Parkinson's disease (PD) is the second most common type of neurogenerative disease among middleaged and older people, characterized by aggregation of alpha-synuclein and dopaminergic neuron loss. The microbiota- gut-brain axis is a dynamic bidirectional communication network and is involved in the pathogenesis of PD. The aggregation of misfolded protein alpha-synuclein is a neuropathological characteristic of PD, originates in the gut and migrates to the central nervous system (CNS) through the vagus nerve and olfactory bulb. The change in the architecture of gut microbiota increases the level…

Various human diseases are known to occur as a result of gene-environment interactions. Amongst such diseases, neurodegenerative Parkinson's disease (PD) is a complex disorder in which genetics and exposure to toxins constitute the main determinants in the onset of the disease. Many studies have reported on a link between pesticide exposure and increased risk of PD, however the role of different classes of pesticides vis-à-vis Parkinsonism has not been well elucidated. We carried out the present study to explore the role of six groups of pesticides viz botanicals, herbicides, fungicides, organ

Parkinson's disease is increasingly recognized as a disorder that extends beyond neurons. In this issue of JEM, Stevenson et al. (https://doi.org/10.1084/jem.20251245) highlight the functional consequence of α-synuclein aggregates at the brain border, uncovering their active role in reshaping lymphatic integrity and the neuroimmune macroenvironment.

α-Synuclein (αSyn) is a neuronal protein predominantly found at the synapse, involved in vesicle trafficking. αSyn aggregates are also the main component of Lewy bodies, the hallmarks of Parkinson's disease. Interactions between the N- and C-termini of αSyn play crucial roles in its behavior in solution, membrane binding, and aggregation. Studying these interactions provides valuable insights into the physiological and pathological functions of αSyn. Here, we employed photoinduced cross-linking of unmodified proteins (PICUP) to identify the transient contacts of αSyn in different conformationa

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by the pathological misfolding and aggregation of α-synuclein (α-syn), which leads to dopaminergic neuronal loss and multisystem dysfunction. Conventional clinical diagnosis is often delayed until after substantial neuronal degeneration has occurred. Recent advances in α-syn seed amplification assays (α-syn SAAs), including real-time quaking-induced conversion (RT-QuIC) and protein misfolding cyclic amplification (PMCA), have revolutionized PD biomarker research. These assays exploit the prion-like seeding and p

BACKGROUND AND AIMS: Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by the loss of dopaminergic neurons and neurological dysfunctions. The aggregation of abnormal α-synuclein (α-Syn) is closely associated with neuronal damage and acts as the major pathogenic driver mediating neurotoxicity. As critical intermediates in the aggregation cascade, α-Syn oligomers are regarded as the most neurotoxic species, which are widely distributed in both the central and peripheral nervous systems and contribute to the initiation and progression of PD. The gut-brain axis has

The definition of Parkinson's disease (PD) is undergoing a profound transformation from a "clinical syndrome" to a "biological entity", with development of two objective biological classification systems, the NSD-ISS (Neuronal Alpha-Synuclein Disease Integrated Staging System) and the SynNeurGe framework. Multimodal diagnostic tools further improve the detection of PD. This review synthesizes advances in three key domains of PD detection. First, fluid and tissue biomarkers, particularly using α-synuclein (αSyn) seed amplification assays, allow detection of synucleinopathy in cerebrospinal flui…

Disease progression in Parkinson's disease has been driven by extracellular α-synuclein prion-like seeding throughout the course of the disease and therefore not just by the intracellular accumulation of the protein in isolated aggregates. Current therapies utilizing PROTACs cannot address the extra-cellular effects of α-synuclein spreading in this manner. This article proposes PolyTACs (Polymeric Lysosome-Targeting Chimeras) as hybrid antibody-polymer conjugates which use neuronal exofacial thiol groups produced because of DJ-1/GSH dysregulation to capture α-synuclein pathological conformers

Dramatic advances have been seen over the past 5 years in neurodegenerative disease medicine. In Parkinson's disease (PD), significant advances have been made in molecular biomarkers and imaging tools. These have led to improved diagnostic accuracy for clinical trial recruitment and stratification, as well as contributing fundamentally to biomarker-based disease frameworks that define PD by its biology. In combination, these advances may enable the tracking of disease progression and improved evaluation of disease-modifying therapies in the research setting. Biofluid-based biomarkers currently

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by the loss of dopaminergic neurons and the accumulation of misfolded α-synuclein. Current treatments, including dopaminergic medications and deep brain stimulation, provide symptomatic relief but do not halt disease progression. Recent advances in molecular research have enabled the development of disease-modifying strategies targeting key pathogenic mechanisms, such as α-synuclein aggregation, mitochondrial dysfunction, and genetic mutations, including LRRK2 and GBA1. In parallel, pluripotent stem cell-derived

Parkinson's disease is the most prevalent disorder among a group of conditions known as synucleinopathies. It is characterized by the presence of intracellular inclusions composed of misfolded α-synuclein (α-Syn) protein within neurons of the central and peripheral nervous systems. The antibody-based immunotherapeutic approach has substantial promise in treating various types of synucleinopathies, including Parkinson's disease. This review critically examines the pathomechanistic impact of α-Syn on the cellular environment, with a focus on neuroinflammation and immune responses. Various strate

In this issue of Structure, Connor et al.1 combine topology and residue-level energetics of α-synuclein and tau fibrils to identify isoenergetic fold families. They reveal how conserved hotspot residues, mutations, and, potentially, environmental cues steer polymorph selection, providing a unified framework for comparing amyloid strains and classifying emergent structures.

Multiple system atrophy (MSA) is a fatal neurodegenerative disease characterized by autonomic failure and motor impairment. The hallmark pathologic finding in MSA is widespread oligodendroglial cytoplasmic inclusions rich in aggregated α-synuclein (αSyn). MSA is widely held to be an oligodendroglial synucleinopathy, and we outline lines of evidence to support this assertion, including the presence of early myelin loss. However, we also consider emerging data that support the possibility of neuronal or immune dysfunction as a primary driver of MSA. These hypotheses are placed in the context of

Amyloid fibril formation is associated with various amyloidoses, including neurodegenerative diseases such as Alzheimer's and Parkinson's diseases. Despite the numerous studies on the inhibition of amyloid formation, the prevention and treatment of a majority of amyloid-related disorders are still challenging. In this study, we investigated the effects of various plant extracts on amyloid formation of α-synuclein. We found that the extracts from Eucalyptus gunnii are able to inhibit amyloid formation, and to disaggregate preformed fibrils, in vitro. The extract itself did not lead to cell dama

The clinical definition of neurodegenerative diseases is based on symptoms that reflect terminal damage of specific brain regions. This is misleading as it tells little about the initial disease processes. Circuitry failures that underlie the clinical symptomatology are themselves preceded by clinically mostly silent, slowly progressing multicellular processes that trigger or are triggered by the accumulation of abnormally folded proteins such as Aβ, Tau, TDP-43, and α-synuclein, among others. Methodological advances in single-cell omics, combined with complex genetics and novel ways to model

Beta-glucocerebrosidase is a lysosomal hydrolase, encoded by GBA1 that represents the most common risk gene associated with Parkinson's disease (PD) and Lewy Body Dementia. Glucocerebrosidase dysfunction has been also observed in the absence of GBA1 mutations across different genetic and sporadic forms of PD and related disorders, suggesting a broader role of glucocerebrosidase in neurodegeneration. In this review, we highlight recent advances in mechanistic characterization of glucocerebrosidase function as the foundation for development of novel therapeutics targeting glucocerebrosidase in P

Pathogenic α-synuclein and tau are critical drivers of neurodegeneration, and their mutations cause neuronal loss in patients. Whether the underlying preferential neuronal vulnerability is a cell-type-intrinsic property or a consequence of increased expression levels remains elusive. Here, we explore cell-type-specific α-synuclein and tau expression in human brain datasets and use deep phenotyping as well as brain-wide single-cell RNA sequencing of >200 live neuron types in fruit flies to determine which cellular environments react most to α-synuclein or tau toxicity. We detect phenotypic and

Cognitive impairment occurs in most individuals with Parkinson's disease (PD), exacting a high toll on patients, their caregivers, and the healthcare system. In this review, we begin by summarizing the current clinical landscape surrounding cognition in PD. We then discuss how cognitive impairment and dementia may develop in PD based on the spread of the pathological protein alpha-synuclein (aSyn) from neurons in brainstem regions to those in the cortical regions of the brain responsible for higher cognitive functions, as first proposed in the Braak hypothesis. We appraise the Braak hypothesis…

The GBA1 gene encodes the lysosomal enzyme glucocerebrosidase (GCase), which is involved in sphingolipid metabolism. Biallelic variants in GBA1 cause Gaucher disease (GD), a lysosomal storage disorder characterised by loss of GCase activity and aberrant intracellular accumulation of GCase substrates. Carriers of GBA1 variants have an increased risk of developing Parkinson disease (PD), with odds ratio ranging from 2.2 to 30 according to variant severity. GBA1 variants which do not cause GD in homozygosis can also increase PD risk. Patients with PD carrying GBA1 variants show a more rapidly pro
