Local production of Aspergillus fumigatus specific immunoglobulin E in nasal polyps.

BACKGROUND: Aspergillus spp. play a significant role in the etiology of immunoglobulin (Ig)E mediated allergic fungal sinusitis (AFS). It is unclear whether Aspergillus spp. are also involved in nasal polyps without the characteristic clinical features of AFS. OBJECTIVES: The frequency of Aspergillus spp. and Aspergillus-specific IgE in nasal lavages and serum of patients with severe nasal polyps (n = 33) without clinical features of AFS should be investigated. STUDY DESIGN: Prospective study. METHODS: An aliquot of nasal lavage fluid was treated with dithiothreitol and examined for Aspergi…

Toxicological studies on Laccase from Myceliophthora thermophila expressed in Aspergillus oryzae.

The bioindustrially produced enzyme laccase can be used in different technical and food applications to facilitate processes. It can be added to different oral care products such as mouthwash, toothpaste, mints, and gums to prevent halitosis. Laccase, produced by submerged fermentation of Aspergillus oryzae, containing a gene originating from Myceliophthora thermophila, was subject to a series of toxicological tests to document its safety in use. It was not found to be mutagenic in the Salmonella typhimurium reverse mutation assay, nor did it cause chromosomal aberrations in cultured human lym…

Aspergillus meningitis: diagnosis by non-culture-based microbiological methods and management.

The performance of antibody detection, antigen detection, and Aspergillus genus-specific PCR for diagnosing Aspergillus meningitis was investigated with 26 cerebrospinal fluid (CSF) samples obtained from a single patient with proven infection caused by Aspergillus fumigatus. Immunoglobulin G antibodies directed against Aspergillus were not detected by enzyme-linked immunosorbent assay in CSF or serum. The antigen galactomannan was detected in the CSF 45 days before a culture became positive, and Aspergillus DNA was detected 4 days prior to culture. Decline of the galactomannan antigen titer in…

Hospital-acquired candida and aspergillus pneumonia--diagnostic approaches and clinical findings.

Bronchoscopy with bronchoalveolar lavage (BAL), collection of bronchial secretions (BS) and/or high resolution computed tomography (CT) of the lungs was performed in 70 patients with candida and/or aspergillus pneumonia. The sensitivity of bronchoscopy in detecting histologically proven fungal disease was 59%. Characteristic CT signs were found in 11 of 14 patients with candida pneumonia and 16 of 19 patients with aspergillosis. The more frequent use of bronchoscopy and CT scans between 1990 and 1992 compared with 1986-1989 for the differential diagnosis of new pulmonary infiltrates in immunoc…

Exogenous lipoid pneumonia complicated by Mycobacterium fortuitum and Aspergillus fumigatus infections.

We report the case of a nonimmunocompromised female patient, who developed exogenous lipoid pneumonia with Mycobacterium fortuitum infection at diagnosis, later followed by Aspergillus fumigatus infection. The association of exogenous lipoid pneumonia with atypical mycobacterial infection is uncommon but well-recognized, but, to our knowledge, association with A. fumigatus infection has not previously been reported.

Pneumonia caused by Nocardia nova and Aspergillus fumigatus after cardiac transplantation.

Nocardia nova, a newly established species of the Nocardia asteroides complex, has recently been characterized as a human pathogen. This report of a case of pneumonia caused by Nocardia nova and Aspergillus fumigatus in a patient after cardiac transplantation is the first reported infection caused by Nocardia nova following its detailed description. Accurate identification and susceptibility testing of the Nocardia nova isolate allowed successful oral therapy with clarithromycin when therapy with sulfisoxazole was not tolerated.

Mycotic (Aspergillus) arteritis resulting in fatal subarachnoid hemorrhage: a case report.

A seventy-one year-old Japanese man suffering from carcinoma of the common bile duct died from subarachnoid hemorrhage secondary in intracranial mycotic arteritis (MA). Repeated cultures of the discharge from the draining tubes, the tip of intravenous hyperalimentation catheters, blood, sputum, and urine failed to grow any fungus. Autopsy disclosed MA due to Aspergillus at the terminal portion of the right internal carotid artery close to the posterior communicating artery.

Aspergillus pneumonia in chronic granulomatous disease: recurrence and long-term outcome.

Four of 13 patients followed with chronic granulomatous disease (CGD) developed biopsy-proven Aspergillus pneumonia. Two patients died of disseminated aspergillosis despite intensive treatment; delays in diagnosis appear to have been a major contributing cause. Two patients survived two episodes each of Aspergillus pneumonia, three and six years apart. At 12 and 18 years of age, both are well and active, although their chest X-rays show residual scarring and pulmonary function tests indicate obstructive lung disease. These cases demonstrate that in patients with CGD, 1) multiple episodes of As…

Biochemical Properties of Tyrosinase from Aspergillus terreus and Penicillium copticola; Undecanoic Acid from Aspergillus flavus, an Endophyte of Moringa oleifera, Is a Novel Potent Tyrosinase Inhibitor.

Tyrosinase is a copper-containing monooxygenase catalyzing the O-hydroxylation of tyrosine to 3,4-dihydroxyphenylalanine then to dopaquinone … hibitor, and evaluating the mechanistics of enzyme inhibition are the main objectives of this work. Aspergillus terreus and Penicillium copticola were reported as the most potential tyrosinase producers. The biochemical properties suggest that this enzyme displays a higher structural and catalytic proximity to human tyrosinase. Upon nutritional bioprocessing …

Efficient cyanide degradation by Aspergillus welwitschiae LOT1 isolated from industrial waste.

Cyanide is a potent cytotoxin and neurotoxin largely derived from anthropogenic activities and can accumulate in the environment through industrial wastewater, posing serious ecological and health risks. This study evaluated the cyanide degradation capacity of intact Aspergillus welwitschiae LOT1 biomass under varying physicochemical and nutritional conditions. Cyanide degradation refers to the biochemical breakdown of cyanide into simpler compounds, whereas detoxification describes the conversion of cyanide into less toxic metabolites. Cyanide removal by A. welwitschiae LOT1 was influenced by…

Thermal adaptation crosstalk with azole response through lncRNA in Aspergillus fumigatus.

Fungal pathogens are adapting to increased temperatures altering host-pathogen interactions, disease patterns, and response to the antimicrobial drugs. Here, we show that thermal adaptation to 42°C leads to reversible changes in fungal colony size and azole drug response in the human pathogenic fungus Aspergillus fumigatus. Importantly, this adaptation is mediated by the lncRNA, afu-182, whose RNA levels negatively correlate with temperature. Growth at a lower temperature or ectopic upregulation of afu-182 RNA levels reverses the temperature adaptation. Previously, we have shown that Δafu-182 …

Features of Invasive Aspergillosis Caused by Aspergillus flavus, France, 2012-2018.

Invasive aspergillosis (IA) caused by Aspergillus flavus remains poorly described. We retrospectively analyzed 54 cases of IA caused by A. flavus reported in France during 2012-2018. Among cases, underlying IA risk factors were malignancy, solid organ transplantation, and diabetes. Most (87%, 47/54) infections were localized, of which 33 were pleuropulmonary and 13 were ear-nose-throat (ENT) infection sites. Malignancy (70% [23/33]) and solid organ transplantation (21% [7/33]) were the main risk factors in localized pulmonary infections, and diabetes mellitus was associated with localized ENT …

Steroids, peniciversiols and aspergilosidols from endophytic fungus Aspergillus sp. TZS-Y4.

Endophytic fungi have emerged as a crucial resource for medicinal drug development. The genus Aspergillus, in particular, has yielded diverse bioactive secondary metabolites. In this study, we isolated nine steroids, five peniciversiols, and two aspergilosidols from the endophytic fungus Aspergillus sp. TZS-Y4. Among these, six steroids-camphoratins K-O (1-6) and one aspergilosidol, eupenoxide B (15), were identified as novel compounds. Their structures were elucidated through comprehensive NMR spectroscopy, HR-MS/MS analysis, experimental ECD, and calculated ECD data. Anti-inflammatory activi…

Biochemical characterization of recombinant Aspergillus niger GZUF36 acetylcholinesterase for pesticide monitoring.

An acetylcholinesterase from Aspergillus niger GZUF36 (AnAChE) was expressed in Escherichia coli BL21(DE3) and comprehensively characterized with respect to its biochemical properties and suitability for pesticide analysis. The recombinant enzyme (rAnAChE) exhibited typical AChE substrate specificity, with optimal activity at pH 7.0 and 30-35 °C. rAnAChE tolerated hydrophobic solvents and methanol, supporting its use in practical assays. The enzyme was highly sensitive to methomyl, carbofuran, trichlorfon, fenitrothion, and carbaryl. For the 12 pesticides tested, the limits of detection of mos…
