Synthetic camphor derivative (E)-2-((1,7,7-trimethylbicyclo[2.2.1]heptan-2-ylidene)amino)phenol: A novel anthelmintic drug candidate for visceral toxocariasis.

Human toxocariasis is a neglected parasitic disease with a global distribution, treated with current anthelmintics that have low to moderate efficacy, and requires the discovery of novel drugs. Camphor derivatives have antimicrobial properties against various pathogens such as fungi and bacteria. This study aimed to identify a camphor derivative with activity against Toxocara canis larvae and evaluate its cytotoxicity, in silico bioavailability, and in vivo activity in Swiss mice infected with this parasite. Three compounds were tested in vitro in duplicate at a concentration of 1.0 to 0.05 mg…

Impact of Anthelmintic Therapy for Invasive Helminth Infection on Microbial Translocation, Inflammation, and Immune Response among Ugandans Living with HIV: A Randomized Proof of Concept Study.

Microbial translocation drives chronic immune activation that is linked to HIV disease progression. Invasive parasitic gut nematodes induce microbial translocation and downregulate the expression of pro-inflammatory cytokines. We evaluated the impact of albendazole anthelmintic therapy on serum markers of microbial translocation and inflammation among helminth-infected Ugandans living with HIV. Participants were randomized to an immediate or delayed 400 mg of daily albendazole and followed for 1 month. Baseline stool analysis, conducted via multiparallel real-time quantitative polymerase chain

Patterns of individual compliance with anthelmintic treatment for soil-transmitted helminth infections in southern Ethiopia over six rounds of community-wide mass drug administration.

BACKGROUND: The mainstay of soil-transmitted helminth (STH) control is repeated mass drug administration (MDA) of anthelmintics to endemic populations. Individual longitudinal compliance treatment patterns are important for identifying pockets of infected individuals who remain untreated and serve as infection reservoirs. METHODS: The Geshiyaro Project censused the study population in Wolaita, Ethiopia at baseline in 2018. Individual longitudinal compliance was recorded for six rounds of community-wide MDA (cMDA). The probability distribution of treatment frequency was analysed by age and gen…

Piloting a surveillance system to monitor the global patterns of drug efficacy and the emergence of anthelmintic resistance in soil-transmitted helminth control programs: a Starworms study protocol.

To eliminate soil-transmitted helminth (STH) infections as a public health problem, the administration of benzimidazole (BZ) drugs to children has recently intensified. But, as drug pressure increases, the development of anthelmintic drug resistance (AR) becomes a major concern. Currently, there is no global surveillance system to monitor drug efficacy and the emergence of AR. Consequently, it is unclear what the current efficacy of the used drugs is and whether AR is already present. The aim of this study is to pilot a global surveillance system to assess anthelmintic drug efficacy and the em

The anthelmintic effects of a new compound "E" (Friedheim) on Onchocerca gutturosa in the cow--a possible tertiary screening system for drug action against O. volvulus in man.

Three cows were treated with different levels of compound "E" which is known to have strong filaricidal activity. In all three animals the number of microfilariae of Onchocerca gutturosa declined after treatment. At autopsy most of the adult worms found were dead and in those still living embryogenesis appeared to have ceased. It is concluded that, despite the disadvantages, this system represents a possible way of assessing onchocercicidal activity before undertaking human trials.

[Approaches to the combined therapy of helminthiases with leukinferon and etiotropic preparations].

The results of therapy with anthelmintics in combination with leukinferon show either higher efficiency of anthelmintics (medamin) or their same efficiency (albendazole). The outcomes of combined treatment of infected animals with anthelmintics and leukinferon are different and associated with the time of leukinferon administration as to the tissue stages of parasitic development, with the host's sensitivity and susceptibility to helminth.

Old world camels in Germany: parasitic nematode communities characterized by nemabiome analysis showed reduced anthelmintic efficacy according to the fecal egg count reduction test.

BACKGROUND: Gastrointestinal nematodes pose a significant health risk to grazing livestock and cause economic losses, which are further increased by anthelmintic resistance. This study examined the gastrointestinal parasite fauna of Old World Camels (OWCs) in Germany and evaluated the efficacy of anthelmintic treatment. METHODS: In total, nine German OWC-keeping farms that dewormed their stock in spring 2023 were examined. The farms with their veterinarians independently selected the drug for treatment. The number of strongyle eggs per gram (EPG) feces was determined in 107 OWCs, Camelus bact

Anthelmintic niclosamide suppresses transcription of BCR-ABL fusion oncogene via disabling Sp1 and induces apoptosis in imatinib-resistant CML cells harboring T315I mutant.

Tyrosine kinase BCR-ABL fusion protein is the driver in patients with chronic myeloid leukemia (CML). The gate-keeper mutation T315I is the most challenging mutant due to its resistance to most tyrosine kinase inhibitors (TKIs). The third generation TKI ponatinib is the only effective TKI to treat CML patients harboring T315I-BCR-ABL mutation, but with high rate of major arterial thrombotic events. Alternative strategies to specifically target T315I-BCR-ABL are needed for the treatment of CML patients harboring such a mutation. Given that Sp1 is a fundamental transcriptional factor to positive

Usnic acid potassium salt from Cladonia substellata (Lichen): Synthesis, cytotoxicity and in vitro anthelmintic activity and ultrastructural analysis against adult worms of Schistosoma mansoni.

We report for the first time the in vitro effect of Potassium Salt, derived from Usnic Acid (PS-UA), isolated from the lichen Cladonia substellata Vanio, on couples of Schistosoma mansoni. As schistosomicide parameters, we evaluated mortality, motility, cell viability of the worms and tegument changes by scanning electron microscopy (SEM). Exposure to a concentration of 100 μM caused 75% mortality after 3 h. After 6 h, changes in motility in concentrations of 50 and 25 μM are evidenced. After 12 h and 24h, the concentrations of 50 and 100 μM caused 6.25% and 87.5% and 50% and 100% mortality, r

Phytochemical analysis and in vitro anthelmintic activity of Lophira lanceolata (Ochnaceae) on the bovine parasite Onchocerca ochengi and on drug resistant strains of the free-living nematode Caenorhabditis elegans.

BACKGROUND: Onchocerciasis is one of the tropical neglected diseases (NTDs) caused by the nematode Onchocerca volvulus. Control strategies currently in use rely on mass administration of ivermectin, which has marked activity against microfilariae. Furthermore, the development of resistance to ivermectin was observed. Since vaccine and safe macrofilaricidal treatment against onchocerciasis are still lacking, there is an urgent need to discover novel drugs. This study was undertaken to investigate the anthelmintic activity of Lophira lanceolata on the cattle parasite Onchocerca ochengi and the a
