In the course of immunoscreening of Clonorchis sinensis cDNA library, a cDNA CsRP12 containing a tandem repeat was isolated. The cDNA CsRP12 encodes two putative peptides of open reading frames (ORFs) 1 and 2 (CsRP12-1 and -2). The repetitive region is composed of 15 repeats of 10 amino acids. Of the two putative peptides, CsRP12-1 was proline-rich and found to have homologues in several organisms. Recombinant proteins of the putative peptides were bacterially produced and purified by an affinity chromatography. Recombinant CsRP12-1 protein was recognized by sera of clonorchiasis patients and

The aim of the study was to purify and characterise recombinant proteins with the potential as an anti-parasite vaccine. Full-length cDNAs encoding seryl-tRNA synthetase (srs-2) were cloned from Haemonchus contortus (HcSRS-2) and Teladorsagia circumcincta (TcSRS-2). TcSRS-2 and HcSRS-2 cDNA (1458bp) encoded proteins of 486 amino acids, each of which was present as a single band of about 55 kDa on SDS-PAGE. Multiple alignments of the protein sequences showed homology of 94% between TcSRS-2 and HcSRS-2, 76-93% with SRS-2s of eight nematodes and 68% with Mus musculus SRS-2. The predicted three-di

BACKGROUND: Cysteine proteinases of Fasciola hepatica are important candidates for vaccine antigens because of their role in fluke biology and host-parasite relationships. In our previous experiments, we found that a recombinant cysteine proteinase cloned from adult F. hepatica (CPFhW) can protect rats against liver fluke infections when it is administered intramuscularly or intranasally in the form of cDNA. We also observed considerable protection upon challenge following mucosal vaccination with inclusion bodies containing recombinant CPFhW produced in Escherichia coli. In this study, we exp

The tegumental membrane of platyhelminth parasites is of crucial importance for modulation of the host response and parasite survival. A cDNA encoding a novel tegumental protein 20.8 kDa (TP20.8) was found by large-scale sequencing of a Clonorchis sinensis cDNA library. This new cDNA was 755 bp long containing an open reading frame of 555 bp, which encoded a 20.8-kDa protein with an isoelectric point of 4.33. The deduced amino acid sequence exhibits 40 and 37% identity to Schistosoma japonicum sj20.8 and Schistosoma mansoni Sm 20.8, respectively. TP20.8 transcripts were detected in the adult w

The heterotrimeric guanine nucleotide-binding proteins (G proteins) act as switches in the signal transduction from cell surface receptors to a variety of effectors. Among them, Gs proteins stimulate adenylate cyclase activities and regulate ion channels in mammals. We identified the gsa-1 gene encoding a G protein alpha subunit in the nematode Caenorhabditis elegans. The predicted product consists of 375 amino acid residues, 66% of which are identical with those of a mammalian Gs(alpha) subunit. The gsa-1 gene was physically mapped near the left end of chromosome I. A gsa-1/lacZ fusion gene w

Infection with the helminth Schistosoma mansoni can cause exacerbated morbidity and mortality via a pathogenic host CD4 T cell-mediated immune response directed against parasite egg antigens, with T helper (Th) 17 cells playing a major role in the development of severe granulomatous hepatic immunopathology. The role of inflammasomes in intensifying disease has been reported; however, neither the types of caspases and inflammasomes involved, nor their impact on the Th17 response are known. Here we show that enhanced egg-induced IL-1β secretion and pyroptotic cell death required both caspase-1 a

One female specimen of the cetacean harbour porpoise Phocoena Phocoena (Linnaeus, 1758) was found dead on the North Sea coast of Denmark (Jutland) in May 2025. During necropsy severe pathological changes of the liver were observed (hepatic fibrosis, enlarged and thickened biliary duct walls) and when dissecting the biliary ducts six intact trematodes (total length 6.9-7.9 mm, width 1.9-2.0 mm) were recovered. Following conservation in formalin four parasites were haematoxylin stained, one specimen kept unstained but all five mounted for morphological analysis. The sixth specimen conserved in 7

Here, we present a comprehensive morphological and molecular phylogenetic analysis of Clinostomum sp. (Digenea: Clinostomidae) metacercariae parasitizing two freshwater fish species from Southeast Brazil: Serrasalmus spilopleura (piranha) and Callichthys callichthys (tambuatá). The morphological examination revealed distinct characteristics of metacercariae in each host. Using the cytochrome c oxidase I (COI) gene barcode region, we obtained DNA sequences that allowed for accurate phylogenetic placement. Phylogenetic analyses revealed that Clinostomum sp. HM41 (metacercariae), isolated from S.

Fasciolosis is a parasitic disease affecting humans and livestock, caused by digenean trematodes of the genus Fasciola, primarily F. hepatica and F. gigantica. This study investigates the coexistence of these species and their hybrids in yaks from Tibet, China. We analyzed the nuclear rDNA internal transcribed spacer (ITS) regions, including ITS1 and ITS2, through Sanger sequencing and Next-Generation Sequencing (NGS) to assess single-nucleotide polymorphisms (SNPs). Our results reveal that one specimen (NM008B) is identical to pure F. hepatica, while another (NM008A) contains genetic informat

A new species of bothriocephalid cestode in the genus Anantrum is described from the intestine of the shorthead lizardfish Synodus scituliceps from the north Pacific coast of Costa Rica. The new species is described based on an integrative taxonomic approach that includes the use of light and scanning electron microscopy, 28S rDNA sequencing, and phylogenetic analysis. Anantrum gallopintoi sp. nov. is the third known member of this genus and can be distinguished from A. tortum (Linton, 1905) and A. histocephalum Jensen & Heckmann, 1977 by a combination of morphological and ecological traits an

The aim of the present study was to genetically characterize Fasciola hepatica strains from diverse ecogeographical regions (America and Europe), susceptible and resistant to Triclabendazole, using the random amplified polymorphic DNA fragments (RAPDs-PCR) technique to elucidate genetic variability between the different isolates. Ten different oligonucleotide primers of 10 bases with GC content varying from 50-70% were used. A polymerase chain reaction (PCR) was carried out in 25 μl of total volume. Duplicate PCR reactions on each individual template DNA were performed to test the reproducibil

Harmful parasites of the wild northern boreal mammals are still surprisingly poorly studied. In 2003-2006, a peritonitis outbreak caused by the filarioid nematode, Setaria tundra, emerged in Finland's reindeer population. In order to gain knowledge about the basic biology, epidemiology, and transmission dynamics of this parasite, samples for S. tundra were collected from reindeer and other cervids during the follow-up period 2004-2006. Using morphology and molecular biology methods, we describe here S. tundra's first larval stage, microfilaria (smf), for the first time scientifically. The prev

Human opisthorchiais caused by the liver fluke Opisthorchis viverrini is a major fish-borne trematode infection endemic in the Southeast Asian countries. The infection is acquired through consumption of raw fish harboring metacercariae of O. viverrini. Owing to potential risk of transmission of opisthorchiasis through fish trade, rapid and reliable detection methods have gained importance to ensure food safety. In the study described here, we report successful development and evaluation of a polymerase chain reaction (PCR) assay for the detection of O. viverrini, based on the nucleotide sequen

The diversity of Rhabdias includes 101 species, 71 of which parasitize the lungs of anurans, caudates, gymnophionans, and some occur in reptiles worldwide. Currently, 26 species are found in the Neotropical region, and in Brazil, there are 16 nominal species, a relatively low number considering the high diversity of potential hosts. Here, we describe a new species of Rhabdias found in Physalaemus albonotatus, with morphological and molecular data, as well as phylogenetic analyses using sequences of the mitochondrial gene Cytochrome Oxidase Subunit I (COI). Rhabdias taquariensis n. sp. differs

Background: Cystic echinococcosis (CE) caused by the parasite Echinococcus granulosus sensu lato is one of the most important zoonoses worldwide, causing a great impact on human and animal health. Despite its impact, control programs have not been able to eradicate the parasite; in part, because its transmission cycles, its phylogeny, and the host-parasite relationships are still not well known. In order to deepen knowledge about Echinococcus spp. populations in the endemic areas of southern Chile, the present study was carried out. Materials and Methods: Ten isolates of CE cysts were analyzed

Saccocoelioides is a genus within the family Haploporidae and is distributed throughout the Americas. The recent application of molecular techniques has facilitated the reorganization of this genus and the description of new species, resulting in a current total of 28 species. In Argentina, 11 species have been identified; however, the validity of Saccocoelioides bacilliformis and Saccocoelioides octavus remains controversial. Recently, specimens of digeneans were collected from two distinct geographical locations: the Paraná River basin (Misiones province) and the Río de la Plata basin (Bueno

Abdominal angiostrongyliasis (AA) is a zoonotic and severe parasitic infection caused by Angiostrongylus costaricensis. AA is currently diagnosed by the observation of A. costaricensis-compatible structures in biopsies or the detection of antibodies in serological tests. However, molecular methods targeting homologous sequences of A. costaricensis have not been designed before, and therefore, an HRM-coupled qPCR was developed to detect the internal transcribed spacer 1 (ITS1) of the parasite. The present assay successfully amplified DNA of A. costaricensis obtained from different hosts and ide
