Dendritic cells control tertiary lymphoid structure development and maintenance in cancer.

Tertiary lymphoid structures (TLSs) are associated with immunotherapy response, yet the mechanisms controlling their formation and maintenance remain unclear. Using spatial transcriptomics and multiplex imaging across human tumors, we found that CCR7+ mature dendritic cells (DCs) accumulate in TLSs. In a mouse non-small cell lung cancer model that forms mature TLSs, we show that early TLS development requires interferon-γ (IFN-γ)-driven type 1 conventional dendritic cell (cDC1) maturation, migration to tumor-draining lymph nodes (tdLNs), and T cell recruitment. As tumors progress, TLSs persist

Biomaterial-Focused Strategies Targeting Dendritic Cells for Autoimmune Disease Treatment.

Autoimmune diseases encompass over 100 distinct diseases where the immune cells betray our body by attacking the tissues they are meant to protect. Even though rare, 15 million Americans are collectively affected by autoimmune diseases, a number that continues to rise every year. Current treatment modalities primarily focus on alleviating symptoms rather than providing prevention or a cure. Tolerogenic dendritic cells (tolDCs) have gained popularity for autoimmune disease treatment as an alternative to traditional systemic immunosuppressive therapies due to their ability to restore immune home

Thermodynamics and Kinetics in Host Design for Dendrite-Free Zinc Metal Anodes.

Aqueous zinc metal batteries (AZMBs) have emerged as promising candidates for grid-scale energy storage due to their intrinsic safety, cost-effectiveness, and environmental friendliness. Nonetheless, the practical application of AZMBs is restricted by the uncontrolled Zn dendrite growth arising from undesirable reaction kinetics, as well as severe parasitic reactions (e.g., hydrogen evolution reaction, corrosion, and passivation) caused by the thermodynamic instability of Zn metal anodes (ZMAs) in aqueous electrolytes. Among various strategies, host design enables the effective mitigation of t

RETREG1-Mediated Reticulophagy is Essential for Dendritic Cell Maturation and Function in Sepsis.

Dendritic cells (DCs) are crucial antigen-presenting cells that mediate the interplay between innate and adaptive immunity during lethal infections. Here, we report the key role of reticulophagy regulator 1 (RETREG1), a selective autophagy receptor, in maintaining DC maturation and function in the early stage of sepsis. Mechanistically, activating transcription factor 6 (ATF6) acts as a direct transcription factor regulating RETREG1 expression in response to bacterial lipopolysaccharide-induced endoplasmic reticulum (ER) stress. RETREG1-mediated reticulophagy reduces excessive ER stress via th

Development of Autologous Dendritic Cell Vaccine Therapeutics for Canine Mammary Cancer.

Canine mammary tumors have been investigated to determine the causes of malignancy and to promote the development of more effective therapies. The current standard of care, surgical resection where possible, often still results in recurrence of disease. Thus, there is an unmet need for better, more effective therapies that can suppress recurrence in canine patients. Because canine and human mammary cancers, particularly carcinomas and adenocarcinomas, share many similarities in genetic defects, etiology, natural history, and environment, canine mammary cancer cell lines have also been used as

Case Report: Primed dendritic cell immunotherapy for multiple solid tumors.

Many patients with advanced solid tumors ultimately develop resistance to conventional therapies, with especially limited options for those experiencing rapid progression or poor performance status, highlighting the need for personalized immunotherapies capable of inducing durable responses. In this retrospective case series, we describe the clinical course and outcomes of five adults (4 male, 1 female; median age 51 years [range, 39-62]) with metastatic solid tumors who achieved complete and durable remission following doubly loaded autologous dendritic cell therapy after failure of standard

CXCR4 marks homeostatically activated dendritic cells in the steady state and in cancer.

Conventional dendritic cells (cDCs) can be activated by pathogen signals and inflammation to drive T cell immunity to infection, but they can also undergo "homeostatic activation" at steady state. However, homeostatically activated cDCs closely resemble those activated by microbial or viral stimuli, hindering their study. Here, we identify the chemokine receptor CXCR4 as a specific marker of homeostatically activated cDCs across mouse tissues. CXCR4 is induced in cDCs in the steady state but not following stimulation with Toll-like receptor agonists or type I interferons. In tumors, CXCR4 expr

Extracellular GPX4 impairs antitumor immunity via dendritic ZP3 receptors.

Understanding the immunogenic properties of different forms of cell death is critical for rationalized antineoplastic therapeutic development. Here, we identify a regulatory axis that suppresses the immunogenicity of ferroptosis. During ferroptosis, but not apoptosis, cuproptosis, or necroptosis, cancer cells release glutathione peroxidase 4 (GPX4), which binds to zona pellucida glycoprotein 3 (ZP3) on the surface of dendritic cells (DCs), activates the 3',5'-cyclic adenosine monophosphate (cAMP)-protein kinase AMP-activated (PRKA) signaling cascade, inhibits glycolysis, and impairs maturation

A Key Regulator of Dendritic Morphology in Supragranular Neocortex Impacts Mismatch Negativity.

BACKGROUND: Cytoskeletal structure and neuronal function are heavily intertwined, and altered pyramidal neuron morphology (e.g., reduced dendritic arbors) has been consistently identified in the neocortex of individuals with psychiatric disorders. A missense mutation in Kalrn (Kalrn-PT) enhances activation of RhoA, a cytoskeletal regulator, and leads to adolescent-onset dendritic regression in supragranular auditory cortex. To investigate the relationship between this spatiotemporally altered cytoskeletal structure and psychiatric-relevant dysfunction, we examine the functional impacts of this

The transcriptome of dendritic cells redraws the boundaries between pathogenicity and commensalism in yeast.

Our immune system is constantly exposed to fungi, but it mounts a consistent, disease-related, and species-specific inflammatory response only against a few fungal species. Most of the current understanding of fungus-host interactions is based on a limited number of strains, hence neglecting the fungal intra-specific genetic and phenotypic diversity. To expand our knowledge of the spectrum of immune responses to pathogenic and non-pathogenic fungi, we compared the cytokine and transcriptional profiles of human monocyte-derived dendritic cells exposed to Aspergillus fumigatus, Candida albicans,

The emerging role of plasmacytoid dendritic cell proliferation in acute myeloid leukemia.

The mature plasmacytoid dendritic cell proliferation associated with myeloid neoplasm represents a clonal proliferation of plasmacytoid dendritic cells within myeloproliferative and myelodysplastic disorders. This entity was recently recognized as a distinct condition in the fifth edition of the World Health Organization classification of hematolymphoid tumors. It occurs in approximately 4.9% of acute myeloid leukemia cases. The pathogenic mechanisms underlying this proliferation and the role of these cells in disease progression remain poorly understood. Nevertheless, the plasmacytoid dendrit

Monocytoid dendritic cells might be a specific marker for lung involvement in dermatomyositis.

Dermatomyositis (DM) is a rare autoimmune disease leading to inflammatory and degenerative changes affecting skin and muscles. Interstitial lung disease is a major risk factor for morbidity and mortality in DM. The precise immune mechanism underlying DM, particularly in the context of lung involvement, remains poorly understood. This research focuses on the significance of dendritic cell subsets and Langerhans cells in the immune irregularities observed in DM.The study analyzed 12 cases of DM with lung involvement, and compared the findings to 12 cases with skin involvement. All subjects were

The histopathological differential diagnosis of BPDCN and other plasmacytoid dendritic cell proliferations.

Blastic plasmacytoid dendritic cell neoplasm (BPDCN) is a rare, aggressive precursor cell hematological malignancy with a dismal prognosis, necessitating prompt and accurate diagnosis. However, its blastoid morphology and immunophenotype may lead to diagnostic confusion with a broad spectrum of hematological neoplasms. This systematic review delineates the critical morphological, immunophenotypic, and molecular features required to distinguish BPDCN from its mimics across common sites of involvement (skin, bone marrow, and lymph nodes). We reinforce the diagnostic criteria for BPDCN and provid

PTEN regulates starburst amacrine cell dendrite morphology during development.

Neurons are subject to extensive developmental regulation to ensure precise subtype-specific morphologies that are intimately tied to their function. Starburst amacrine cells (SACs) in the mammalian retina have a highly stereotyped, radially symmetric dendritic arbor that is essential for their role in direction-selective circuits in the retina. We show that PTEN, the primary negative regulator of the PI3K-AKT-mTOR pathway that is highly implicated in neurodevelopmental disorders, regulates SAC morphology in a cell-autonomous manner. Pten-deficient SACs show a nearly twofold increase in the nu

Nootropic benzothiazoles promote dendritic spine formation by targeting fascin-1.

Identifying new drug targets to enhance memory and learning is essential for treating neurodegenerative diseases, such as Alzheimer's disease. We previously showed that benzothiazole amphiphiles can improve memory and learning by increasing dendritic spine and synaptic density in both WT mice and in a transgenic Alzheimer's model mice. The cellular target for this class of compounds, however, was unknown. Using a photoaffinity-labeling approach, we identify fascin-1 as the major protein target in neurons for these compounds. These compounds enhance spine density by directly modulating actin dy

Revolutionizing cancer treatment: The power of dendritic cell-based vaccines in immunotherapy.

In the modern time, cancer immunotherapies have increasingly become vital treatment options, joining long-established methods like surgery, chemotherapy, and radiotherapy treatment. Central to this emerging approach are dendritic cells (DCs), which boast a remarkable ability for antigen presentation. This ability is being leveraged to modulate T and B cell immunity, offering a groundbreaking strategy for tackling cancer. However, the percentage of patients experiencing meaningful benefits from this treatment remains relatively low, underscoring the ongoing necessity for further research and de

Dendritic retraction contributes to spine density recovery following denervation in vitro.

Neurodegenerative diseases cause local brain lesions as well as denervation of neurons in areas connected to the lesion site. In part, the denervation-induced damage associated with brain lesions can be homeostatically compensated by network reorganization. To study the dynamics of compensatory changes occurring in denervated brain areas, we established an in vitro denervation model based on organotypic slice cultures of mouse entorhinal cortex and hippocampus. In these cultures, the entorhino-dentate projection was transected and denervation-induced changes of tdTomato-labeled dentate granule

Dendrite cross attention for high-dose-rate brachytherapy distribution planning.

Cervical cancer is a significant global health issue, and high-dose-rate brachytherapy (HDR-BT) is crucial for its treatment. However, manually creating HDR-BT plans is time-consuming and heavily relies on the planner's expertise, making standardization difficult. This study introduces two advanced deep learning models to address this need: Bi-branch Cross-Attention UNet (BiCA-UNet) and Dendrite Cross-Attention UNet (DCA-UNet). BiCA-UNet enhances the correlation between the CT scan and segmentation maps of the clinical target volume (CTV), applicator, bladder, and rectum. It uses two branches:

Regional Specificity of Cortical Layer 3 Dendritic Spine Deficits in Schizophrenia.

IMPORTANCE: Schizophrenia (SZ) is characterized by deficits in visual-spatial working memory, a function dependent on a distributed cortical network that includes nodes in the primary visual (V1), posterior parietal (PPC), and dorsolateral prefrontal (DLPFC) cortices. The connections across these regions involve excitatory synapses on the dendritic spines of pyramidal neurons in layer 3 (L3). OBJECTIVE: To assess whether SZ is associated with regional differences in the magnitude or nature of L3 spine alterations. DESIGN, SETTING, AND PARTICIPANTS: This case-control study examined brain tiss

Direct evidence for dendritic spine compensation and regeneration in Alzheimer's disease models.

INTRODUCTION: Dendritic spine loss in Alzheimer's disease (AD) strongly correlates with cognitive decline, whereas spine preservation is associated with cognitive resilience. Yet, whether and how neurons compensate for spine loss in AD remains largely unknown. METHODS: We developed a chromophore-assisted light inactivation (CALI) strategy to selectively eliminate dendritic spines to model this key feature of AD. Two-photon microscopy was used to monitor the structural plasticity of spines over time after spine elimination. Validation experiments were conducted in amyloid beta (Aβ)-driven mode
