Rapid screening of polysaccharide-based plasma volume expanders dextran and hydroxyethyl starch in human urine by liquid chromatography-tandem mass spectrometry.

The increasing number of samples and target substances in doping control requires continuously improved screening methods, combining high-throughput analysis, simplified sample preparation, robustness and reliability. Hence, a rapid screening procedure based on liquid chromatography-electrospray ionization-tandem mass spectrometry with in-source collision-induced dissociation was developed. The detection of the polysaccharide-based plasma volume expanders dextran and hydroxyethyl starch (HES) in human urine was established without further sample preparation. The in-source fragmentation strateg

Detection of diffuse glomerular lesions in rats: II. Comparison of indium-111 cationic small macromolecules with technetium-99m DTPA.

Dextrans with average molecular weights of 5,000, 10,000, and 17,500 and inulin were rendered cationic by amination with 2-bromoethylamine hydrobromide. After limited coupling with DTPA cyclic dianhydride, they were labeled with 111In. A good correlation was found between their early renal uptake quantitated by camera-computer techniques and their renal clearance from multiple plasma samples in rats with glomerular damage induced by puromycin aminonucleoside and controls. However, there was poor correlation between the early renal uptake of these agents and the clearance of simultaneously inje…

[Transfusion therapy as a method of correcting peripheral microcirculatory disorders in severe cranio-cerebral trauma].

Photoplethysmographic and capillaroscopic studies of peripheral microcirculation conducted in patients with severe craniocerebral injuries during the acute period revealed some regularities of its changes depending on the type of the transfusion medium. While transfusions of glucose-novocain mixtures and, especially, of low molecular weight dextrans increase the volume pulse and improve the capillary flow in patients with supratentorial pathology, and blood and plasma transfusions reduce the volume pulse and enhance the capillary disorders, patients with brain stem insufficiency display a reve…

Daikenchuto ameliorates dextran sulfate sodium-induced acute and chronic ulcerative colitis by regulating gut microbiota-derived indoles to activate AhR signaling.

BACKGROUND: Ulcerative colitis (UC), a chronic-relapsing inflammatory disease with rising prevalence worldwide, is primarily driven by intestinal epithelial barrier dysfunction resulting from gut microbial dysbiosis and metabolic disturbances. Daikenchuto (DKT), a traditional Chinese medicine formulation, is commonly used for digestive disorders. Although DKT has demonstrated therapeutic potential for gut inflammation by modulating gut microbiota, its therapeutic effects on chronic ulcerative colitis (CUC) and the related mechanisms remain elusive. METHODS: The main components of DKT were ten

Noble metal/dextran-graft-polyacrylamide branched polymer nanocomposites induce ROS-dependent apoptosis in diffuse large B-cell lymphoma cells.

In general, polymer/metal nanocomposites might have better anti-cancer properties in comparison to standalone metal nanoparticles (NPs). This emerging class of functional nanomaterials shows improved and more selective tumor delivery with lower toxicity against non-tumor tissues, providing a better biocompatibility and higher effectiveness. Herein, the cytotoxicity of four two-component nanocomposites comprising a metal NP (Au or Ag) and a star-like polymer (dextran-graft-polyacrylamide, D-PAA, or its anionic form, D-PAAan) was tested against multiple diffuse large B-cell lymphoma (DLBCL) cell

Generation of MCF-7 Spheroids in Polyethylene Glycol-Dextran Droplets for Cancer Niche Studies Using Aqueous Two-Phase System-3D Platforms.

Cell-based aqueous two-phase system (ATPS) applications involve the construction of 3D cultures encapsulating cells in polyethylene glycol (PEG)-dextran (DEX) droplets due to their low interfacial tension. This technology has enabled cell patterning in a controlled-defined shape giving rise to specific cell microenvironments. The present work aims to encapsulate MCF-7 cancer cells in ATPS droplets evaluating four different compositions of PEG-DEX, construction Strategies A (droplet added), B (immersed), or C (covered), and two cell densities (5000 and 10,000 cells/µL) during a 24 h incubation

4-octyl itaconate-functionalized dextran-polyethylene glycol hydrogel prevents postoperative peritoneal adhesion via regulating macrophage function and mesothelial differentiation.

Hydrogel barrier is considered to be a promising strategy to prevent postoperative peritoneal adhesion (PPA) formation. However, a simple physical barrier alone might not satisfactorily prevent adhesion. Herein, four-octyl itaconate (4-OI), a cell-permeable itaconate derivative with antiinflammatory and antioxidant properties, was encapsulated into micelles and then loaded into the dextran-polyethylene glycol hydrogel via the Schiff's base reaction between the aldehyde group of oxidized dextran and the amino group of four-arm polyethylene glycol amine (4-arm-PEG-NH2). The fabricated hydrogels

Anti-inflammatory effects of ethanolic extract from Abeliophyllum distichum (Miseon Tree) leaves in mice with dextran sulfate sodium-induced ulcerative colitis.

Abeliophyllum distichum (Miseon tree), a native Korean plant, is known for the high phenolic content in its leaves. The ethanolic leaf extracts of A. distichum have shown antioxidant, anti-obesity, and anti-inflammatory properties. However, studies on its potential to improve colitis are limited. This study aimed to determine whether the ethanolic extract of A. distichum leaves (ADE) could alleviate dextran sulfate sodium (DSS)-induced colitis in mice. The mice were divided into three groups, and the experimental group was given 300 mg/kg ADE for 4 weeks. One week before the end of the experim

Diallyl trisulfide alleviates dextran sulphate sodium-induced colitis in mice by inhibiting NLRP3 inflammasome activation via ROS/Trx-1 pathway.

Diallyl trisulfide (DATS), a sulphur-containing compound isolated from the medicinal food plant garlic, has been previously reported to attenuate experimental colitis induced by either dextran sodium sulphate (DSS) or 2,4,6-trinitrobenzenesulfonic acid (TNBS) in mice; however, the underlying mechanism remains to be identified. In this study, we deciphered the key mechanism by which DATS alleviates ulcerative colitis (UC). We showed that oral administration of DATS for 10 consecutive days greatly restrained the infiltration of macrophages and the pathological changes in colonic tissues of mice

Oxidative Stress-Responsive Apoptosis Inducing Protein (ORAIP) Plays a Critical Role in Dextran Sulfate Sodium-Induced Murine Model of Ulcerative Colitis.

Oxidative stress is implicated in the pathogenesis of various acute disorders including ischemia/reperfusion injury, ultraviolet/radiation burn, as well as chronic disorders such as dyslipidemia, atherosclerosis, diabetes mellitus, chronic renal disease, and inflammatory bowel disease (IBD). However, the precise mechanism involved remains to be clarified. We formerly identified a novel apoptosis-inducing humoral protein, in a hypoxia/reoxygenation-conditioned medium of cardiac myocytes, which proved to be 69th tyrosine-sulfated eukaryotic translation initiation factor 5A (eIF5A). We named this

Sustained delivery of CpG oligodeoxynucleotide by acetalated dextran microparticles augments effector response to Computationally Optimized Broadly Reactive Antigen (COBRA) influenza hemagglutinin.

A subunit or protein-based influenza vaccine can be a safer alternative to live attenuated vaccine (Flumist) and require fewer boosts than an inactivated vaccine (e.g. Fluzone). However, to form an effective subunit vaccine, an adjuvant is often needed. In this work we used electrospray to encapsulate the hydrophilic adjuvant CpG into microparticles made from the hydrophobic biodegradable polymer acetalated dextran. To understand the rate of particle degradation on CpG release, polymer that was slow (21 h at phagosomal pH 5) and fast (0.25 h at pH 5) degrading was used to encapsulate the adjuv

Deinococcus radiodurans R1 Lysate Induces Tolerogenic Maturation in Lipopolysaccharide-Stimulated Dendritic Cells and Protects Dextran Sulfate Sodium-Induced Colitis in Mice.

Deinococcus radiodurans is an extremophilic bacterium that can thrive in harsh environments. This property can be attributed to its unique metabolites that possess strong antioxidants and other pharmacological properties. To determine the potential of D. radiodurans R1 lysate (DeinoLys) as a pharmacological candidate for inflammatory bowel disease (IBD), we investigated the anti-inflammatory activity of DeinoLys in bone marrow-derived dendritic cells (BMDCs) and a colitis mice model. Lipopolysaccharide (LPS)-stimulated BMDCs treated with DeinoLys exhibited alterations in their phenotypic and f
