ALDH2 Deficiency Drives Proliferative Mammary Morphogenesis and Epithelial Cell Stemness via Oxidative Stress and Estrogen Receptor Activation.

Alcohol consumption has been linked to breast cancer, partly due to the accumulation of toxic aldehydes like acetaldehyde, a carcinogenic byproduct of ethanol metabolism. ALDH2, a key mitochondrial enzyme, detoxifies acetaldehyde and other harmful aldehydes that drive oxidative stress, DNA damage, and hormonal dysregulation, key factors in carcinogenesis. Despite the possible link between alcohol consumption and breast cancer risk, little is known about how ALDH2 deficiency itself, independent of alcohol exposure, affects mammary gland biology and cancer susceptibility. Genetic variants leadin…

ALDH2 deficiency promotes alcohol-associated liver cancer by activating oncogenic pathways via oxidized DNA-enriched extracellular vesicles.

BACKGROUND & AIMS: Excessive alcohol consumption is one of the major causes of hepatocellular carcinoma (HCC). Approximately 30-40% of the Asian population are deficient for aldehyde dehydrogenase 2 (ALDH2), a key enzyme that detoxifies the ethanol metabolite acetaldehyde. However, how ALDH2 deficiency affects alcohol-related HCC remains unclear. METHODS: ALDH2 polymorphisms were studied in 646 patients with viral hepatitis B (HBV) infection, who did or did not drink alcohol. A new model of HCC induced by chronic carbon tetrachloride (CCl4) and alcohol administration was developed and studied…

Ethylene Glycol Poisoning With Oxalate Nephropathy and Acute Kidney Injury: A Case Report and Review of the Literature.

Ethylene glycol (EG) is a hazardous alcohol present in various household and industrial products. After being metabolized by ethanol dehydrogenase and aldehyde dehydrogenase, it can produce glycolate, acetaldehyde, and oxalate, leading to acute kidney injury. Renal failure is mainly caused by tubular damage induced by metabolites such as glyoxylate and oxalate, and may also be related to tubular obstruction caused by oxalate crystal precipitation. A 35-year-old Chinese man was brought to the emergency department in an unconscious state with a suspected history of EG ingestion. Laboratory exami…

A gas-permeable edible coating alleviates CO2-induced off-flavor formation and reduces weight loss in postharvest citrus fruit.

Coating can reduce fruit weight loss and delay wrinkling, but often induces off-flavor formation. This study aims to develop coatings for reducing off-flavor formation in citrus fruits. Initially, nine single-component edible coatings were screened, identifying lipid-based carnauba (CNW) and candelilla wax (CW) as the most effective in reducing off-flavor and weight loss, respectively. Twenty CW-CNW composite coatings (ECs) were then formulated, with EC14 (2% CNW + 8% CW) showing optimal performance. The ethanol, acetaldehyde, and weight loss in EC14-coated Satsuma mandarin were 20.25%, 28.78%…

Low-oxygen storage modulates fruit flavor formation: Metabolic and regulatory insights.

Controlled atmosphere prolongs fruit storage period by reducing oxygen availability, thereby suppressing respiration, ethylene production, s … bition of fatty acid oxidation and lipoxygenase activity. Anaerobic metabolites such as ethanol and acetaldehyde may provide temporary fruity notes but often reduce aroma complexity during prolonged storage. These shifts are driven by hypoxia-responsive genes including but not limited to alcohol dehydrogenase and pyruvate decarboxylase, which are probably regu …

Mechanisms of formic acid stress response in evolved yeast for microbial-electrocatalytic systems.

Integrated electrocatalytic microbial systems (iEMS) offers a sustainable route for carbon cycling. However, formic acid by-product stress severely hinders microbial conversion. Herein, we reveal the adaptive mechanisms of Saccharomyces cerevisiae under formic acid stress during ethanol cultivation through adaptive laboratory evolution and transcriptomic analysis. Formic acid exposure systematically downregulates central carbon metabolism while upregulating the acetaldehyde branch to replenish carbon skeletons. Concurrently, cell cycle genes are activated via MAPK signaling, and ribosomal biog…

ALDH1A3 promotes aggressive basal-like pancreatic cancer through an AP-1/RUNX2 enhancer network.

The basal-like transcriptional subtype of pancreatic ductal adenocarcinoma (PDAC) is linked to therapy resistance and poor prognosis. The cancer stem cell marker aldehyde dehydrogenase 1A3 (ALDH1A3) is a critical enzyme in acetaldehyde metabolism, but the interconnection to the basal-like subtype is poorly understood. Here, we identified ALDH1A3 as a key gene, which correlates with reduced survival and increased tumor growth. Functional studies revealed interaction of ALDH1A3 with genes like FAM3C, MCC, PMEPA1, and IRS2, forming a network driving PDAC progression. Chromatin profiling showed th…

Carbon-monoxide-driven bioethanol production operates through a tungsten-dependent catalyst.

Microbial alcohol production from waste gases is a game changer for sustainable carbon cycling and remediation. While the biotechnological process using Clostridium autoethanogenum to transform syngas (H2, CO2 and CO) is blooming, scientific debates remain on the ethanol biosynthesis pathway. Here, we experimentally validated that ethanol production is initiated through a tungsten-dependent aldehyde:ferredoxin oxidoreductase (AFOR), which reduces acetate to acetaldehyde. The reaction, thermodynamically unfavorable under standard conditions, has been considered by many as unsuitable in vivo but…

Comparative Physiological and Transcriptomic Analyses Revealed Key Metabolic Pathways Involved in Ethanol Fermentation in Nanguo Pears during Ripening.

Nanguo pears produce a distinctive "alcoholic taste" during ripening, while Jingbai pears do not. To better understand the regulatory mechanism of "alcohol taste" formation in Nanguo pears, differences in ethanol fermentation-related metabolisms between the two cultivars were investigated. Compared to Jingbai pears, Nanguo pears had higher activities of pyruvate decarboxylase (PDC) and alcohol dehydrogenase (ADH) and contents of pyruvate, acetaldehyde, and ethanol during ripening. Nanguo pears generally showed higher starch degradation and higher levels of acetyl-CoA and citric acid but lower …

Toxic aldehydes in cooking vegetable oils: Generation, toxicity and disposal methods.

The generation of toxic aldehydes in vegetable oils subjected to high-temperature cooking processes, such as frying, poses significant health risks due to their high reactivity and potential to form carcinogenic and mutagenic compounds. This review discusses the mechanisms of aldehydes formation in vegetable oils, focusing on key factors such as oil composition, cooking temperature, and heating time. The major toxic aldehydes identified include acrolein, acetaldehyde, formaldehyde, t,t-2,4-decadienal (t,t-2,4-DDE), 4-hydroxy-2-hexenal (4-HHE), and 4-hydroxynonenal (4-HNE), which have been asso…

A Case of Cyanamide-Ethanol Reaction Leading to Atrial Fibrillation and Heart Failure.

The cyanamide-ethanol reaction (CER) is a rare but potentially life-threatening adverse event caused by the inhibition of aldehyde dehydrogenase (ADLH), leading to the accumulation of acetaldehyde after alcohol intake. This reaction is characterized by facial flushing, hypotension, tachycardia, nausea, and vomiting. We report the case of a 65-year-old male who developed new-onset atrial fibrillation (AF) and congestive heart failure (CHF) following moderate alcohol consumption while on cyanamide therapy for alcohol dependence. About twelve hours after taking cyanamide, he consumed alcohol equi…

Revisiting the Disulfiram-Like Reaction Between Alcohol and Oral Metronidazole.

Oral metronidazole is a widely used drug in the 5-nitroimidazole class effective against various anaerobic bacteria and protozoa. It has long been thought to cause a disulfiram-like reaction when combined with alcohol, believed to result from inhibition of aldehyde dehydrogenase, leading to acetaldehyde accumulation and symptoms such as nausea, flushing, vomiting, and tachycardia. To assess the validity of this association, we reviewed the English-language literature in PubMed, EMBASE, and the Cochrane Central Register of Controlled Trials from January 1, 1970, to November 22, 2024, using the …
