[Pharmacokinetics of (-)-, (+)- and (+/-)-norephedrine. Plasma concentrations, serum protein binding and urinary excretions].

The pharmacokinetics of norephedrine enantiomers were determined after the independent i.v. administration of (+/-)-norephedrine (20 mg/kg), … nistered as a racemate showed a tendency to decrease. There was no difference in the in vitro serum protein binding of (-)- and (+)-norephedrine. The data from this study reveal that pharmacokinetic interactions exist between the norephedrine enantiomers and also reveal that the serum protein binding is not concerned with those interactions. The differences in th …

Identification of a new high-affinity binding protein for neurotoxic phospholipases A2.

Ammodytoxin C is a neurotoxic phospholipase A2 which blocks the release of neurotransmitter from the nerve terminal. Using a radioiodinated derivative of the toxin, we located its specific high-affinity binding site in the demyelinated P2 fraction of porcine cerebral cortex (Kd = 15 nM; Bmax = 1.5 pmol/mg membrane protein). In cross-linking experiments on a membrane preparation, 125I-ammodytoxin C labeled a protein of 25 kDa. The formation of a specific adduct was not inhibited by nontoxic phospholipases A2 or even by neurotoxic phospholipases A2 which have practically identical pathophysiolog

Insulin-like growth factor binding protein-1: recent findings and new directions.

In 1988, insulin-like growth factor-binding protein-1 (IGFBP-1) became the first characterized member of a group of structurally related soluble proteins which specifically bind and modulate the actions of the IGFs. Since then, a wealth of information has accumulated regarding the physiology of this dynamic serum protein. In this review, we update our 1993 summary (Lee PDK et al. Proc Soc Exp Biol Med 204:4-29) of the status of IGFBP-1 research. The IGFBP-1 protein sequence contains 12 N-terminal and 6 C-terminal cysteine residues which are conserved in other mammalian IGFBP-1 sequences and am

Rab11 is an apically located small GTP-binding protein in epithelial tissues.

Rab proteins are involved in many aspects of dynamic vesicle processing within eukaryotic cells. We have previously identified Rab11 in gastric parietal cell tubulovesicle membranes. We have produced a monoclonal antibody that is specific for Rab11. In all rabbit tissues examined, Rab11 immunoreactivity was highly enriched in epithelial cells. In the gastric fundus, parietal cells were stained in a pattern consistent with localization on tubulovesicles. Surface mucous cells of both the fundus and antrum demonstrated punctate subapical staining. Ileal and proximal colonic enterocyte labeling wa

Regulation of corticotropin-releasing factor-binding protein expression in cultured rat astrocytes.

The regulation of brain corticotropin-releasing factor (CRF)-binding protein (BP), an endogenous modulator of the CRF family of neuropeptides, has been difficult to pursue due to a lack of basal expression in a known cell line or primary cells in vitro. In light of the ability of intracellular factors to modulate neuronal and glial function, we examined the effects of a variety of signal transduction modulators on CRF-BP expression in cultured astrocytes. In particular, the effect of agents that stimulate protein kinase A and protein kinase C pathways was evaluated. CRF-BP was measured using a

Isolation and biochemical characterization of a Ca2+-independent alpha-latrotoxin-binding protein.

alpha-Latrotoxin, a black widow spider neurotoxin, can bind to high affinity receptors on the presynaptic plasma membrane and stimulate massive neurotransmitter release in the absence of Ca2+. Neurexins, previously isolated as alpha-latrotoxin receptors, require Ca2+ for their interaction with the toxin and, thus, may not participate in the Ca2+-independent alpha-latrotoxin activity. We now report the isolation of a novel protein that binds alpha-latrotoxin with high affinity in the presence of various divalent cations (Ca2+, Mg2+, Ba2+, and Sr2+) as well as in EDTA. This protein, termed here

In vitro plasma protein binding of zileuton and its N-dehydroxylated metabolite.

An ultrafiltration technique or equilibrium dialysis has been used to study the in vitro human plasma protein binding of racemic zileuton, its individual enantiomers, and its pharmacologically inactive metabolite N-dehydroxyzileuton. The plasma protein binding of zileuton and N-dehydroxyzileuton over the concentration range of 0.1 to 100 mg/L averaged 93.1 +/- 0.22 and 92.0 +/- 0.12%, respectively. However, there appeared to be a stereoselective effect, with the R(+) enantiomer of zileuton demonstrating greater binding to plasma proteins than the S(-) enantiomer (96 vs 88%, respectively). Zile…

Penicillin-binding protein 2b of Streptococcus pneumoniae in piperacillin-resistant laboratory mutants.

In Streptococcus pneumoniae, alterations in penicillin-binding protein 2b (PBP 2b) that reduce the affinity for penicillin binding are observed during development of beta-lactam resistance. The development of resistance was now studied in three independently obtained piperacillin-resistant laboratory mutants isolated after several selection steps on increasing concentrations of the antibiotic. The mutants differed from the clinical isolates in major aspects: first-level resistance could not be correlated with alterations in the known PBP genes, and the first PBP altered was PBP 2b. The point m

Characterization of glycoprotein gp43, the major laminin-binding protein of Paracoccidioides brasiliensis.

We have demonstrated that laminin mediates the adhesion of P. brasiliensis to monolayers of epithelial cells through specific binding to the surface glycoprotein gp43. This binding seems to be related to the fungal pathogenesis. We now report the confirmation of these findings by scanning electron microscopy and show that some isolates that do not secrete gp43 do express the protein as seen by studying whole cell extracts. These results confirm the ability of these strains to produce paracoccidioidomycosis but should not be used for serological purposes since the absence of gp43 in exoantigens

Rat heart fatty acid-binding protein content is increased in experimental diabetes.

Cytoplasmic fatty acid-binding protein (FABP) was assayed immunochemically in hearts from rats with insulin-dependent (IDDM) or non-insulin-dependent diabetes mellitus (NIDDM). FABP contents were 34% higher in IDDM and 103% higher in NIDDM hearts than in respective age-matched controls. FABP levels returned to control values when islets of Langerhans were transplanted into diabetic IDDM animals. In the diabetic hearts the activity of fructose-6-phosphate kinase decreased (IDDM and NIDDM animals), while that of 3-hydroxyacyl-CoA dehydrogenase increased (NIDDM animals only). These data indicate

Purification and characterization of a soluble salicylic acid-binding protein from tobacco.

Previously, we identified a soluble salicylic acid (SA)-binding protein (SABP) in tobacco whose properties suggest that it may play a role in transmitting the SA signal during plant defense responses. This SA-binding activity has been purified 250-fold by conventional chromatography and was found to copurify with a 280-kDa protein. Monoclonal antibodies capable of immunoprecipitating the SA-binding activity also immunoprecipitated the 280-kDa protein, indicating that it was responsible for binding SA. These antibodies also recognized the 280-kDa protein in immunoblots of the partially purified

Hypoxia up-regulates the activity of a novel erythropoietin mRNA binding protein.

The mechanisms which control the production of erythropoietin (Epo) remain enigmatic. Recent data suggest that the half-time of Epo messenger RNA (mRNA) is increased by hypoxia in Hep 3B cells, a human hepatoma line. The post-transcriptional regulation of other rapidly degraded mRNAs is mediated by sequence-specific mRNA binding proteins. In order to determine if Epo mRNA specific binding proteins exist, we probed cytosolic lysates from Hep 3B cells and mouse tissues with radiolabeled Epo RNA. A cytosolic protein that binds specifically to Epo RNA was identified in the Epo-producing, hepatobla

Adoptive transfer of experimental autoimmune uveoretinitis induced by interphotoreceptor retinoid-binding protein.

The immunopathogenic mechanisms of experimental autoimmune uveoretinitis (EAU) induced by interphotoreceptor retinoid-binding protein (IRBP) were studied. Lymph node (LN) cells or spleen cells were collected from donor rats 14 days after the immunization with IRBP. After the 3-day pre-incubation of these cells with either IRBP or Concanavalin A (Con A), the cells were transferred to naive syngeneic rats intraperitoneally. EAU was successfully transferred by LN cells pre-cultured with IRBP, while EAU was poorly transferred by LN cells pre-cultured with Con A. On the other hand, spleen cells pre

Plasma sex-steroid binding protein in a seasonally breeding reptile, Alligator mississippiensis.

The properties of a sex-steroid binding protein (SSBP) in the plasma of the American alligator, Alligator mississippiensis, were partially characterized. Alligator SSBP has a sedimentation coefficient of 4S in a 5-20% sucrose gradient. It binds to estradiol-17 beta (E2) and testosterone (T) with limited capacities and moderate affinities (association constant for [3H]E2 is 4.70 +/- 0.09 X 10(8) M-1 and for [3H]T is 1.05 +/- 0.07 X 10(8) M-1, mean +/- SEM of six determinations). Plasma SSBP level, as measured by plasma [3H]E2 binding capacity, varies from 30 to 140 nmol per liter plasma (nM) an

Characterization of multiple forms of folate-binding protein from human leukemia cells.

Folate-binding proteins were isolated from the particulate fraction (44,000 X g pellet) and the soluble fraction (44,000 X g supernate) of the homogenate of a spleen obtained from a patient who had an acute leukemic (blast) transformation of chronic myelogenous leukemia. The folate-binding activity which was obtained from the particulate fraction by solubilization with 1% Triton X-100 could be resolved into two binding proteins (Mr 310,000 and 28,000) by gel filtration through Sephadex G-200 after incubation with excess [3H]pteroylglutamic acid (PteGlu). The folate-binding protein in the solub

Pityriasis rubra pilaris, vitamin A and retinol-binding protein: a case study.

A patient with longstanding erythroderma and decreased sweat secretion due to the classical adult form of pityriasis rubra pilaris is described. The patient did not respond to oral megadoses of Vitamin A, even though a large increase of liver content of Vitamin A was demonstrated. Retinol-binding protein levels in serum of this patient and his relatives were normal. Danazol (Danatrol, Winthrop) therapy caused an increase of retinol-binding protein level, but clinical improvement did not occur.

Protein binding of some non-steroidal anti-inflammatory drugs in rheumatoid arthritis.

The protein binding of 4 non-steroidal anti-inflammatory drugs and warfarin was investigated in 10 patients with rheumatoid arthritis and in a matched control group of patients with osteoarthritis. There were no differences between the groups in the free fractions of the 5 drugs studied. Fluorescent probe studies also showed only minor differences between groups. The results do not support previous suggestions that drug protein binding is altered in rheumatoid arthritis. Albumin concentration was positively correlated with the binding of salicylate, but not with binding of the other 4 drugs. F…

Hyperthyroidism with normal serum total T4, T3, and thyroid binding protein concentration.

A 60-year-old woman, free of other disease, with severe clinical hyperthyroidism had normal total serum T4 with elevated free T4 concentration and T3 resin uptake characteristic of hyperthyroidism, with thyroid binding globulin (TBG) deficiency. However, serum TBG concentration as determined by immunoassay was normal, suggesting that the biochemical defect affecting the T4 binding sites did not affect is antigenicity. Her total and free serum T3 levels were low-normal with a markedly elevated reverse T3 concentration. These findings, in the presence of clinical hyperthyroidism, are consistent
