Effects of environmental concentrations of toxins BMAA and its isomers DAB and AEG on zebrafish larvae.

The increasing concern over the environmental presence of β-N-Methylamino-L-alanine (BMAA), a toxin primarily produced by cyanobacteria and diatoms, has stimulated numerous studies to evaluate the risk for exposed populations, mainly aquatic organisms and humans. This study focuses on the toxicity of environmental concentrations of BMAA and its isomers, l-2,4 diaminobutyric acid dihydrochloride (DAB) and N-(2-aminoethyl) glycine (AEG) on zebrafish embryo development (ng.L-1). Presence of BMAA in various environments, including aquatic sources, air, and desert crusts, has raised concerns due to…

Cyanotoxins Increase Cytotoxicity and Promote Nonalcoholic Fatty Liver Disease Progression by Enhancing Cell Steatosis.

Freshwater prokaryotic cyanobacteria within harmful algal blooms produce cyanotoxins which are considered major pollutants in the aquatic system. Direct exposure to cyanotoxins through inhalation, skin contact, or ingestion of contaminated drinking water can target the liver and may cause hepatotoxicity. In the current study, we investigated the effect of low concentrations of cyanotoxins on cytotoxicity, inflammation, modulation of unfolded protein response (UPR), steatosis, and fibrosis signaling in human hepatocytes and liver cell models. Exposure to low concentrations of microcystin-LR (MC…

Abundant iron and sulfur oxidizers in the stratified sediment of a eutrophic freshwater reservoir with annual cyanobacterial blooms.

The microbial community in eutrophic freshwater sediment was investigated from a 67-cm-deep sediment core collected from the Daechung Reservoir in South Korea, where cyanobacterial blooms have occurred annually for the past 30 years. The majority of core sediments were characterized by dark-grayish, fine-grained mud with abundant gas-escaped and thinly laminated layers. Intervals of summer and winter seasons were represented by periodic peaks of geochemical profiles of parameters such as grain size and relative carbon mass ratios to various nutrients such as nitrogen, sulfur, and phosphorus. I…

Growth phase-dependent activation of nitrogen-related genes by a control network of group 1 and group 2 sigma factors in a cyanobacterium.

It has been reported that an RNA polymerase sigma factor, SigC, mainly contributes to specific transcription from the promoter PglnB-54,-53 under nitrogen-deprived conditions during the stationary phase of cell growth in the cyanobacterium Synechocystis sp. strain PCC 6803 (Asayama, M., Imamura, S., Yoshihara, S., Miyazaki, A., Yoshida, N., Sazuka, T., Kaneko, T., Ohara, O., Tabata, S., Osanai, T., Tanaka, K., Takahashi, H., and Shirai, M. Expression of the psbA gene during photoinhibition and recovery in Synechocystis PCC 6714: inhibition and damage of transcriptional and translational machinery prevent the restoration of photosystem II activity.

The D1 reaction center protein of the photosystem II complex is very sensitive to light. It is continuously being damaged, degraded and resynthesized. Under high light, photosystem II inactivation is observed. This is because the rate of D1 damage is faster than that of its replacement. This process can be reversed if exposure to high light is not too long. In this work we study the changes that occur in the transcriptional and translational machinery that could lead to irreversible photoinhibition in Synechocystis PCC 6714. In the first minutes of photoinhibition, high light induced an accumu

Unveiling the 12-Hour Ultradian Rhythm: Biological Foundations, Mechanistic Insights, and Potential Applications.

The ~12-h ultradian rhythm (circasemidian) represents an evolutionarily conserved temporal architecture that complements the canonical 24-h circadian clock. Over the past 5 years, mounting evidence has revealed its ubiquity across biological kingdoms, from tidal marine organisms and cyanobacteria to plants, microbiomes, and mammals, including humans, manifesting as intrinsic oscillations in gene expression, metabolism, and behavior that often persist independently of circadian control. In mammals, this rhythm is driven by a cell-autonomous oscillator centered on the XBP1s (X-box binding protei…

Deciphering the directed iron-stress regulatory network of Synechocystis sp. PCC 6803 reveals novel master regulators.

Iron deficiency elicits extensive transcriptional reprogramming in cyanobacteria; however, the directed regulatory hierarchy that coordinates this response remains insufficiently resolved. In this study, we integrated 34 iron-stress transcriptomic samples into a customized deep graph transformer (DGT) framework to infer a directed, context-specific gene regulatory network (GRN) for Synechocystis sp. PCC 6803 under iron limitation. This analysis identified 82 differentially expressed genes, with isiB and the antisense RNA IsrR exhibiting the strongest transcriptional shifts. The resulting GRN d…

A novel synergistic phycobiliprotein combination against HIV-1 shows potent antiviral activity.

Human immunodeficiency virus type 1 (HIV-1) remains a major global public health challenge due to lifelong persistence, latency, and the emergence of drug resistance. Natural bioactive compounds are increasingly being explored as alternative or adjunct antiviral strategies. In this study, we evaluated the antiviral efficacy and cytotoxicity of a synergistic pharmaceutical composition comprising three phycobiliproteins: C-phycocyanin (C-PC), B-phycoerythrin (B-PE), and allophycocyanin (APC), derived from cyanobacteria and marine algae. Individual and combinatorial effects were assessed against …

Glycogen deficiency impairs diurnal energy metabolism and cell division in Synechocystis.

Diurnal changes in light availability are a defining feature of life on Earth. Photoautotrophic organisms therefore store reduced carbon during the day to sustain energy metabolism at night. In cyanobacteria, glycogen is the primary carbon storage compound and supports both energy homeostasis and stress responses. Although glycogen-deficient Synechocystis strains have been studied previously, how these mutants cope with the loss of the major daytime carbon sink and can sustain themselves during the night remains unclear. Using single-cell microfluidics, transcriptomics, and metabolomics, we sh…

CurT dosage quantitatively affects thylakoid structure and PSII performance in Synechocystis sp. PCC 6803.

The photosynthetic thylakoid membrane systems of many oxygenic photosynthesizers form elaborate three-dimensional structures. Curvature Thylakoid (CurT/CURT1) proteins are central regulators of thylakoid architecture in cyanobacteria and plants, driving both grana stacking and the formation of thylakoid convergence zones (TCZs) by promoting membrane curvature while also contributing to green-algal chloroplast and cyanobacterial cell division. While the functional role of grana stacks in terrestrial photosynthesis is well established, the physiological significance of cyanobacterial TCZs remain…

Pathogenic bacteria contribute to antibiotic resistance genes dissemination in microcystin-LR exposed intestine of zebrafish.

Cyanobacterial blooms and antibiotic resistance genes (ARGs) in aquatic ecosystems are emerging public health concerns. Microcystin-LR (MC-LR), a representative cyanotoxin released from cyanobacterial blooms, has poorly understood mechanisms regarding its influence on the dissemination of ARGs. In this study, zebrafish were exposed to MC-LR at environmentally relevant concentrations to investigate the mechanisms underlying the influence of MC-LR on the prevalence and dissemination of ARGs in the intestine, with a focus on intestinal microbiota and pathogenic bacteria. Overall, MC-LR exposure s…

Assessing the risk of cyanobacterial toxins in freshwater ecosystems: microcystin-LR exposure in Chironomus riparius larvae.

Cyanobacteria produce a variety of secondary metabolites, including the potent and widespread toxin microcystin-LR (MC-LR). While the toxic effects of MC-LR on aquatic species are well documented, its impact on Chironomid larvae, key organisms in freshwater food chains, remains insufficiently understood. This study investigated the acute toxicity of MC-LR at environmentally relevant concentrations (5, 10, 15, and 30 µg/L) on Chironomus riparius (Diptera, Chironomidae). The effects of the tested concentrations were evaluated using a multi-biomarker approach, including hemoglobin levels, oxidati…

Cyanotoxin cylindrospermopsin disrupts lipid homeostasis and metabolism in a 3D in vitro model of the human liver.

Cylindrospermopsin, a potent hepatotoxin produced by harmful cyanobacterial blooms, poses environmental and human health concerns. We used a 3D human liver in vitro model based on spheroids of HepG2 cells, in combination with molecular and biochemical assays, automated imaging, targeted LC-MS-based proteomics, and lipidomics, to explore cylindrospermopsin effects on lipid metabolism and the processes implicated in hepatic steatosis. Cylindrospermopsin (1 μM, 48 h) did not significantly affect cell viability but partially reduced albumin secretion. However, it increased neutral lipid accumulati…

Degradation of Cylindrospermopsin Spiked in Natural Water (Paranoá Lake, Brasília, Brazil) by Fenton Process: A Bench-Scale Study.

The frequency and intensity of harmful cyanobacterial blooms have increased in the last decades, posing a risk to public health since conventional water treatments do not effectively remove extracellular cyanotoxins. Consequently, advanced technologies such as the Fenton process are required to ensure water safety. The cyanotoxin cylindrospermopsin (CYN) demands special attention, as it is abundant in the extracellular fraction and has a high toxicological potential. Hence, this study aimed to assess the application of the Fenton process for the oxidation of CYN spiked in natural water from Pa…

Potential oestrogenic effects (following the OECD test guideline 440) and thyroid dysfunction induced by pure cyanotoxins (microcystin-LR, cylindrospermopsin) in rats.

Potential endocrine-disrupting properties of cyanotoxins, such as microcystin-LR (MC-LR) and cylindrospermopsin (CYN) are of concern due to their increasing occurrence, the scarcity of reports on the topic (particularly for CYN) and the impact of human's health at different levels. Thus, this work performed for the first time the uterotrophic bioassay in rats, following the Organization for Economic Cooperation and Development (OECD) Test Guideline 440, to explore the oestrogenic properties of CYN and MC-LR (75, 150, 300 μg/kg b.w./day) in ovariectomized (OVX) rats.
