Contractile and electrophysiologic characterization of optimized self-organizing engineered heart tissue.

BACKGROUND: Engineered heart tissue (EHT) is being developed for clinical implantation in heart failure or congenital heart disease and therefore requires a comprehensive functional characterization and scale-up of EHT. Here we explored the effects of scale-up of self-organizing EHT and present detailed electrophysiologic and contractile functional characterization. METHODS: Fibers from EHT were generated from self-organizing neonatal rat cardiac cells (0.5×10(6) to 3×10(6)/fiber) on fibrin. We characterized contractile patterns and measured contractile function using a force transducer, and

The effect of a bioactive tissue-engineered sling in a rat of stress incontinence model.

In this study, we attempt to examine the feasibility of the bioactive tissue-engineered sling by using muscle precursor cells (MPCs)-seeded Poly(ε-caprolactone) (PCL) nanofiber sheet in a rat model of stress urinary incontinence (SUI). In vitro, MPCs were cultured on a PCL nanofiber sheet for one week, where the MPCs-seeded PCL nanofiber sheet showed constant twitching contraction by electrical field stimulation in an organ bath. In vivo, MPCs-seeded PCL nanofiber sheet was placed under the female rat's urethra after pudendal nerve denervation (animal model of SUI). The leak point pressure (LP

Tissue engineered bone versus alloplastic commercial biomaterials in craniofacial reconstruction.

This research was developed in order to demonstrate the tissue engineering method as an alternative to conventional methods for bone reconstruction, in order to overcome the frequent failures of alloplastic commercial biomaterials, allografts and autografts. Tissue engineering is an in vitro method used to obtain cell based osteoinductive bone grafts. This study evaluated the feasibility of creating tissue-engineered bone using mesenchymal cells seeded on a scaffold obtained from the deciduous red deer antler. We have chosen mesenchymal stem cells because they are easy to obtain, capable to di…

Biodegradable polymer coating promotes the epithelization of tissue-engineered airway prostheses.

OBJECTIVE: We have developed a prosthesis that includes a collagen layer for tracheobronchial reconstruction and applied it in a canine model. In previous studies luminal epithelization remained partial or rather slow because of the early disintegration of the collagen layer. We have improved this type of prosthesis by coating the luminal surface with a biodegradable polymer, which serves to protect the collagen layer. The effect of the polymer coating on the epithelization of the luminal surface of the prosthesis was examined. METHODS: The main frame consisted of a polypropylene mesh tube, m

Replacement of the left main bronchus with a tissue-engineered prosthesis in a canine model.

BACKGROUND: Stenosis of the left main bronchus caused by inflammatory diseases and neoplasms is a serious clinical problem because it can cause obstructive pneumonia and may require pneumonectomy. As an alternative to various treatments currently available, including balloon dilatation, stenting, and bronchoplasty, we propose the use of a prosthesis developed based on the concept of in situ tissue engineering for replacement of the left main bronchus. METHODS: The main frame of the tissue-engineered prosthesis is a polypropylene mesh tube, 12 to 15 mm in inner diameter and 30 mm in length, wi…

Repairing peripheral nerve defects with tissue engineered artificial nerves in rats.

OBJECTIVE: To observe the effect of tissue engineered nerves in repairing peripheral nerve defects (about 1.5 cm in length) in rats to provide data for clinical application. METHODS: Glycerinated sciatic nerves (2 cm in length) from 10 Sprague Dawley (SD) rats (aged 4 months) were used to prepare homologous dermal acellular matrix. Other 10 neonate SD rats (aged 5-7 days) were killed by neck dislocation. After removing the epineurium, the separated sciatic nerve tracts were cut into small pieces, then digested by 2.5 g/L trypsin and 625 U/ml collagenase and cultured in Dulbecco's modified Eag

Survival and integration of tissue-engineered corneal stroma in a model of corneal ulcer.

Tissue-engineered replacement of diseased or damaged tissue has become a reality for some types of tissue, such as skin and cartilage. Tissue-engineered corneal stroma represents a promising concept to overcome the limitations of cornea replacement with allograft. In this study, porcine cornea was decellularized by a series of extraction methods, and the in vivo biocompatibility of the scaffold was measured subcutaneously in rabbits (n = 8). These were not acutely rejected and no abscesses were observed by hematoxylin and eosin staining at the 8th week, indicating that the scaffolds had good b

Tissue-engineered cartilage as a graft source for laryngotracheal reconstruction: a pig model.

OBJECTIVE: To evaluate the feasibility of using tissue-engineered cartilage for laryngotracheal reconstruction in the pig model. DESIGN: Auricular cartilage was harvested from 3 young swine. The cartilage was digested, processed, and suspended and a cell culture was obtained. The cells were then suspended in 3 mL of a 30% solution of a biodegradable polymer (Pluronic F-127) (polyethylene oxide/polypropylene oxide copolymer) at a cellular concentration of 50 x 10(6) cells/mL. This suspension was then implanted subcutaneously into each pig's dorsum. Eight weeks after implantation, the cartilage

Effective management of difficult surgical defects using tissue-engineered skin.

BACKGROUND: Graftskin, a bilayered living skin construct, is an effective therapeutic option in the management of chronic venous ulcerations and simple acute surgical excisions. However, it is not routinely used in the management of complicated surgical wound defects. OBJECTIVE: To determine the effectiveness of graftskin as a therapeutic modality in difficult surgical defects. METHODS: Two patients with complex surgical wound defects after Mohs micrographic surgery underwent a single application of graftskin. The engineered skin was fenestrated and sutured in place. The wounds were examined

The use of tissue-engineered skin (Apligraf) to treat a newborn with epidermolysis bullosa.

BACKGROUND: Inherited epidermolysis bullosa (EB) is a mechanobullous disorder. The Dowling-Meara variant, a subtype of EB, is characterized by widespread blister formation that may include the oral cavity and nails. Many patients with the Dowling-Meara phenotype are at increased risk of sepsis and death during infancy. The treatment of EB is generally supportive. The tissue-engineered skin used (Apligraf) is a bilayered human skin equivalent developed from foreskin. It is the only Food and Drug Administration-approved skin equivalent of its kind. It is approved for the treatment of venous ulce

Tissue engineered skin: Apligraf, a bi-layered living skin equivalent.

Driven by the need for a readily available, non-immunological tissue that possesses many of the characteristics of normal human skin, tissue-engineered skin has been developed. For over a decade, laboratory grown or processed skin has been under investigation and, in some cases, available as an alternative to autologous grafts. Apligraf, derived from neonatal foreskin and bovine type I collagen, is the first bi-layered living skin equivalent approved in the US and other countries for use in venous ulcers. Apligraf is effective both in the treatment of refractory venous ulcers and for acute wou

From Design to Application: Advanced Cellulose Scaffolds for Engineered Tissue Regeneration.

The regeneration of complex tissues demands advanced scaffolds that offer biomimetic support and tissue-specific bioactive guidance. However, the materials in clinic face big challenges with immune rejection, limited donors, and unsatisfactory inductive activity. Fortunately, cellulose-based scaffolds have risen as a leading sustainable platform, considering their natural abundance, inherent biocompatibility, and highly tunable properties. This review comprehensively presented their evolution from rational design to potential clinical application. The primary cellulose sources and key detailed

Early economic evaluation of tissue engineered vascular access grafts for hemodialysis.

AIMS: Vascular access (VA) is essential for hemodialysis in patients with end-stage kidney disease. Expanded polytetrafluoroethylene (ePTFE) grafts are used for VA but are associated with high intervention rates. Tissue-engineered vascular grafts (TEVGs) are emerging as a promising alternative. However, performance and cost thresholds for TEVGs to become clinically and economically competitive to ePTFE grafts remain unclear. This early economic evaluation aims to quantify the clinical and economic impact TEVGs must achieve to be viable in routine hemodialysis care. METHODS: The Dutch VA care

Unified platform for multiplex immunofluorescence across liver tissues and engineered models.

BACKGROUND: Multiplex immunostaining combined with digital image analysis has become a central tool in hepatology research because it allows for the simultaneous visualisation and spatial mapping of multiple cellular markers within a single tissue section, providing critical insights into the complex, heterogeneous microenvironments of liver diseases like cholangiopathies, hepatocellular carcinoma, metabolic dysfunction-associated steatohepatitis or hepatic fibrosis. Nevertheless, significant barriers prevent the widespread adoption of most multiplexing platforms, including the technological c

Bioceramic bone tissue-engineered substitutes with anti-inflammatory effects in periodontitis.

Periodontitis is a chronic inflammatory oral disease characterized by irreversible alveolar bone resorption, which severely impairs oral health and even leads to tooth loss. Clinical bone augmentation therapy for alveolar bone defects mainly relies on exogenous bone substitute materials. However, traditional materials are prone to implantation failure due to the persistent inflammatory microenvironment and bacterial infection in the periodontal area. The addition of antibiotics to improve antibacterial properties not only induces bacterial resistance but also causes systemic toxic and side eff
