Hypoxia Induces Alterations in the Circadian Rhythm in Patients with Chronic Respiratory Diseases.

The function of the circadian cycle is to determine the natural 24 h biological rhythm, which includes physiological, metabolic, and hormonal changes that occur daily in the body. This cycle is controlled by an internal biological clock that is present in the body's tissues and helps regulate various processes such as sleeping, eating, and others. Interestingly, animal models have provided enough evidence to assume that the alteration in the circadian system leads to the appearance of numerous diseases. Alterations in breathing patterns in lung diseases can modify oxygenation and the circadian

Pituitary stalk interruption syndrome: a rare and severe cause of pituitary deficiency Laboratory diagnosis of a newborn case.

We report the case of a newborn with neonatal hypotonia associated to a micropenis and a bilateral cryptorchidia. The discovery of severe hypoglycemia at 0.22 mmol/L led to further biological investigations that revealed sharply decreased levels of several hypophyseal hormones. Altered corticotropic, somatotropic, thyreotropic, and gonadotropic axes finally suggested congenital hypophyseal insufficiency. This diagnostic was confirmed by a brain MRI (magnectic resonance imaging), which revealed a total interruption of the pituitary stalk. Immediate substitutive hormonal treatment allowed a clin

[Rhythms, depressions and light].

Many aspects of life in plants, animals and humans are controlled by light. Endogenous, so-called circadian rhythms in the body deviate from the exact 24-hour day and have typically a period of around 25.5 hours in man. Normally these rhythms adapt to the external 24-hour day-and night changes but under constant conditions the rhythms can free run. Many studies show how important the interplay between light and the circadian rhythms are for man as well as for other organisms. The control of these rhythms by light is mediated via the retina and the melatonin system in man. The adaptation of the

Adaptive Salvage Stereotactic Radiation for Locoregional Recurrence of Pancreatic Adenocarcinoma After Surgery.

PURPOSE: Isolated locoregional recurrence of pancreatic adenocarcinoma occurs in 25% to 50% of cases after surgical resection and has limited treatment options. Data on ablative stereotactic body radiation therapy (SBRT) with daily online adaptation for this condition are limited. Our objective was to illustrate the efficacy and safety of adaptive radiation for isolated locoregional recurrences of pancreatic adenocarcinoma. METHODS AND MATERIALS: This was a retrospective cohort study of 46 patients treated with adaptive 5-fraction SBRT for isolated locoregional recurrence of pancreatic adenoc

Towards Post-Genomic Oncology: Embracing Cancer Complexity via Artificial Intelligence, Multi-Targeted Therapeutics, Drug Repurposing, and Innovative Study Designs.

Recent advances in precision oncology have led to significant breakthroughs through the targeting of defined oncogenic drivers. However, the clinical efficacy of single-target therapies is increasingly constrained by the intrinsic complexity and adaptability of cancer. Solid tumors frequently arise from multifactorial oncogenic processes and adapt via diverse resistance mechanisms, ultimately limiting the durability of monotherapies. This review advocates for a paradigm shift toward multi-targeted, AI-enhanced strategies that harness high-throughput multi-omic data to inform the rational desig

Emerging Roles for Dendritic Cells in Heart Failure.

The field of cardio-immunology has emerged from discoveries that define roles for innate and adaptive immune responses associated with myocardial inflammation and heart failure. Dendritic cells (DCs) comprise an important cellular compartment that contributes to systemic immune surveillance at the junction of innate and adaptive immunity. Once described as a singular immune subset, we now appreciate that DCs consist of a heterogeneous pool of subpopulations, each with distinct effector functions that can uniquely regulate the acute and chronic inflammatory response. Nevertheless, the cardiovas

Does perception of biological motion rely on specific brain regions?

Perception of biological motions plays a major adaptive role in identifying, interpreting, and predicting the actions of others. It may therefore be hypothesized that the perception of biological motions is subserved by a specific neural network. Here we used fMRI to verify this hypothesis. In a group of 10 healthy volunteers, we explored the hemodynamic responses to seven types of visual motion displays: drifting random dots, random dot cube, random dot cube with masking elements, upright point-light walker, inverted point-light walker, upright point-light walker display with masking elements

SERENA-6 and the regulatory limits of radiologic progression in modern oncology.

BACKGROUND: The emergence of real-time molecular monitoring has challenged traditional oncology paradigms, where radiologic progression has historically represented the primary trigger for therapeutic intervention. The SERENA-6 clinical trial introduced a novel treatment approach by adapting therapy based on ctDNA-detected molecular progression before radiologic progression in patients with HR-positive, HER2-negative advanced breast cancer with emergent ESR1 mutations. The trial has prompted important discussion regarding the integration of molecular biomarkers into treatment decision-making a

PET-guided therapy: synergies, strategies, and future directions in external beam radiation therapy and radiopharmaceutical therapy.

Positron Emission Tomography (PET) has evolved from a purely diagnostic modality into a cornerstone of precision radiation oncology. PET now informs patient selection, target delineation, treatment personalization, and post-therapy evaluation across External Beam Radiation Therapy (EBRT) and Radiopharmaceutical Therapy (RPT). Radiotracers provide quantitative data on tumor biology, heterogeneity, receptor expression, and therapeutic response, enabling a shift from morphology-based to biology-driven oncology. PET-guided therapy is increasingly used to select patients for molecular radiotherapy,

How can innovative trials in juvenile idiopathic arthritis serve the unmet needs of patients?

Juvenile idiopathic arthritis (JIA) comprises a heterogeneous group of childhood-onset inflammatory arthritides that remain challenging to treat. An expanding range of biologic and targeted synthetic disease-modifying antirheumatic drugs has improved outcomes, yet major unmet needs persist including predicting treatment response and identifying optimal drug sequences. Traditional frequentist randomised controlled trials (RCTs) are often infeasible in JIA because of small sample sizes and ethical concerns about placebo use in children. Modified RCTs - including randomised withdrawal, placebo-ph

Integrated Multi-Omics Network by Physical Exercise Across Aging: Molecular Adaptation Beyond Single Signaling Pathways.

Aging is characterized by progressive molecular dysregulation across interconnected biological systems, involving alterations in epigenetic regulation, gene expression, protein homeostasis, and metabolic function. Although physical exercise is widely recognized as one of the most effective non-pharmacological interventions to promote healthy aging, the integrative molecular mechanisms underlying its systemic effects remain incompletely understood. This review proposes a multi-omic framework in which exercise acts as a coordinated biological stimulus capable of remodeling age-associated molecul

Chronology of H3N2 human influenza virus surface glycoprotein adaptation from 1968 to 2019 reveals a surge of adaptation between 1997 and 2002.

Subtype H3N2 influenza A viruses (IAVs), which emerged in 1968 to cause a pandemic, have shown continual circulation and adaptation that has necessitated frequent updates of candidate vaccine viruses. Here, we sought to determine how genetic changes in the hemagglutinin (HA) and neuraminidase of 21 antigenically distinct H3N2 IAVs isolated from 1968 to 2019 correlate with mammalian fitness and adaptation. We found a surge of adaptation between 1997 and 2002, resulting in the emergence of A/Fujian/411/2002 (H3N2) and poor vaccine efficacy, leading to an epidemic during the 2003-2004 season. Thi

Soft overcomes the hard: Flexible materials adapt to cell adhesion to promote cell mechanotransduction.

Cell behaviors and functions show distinct contrast in different mechanical microenvironment. Numerous materials with varied rigidity have been developed to mimic the interactions between cells and their surroundings. However, the conventional static materials cannot fully capture the dynamic alterations at the bio-interface, especially for the molecular motion and the local mechanical changes in nanoscale. As an alternative, flexible materials have great potential to sense and adapt to mechanical changes in such complex microenvironment. The flexible materials could promote the cellular mecha

Protein Intrinsic Disorder and Adaptation to Extreme Environments: Resilience of Chaos.

Extremophiles are organisms adapted not only to survive in harsh conditions but thrive in environments considered extreme or uninhabitable by most life forms. They serve as a living reflection of a popular motto "What doesn't kill you makes you stronger" and show that life on Earth is spread well outside the human-centric comfort zone. In fact, extremophiles, being found in deep crustal and oceanic depths, outer space, highly acidic and basic conditions, extreme temperatures (+122 °C to -20 °C), and in the presence of toxins and high radiation, demonstrate life's remarkable adaptability. Cutaneous Manifestations in Biological-Treated Inflammatory Bowel Disease Patients: A Narrative Review.

The biologic era has greatly improved the treatment of Crohn's disease and ulcerative colitis. Biologics can however induce a wide variety of skin eruptions, especially those targeting the TNF-α and Th17 pathway. These include infusion reactions, eczema, psoriasis, lupus, alopecia areata, vitiligo, lichenoid reactions, granulomatous disorders, vasculitis, skin cancer, and cutaneous infections. It is important to recognize these conditions as treatment-induced adverse reactions and adapt the treatment strategy accordingly. Some conditions can be treated topically while others require cessation

[Burnout : ten years later.].

The important literature on burn-out or professionnal exhaustion is examined here from three aspects: socio-cultural anthropology, biological anthropology and ethnopsychiatry. On the socio-cultural level, the emergence of burn-out is seen as an epiphenomenon arising conjointly from a new work ethic and a new concept of the person; on the biological level, the burnout is presented in relation to two main areas of study: as an adaptative strategy used by individuals to face particularly demanding situations in their working environment and as a particular modality of the interrelations between p

Chemically Fueled Interfacial Supramolecular Polymerization.

Systems capable of dissipative self-assembly emulate the dynamic, adaptive behavior of biological systems, yet spatial control over this behavior remains a challenge. Here, we show how to generate chemically fueled supramolecular polymers at liquid-liquid interfaces by coupling a chemical reaction network with an orthogonal self-assembly based on host-guest interactions and metal-ligand coordination. Confinement to the interface yields readily tunable lifetimes for the polymers as well as an ability to undergo controlled depolymerization in response to stimuli, including redox cues or competit

Colorimetric Determination of Tungsten and Molybdenum in Biological Samples.

Molybdenum (Mo) and tungsten (W) are elements that are utilized in biological systems. They are typically incorporated into the catalytic sites of enzymes coordinated to an organic pyranopterin cofactor; Mo may also be present in the form of a FeMo cofactor. While Mo is used by all branches of life, only a few microbes are able to utilize W. In order to study Mo- and W-dependent enzymes, it is important to be able to measure Mo and W in biological samples. Methods for determining Mo and W content in biological samples currently involve expensive and time-consuming processes like inductively co

Apalutamide in prostate cancer: clinical evidence, disease modification hypothesis, and emerging treatment de‑intensification strategies.

INTRODUCTION: Apalutamide is a potent androgen receptor pathway inhibitor with established efficacy in non-metastatic castration-resistant and metastatic hormone-sensitive prostate cancer. Emerging evidence has also raised questions regarding the biological impact of early treatment, depth of response, and the potential for response-adapted treatment strategies. AREAS COVERED: This narrative review summarizes randomized clinical trials and real-world evidence evaluating apalutamide across the prostate cancer continuum, with particular emphasis on efficacy, safety, PSA kinetics, comparative ef

Bioinspired Sensory Transduction for Magnetic Profile Recognition and Encryption.

In biological systems, adaptive responses to environmental stimuli are facilitated by sensory transduction, where receptors transform stimuli into dynamic intermediate electrical signals for further processing. For bioinspired artificial systems, this suggests the need to develop concepts that transduce various stimuli into electrical intermediates for recognition. Inspired by biological magnetoreception in elasmobranchs, which sense magnetic environmental profiles for navigation, we introduce an artificial sensory transduction system for magnetic profile recognition of objects using electroma
