Interferon type I receptor-deficient mice have altered disease symptoms in response to influenza virus.

The role of type I interferons (IFNs) in mediation of acute viral symptoms (fever, somnolence, anorexia, etc.) is unknown. To determine the role of type I IFN in selected symptom development, body temperature and sleep responses to a marginally lethal dose of X-31 influenza virus were examined in mice with a targeted mutation of the IFN receptor type I (IFN-RI knockouts) and compared to wild-type 129 SvEv control mice. Mice were monitored for 48 h to determine baseline temperature and sleep profiles prior to infection, and then for 9 days following infection. Hypothermic responses to virus wer

Altered wakeful theta activity characterizes levodopa-induced dyskinesia in Parkinson's disease.

Slow-wave activity during sleep facilitates synaptic downscaling, while theta activity during wakefulness reflects synaptic upscaling, and both processes may be altered in levodopa-induced dyskinesia (LID) in Parkinson's disease (PD). We compared actigraphy and high-density EEG in 12 healthy volunteers and three PD cohorts: early stage (EPD, n = 12), advanced non-dyskinetic (ADV, n = 13), and advanced dyskinetic (DYS, n = 11). Participants completed one week of actigraphy monitoring, followed by two resting-state EEG recordings conducted separately in the morning and evening. Wake-theta activi

[THE EFFECT OF MOTION SICKNESS ON THE SLEEP-WAKE CYCLE IN RATS EXPOSED TO PRENATAL HYPOXIA].

This study is a continuation of our research of phenomenology and mechanisms of motion sickness (MS) and the relation of this phenomenon to features of sleep-wake cycle (SWC) changes. It presents data about the effect of MS on SWC in 30-day-old intact rats and rats exposed previously to prenatal hypoxia on days 13 and 19 of gestation. In all groups of animals MS was shown to reduce significantly the waking time and to increase the time of paradoxical sleep (PS). For the first time it became possible to reveal a connection between hypothalamic mechanisms controlling MS and SWC and to suggest th

Twenty-four hour quantitative-EEG and in-vivo glutamate biosensor detects activity and circadian rhythm dependent biomarkers of pathogenesis in Mecp2 null mice.

Mutations in the X-linked gene encoding methyl-CpG-binding protein 2 (Mecp2) cause most cases of Rett syndrome (RTT). Currently there is no cure for RTT. Abnormal EEGs are found in 100% of RTT cases and are associated with severe sleep dysfunction, the cause of which is not well understood. Mice deficient in MeCP2 protein have been studied and characterized for their neuropathological and behavioral deficits to better understand RTT. With the goal to study the non-ictal EEG correlates in symptomatic Mecp2 KO mice (Mecp2(tm1.1Bird/y)), and determine novel EEG biomarkers of their reported progre

TNF-NF-kappaB signaling mediates excessive somnolence in hemiparkinsonian rats.

Daytime somnolence is common in patients with Parkinson's disease (PD); however there is a lack of understanding of the cellular mechanisms involved in mediating these effects. It has been hypothesized that microglial activation and the subsequent increase of pro-inflammatory cytokines play an important role in the pathogenesis of PD. Because some cytokines are involved in the regulation of sleep, this study was designed to determine if tumor necrosis factor (TNF) and interleukin-1beta (IL-1beta), mediate daytime somnolence in the proteasome inhibitor (MG-132)-induced hemiparkinsonian rat mode

Anesthesia-Informed Approaches to Insomnia: Shared Neural Circuits, Biomimetic Sleep, and Emerging Interventional Strategies.

BACKGROUND: Insomnia affects approximately 10-30% of adults, burdening quality of life, cognition, and health. Although cognitive behavioral therapy for insomnia (CBT-I) and approved pharmacotherapies remain standard care, limited accessibility, durability, and efficacy in refractory populations have motivated interest in mechanism-based alternatives, including approaches derived from anesthetic pharmacology. OBJECTIVE: This narrative review examines the convergent neural circuits underlying anesthesia-induced and physiological sleep, critically appraises evidence for anesthetic agents-dexmed

Role and mechanism of gut microbiota and metabolites in schizophrenia complicated with sleep disorder.

Schizophrenia (SCZ) is a major mental disorder with a high disability rate, and its pathogenesis involves the interaction of multiple factors such as genetics, environment, immunity, and neurodevelopment. Most SCZ patients are complicated with significant sleep disorder (SD), manifested as insomnia, sleep fragmentation, reduction in slow-wave sleep, and circadian rhythm disturbance. This comorbidity not only aggravates the severity of psychiatric symptoms but also significantly impacts treatment adherence and long-term prognosis. In recent years, the role of gut microbiota and its metabolites

Extended evaluation of the efficacy of a proactive forced oscillation technique-based auto-CPAP algorithm.

PURPOSE: Previous studies revealed substantially varying therapy efficacy of automatic continuous positive airway pressure (APAP) devices in the treatment of obstructive sleep apnea (OSA). We evaluated the efficacy of a new APAP device using the forced oscillation technique (FOT) to evaluate upper airway obstruction during apneas and flow contour analyses during hypopneas. METHODS: Forty-six initially diagnosed OSA patients were included and the pressure range was set from 5 to 20 hPa. Therapy efficacy was assessed based on the reduction of apnea-hypopnea index (AHI), improvement of objective

Bed Rest and Hypoxic Exposure Affect Sleep Architecture and Breathing Stability.

Objective: Despite over 50 years of research on the physiological effects of sustained bed rest, data characterizing its effects on sleep macrostructure and breathing stability in humans are scarce. This study was conducted to determine the effects of continuous exposure to hypoxia and sustained best rest, both individually and combined, on nocturnal sleep and breathing stability. Methods: Eleven participants completed three randomized, counter-balanced, 21-days trials of: (1) normoxic bed rest (NBR, PIO2 = 133.1 ± 0.3), (2) hypoxic ambulatory confinement (HAMB, PIO2 = 90.0 ± 0.4) and (3) hypo

Very early screening for sleep-disordered breathing in acute coronary syndrome in patients without acute heart failure.

BACKGROUND: Obstructive sleep apnea (OSA) is frequently associated with acute coronary syndrome (ACS). Screening of sleep-disordered breathing (SDB) has not been previously evaluated in ACS within 72 h in intensive care settings and its management could potentially enhance patients' prognosis. This pilot study assessed the feasibility of SDB screening at the early phase of ACS. METHODS: All consecutive patients admitted to the coronary care unit (CCU) for ACS without acute heart failure underwent one overnight-attended polysomnography (PSG) within 72 h after admission. A telemonitoring (TM) s

Dynamic changes of gamma activities of somatic cortical evoked potentials during wake-sleep states in rats.

Somatic evoked potentials (SEPs) from three brain sites elicited by electrical stimulation in 10 rats were recorded throughout wake-sleep states with intrinsic changes in temporal architectures under different vigilance states. Based on the patterns of spontaneous brain and muscle activities, three characteristic vigilance states could be classified: awake, slow-wave sleep (SWS), and paradoxical sleep (PS). Spontaneous gamma activities prominently appeared under awake and PS states, but less under SWS. SEP was filtered out via a zero-phase highpass filter (20 Hz) to extract the gamma activity

Excessive daytime sleepiness in patients suffering from different levels of obstructive sleep apnoea syndrome.

Excessive daytime sleepiness (EDS) is a frequent symptom of patients with obstructive sleep apnoea (OSA). EDS is a high-risk factor for accidents at work and on the road. Thirty untreated patients with different levels of severity of OSA were studied concerning night sleep and EDS. The criterion for severity was the respiratory disturbance index (RDI): 15 patients were classified as 'moderately' apnoeic (RDI < 40), 15 as 'severely' apnoeic (RDI > 40). Following night-time polysomnography, objective and subjective aspects of EDS were studied. To assess objective EDS the Maintenance of Wakefulne

Dietary palmitic acid to oleic acid ratio modulates energy metabolism and biological rhythms in young healthy Japanese males.

The present study investigated the potential role of the composition of dietary fatty acids in the regulation of biological rhythms, such as the sleep architecture, core body temperature and leukocyte clock gene expression, in subjects fed meals rich in palmitic acid (PA) or oleic acid (OA). Eleven males participated in two sessions of indirect calorimetry in a whole-room metabolic chamber. In each session, subjects consumed three meals rich in PA (44·3 % of total fat as PA and 42·3 % as OA) or OA (11·7 % of total fat as PA and 59·3 % as OA) in the metabolic chamber. The ratio of PA to OA in p

Selection on PER2 Phosphorylation Regulation Underlies Atypical Mammalian Circadian Rhythms.

Circadian rhythms are fundamental biological processes that enhance survival and reproductive fitness by coordinating physiology and behavior with predictable environmental cycles. However, in some species, circadian rhythms are weakened, irregular, or even undetectable, likely reflecting evolutionary responses to extreme environments or specialized ecological niches. Nevertheless, the genetic basis of these atypical phenotypes remains incompletely understood. Here, we investigated the molecular evolution of 20 PER2-associated genes across 71 representative mammalian species. We identified fou

Adaptive pressure support servo-ventilation: a novel treatment for Cheyne-Stokes respiration in heart failure.

Adaptive servo-ventilation (ASV) is a novel method of ventilatory support designed for Cheyne-Stokes respiration (CSR) in heart failure. The aim of our study was to compare the effect of one night of ASV on sleep and breathing with the effect of other treatments. Fourteen subjects with stable cardiac failure and receiving optimal medical treatment were tested untreated and on four treatment nights in random order: nasal oxygen (2 L/min), continuous positive airway pressure (CPAP) (mean 9.25 cm H(2)O), bilevel (mean 13.5/5.2 cm H(2)O), or ASV largely at the default settings (mean pressure 7 to

Rat strain differences suggest a role for corticotropin-releasing hormone in modulating sleep.

Corticotropin-releasing hormone (CRH) mediates many of the hormonal, behavioral, and autonomic responses to a variety of stressors. There is also evidence suggesting that CRH may be involved in the modulation of physiologic waking. Lewis (LEW) rats possess a hypothalamic gene defect that results in reduced synthesis and secretion of CRH relative to genetically related Fischer 344 (F344) and Sprague-Dawley (Sp-D) rat strains. We therefore hypothesized that LEW rats would spend less time awake, and more time asleep, than either F344 or Sp-D rats. Adult male LEW, F344, and Sp-D rats were surgical

Shooting a high-density electroencephalographic picture on sleep in children with attention-deficit/hyperactivity disorder.

STUDY OBJECTIVES: Sleep-related slow-wave activity (SWA) has been recognized as a marker of synaptic plasticity. In children affected by attention deficit hyperactivity disorder (ADHD), SWA is mainly located in the central rather than frontal regions, reflecting a maturational delay. A detailed subjective and objective sleep investigation, including a full night video-polysomnography (PSG-HD-EEG), was performed on 30 consecutive drug naïve outpatients with a diagnosis of ADHD. They received a diagnosis of sleep disorders in 29/30 cases, and most of them had a past history of sleep problems. Th
