic poly(acrylamide-co-acrylonitrile) with an upper critical solution temperature (UCST) to build up self-localizing microcages by inverse emulsification. The results showed that these UCST-type microcages behaved with the appropriate phase-transition threshold value around 40 °C, and spontaneously underwent an expansion-fusion-fission cycle under the stimulus of mild temper

The L1 protein as a new indicator of inflammatory activity in patients with juvenile rheumatoid arthritis.

L1 is a major granulocyte and monocyte protein with Mr 36.5 kDa. It is released during leukocyte activation and detected in plasma by use of an enzyme immunoassay. In our study, L1, erythrocyte sedimentation rate (ESR), C-reactive protein (CRP), orosomucoid and haptoglobin were analyzed in 127 patients with juvenile rheumatoid arthritis (79 pauci, 33 poly and 15 systemic). L1, ESR and the acute phase proteins were all found to have positive correlations with the clinical joint assessments, with L1 having the strongest correlations. High correlation coefficients were found between L1 and orosom

Maternal Immune Activation Leads to Mitochondrial Dysfunction and a Social Deficit in Offspring That Is Reversed by Nicotinamide Riboside.

Background: Maternal immune activation (MIA) during pregnancy is a known risk factor for several neurodevelopmental and psychiatric disorders, including schizophrenia. In rodent models, MIA is commonly induced using polyinosinic/polycytidylic acid (Poly(I/C)), a viral mimetic that activates Toll-like receptor 3 (TLR3) signaling and elicits an inflammatory response in both the dam and the fetuses. MIA results in various behavioral abnormalities in offspring, including deficits in social interaction. Recent studies have shown that MIA decreases the ability to maintain mitochondrial membrane pote

Golden vesicular nanoadjuvant-loaded injectable hydrogel for locoregional photothermal-immunotherapy of unresectable glioblastoma.

Glioblastoma (GBM) as the most malicious primary brain tumor is infiltrative and often unresectable, which coupled with a profoundly immunosuppressive tumor microenvironment (TME) lead to a poor prognosis for patients. Here, we developed a golden vesicular nanoadjuvant (Au/TLR3-V) comprising gold nanoclusters and TLR3 agonist poly(I:C), formulated within an injectable hydrogel (Au/TLR3-V@gel) for locoregional photothermal-immunotherapy of large orthotopic GBM. Upon near-infrared (NIR) irradiation, Au/TLR3-V generated a potent photothermal response that induced immunogenic cell death and promot

Prenatal immune activation and adult Poly(I:C) re-challenge promote neuroimmune priming and AD-related behavioural, cellular and molecular alterations in wild-type mice.

OBJECTIVE: Alzheimer's Disease (AD) is neurodegenerative disorder characterized by deposition of Aβ plaques, tau-positive neurofibrillary tangles, neuroinflammation and clinical dementia. Epidemiological and experimental evidence suggest that peripheral immune inflammation is a risk factor for age-related neurodegeneration but whether its sustained activation is sufficient to drive behavioural, molecular and cellular changes consistent with AD-associated neurodegenerative vulnerability remains unclear. Here we investigated whether prenatal immune stimulation followed by an adult systemic re-ch

Characteristics and functions of interleukin 21 in channel catfish (Ictalurus punctatus).

Interleukin-21 (IL-21) is mainly produced by CD4+ T cells and NKT cells, belonging to the gamma chain (γc) family. It promotes the proliferation, activation, and differentiation of immune cells and activates the JAK-STAT, MAPK signaling, and PI3K pathways to regulate immune responses, crucially impacting anti-infectious inflammatory responses. In this study, IL-21 was successfully identified in channel catfish (Ictalurus punctatus, Ip), with a coding sequence (CDS) length of 438 bp encoding a 145-amino acid (aa) protein. The signal peptide comprised 19 aa, while the mature peptide consists of

Reversal learning impairments in the maternal immune activation rat model of schizophrenia.

One of the cognitive symptoms observed in schizophrenia is decreased flexibility in several tasks, including reversal learning. Reversal learning has previously been tested in rats following maternal immune activation (MIA), a risk factor for schizophrenia, with varying results. Whereas some previous studies have shown that MIA rats are slower to learn a reversal, others have reported more rapid learning compared with controls. Several of these latter studies have, however, used a T-maze task with aversive, negative reinforcement as a motivating factor. Because most human studies use positive

Double-stranded RNA poly(I:C) enhances matrix metalloproteinase mRNA expression in human nasal polyp epithelial cells.

CONCLUSION: The significant up-regulation of matrix metalloproteinase (MMP)-9 mRNA, which is not modulated by tissue inhibitor of metalloproteinase (TIMP)-1, is an additional source of increased proteolytic activity in virus-infected upper airways that might contribute to the exacerbation of chronic rhinosinusitis with nasal polyps. OBJECTIVES: Chronic rhinosinusitis is often exacerbated by viral infection. We hypothesized that a disruption of the mechanisms that regulate the activity of MMPs during viral infection is one possible mechanism responsible for the exacerbation. In the present stu

Effects of poly I:C in porcine iron deficient neutropenia.

Withholding iron dextran treatment normally given to pigs at 1-3 days of age to prevent anemia resulted also in neutropenia. Polyinosinic acid:polycytidylic acid (poly I:C) at 0.5 mg/kg IV at 25 days of age resulted in induction of putative interferon 2 to 24 hours later, with significantly (P less than 0.05) lower concentrations in iron-deficient (Fe-) female pigs than in iron-supplemented (Fe+) female pigs. Poly I:C caused several transient toxic manifestations, including elevations in blood urea nitrogen, creatinine, aspartate aminotransferase, potassium (K), total bilirubin and phosphorus

[Characteristics of the factors that suppress and stimulate interferon production].

A repressor of interferon production is contained in the hyaloplasm of the chick embryo fibroblast cells in the state of hyporeactivity. A factor stimulating interferon production (FSIP) was found in the hyaloplasm of the cells subjected to single or double induction with poly (pI) x poly (pC) followed by exposure to actinomycin D. Both preparations were of the protein nature and possessed high biological activity. They repressed or stimulatd interferon production at a dilution of 1:512-1:1024. The preparations had tissue and inductor specificity, did not affect the Venezuelan equine encephalo

A comparison of the prophylactic and therapeutic effects of poly I:C and endotoxin in mice infected with Mengo virus.

We have compared the protective effect in AKR mice of poly I:C and bacterial endotoxins against lethal doses of Mengo virus. Administered intravenously or intraperitoneally, both interferon inducers protected mice to about the same extent from virus challenges of 2-3 LD50's. Endotoxin, however, was unable to protect the mice effectively against higher challenge doses of virus. Evidence is presented that the level of protection afforded by both inducers is related to the level of circulating interferon produced. We have also shown that a single intravenous dose of poly I:C results in the appear

PARP1 rs1136410 C/C genotype associated with an increased risk of esophageal cancer in smokers.

DNA repair system plays a crucial role in maintaining genomic integrity and stability and in protecting against cancer. Poly(ADP-ribose) polymerase 1 (PARP1) functions as a key enzyme in the base excision repair (BER) pathway. Single nucleotide polymorphism (SNP) that could affect the function or expression of PARP1 gene might be associated with the risk of cancer. This study was designed to evaluate the association between PARP1 SNPs and the susceptibility to esophageal squamous cell carcinoma (ESCC) in a population from Cixian, a high incidence region from northern China. In 574 ESCC patient

Microbial synthesis and characterization of poly(3-mercaptoproprionate-co-3-mercapto-2-methylproprionate) [P(3MP-co-3M2MP)]: Co-polythioesters consisting of achiral 3MP unit and (RS)-3M2MP unit.

Polythioesters (PTEs) can be microbially polymerized from thiol carboxylate precursors by polyhydroxyalkanoate (PHA)-producing bacteria. Poly[(RS)-3-mercapto-2-methylpropionate] [P(3M2MP)], characterized in our previous study, is an amorphous and highly elastic polymer. In this study, achiral 3-mercaptopropionate (3MP) was polymerized in recombinant Escherichia coli DH5α, and the copolymerization of 3MP with (RS)-3M2MP was subsequently investigated to characterize a PTE copolymer. P(3MP) was hard and brittle, and insoluble in most solvents, but its amorphous form immediately after extraction f

Solubility-Difference-Driven Strategy for Constructing Core-Shell Structure Si@C Anodes: Ammonia-Assisted Carbon Shell Embedded with Copper Nanoparticles.

Compounding with carbon materials is an effective strategy to mitigate the intrinsic challenges of silicon anodes, particularly their severe volume variation and poor electrical conductivity, thereby demonstrating significant potential for commercial lithium-ion battery applications. In this study, a solubility-difference-driven approach is proposed for constructing core-shell structured Si@C anodes. Utilizing the distinct solubility difference of poly(vinyl alcohol) and basic copper carbonate in ammonia-water and ethanol, a uniform carbon shell embedded with copper nanoparticles is successful

Moderate temperatures affect Escherichia coli inactivation by high-pressure homogenization only through fluid viscosity.

The inactivation of suspensions of Escherichia coli MG1655 by high-pressure homogenization was studied over a wide range of pressures (100-300 MPa) and initial temperatures of the samples (5-50 degrees C). Bacterial inactivation was positively correlated with the applied pressure and with the initial temperature. When samples were adjusted to different concentrations of poly(ethylene glycol) to have the same viscosity at different temperatures below 45 degrees C and then homogenized at these temperatures, no difference in inactivation was observed. These observations strongly suggest, for the

Mercuric chloride induces apoptosis via a mitochondrial-dependent pathway in human leukemia cells.

Mercurial compounds modulate immunologic functions by inducing cytotoxicity. Although mercury chloride (HgCl(2)) is known to induce apoptosis in various immune system cells, the mechanism of the induction of apoptosis is poorly understood. In this study, we examined the activation of caspase-3, an important cysteine aspartic protease, during HgCl(2)-induced apoptosis in a human leukemia cell line (HL-60 cells). Both DNA fragmentation, a characteristic of apoptotic cells, and proteolysis of poly(ADP-ribose) polymerase (PARP), a substrate of caspase-3, occurred at 6 h after HgCl(2) treatment in

3D printed autoclavable biocompatible biodegradable bioreactor vessels with integrated sparger made from poly-lactic acid.

3D printing has become widespread for the manufacture of parts in various industries and enabled radically new designs. This trend has not spread to bioprocess development yet, due to a lack of material suitable for the current workflow, including sterilization by autoclaving. This work demonstrates that commercially available heat temperature stable poly-lactic acid (PLA) can be used to easily manufacture novel bioreactor vessels with included features like harvest tubes and 3D printed spargers. Temperature responsiveness was tested for PLA, temperature stable PLA (PLA-HP) and glass for tempe
