Downstream activation of NF-κB in the EDA-A1/EDAR signalling in Sjögren's syndrome and its regulation by the ubiquitin-editing enzyme A20.

Sjögren's syndrome (SS) is an autoimmune disease and the second most common chronic systemic rheumatic disorder. Prevalence of primary SS in the general population has been estimated to be approximately 1-3%, whereas secondary SS has been observed in 10-20% of patients with rheumatoid arthritis, systemic lupus erythematosus (SLE) and scleroderma. Despite this, its exact aetiology and pathogenesis are largely unexplored. Nuclear factor-kappa B (NF-κB) signalling mechanisms provide central controls in SS, but how these pathways intersect the pathological features of this disease is unclear. The

Hofbauer cell activation and its increased glucose-6-phosphate dehydrogenase activity in second trimester-spontaneous abortion: an ultrastructural dual staining enzyme-cytochemical study.

PROBLEM: While activated/phagocytosing phagocytes infiltrating to the chorioamnion are considered to be one of the causal agents of preterm labor onset, whether placental villous macrophages (Hofbauer cells) are activated/phagocytosing in this condition is not known. METHOD OF STUDY: We concomitantly localized two important phagocytosis-related enzymes, acid phosphatase (ACP) and glucose-6-phosphate dehydrogenase (G6PD), in Hofbauer cells in second trimester placental villi, and compared them with those from infection-related second trimester-spontaneous abortion (miscarriage) placentas. N-thiocarboxyanhydrides, amino acid-derived enzyme-activated H2S donors, enhance sperm mitochondrial activity in presence and absence of oxidative stress.

BACKGROUND: Hydrogen sulfide (H2S) donors are crucial tools not only for understanding the role of H2S in cellular function but also as promising therapeutic agents for oxidative stress-related diseases. This study aimed to explore the effect of amino acid-derived N-thiocarboxyanhydrides (NTAs), which release physiological H2S levels in the presence of carbonic anhydrase, on porcine sperm function during short-term incubation with and without induced oxidative stress. For this purpose, we employed two H2S-releasing NTAs with release half-lives (t1/2) in the range of hours that derived from the

Advancement and prospects of production, transport, functional activity and structure-activity relationship of food-derived angiotensin converting enzyme (ACE) inhibitory peptides.

Food-derived antihypertensive peptides have attracted increasing attention in functional foods for health promotion, due to their high biological activity, low toxicity and easy metabolism in the human body. Angiotensin converting enzyme (ACE) is a key enzyme that causes the increase in blood pressure in mammals. However, few reviews have summarized the current understanding of ACE inhibitory peptides and their knowledge gaps. This paper focuses on the food origins and production methods of ACE inhibitory peptides. Compared with conventional methods, the advanced technologies and emerging bioi

Anti-ulcer activity of curcumin on experimental gastric ulcer in rats and its effect on oxidative stress/antioxidant, IL-6 and enzyme activities.

OBJECTIVE: To investigate the possible mechanism by which curcumin protects stomach during the acute chronic phase of gastric ulcer disease. METHODS: The rats were divided into four groups and fasted for 2 days with free access to water. On the third day, the animals were fasted for a further 24 h with no access to water followed by surgery. Rats received different doses of curcumin (20, 40, and 80 mg/kg) or vehicle by oral gavage. Nineteen hours after ulcer induction, the rats were killed by decapitation. Stomach was opened along the greater curvature and ulcerative lesions were counted. Tot

Activity-Guided Identification of Pro-inflammatory Neutrophils with Dual Enzyme- and Metabolite-Activatable Fluorescent Probes.

Neutrophils are the most abundant circulating leukocytes in the innate immune system. Primary neutrophils comprise multiple distinct subtypes that regulate antimicrobial defense and inflammatory diseases. Current fluorescent probes for imaging neutrophils cannot distinguish between activation mechanisms. In this work, we present dual enzyme- and metabolite-activatable fluorogenic probes that are selective for activated neutrophils prone to inflammatory responses. Our probes integrate, within a single scaffold, two neutrophil-associated activation mechanisms, that is, cathepsin G-mediated cleav

Dual-functionalized carbon dots with precise antibacterial activity through specific recognition and enzyme inactivation.

The rise of drug-resistant bacteria, coupled with the toxicity of existing therapies, has underscored an urgent demand for novel, precise, and resistance-resistant antimicrobial agents. Carbon dots (CDs), an emerging class of carbon nanomaterials, offer a promising platform due to their excellent biocompatibility and tunable surface chemistry. In this study, we design and synthesize functional CDs capable of selective bacterial recognition coupled with potent antibacterial activity. These CDs were prepared from phytic acid (PA) and citric acid (CA) via electro-polymerization, which features du

UV-Mediated Enzyme-Like Activity Reversal Strategy To Regulate Nanozymes for Detection and Elimination of Bacteria.

Nanozyme-based biosensing has gained a great deal of attention in various fields. However, most peroxidase-like (POD-like) nanozymes exhibit excellent enzyme-like activity, relying heavily on acidic environments, and their activity decays sharply in neutral or alkaline scenarios, posing a substantive challenge in more extensive applications. Herein, we propose an ultraviolet (UV)-mediated enzyme-like activity reversal strategy to regulate Fe3O4/CDs nanozyme, greatly recovering the enzyme-like activity of nanozymes in neutral/weakly alkaline environments. Specifically, UV irradiation of the nan

Neohesperidin and neohesperidin dihydrochalcone alleviated benzo[a]pyrene-induced toxicity: By inhibiting the activity of CYP1A1 enzyme.

Benzo(a)pyrene (BaP), a widespread environmental and food pollutant, induces cellular toxicity through CYP1A1-mediated metabolic activation. This study demonstrated that neohesperidin (NH) and neohesperidin dihydrochalcone (NHDC) effectively inhibited CYP1A1 activity. The recombinant CYP1A1 protein expressed in E. coli confirmed these inhibitory effects in vitro. Using a HepG2 cell model, both NH and NHDC significantly alleviated BaP-induced cytotoxicity and oxidative stress, as evidenced by enhanced cell viability and upregulated HO-1, NQO-1, and Nrf-2 expression. Comet assay results further

Electro-activated nanozyme for in situ tumor vaccination: Synergistic multi-enzyme catalysis and electrodynamic therapy.

Oral squamous cell carcinoma (OSCC) is a highly aggressive malignancy with poor prognosis and limited treatment options. To address the challenges of conventional therapy resistance, antioxidant defence, and immune evasion, we developed a multifunctional nanozyme platform, MIL-100(Fe)@Pt@R837@HA (HMPR), that integrates electrodynamic therapy (EDT), chemodynamic therapy, and immunotherapy. The multi-enzyme activities of HMPR allow for the oxygen generation, hydroxyl radicals production, and glutathione depletion, thereby alleviating hypoxia and triggering both apoptosis and ferroptosis. IQGAP3 deficiency leads to non-syndromic hearing loss via inhibition of CDC42 enzyme activity.

BACKGROUND: Genetic factors are important causes of congenital hearing loss. To better understand hereditary hearing loss, we performed in-depth clinical and molecular analysis of families with congenital hearing loss and a new disease-related gene, IQGAP3, was identified in this process. This gene encodes a protein that belongs to the IQGAP family which is well known as a GTPase-activating protein involved in various cellular functions. However, there is no research on the relationship between IQGAP3 and the auditory system. METHOD: This study was conducted at Guangzhou Women and Children's

Activity-based protein profiling reveals both canonical and novel ubiquitin pathway enzymes in Plasmodium.

The ubiquitin-proteasome system (UPS) is essential for Plasmodium falciparum survival and represents a potential target for antimalarial therapies. We utilised a ubiquitin- activity based probe (Ub-Dha) to capture active components of the ubiquitin conjugating machinery during asexual blood-stage development. Several E2 ubiquitin-conjugating enzymes, the E1 activating enzyme, and the HECT E3 ligase PfHEUL were identified and validated through in vitro ubiquitination assays. We also demonstrate selective functional interactions between PfHEUL and a subset of both human and P. falciparum E2s. Ad

Evaluation of Insecticide Resistance and Associated Enzyme Activity in Field Populations of Diamondback Moth (Plutella xylostella).

The diamondback moth, Plutella xylostella (L.), is a major pest of cruciferous crops with a remarkable capacity to develop insecticide resistance. The present study evaluated the susceptibility, resistance ratios, and detoxification mechanisms of field populations collected from Andhra Pradesh, India, against tolfenpyrad 15 EC and emamectin benzoate 5 SG. Laboratory selection studies revealed declining LC50 values over successive generations reared without insecticide pressure, indicating partial recovery of susceptibility. Field bioassays showed resistance ratios ranging from 9.8 to 13.1 for
