The relationship between variability of the heterochromatic regions of chromosomes 1, 9, 16, and Y, and anthropometric characteristics (length and mass of body, shoulder diameter) in 70 children with embryopathies of unknown etiology and in 40 children with Down syndrome was studied. The positive statistically significant correlations of the C segment lengths of chromosomes 1, 9, 16, their sum included, and the above characteristics were found. The correlation coefficients of Y chromosome were not significant. The questions of the functional role of the structural heterochromatin and its influ

The examination of the chromosomes of a seven year old child with severe mental retardation, allowed us to bring to light, beside a majority of cells with a normal karyotype, a population of about 10% of cells in which chromosome 7 showed a gap in the juxtacentromeric area of its short arm. The degree of this alteration varied from the simple angulation to the complete break with short arm's loss. The functional and structural signification of this abnormality is discussed and this case is compared with previous observations concerning the abnormalities of this area of chromosome 7 (specially

Chromosomes of 52 cases of mouse myeloid leukemia were examined. There were 5 myeloblastic leukemias, 22 granulocytic leukemias, 17 myelomonocytic leukemias, and 8 monocytic leukemias. Fifty cases were radiation induced and the other 2 were nonirradiated. Each case had leukemic cells with 1 to 10 marker chromosomes. Partially deleted No. 2 chromosomes appeared in 49 cases, including 2 nonirradiated cases. These deleted No. 2 chromosomes were varied in size, and they were classified into 7 types according to morphological features. There was no type-dependent difference in histological or cytol

An estimated one billion people are at risk of acquiring the disease leishmaniasis annually. This disease, among others, is transmitted to humans via sandfly vectors of the family Psychodidae. In the Neotropics, Lutzomyia longipalpis is one of the most common vectors of leishmaniasis, but little is known about its genetics and population structure, hampering control efforts. We generated sex-specific genome assemblies of Lu. longipalpis from Colombia, and examined population genomics of this species. We found that sex accounts for ~11% of genome-wide variation, primarily driven by differentiat

Sex chromosomes have independently evolved in various species, displaying unique evolutionary patterns, including differentiation, degeneration, and repetitive DNA accumulation. Pentatomidae hemipterans are characterized by a highly conserved diploid number of 2n = 14 with a XX/XY sex chromosome system, i.e. 2n = 14, 12 A + XY. Thus, it represents an interesting group for investigating the reorganization of repeats in conserved karyotypes, i.e. the absence of large chromosomal rearrangements. Using comparative genomic hybridization (CGH) with male and female genomic DNAs (gDNA), this study exa

While sex chromosomes carry sex-determining genes, they also often differ from autosomes in size and composition, consisting mainly of silenced heterochromatic repetitive DNA. Even though Y chromosomes show structural heteromorphism, the functional significance of such differences remains elusive. Correlative studies suggest that the amount of Y chromosome heterochromatin might be responsible for several male-specific traits, including sex-specific differences in longevity observed across a wide spectrum of species, including humans. However, experimental models to test this hypothesis have be

Trisomy is the presence of one extra copy of an entire chromosome or its part in a cell nucleus. In humans, autosomal trisomies are associated with severe developmental abnormalities leading to embryonic lethality, miscarriage or pronounced deviations of various organs and systems at birth. Trisomies are characterized by alterations in gene expression level, not exclusively on the trisomic chromosome, but throughout the genome. Here, we applied the high-throughput chromosome conformation capture technique (Hi-C) to study chromatin 3D structure in human chorion cells carrying either additional

BACKGROUND: Structural variants (SVs) are genomic rearrangements of 50 bp or more and contribute substantially to human genetic disorders. They are often incompletely resolved using cytogenetic and short-read sequencing approaches. Emerging technologies such as long-read sequencing (LRS) and optical genome mapping (OGM) provide complementary advantages for SV detection. This study aimed to evaluate the combined utility of OGM and LRS for the precise delineation of clinically relevant chromosomal rearrangements.
METHODS: Five patients with suspected chromosomal abnormalities were investigated

Congenital right atrial appendage aneurysm (RAAA) represents a rare cardiac anomaly whose exact etiology, prognostic implications, and optimal management remain undetermined. RAAA characteristic echocardiographic features include a cystic structure demonstrating continuity with the right atrial chamber, with confirmed hemodynamic communication between the two structures. While fetal RAAA is typically detected during the third trimester of pregnancy, our case represents an unusually early diagnosis during the first trimester, with comprehensive longitudinal follow-up extending to 2 years postna

NUT carcinoma (NUTc) is a rare, aggressive cancer characterized by rearrangement of the NUTM1 gene on chromosome 15q14. We present the case of a 32-year-old man with a persisting cough, reduced general condition, and B symptoms for 2 months. CT imaging revealed a highly suspicious mass in the right lung, infiltrating hilar structures, and the mediastinum. Staging with 18 F-FDG PET/CT demonstrated pathologic 18 F-FDG uptake within the tumor. Histology, immunohistochemistry, and molecular analysis confirmed NUTc of the lung. Metabolic imaging modalities may play a key role in staging, restaging,

Chromatin is organized into multiscale three-dimensional structures, including chromosome territories, A/B compartments, topologically associating domains, and chromatin loops. This hierarchically organized genomic architecture regulates gene transcription, which, in turn, is essential for various biological processes during brain development and adult plasticity. Here, we review different aspects of spatial genome organization and their functions in regulating gene expression in the nervous system, as well as their dysregulation in brain disorders. We also highlight new technologies to probe

Huntington disease (HD) is an autosomal dominant, adult-onset, progressive neurodegenerative disease characterized by the triad of abnormal movements (typically chorea), cognitive impairment, and psychiatric problems. It is caused by an expanded CAG repeat in the gene encoding the protein huntingtin on chromosome 4 and causes progressive atrophy of the striatum as well as cortical and other extrastriatal structures. Genetic testing has been available since 1993 to confirm diagnosis in affected adults and for presymptomatic testing in at-risk individuals. This review covers HD signs, symptoms,

We present three cases of fetal holoprosencephaly diagnosed by transabdominal and transvaginal ultrasound examinations at 10 and 13 weeks' gestation. The diagnosis was based on two sonographic criteria: first, the intracranial finding of a single ventricle with a cerebral mantle and no visible midline structures but fusion of the thalami and corpus striatum; and, second, facial abnormalities, including hypotelorism. The ultrasound findings were confirmed by embryoscopy before abortion in one case and by pathological examination after abortion in two cases. Chromosome study of the three fetuses

Evolution of microsatellites (or simple sequence repeats, SSRs) is a complex process that converts perfect repeats to novel structural elements with functions poorly understood, such as imperfect and compound microsatellites. An in silico analysis often Burkholderia pseudomallei genomes revealed 215683 micro-satellites, and more than 98% of them proved imperfect. The density of microsatellites in the genome ranged from 2922.7 to 3022.6 per Mbp. Approximately 10.20-10.67% of the repeats were parts of compound micro-satellites. The of compound microsatellite density varied from 144.7 to 150.6 pe

We recently cloned three membrane guanylyl cyclases, designated GC-D, GC-E, and GC-F, from rat olfactory tissue and eye. Amino acid sequence homology suggests that they may compose a new gene subfamily of guanylyl cyclase receptors specifically expressed in sensory tissues. Their chromosomal localization was determined by mouse interspecific backcross analysis. The GC-D, GC-E, and GC-F genes (Gucy2d, Gucy2e, and Gucy2f) are dispersed through the mouse genome in that they map to chromosomes 7, 11, and X, respectively. Close proximity of the mouse GC-D gene to Omp (olfactory marker protein) and

Supernumerary B chromosomes contribute to intraspecific karyotypic variation. B chromosomes have been detected in more than 2000 organisms; they possess unique and diverse features, including non-Mendelian inheritance. Here, we report one or more B chromosomes in the gynodioecious plant Atractylodes lancea. Among 54 A. lancea lines, 0-2 B chromosomes were detected in both hermaphroditic and female plants, with the B chromosomes appearing as DAPI-bright regions within the nuclei. Genomic in situ hybridization revealed that the B chromosomes had no conserved A chromosome DNA sequences, confirmed

Primary amenorrhea is one of the common reproductive disorder affecting females. It leads to the absence of menarche in the reproductive age group in females and/or complete absence of reproductive organs. The physiology of menstruation and reproduction has a strong correlation with the expression of the X chromosome. Thus, the role of the clinical geneticists in terms of diagnosis, risk assessment, genetic counseling and management of patients with primary amenorrhea and their families is essential. The genetic contribution to amenorrhea is studied both at the cellular and molecular level aim

Invasive cardiology laboratory workers are occupationally exposed to chronic ionizing radiation. It is known that ionizing radiation has a damaging effect on chromosomes. In present study. we investigated the frequency of sister chromatid exchange (SCE) and chromosomal aberrations in 11 invasive cardiology laboratory workers and 11 healthy controls. After a vacation period, we took blood samples for chromosome analysis in months 0, 4, 8 and 12 (last two month period was the nonradiation time). The SCE frequencies did not change significantly after exposure to ionizing radiation in any worker.
