Regulation of Intestinal Epithelial Barrier Function by Long Noncoding RNA uc.173 through Interaction with MicroRNA 29b.

The mammalian intestinal epithelium establishes a selectively permeable barrier that supports nutrient absorption and prevents intrusion by noxious luminal substances and microbiota. The effectiveness and integrity of the barrier function are tightly regulated via well-controlled mechanisms. Long noncoding RNAs transcribed from ultraconserved regions (T-UCRs) control diverse cellular processes, but their roles in the regulation of gut permeability remain largely unknown. Here we report that the T-UCR uc.173 enhances intestinal epithelial barrier function by antagonizing microRNA 29b (miR-29b).

Zc3h12a is an RNase essential for controlling immune responses by regulating mRNA decay.

Toll-like receptors (TLRs) recognize microbial components, and evoke inflammation and immune responses. TLR stimulation activates complex gene expression networks that regulate the magnitude and duration of the immune reaction. Here we identify the TLR-inducible gene Zc3h12a as an immune response modifier that has an essential role in preventing immune disorders. Zc3h12a-deficient mice suffered from severe anaemia, and most died within 12 weeks. Zc3h12a(-/-) mice also showed augmented serum immunoglobulin levels and autoantibody production, together with a greatly increased number of plasma ce

Increased expression and localization of the RNA-binding protein HuD and GAP-43 mRNA to cytoplasmic granules in DRG neurons during nerve regeneration.

The neuronal-specific RNA-binding protein, HuD, binds to a U-rich regulatory element of the 3' untranslated region (3' UTR) of the GAP-43 mRNA and delays the onset of its degradation. We have recently shown that overexpression of HuD in embryonic rat cortical cells accelerated the time course of normal neurite outgrowth and resulted in a twofold increase in GAP-43 mRNA levels. Given this evidence, we sought to investigate the involvement of HuD during nerve regeneration. It is known that HuD protein and GAP-43 mRNA are expressed in the dorsal root ganglia (DRG) of adult rat and that GAP-43 is

The 5'-untranslated region of picornaviral genomes.

Picornaviruses are small naked icosahedral viruses with a single-stranded RNA genome of positive polarity. According to current taxonomy, the family includes four genera: Enterouirus (polioviruses, coxsackieviruses, echoviruses, and other enteroviruses), Rhinovirus, Curdiouirus [encephalomyocarditis virus (EMCV), mengovirus, Theiler's murine encephalomyelitis virus (TMEV)], and Aphthouirus [foot-and-mouth disease viruses (FMDV)]. There are also some, as yet, unclassified picornaviruses [e.g., hepatitis A virus (HAW] that should certainly be assessed as a separate genus. Studies on the molecula

Dengue virus structural differences that correlate with pathogenesis.

The understanding of dengue virus pathogenesis has been hampered by the lack of in vitro and in vivo models of disease. The study of viral factors involved in the production of severe dengue, dengue hemorrhagic fever (DHF), versus the more common dengue fever (DF), have been limited to indirect clinical and epidemiologic associations. In an effort to identify viral determinants of DHF, we have developed a method for comparing dengue type 2 genomes (reverse transcriptase PCR in six fragments) directly from patient plasma. Samples for comparison were selected from two previously described dengue

miR-29b suppresses CML cell proliferation and induces apoptosis via regulation of BCR/ABL1 protein.

MicroRNAs (miRNAs) are small RNAs that regulate gene expression posttranscriptionally and are critical for many cellular pathways. Recent evidence has shown that aberrant miRNA expression profiles and unique miRNA signaling pathways are present in many cancers. Here, we demonstrate that miR-29b is markedly lower expressed in CML patient samples. Bioinformatics analysis reveals a conserved target site for miR-29b in the 3'-untranslated region (UTR) of ABL1. miR-29b significantly suppresses the activity of a luciferase reporter containing ABL1-3'UTR and this activity is not observed in cells tra

Temperature triggers immune evasion by Neisseria meningitidis.

Neisseria meningitidis has several strategies to evade complement-mediated killing, and these contribute to its ability to cause septicaemic disease and meningitis. However, the meningococcus is primarily an obligate commensal of the human nasopharynx, and it is unclear why the bacterium has evolved exquisite mechanisms to avoid host immunity. Here we demonstrate that mechanisms of meningococcal immune evasion and resistance against complement increase in response to an increase in ambient temperature. We have identified three independent RNA thermosensors located in the 5' untranslated region

microRNAs and muscle disorders.

MicroRNAs (miRNAs) are a class of non-coding regulatory RNAs of approximately 22 nucleotides in length. miRNAs are highly conserved across a number of species, including plants, worms and humans. miRNAs regulate gene expression post-transcriptionally, primarily by associating with the 3' untranslated region (UTR) of their regulatory target mRNAs. Recent work has begun to reveal roles for miRNAs in a wide range of biological processes, including cell proliferation, differentiation and apoptosis. miRNAs are expressed in cardiac and skeletal muscle, and dysregulated miRNA expression has been corr

Suppression of cell growth and invasion by miR-205 in breast cancer.

MicroRNAs (miRNAs) are endogenous, small, non-coding RNAs, which are capable of silencing gene expression at the post-transcriptional level. In this study, we report that miR-205 is significantly underexpressed in breast tumor compared to the matched normal breast tissue. Similarly, breast cancer cell lines, including MCF-7 and MDA-MB-231, express a lower level miR-205 than the non-malignant MCF-10A cells. Of interest, ectopic expression of miR-205 significantly inhibits cell proliferation and anchorage independent growth, as well as cell invasion. Furthermore, miR-205 was shown to suppress lu

Aberrant neuronal differentiation and splicing defects in Congenital Myotonic Dystrophy (DM1) iPSC models.

Myotonic Dystrophy type 1 (DM1) is an autosomal multisystem disorder manifested due to unstable CTG nucleotide repeat expansion within the 3'-untranslated region of the dystrophia myotonica protein kinase (DMPK) gene. Although progress towards understanding of molecular pathogenesis in muscle and heart has been made, the pathways that affect the brain in DM1 is fundamentally unknown. In addition, the congenital DM1 manifest even more complicated brain abnormalities. Despite the wealth of existing cellular and animal models, iPSCs based studies are being fostered as they replicate the human mod

Disruption of Zika Virus xrRNA1-Dependent sfRNA1 Production Results in Tissue-Specific Attenuated Viral Replication.

The Zika virus (ZIKV), like other flaviviruses, produces several species of sub-genomic RNAs (sfRNAs) during infection, corresponding to noncoding RNA fragments of different lengths that result from the exonuclease degradation of the viral 3' untranslated region (UTR). Over the course of infection, these sfRNAs accumulate in the cell as a result of an incomplete viral genome degradation of the 3' UTR by the host 5' to 3' exoribonuclease, Xrn1. The halting of Xrn1 in the 3' UTR is due to two RNA pseudoknot structures in the 3' UTR, termed exoribonuclease-resistant RNA1 and 2 (xrRNA1&2). Studies

BACKGROUND: Circular RNAs (circRNAs) represent a class of non-coding RNAs (ncRNAs) which are widely expressed in mammals and tissue-specific, of which some could act as critical regulators in the atherogenesis of cerebrovascular disease. However, the underlying mechanisms by which circRNA regulates the ectopic phenotype of endothelial cells (ECs) in atherosclerosis remain largely elusive. METHODS: CCK-8, transwell, wound healing and Matrigel assays were used to assess cell viability, migration and tube formation. QRT-qPCR and Immunoblotting were used to examine targeted gene expression in dif

A HuD-ZBP1 ribonucleoprotein complex localizes GAP-43 mRNA into axons through its 3' untranslated region AU-rich regulatory element.

Localized translation of axonal mRNAs contributes to developmental and regenerative axon growth. Although untranslated regions (UTRs) of many different axonal mRNAs appear to drive their localization, there has been no consensus RNA structure responsible for this localization. We recently showed that limited expression of ZBP1 protein restricts axonal localization of both β-actin and GAP-43 mRNAs. β-actin 3'UTR has a defined element for interaction with ZBP1, but GAP-43 mRNA shows no homology to this RNA sequence. Here, we show that an AU-rich regulatory element (ARE) in GAP-43's 3'UTR is nece

Non-coding RNAs regulating autophagy in Osteoporosis: mechanistic insights and therapeutic potential.

Osteoporosis (OP) is a systemic skeletal disease, defined by low bone mass and microarchitectural deterioration of bone, which increases the risk of fragility fractures. Autophagy is a dynamic and evolutionarily conserved process that regulates the degradation and recycling of cellular proteins and organelles in response to various physiological and pathophysiological conditions. Autophagy is a major stress-response pathway and it is critically involved in regulating the metabolic and homeostatic functions of bone cells. Non-coding RNAs (ncRNAs) are a diverse group of RNAs that do not code for

Roles of Cellular Noncoding RNAs in Productive Infection of Influenza A Virus.

Influenza A virus (IAV) causes annual epidemics and occasional pandemics, remaining a threat to public health and economy worldwide. It has been established that many cellular noncoding RNAs (ncRNAs) are differentially expressed during IAV infection. These ncRNAs vigorously modulate the productive infection of the virus through diverse mechanisms, such as directly affecting the virus life cycle, altering the cellular signaling pathways, or regulating the host's innate immune response to viral infection. Some of the ncRNAs are deemed to be essential for IAV replication, while others, conversely

The SL-I structural element of the 3' UTR region of West Nile virus participates in the regulation of viral translation.

The genome of West Nile virus (WNV) is a positive, ssRNA molecule, which encodes a single ORF flanked by UTRs (5' and 3' UTRs). These UTRs are enriched with structural RNA elements that play critical roles in the viral cycle, including translation, replication and encapsidation. The 3' UTR is crucial for translation control by the combined enhancer and repressor activities of different structural elements. This manuscript provides new roles for the 3' UTR SL-I element as a translation inhibitor in WNV in cis. This inhibitory effect is primarily observed in a cap-dependent translation context,

Emerging roles of epigenetics in the pathogenesis of sarcopenia.

Sarcopenia is an age-associated degenerative disorder of skeletal muscle, characterized by the progressive decline in muscle mass and function, which increases the risk of falls, injuries, and accidental death in older adults. Deciphering its pathogenic mechanisms is crucial for advancing early detection, precision prevention, and ultimately improving the quality of life for the elderly population. Increasing evidence suggests that epigenetic regulation plays a central role in driving sarcopenia. DNA methylation, chromatin remodeling, and noncoding RNA regulation collectively accelerate the de

Exploring shared hypoxic pathways in cancer and obstructive sleep apnea: A critical review of extracellular vesicles-non-coding RNA signaling and biological plausibility.

Hypoxia plays a central role in the pathophysiology of both cancer and obstructive sleep apnea (OSA), while through biologically distinct mechanisms. Chronic hypoxia (CH), typical of the tumor microenvironment (TME), drives angiogenesis, metabolic rewiring, and malignant progression, whereas intermittent hypoxia (IH), a hallmark of OSA, triggers fluctuating oxidative stress, sympathetic activation, and inflammatory signaling. In recent years, extracellular vesicles (EVs) and their non-coding RNA (ncRNA) cargo, particularly microRNAs (miRNAs) and circular RNAs (circRNAs), have emerged as critic

Exploring the role of epigenetics in the processes related to the development of endometrosis in the mare.

Endometrosis is a chronic degenerative condition of the mare endometrium characterized by progressive fibrosis and glandular alterations that impair uterine function and fertility. Its pathogenesis involves persistent inflammation, the activation of myofibroblasts, and the accumulation of extracellular matrix (ECM), leading to disrupted glandular secretion and compromised maintenance of pregnancy. While histopathological studies of endometrosis are well described, the underlying molecular mechanisms remain incompletely understood. Emerging evidence highlights the crucial role of epigenetic reg
