Assessment of the effect of Maytenus ilicifolia (espinheira santa) extract on the labeling of red blood cells and plasma proteins with technetium-99m.

We are trying to develop a model to assess properties of products utilized in popular medicine. Maytenus ilicifolia is used in herbal medicine. Red blood cells (RBC) labeled with technetium-99m (99mTc) are employed in nuclear medicine. This labeling procedure depends on a reducing agent and stannous chloride is used. There is evidence that this labeling may be altered by drugs. We have investigated the possibility of M. ilicifolia extract being capable to alter the labeling of blood elements with 99mTc. Blood was incubated with M. ilicifolia extract. Stannous chloride solution and Tc-99m were

Histologic evaluation of human extraction sockets treated with demineralized freeze-dried bone allograft (DFDBA) and cell occlusive membrane.

This study evaluated new bone formation in human extraction sockets treated with demineralized freeze-dried bone allografts (DFDBA) and celloc occlusive membranes. Hard tissue biopsies of 7 sites in 6 patients were obtained 14 weeks to 13 months following extraction and grafting. Histologic analysis revealed that individual particles of DFDBA were discernible up to 13 months in situ. In all samples, all particles of DFDBA were well incorporated within new bone, which exhibited osteocyte-containing lacunae. Distinct cement lines clearly demarcated the DFDBA particles from the surrounding, intim

Crude extract and recombinant protein of Cryptosporidium parvum oocysts induce proliferation of human peripheral blood mononuclear cells in vitro.

Cryptosporidium parvum preparations were studied for their ability to induce specific proliferation of cultured human peripheral blood mononuclear cells (PBMC) from both immunocompetent and human immunodeficiency virus-positive persons, some of whom had transient cryptosporidiosis. The proliferation of PBMC from sensitized donors induced by C. parvum preparations was due mainly to antigen-specific rather than nonspecific activation, as indicated by the kinetics of the proliferative response, inhibition of the PBMC proliferation by a monoclonal antibody directed against major histocompatibility

Cell-mediated immunity against human retinal extract, S-antigen, and interphotoreceptor retinoid binding protein in onchocercal chorioretinopathy.

Autoimmune mechanisms are thought to be involved in the pathogenesis of onchocercal chorioretinopathy. Cell-mediated immune responses to human retinal S-antigen, interphotoreceptor retinoid binding protein (IRBP), and crude retinal extract were investigated in patients with onchocerciasis from Sierra Leone, West Africa using a two-step migration-inhibition factor assay. Patients were subdivided into three groups: (1) without ocular involvement (n = 10), (2) with ocular onchocerciasis limited to the anterior segment (n = 19), and (3) with onchocercal chorioretinopathy (n = 21). A group of endem

In vitro tapeworm extract-induced proliferative responses of gut-associated lymphoid cells from Hymenolepis diminuta infected mice.

The development of lymphoid cells reactive to tapeworm-associated antigens during the course of Hymenolepis diminuta rejection from mice was studied using an in vitro tapeworm extract (TWE)-induced cell proliferation culture system. Mice infected with three cysticercoids on day 0 developed three adult worms by day 7 but worms were rejected by day 21 post-infection. Concomitant with worm rejection was the development of TWE-sensitized lymphoid cells which responded by proliferation when stimulated in vitro with TWE. Sensitized cells were detected in gut-associated mesenteric lymph nodes but wer

Symptomatology and diagnosis in connective tissue disease. Antibodies to extractable ribonucleoprotein in 123 patients reacting with cell nuclei in the immunofluorescence test.

Sera with an antinuclear immunofluorescence titre of 1/000 were taken consecutively from the diagnostic routine flow and examined for agglutinating antibodies against desoxyribonucleic acid (DNA) and extractable nuclear antigens (ENA). Passive haemagglutination tests with antigen-coated tanned erythrocytes were used and the specificity of the reactions was corroborated by testing against enzyme-treated cells. After the exclusion of the DNA-reacting 15%, three major groups and one minor could be distinguished on a serological basis. The largest group (41%) contained cases with a speckled immuno

Ruscus aculeatus Extract Inhibits Amyloid Beta Production and Upregulates the Active form of ADAM10 in Human Neural Progenitor Cells.

Alzheimer's disease is characterized by the accumulation of cerebral amyloid beta (Aβ) peptides, making the inhibition of Aβ production a key therapeutic strategy. We screened a plant extract library for Aβ42 inhibitors using human neural progenitor cells and identified a Ruscus aculeatus extract as a potential candidate. Although rhizome extracts of R. aculeatus exhibit vasotonic, and anti-inflammatory properties, their effects on amyloid-β precursor protein (APP) processing have not been investigated. We aimed to elucidate how R. aculeatus extract influences Aβ production and its underlying

Beyond the root: licorice (Glycyrrhiza glabra L.) fruit extract modulates oxidative stress and apoptotic markers in PC12 cells.

The rising global prevalence of neurodegenerative disorders underscores the urgent need for novel therapeutic strategies. Oxidative stress is a well-established central driver in the pathogenesis of conditions like Alzheimer's and Parkinson's disease. While the root of Glycyrrhiza glabra L. (licorice) is a renowned source of natural antioxidants, its fruit remains a largely unexplored reservoir of bioactive compounds with potential neuroprotective properties. This in vitro study aimed to systematically evaluate the neuroprotective potential of different licorice fruit extracts and elucidate th

Protective effects of three herbal extracts on mitochondrial dysfunction in VPS13C-knockdown SH-SY5Y cells: implications for Parkinson's disease.

Parkinson's disease (PD) results primarily from the loss of midbrain dopaminergic neurons. Mitochondrial dysfunction may contribute to PD pathogenesis and depletion of vacuolar protein sorting 13 homolog C (VPS13C) leads to mitochondrial dysfunction. Traditional Chinese medicine (TCM) extracts Uncaria rhynchophylla, Gardenia jasminoides, and Scutellaria baicalensis were reported to have anti-PD effects. This study aimed to examine these three extracts for enhancing VPS13C expression and mechanisms of neuroprotection. Using FCCP-mediated oxidative phosphorylation uncoupling or RNA interference-

Mallotus japonicus leaf extract restores TRPM6 magnesium channel expression and suppresses cellular senescence in colonic epithelial cells.

Age-related alterations in intracellular Mg2+ homeostasis can influence cellular bioenergetics and stress responses, yet the epithelial mechanisms linking aging and cellular senescence to impaired magnesium handling remain incompletely defined. Frailty is an emerging public health challenge in aging societies, and age-related magnesium deficiency has been proposed as one contributing factor. We investigated whether aging and cellular senescence impair epithelial magnesium handling in the colon and whether Mallotus japonicus (M. japonicus) leaf extract ameliorates these alterations in mouse col

Astragalus membranaceus Root Extract Improves T Cell Immunity in CTX-Immunosuppressed Mice and Is Associated with Hypermethylation of the Cpt1a Locus.

Astragalus membranaceus (AM) has been reported to support immune function, but its epigenetic mechanism on immune restoration of cyclophosphamide (CTX)-induced immunosuppression remains unclear. To evaluate the recovery of immune function, we assessed the injury of splenic pathology, cytokines levels in serum, proportional changes in CD4+ and CD8+ T cells, and the recall response of Hspx-specific T cell. In addition, to define the underlying epigenetic regulatory mechanism, we measured the expression levels of DNA methyltransferases (Dnmt1, Dnmt3a and Dnmt3b) and the methylation status of the
