Combinatorial engineering approaches for improved lipid production in Saccharomyces cerevisiae.

Triacylglycerols (TAGs) hold substantial medical, nutritional, and industrial value. However, environmental concerns demand more sustainable production alternatives. A promising approach is the fermentation of TAG-accumulating microbes. In this study, we engineered a previously developed high-lipid producing Saccharomyces cerevisiae strain to further increase lipid accumulation through a combinatorial approach. We obtained up to 327 mg∙gCDW-1 of lipid content by deleting CKB1, encoding a subunit of casein kinase 2 (CK2) related to the regulation of lipid metabolism, in combination with the del

Stabilizing Cu-Based Photocathodes: From Interfacial Engineering to Advanced Architectures.

Copper-based oxides, including Cu2O, CuO, CuBi2O4, CuFeO2 and CuFe2O4 have emerged as promising photocathode materials for solar-driven photoelectrochemical (PEC) reduction reactions such as hydrogen evolution (HER), carbon dioxide reduction (CO2RR), and nitrogen reduction (NRR). Their appeal lies in the combination of their earth-abundance, low toxicity, and suitable optoelectronic properties. However, the practical deployment of these materials is hindered by their intrinsic instability under operating conditions, primarily due to photocorrosion, interfacial charge recombination, and limited

Comparative protein engineering redirects the specificity of Clostridium botulinum proteases.

Botulinum neurotoxin serotypes A and E (BoNT/A and BoNT/E) cleave SNAP25 and are widely used in therapeutic applications. Redirecting the substrate specificity of their protease domains, LC/A and LC/E, could expand their utility to new therapeutic targets. Here, we report a comparative protein engineering strategy that integrates prior mutagenesis, bioinformatics, and structural insights to reprogram LC protease specificity. Directed evolution yielded a 14-mutation LC/A variant with 273-fold greater specificity for SNAP23 than a previously reported engineered protease. Insights from LC/A engin…

Salinity-Driven Microbial Community Engineering for Safer Ultrafiltration Water Reuse.

Ultrafiltration is central to water reclamation but faces two critical challenges: microbial regrowth that threatens biostability and pathogen invasion that undermines biosafety. Here, we proposed an ecological strategy that transformed backwash from a cleaning procedure into a microbiome engineering tool, thereby simultaneously addressing both challenges. Our findings provided evidence for the major microbial sources in permeate, including membrane breakthrough, detachment from the membrane permeate side, and from downstream pipeline surfaces. High-salinity backwash (100 mM NaCl) suppressed t…

Microbial Synthesis of Daphnetin by Engineering 2-Oxoglutarate-Dependent Dioxygenases.

Daphnetin is a clinically established coumarin, but its native biosynthetic pathway remains elusive. In this study, a 2-oxoglutarate-dependent dioxygenase (2OGD), AtS8H, was identified that hydroxylates umbelliferone (UMB) to daphnetin , enabling the design of an artificial biosynthetic pathway. However, the pathway efficiency was constrained by the poor solubility of AtS8H and the low catalytic activity of the upstream 2OGD enzyme IbC2'H. To enhance AtS8H solubility, we developed an integrated strategy combining ProteinMPNN-guided sequence redesign with surface charge engineering. The obtaine…

Engineering energy-efficient Saccharomyces cerevisiae for methanol and CO2 assimilation.

Methanol is a promising one-carbon (C1) feedstock for microbial bioconversion; however, engineered Saccharomyces cerevisiae often faces energetic constrains during its assimilation. Here, we develop SC-AOX25, an energy-efficient methylotrophic S. cerevisiae, through engineering of heterologous methanol-formaldehyde-formate (MFF) oxidation pathways coupled with adaptive laboratory evolution. SC-AOX25 efficiently generates adenosine triphosphate (ATP) and nicotinamide adenine dinucleotide (NADH) during methanol metabolism while co-assimilating methanol-derived intermediates (formaldehyde, format…

Cation substitution engineering persistent luminescence for dynamic optical anti-counterfeiting.

Long-persistent phosphors possess the unique ability to emit light long after excitation ceases. However, the tunable luminescence with diverse emission colors remains critically needed for applications such as advanced optical anti-counterfeiting. Herein, we designed a series of Sr1.98-x-y-zCayBazMgSi2O7:0.02Dy3+,xEu2+ (abbreviated as SCyBzMS:0.02Dy3+,xEu2+, 0 ≤ x ≤ 0.04, 0 ≤ y ≤ 2, and 0 ≤ z ≤ 1) phosphors using cation substitution engineering. XRD analysis confirmed the single-phase structure. The phosphors exhibit intense broad excitation bands spanning 250-450 nm. In the initial Sr2MgSi2O…

Armoring CAR-T Cells Against Exhaustion: Engineering Strategies for Glioblastoma.

Glioblastoma (GBM) is the most lethal primary brain tumor, with a median survival of 15 months despite intensive treatment. Chimeric antigen receptor (CAR) T-cell therapy, while transformative in hematological malignancies, consistently fails in GBM because the immunosuppressive tumor microenvironment (TME) drives T-cell exhaustion. We examined transcriptional programs, microenvironmental factors, and metabolic competition that collectively drive exhaustion in this context. Then we reviewed 5 convergent engineering strategies: localized cytokine delivery to bypass autocrine deficits; adjunctiv…

Engineering plant tandem kinase immune receptors expands effector recognition profiles.

Plant intracellular immune receptors are widely deployed in breeding to protect crops from disease. In addition to nucleotide-binding leucine-rich repeat receptors (NLRs), tandem kinase proteins (TKPs) have recently emerged as an important family of immune receptors within staple cereal food crops, but how TKPs recognize effectors and whether they are amenable to engineering is essentially unknown. Here, we show that the barley and wheat TKPs Rmo2 and Rwt7 recognize different blast fungus effectors via their integrated HMA domains using different protein interfaces with nanomolar binding affin…

Engineering dynamic hydrogels to overcome translational bottlenecks in therapeutic delivery.

Hydrogels are promising platforms for the controlled release of drugs, biologics, and living cells, but conventional statically crosslinked networks face important translational limitations, including poor injectability, brittle mechanical behavior, and an inability to recapitulate the time-dependent mechanics of native tissues. Dynamic bonding provides a powerful strategy to overcome these limitations while expanding the design space for controlled release. Non-covalent interactions, including hydrogen bonding, host-guest complexation, metal-ligand coordination, and electrostatics, as well as
