A review of computed tomography patterns of metastatic breast cancer patients undergoing treatment at a private oncology centre in Ghana.

INTRODUCTION: breast cancer is the commonest malignant disease in Ghanaian women and accounts for 17% of cancer-related deaths in the country. It has been classified into molecular subtypes depending on the presence or absence of hormone receptors and the human epidermal growth factor receptor 2. Computed tomography is often the preferred modality for monitoring metastatic disease due to its ability to determine the extent of local and metastatic disease. METHODS: this was a retrospective study conducted at Sweden Ghana Medical Centre (SGMC). Hospital records and chest and abdominal computed

Envisioning how the prototypic molecular machine TFIIH functions in transcription initiation and DNA repair.

Critical for transcription initiation and bulky lesion DNA repair, TFIIH provides an exemplary system to connect molecular mechanisms to biological outcomes due to its strong genetic links to different specific human diseases. Recent advances in structural and computational biology provide a unique opportunity to re-examine biologically relevant molecular structures and develop possible mechanistic insights for the large dynamic TFIIH complex. TFIIH presents many puzzles involving how its two SF2 helicase family enzymes, XPB and XPD, function in transcription initiation and repair: how do they

Baicalin protects against heat-induced multiorgan dysfunction via organ-specific protein modulation: integrative in silico and in vivo evidence.

Heatstroke-induced multiorgan dysfunction represents a life-threatening clinical emergency characterized by systemic oxidative stress, inflammation, and metabolic collapse across vital organs. Despite advances in supportive care, there remains a critical lack of multitarget pharmacological interventions that address the underlying molecular pathology. Here, we evaluated baicalin, a naturally occurring flavone glycoside, for its multiorgan protective efficacy against systemic hyperthermia through an integrated computational-experimental framework. Five key heat-responsive proteins-heat shock pr

Beyond structural cycles: computational mechanisms of biomolecular machines.

Understanding biomolecular machines requires more than determining their structures; it requires identifying the dynamical principles that couple chemical or electrochemical free energy to function. Recent advances in cryo-EM, single-molecule spectroscopy, high-speed atomic force microscopy, and deep mutational scanning provide unprecedented experimental constraints for computational models, while developments in molecular simulation and kinetic modeling increasingly allow mechanistic interpretation of these observations. Here, we discuss recent computational advances that exemplify this trans

Drugst.One - a plug-and-play solution for online systems medicine and network-based drug repurposing.

In recent decades, the development of new drugs has become increasingly expensive and inefficient, and the molecular mechanisms of most pharmaceuticals remain poorly understood. In response, computational systems and network medicine tools have emerged to identify potential drug repurposing candidates. However, these tools often require complex installation and lack intuitive visual network mining capabilities. To tackle these challenges, we introduce Drugst. One, a platform that assists specialized computational medicine tools in becoming user-friendly, web-based utilities for drug repurposing

Integration of mechanics and biology in computer simulation of bone remodeling.

Bone remodeling is a complex physiological process that spans across multiple spatial and temporal scales and is regulated by both mechanical and hormonal cues. An imbalance between bone resorption and bone formation in the process of bone remodeling may lead to various bone pathologies. One powerful and non-invasive approach to gain new insights into mechano-adaptive bone remodeling is computer modeling and simulation. Recent findings in bone physiology and advances in computer modeling have provided a unique opportunity to study the integration of mechanics and biology in bone remodeling. Ou

Gibbs-Thomson mediated ice growth inhibition by MOF-based organic linkers through interfacial clustering.

In this work, we present the first computational investigation of MOF organic linkers as small-molecule ice recrystallisation inhibition (IRI) agents. Guided by previous computational and experimental studies on MOF-based IRI agents, we selected four promising linker candidates: benzene-1,4-dicarboxylic acid (BDC) and its hydroxyl- and carboxyl-functionalized variants, alongside the 2,2'-bipyridine-5,5'-dicarboxylic acid (commonly known as BPYDC) linker. Density functional theory calculations confirmed their structural stability, and molecular dynamics (MD) simulations at 240 K validated their

Tunable triazole-based cholera toxin inhibitors: A QSAR-guided design and evaluation approach.

Cholera toxin B subunit (CTB) is a validated target for anticholera therapeutics, but current inhibitors often suffer from synthetic complexity and limited tunability. This study aimed to develop a compact, tunable triazole scaffold exhibiting low-micromolar potency combined with high synthetic tractability. We applied a unified, QSAR-driven, multiscale computational-experimental workflow integrating molecular docking, induced fit docking (IFD), molecular dynamics (MD), QM/MM calculations, and descriptor-based QSAR modeling to prioritize a focused library of 44 N-sulfonyl triazole inhibitors.

In Silico Analyses Suggest That Exercise-Induced Irisin-Mediated Neuroprotection Supports Non-Pharmacological Preventive Strategies for Alzheimer's Disease in Public Health.

Alzheimer's disease (AD) is the leading cause of dementia and imposes a high economic and social burden on healthcare systems. In Brazil, the consistent increase in costs associated with AD hospitalizations, coupled with the absence of curative therapies and population aging, reinforces the need for low-cost, broadly applicable preventive strategies. This study investigated the role of irisin, a myokine induced by physical activity, in the prevention of AD, integrating epidemiological and bioinformatic analyses. Public data on the nutritional status of the Brazilian population in the early 200

An in silico Approach for Identification of Novel Natural Selective ALR2 Inhibitors from Cynomorium songaricum for Treating Diabetic Complications.

INTRODUCTION: Aldose reductase-2 (ALR2) is a key enzyme in the polyol pathway whose overexpression is implicated in several diabetic complications, including neuropathy, nephropathy, retinopathy, and atherosclerotic plaque formation. Under hyperglycemic conditions, the intracellular accumulation of sorbitol and the depletion of NADPH lead to osmotic imbalance and oxidative stress, driven by the formation of reactive oxygen species and advanced glycation end products. Although various ALR2 inhibitors have been developed, their clinical application has been hampered by nonselective inhibition of

Bioinformatics Approaches for Functional and Structural Annotation and Molecular Docking Study of a Hypothetical Protein From Staphylococcus aureus.

Staphylococcus aureus is a common Gram-positive bacterium that colonizes the skin and mucous membranes of humans. It is a major etiological agent of nosocomial infections and a persistent source of contamination in food and hospitals. The complete sequencing of the S. aureus genome has been accomplished, yet numerous hypothetical proteins encoded within it remain functionally uncharacterized. For this study, we performed a comprehensive functional and structural analysis of a hypothetical protein (accession number OHS92176.1) from the S. aureus HMSC77A05 strain using bioinformatics approaches.

In silico identification for flavor antioxidant compounds in Chrysanthemi flos uncovers the interactions between saccharides and secondary metabolites.

Secondary metabolites and saccharides are responsible for antioxidant activity and flavor of Chrysanthemi flos (CF). However, the flavor antioxidant compounds of CF and their intermolecular interactions remain unclear. Here, we primarily employed in silico methods to identify CF antioxidants. After characterizing by physicochemical properties, FT-NIR and HPLC fingerprint, the "spectrum-effect" fusion correlation was established to select the spectral features of CF antioxidants. Quercetagitrin (QU), chlorogenic acid (CA) and saccharides fragments were clarified based on their characteristic sp
