Medical management of obesity.

Obesity has become a major health problem in many countries because of its high prevalence and causal relationship with serious medical complications. Many of the medical complications associated with obesity improve with intentional weight in a dose-dependent fashion, and even a modest weight loss of 50% of initial weight has beneficial effects. This article reviews the nonsurgical approaches for achieving weight loss in obese persons.

Bidirectional instrumental conditioning.

Three experiments examined bidirectional instrumental conditioning by training hungry rats to push a pole in one direction for food pellets and in the other for either a sugar or a starch solution. In the first study we examined whether the animals learned about the action-reinforcer relations using a specific satiety procedure. Prefeeding one type of reinforcer before an extinction test selectively depressed the performance of the action that had been paired with this reinforcer during training. The second experiment investigated the sensitivity of the bidirectional actions to variations in t

Pyrithiamine-induced thiamine deficiency impairs object recognition in rats.

Pyrithiamine-induced thiamine deficiency (PTD) in rats is used to model the etiology, diencephalic neuropathology, and memory deficits of Korsakoff's amnesia. We assessed the performance of rats exposed to PTD on a test of object recognition--nonrecurring-items delayed nonmatching-to-sample (DNMS). PTD produced thalamic lesions similar to those of Korsakoff's amnesics and similar to those previously observed in PTD rats. PTD rats required more trials to master DNMS at a 4-s retention delay than did controls, and after they had done so, they performed more poorly than controls at delays of 15,

Midazolam administered to rats induces anterograde amnesia for changes in reward magnitude.

L. P. Crespi (1942) showed that rats trained to run an alley for a large food reward slowed down when shifted to a small reward. This effect is usually interpreted as an aversive emotional response to reward reduction (A. Amsel, 1958). Benzodiazepines attenuate the behavioral effects of reward reduction (cf. C. F. Flaherty, 1990), but the emphasis has been on their anxiolytic, not memory-impairing, effects. Researchers trained rats (175-200 g) to run an alley for food until asymptote was reached. Reward magnitude was then either decreased (Experiment 1) or increased (Experiment 2). The benzodi

Depletion of cortical norepinephrine in rats by 6-hydroxydopamine does not impair performance of a delayed-nonmatching-to-sample task.

Rats were trained on a spatial delayed-nonmatching-to-sample (DNMTS) task, matched for performance, and randomly assigned to treatment with dorsal noradrenergic bundle injections of either 6-hydroxydopamine, to deplete cortical norepinephrine (NE), or vehicle, to control for the effects of surgery. After recovery, there were no significant differences between the groups when retrained on the DNMTS task at retention intervals (RI) from 0.1 to 15.0 s. Furthermore, no differences were observed when rats were trained at a 6.0-s RI filled with distracting stimuli or when dummy information runs were

Effects of diazepam and two beta-carbolines on epileptic activity and on EEG and behavior in rats with absence seizures.

In the present series of experiments, effects of a full benzodiazepine receptor agonist (diazepam) are described and compared with those of a partial benzodiazepine receptor agonist (ZK 91296) and an inverse partial benzodiazepine receptor agonist (FG 7142), both compounds of the beta-carboline family. In a rat model for generalized absence epilepsy, the anticonvulsant, the hypnotic and the myorelaxant properties were investigated, as well as effects on on-going behavior and effects on the electroencephalogram (EEG). While diazepam showed all behavioral and electrophysiological changes charact

The appetite suppressant, d-fenfluramine, decreases self-stimulation at a feeding site in the lateral hypothalamus.

In prior studies rats showed a relative shift from self-stimulation to escape (i.e., from reward to aversion) following a large meal, obesity or anorectic doses of insulin. Racemic fenfluramine, on the other hand, decreased both self-stimulation and escape suggesting it had a general behavior suppressant property. To avoid the depressive, antidopaminergic effects of the l-isomer, this study tested the d-isomer which is primarily serotonergic. Rats were screened for stimulation-induced feeding and then trained to self-stimulate with one lever in 5-min periods that alternated with 5-min periods

Effects of anorectic drugs on the topography of feeding behavior in baboons.

Food intake of six adult male baboons (Papio c. anubis) was monitored during daily experimental sessions lasting 22 hr. Food was available under a chain schedule with two-components. After completion of the first "procurement component" response requirement, access to food, i.e., a meal, became available under the second "consumption component" during which each response produced a 1-g food pellet. After 10 min in which no response occurred, the consumption component was terminated. When given p.o. 45 to 60 min before the start of the daily session, anorectic drugs, with the exception of (+/-)

Evidence for opiate receptor involvement in the consumption of a high palatability diet in nondeprived rats.

Nondeprived adult rats were familiarized with a highly palatable diet (powdered small animal diet mixed with sweetened condensed milk and water). The palatability of food was such that it induced vigorous feeding responses, 15-20 g food consumed within the first 30 min of access. In partially-satiated male rats, the kappa receptor agonists EKC and U-50,488 (subcutaneously administered) produced large increases in food consumption in the first 30 min of access, post-injection. In experiments with naloxone and WIN 44,441-3, we found that the effects of naloxone (0.01-10 mg/kg; S.C.) were crucial

[Effect of 3-hydroxymethyl-N-methylpiperidine (4-chlorophenoxyacetate) hydrochloride on feeding behavior. Toxicity and neuropsychopharmacology].

The 3-hydroxymethyl N-methyl piperidine (4-chlorophenoxy) acetate, hydrochloride, A, has been tested for its effect on feeding behavior. This first non-amphetamine substance, with low toxicity and without psychotropic activity, affects the satiety center by reducing gold thioglucose-induced obesity in mice.

The selectivity of the anorectic effect of satietin. II. Ineffectiveness of satietin on the water intake in food deprived rats. Effect of satietin in "conditioned aversion" paradigm.

The effect of satietin, a potent endogenius anorexigenic agent, on the water intake of fed and food deprived rats was studied and compared with the effects of calcitonin. In rats deprived of food for 96 h and supplied with water ad libitum, no change in the water intake of intracerebroventricularly (icvtr) administered satietin (2 units/rat), calcitonin (1 unit/rat) and saline (0.2 ml/rat), respectively, was observed. In animals deprived of food and water for 23 h, satietin (2 units/rat, icvtr) left water consumption unchanged, whereas calcitonin (1 unit/rat, icvtr) inhibited the intake of wat

Weight reduction in schizophrenics by molindone.

The weight-reducing property of molindone, a recently introduced antipsychotic drug, was tested in 9 hospitalized chronic schizophrenic patients. There was an average weight loss of 7.6 kg after 3 months on molindone; most of the loss occurred during the first month. The mechanism producing this weight loss is uncertain, but a central anorexigenic effect may be an important factor.

Disruption of gut bacteria-derived hydrogen sulfide signaling mediates antibacterial-induced acute anorexia.

Numerous commonly used antibacterial agents frequently induce acute anorexia as a clinical side effect, yet the underlying mechanism remains poorly understood. Here, we investigated the role of gut bacteria-derived hydrogen sulfide (H2S) signaling in the regulation of feeding behavior and its involvement in antibiotic-associated anorexia. Using methylene blue colorimetric and fluorescent probe-based detection, we found that 24-h fasting elevated H2S levels in rat feces and serum, which robustly stimulated feeding behavior. 16S rRNA sequencing revealed that fasting reshaped the gut microbiota a

The role of enteroendocrine cells, hormones, and gut microbiota in anorexia nervosa: a complex interplay of the microbiome-gut-brain axis.

Eating disorders (EDs) have a serious public health concern, especially anorexia nervosa (AN), which includes a variety of mental disorders that are frequently defined by appetite dysregulation. The purpose of this review was to clarify the role of enteroendocrine cells (EECs) and hormones secreted from EECs in the fundamental processes of AN, as well as the participation of intestinal microbiota in creating satiety sensations in the brain through the microbiome-gut-brain axis. In the advancement mechanism of AN, the endocrine system has a critical function, especially in relation to EECs and

Low Doses of Ionizing Radiation as a Treatment for Alzheimer's Disease: A Pilot Study.

BACKGROUND: In 2015, a patient in hospice with Alzheimer's disease (AD) was treated with ionizing radiation to her brain using repeated CT scans. Improvement in cognition, speech, movement, and appetite was observed. These improvements were so momentous that she was discharged from the hospice to a long-term care home. Based on this case, we conducted a pilot clinical trial to examine the effect of low-dose ionizing radiation (LDIR) in severe AD. OBJECTIVE: To determine whether the previously reported benefits of LDIR in a single case with AD could be observed again in other cases with AD whe

Assessing Activity-based Anorexia in Mice.

Rodents develop activity-based anorexia (ABA) when exposed to a restricted feeding schedule and allowed free access to a running wheel. These conditions lead to a life-threatening reduction in body weight. However, rodents exposed to only one of these conditions ultimately adapt to re-establish normal body weight. Although increased running coupled with reduction in voluntary food intake appear paradoxical under ABA conditions, ABA behavior is observed across numerous mammalian species. The ABA paradigm provides an animal model for anorexia nervosa (AN), an eating disorder with severe dysregul
