AexB is an aromatic amino acid exporter that functions as a metabolic safety valve.

UNLABELLED: Aromatic amino acids-tryptophan, tyrosine, phenylalanine, and histidine-are essential for bacterial growth and are among the most energetically expensive metabolites to synthesize. Despite this cost, it has been recently shown that bacteria possess exporters for these amino acids. Here, we identify aexB (formerly yvjA) as a gene encoding a novel aromatic amino acid exporter in Bacillus subtilis. Using a transposon-based screen, we found that aexB overexpression confers resistance to the toxic tryptophan analog 5-fluorotryptophan. Additional analog screens revealed that AexB also pr…

Antimicrobial Activities of Ganoderma mbrekobenum Strain EGDA (Agaricomycetes) from Egypt.

Ganoderma is a well-known genus of medicinal mushrooms. The biological activity of the fruiting bodies of G. mbrekobenum (previously identified as Ganoderma sp. EGDA, (AC: LN774971) is scarcely studied. The microorganisms including bacteria and fungi were chosen for screening of the antimicrobial activity produced by G. mbrekobenum strain EGDA. The bioactive compounds were extracted from aqueous, petroleum ether, chloroform, ethyl acetate, and methanol extracts. The higher antibacterial activity produced by methanol extract was against Bacillus subtilis and B. cereus (14.13 ± 0.12 mm, 13.03 ± …

Anti-proliferative activity of surfactins on human cancer cells and their potential use in therapeutics.

Surfactins are cyclic lipopeptides that are isolated from various Bacillus strains. They are made up of heptapeptides and β-hydroxy fatty acids of variable chain lengths of carbon atoms. Therapeutically they are known to inhibit invasion, migration, and colony formation of human breast carcinoma cells. The role of surfactins is also known as anti-proliferative agents against human cancer cells through induction of apoptosis, arrest of the cell cycle, or suppression of survival signaling. The cytotoxic activity of surfactins is also perceived against human chronic myelogenous leukemia cells, hu

Evaluation of different bacterial honey isolates as probiotics and their efficient roles in cholesterol reduction.

Continue to hypothesize that honey is a storehouse of beneficial bacteria, and the majority of these isolates are levansucrase producers. Accordingly, ten bacterial strains were isolated from different honey sources. Four honey isolates that had the highest levansucrase production and levan yield were identified by the partial sequencing of the 16S rRNA gene as Achromobacter sp. (10A), Bacillus paralicheniformis (2M), Bacillus subtilis (9A), and Bacillus paranthracis (13M). The cytotoxicity of the selected isolates showed negative blood hemolysis. Also, they are sensitive to the tested antibio…

Nattokinase (Bac s 1), a subtilisin family serine protease, is a novel allergen contained in the traditional Japanese fermented food natto.

BACKGROUND: Immediate allergy caused by natto, a popular Japanese food prepared by fermenting soybeans with Bacillus subtilis var. natto, has been reported. Polygamma glutamic acid (PGA) in the sticky substance around natto beans has been reported to be a causative allergen of natto allergy. However, some of our patients with natto allergy were negative for PGA in the skin prick test (SPT). The sticky substance of natto beans contains a subtilisin family serine protease, nattokinase, along with PGA. In this study, we aimed to examine the antigenicity of nattokinase in natto allergy. METHODS: …

Production and initial characterization of bionites: materials formed on a bacterial backbone.

The addition of soluble metal salts of calcium, iron, or copper to cultures of Bacillus subtilis grown in web form nucleated precipitation at the surface of the bacterial cell walls. The mineralized cell filaments can be drawn into a fiber that when dried consists of a bacterial thread backbone carrying an inorganic solid. The ratios of organic to inorganic components (by weight) in the stiff brittle materials, called bionites, were: 1.08 for fe(2)bactonite, 1.8 for calbactonite, 2.3 for fe(3)bactonite, and 5 for cu(2)bactonite. X-ray photoelectron spectra suggest that the fe(3)bactonite conta…

[Various ways of standardizing and unifying biological methods of determining the activity of antibiotics].

It was shown possible to use nutrient media containing only salt components and agar-agar for determination of biological activity of tetracyclines (tetracycline, morphocycline, oxytetracycline and chlortetracycline), erythromycin and oleandomycin. A uniform nutrient medium containing ammonium chloride, trisubstituted sodium citrate, disubstituted sodium phosphate and agar-agar was developed. Glucose is added to this medium simultaneously with the test microbe, B. subtilis ATCC 6633. The medium of the above composition provided readily reproducible results.

Weaponizing volatiles to inhibit competitor biofilms from a distance.

The soil bacterium Bacillus subtilis forms beneficial biofilms that induce plant defences and prevent the growth of pathogens. It is naturally found in the rhizosphere, where microorganisms coexist in an extremely competitive environment, and thus have evolved a diverse arsenal of defence mechanisms. In this work, we found that volatile compounds produced by B. subtilis biofilms inhibited the development of competing biofilm colonies, by reducing extracellular matrix gene expression, both within and across species. This effect was dose-dependent, with the structural defects becoming more prono…

Phosphorus-solubilizing bacteria and phosphorus-enriched biochar enhance growth, cannabinoid content, and essential oil yield in salt-stressed hemp (Cannabis sativa L.).

Soil salinity induces phosphorus (P) deficiency, limiting crop productivity, yet the combined use of phosphorus-solubilizing bacteria (PSB) and phosphorus-enriched biochar (P-biochar) under salinity remains unexplored. This study presents a novel integrated PSB+P-biochar approach to improve growth and secondary metabolite production in Cannabis sativa L. under salinity stress. A completely randomized design with three independent replicates (each replicate containing two pots) was employed. A factorial experiment was conducted with two factors: salinity at two levels (0 and 100 mM NaCl) and so

Harnessing Microbes to Weather Native Silicates in Agricultural Soils for Scalable Carbon Dioxide Removal.

Anthropogenic carbon emissions contribute significantly to the greenhouse effect, resulting in global warming and climate change. Thus, addressing this critical issue requires innovative and comprehensive solutions. Silicate weathering moderates atmospheric CO2 levels over geological time, but it occurs too slowly to counteract anthropogenic emissions effectively. Here, we show that the microorganism Bacillus subtilis strain MP1 promotes silicate weathering across different experimental setups with various levels of complexity. First, we found that MP1 was able to form a robust biofilm in the …

Bioaugmentation-induced assimilatory sulfate reduction and chain elongation mitigate H2S and CH4 emissions in sewers.

Controlling H2S and CH4 emissions is critical for ensuring the safety of sewer systems. Bioaugmentation offers a cost-effective and environmentally friendly alternative to chemical dosing, yet its performance and mechanisms in sewers remain unclear. This study investigated a bioaugmentation strategy by inoculating laboratory-scale sewer reactors with Bacillus subtilis and Saccharomyces cerevisiae. A 7-day inoculation (10⁷ CFU/mL) reduced H2S and CH4 emissions by 86.1% and 62.9%, respectively, with half-recovery times of 12 and 22 days. The strategy extended the duration of effective H2S contro…

Exposure to High Dosage of Gold Nanoparticles Accelerates Growth Rate by Modulating Ribosomal Protein Expression.

Gold nanoparticles (AuNPs) have been utilized in various biomedical applications including diagnostics and drug delivery. However, the cellular and metabolic responses of cells to these particles remain poorly characterized. In this study, we used bacteria (Escherichia coli and Bacillus subtilis) and a fungus (Saccharomyces cerevisiae) as model organisms to investigate the cellular and metabolic effects of exposure to different concentrations of citrate-capped spherical AuNPs with diameters of 5 and 10 nm. In different growth media, the synthesized AuNPs displayed stability and microorganisms …

In-Depth Characterization of the Staphylococcus aureus Phosphoproteome Reveals New Targets of Stk1.

Staphylococcus aureus is a major cause of infections worldwide, and infection results in a variety of diseases. As of no surprise, protein phosphorylation is an important game player in signaling cascades and has been shown to be involved in S. aureus virulence. Albeit long neglected, eukaryotic-type serine/threonine kinases in S. aureus have been implicated in this complex signaling cascades. Due to the substoichiometric nature of protein phosphorylation and a lack of suitable analysis tools, the knowledge of these cascades is, however, to date, still limited. Here, were apply an optimized pr
