Stage-specific disruption of host resistance and metabolic pathways following developmental PFAS exposure in marine medaka.

Per- and polyfluoroalkyl substances (PFAS) are persistent contaminants with immunotoxic potential, yet impacts on immune development remain poorly defined. We examined effects of developmental PFAS exposure on host resistance and metabolism in marine medaka (Oryzias melastigma). Larvae were exposed to legacy (PFOA, PFOS) and replacement PFAS (PFHxA, PFHxS, GenX, PFBS) at 0.5 or 5 ng/mL. Exposures occurred during two immune-development windows: 7-11 days post fertilization (dpf) and 3-5 days post hatching (dph). Host resistance was measured as survival following Edwardsiella piscicida infection

High-performance aptapipette sensor for ultrasensitive and specific detection of perfluorooctanoic acid in environmental water samples.

Perfluorooctanoic acid (PFOA) is a synthetic perfluorinated compound widely used in industrial and consumer products. It exhibits strong resistance to natural degradation due to its stable structure, leading to long-term environmental persistence. It poses potential carcinogenic and reproductive toxic risks, prompting global regulatory restrictions and creating an urgent need for highly sensitive detection methods. Here, we developed a PFOA detection technology based on an aptamer-functionalized nanopipette, which is referred to as an aptapipette. Glass nanopipettes were fabricated, infused wi

Perfluorooctanesulfonate (PFOS) exposure in ringed seals (Pusa saimensis) in Lake Saimaa, Finland.

Perfluorooctanesulfonate (PFOS) is common in the environment, highly accumulative, toxic, and thus a good indicator for per- and polyfluorinated alkyl acid (PFAS) contamination. In this study, PFOS was analysed from the tissues of Saimaa ringed seal (Pusa saimensis) pups and adults to investigate the exposure to PFAS in Lake Saimaa. Additionally, a toxicity evaluation was performed for the seals based on the thresholds from the literature, PFOS concentration data from the national exposure database, and the current study. For all studied specimens, the highest PFOS concentrations were found in

A Non-Contact Colorimetric Sensor Based on Bromocresol Green-Nafion Composite for Rapid Pre-Screening of Urinary Tract Infections.

Urinary tract infections (UTIs) are among the most prevalent bacterial infections and require timely detection to prevent complications such as acute kidney injury and bloodstream infections. Conventional diagnostic methods, such as bacterial culture, are time-consuming and resource-intensive, necessitating more efficient alternatives. To address this, a noncontact colorimetric sensor is developed for rapid UTI prescreening based on volatile organic compounds (VOCs) emitted from Escherichia coli (E. coli) - contaminated urine. This system employs a bromocresol green-Nafion composite for the se

Hepatotoxicity of 8:2 Polyfluoroalkyl Phosphate Diesters Exposure via JAK2/STAT3 Pathway Activation and M1 Macrophage Polarization in Mice.

Polyfluoroalkyl phosphate esters (PAPs) are widely used as substitutes for legacy perfluoroalkyl and polyfluoroalkyl substances (PFAS), contributing a non-negligible environmental burden. However, their in vivo hepatotoxicity and underlying mechanisms remain poorly understood. We developed a mouse model to assess the hepatotoxic effects and mechanisms of 8:2 polyfluoroalkyl phosphate diesters (8:2 diPAP). Exposure to 8:2 diPAP at doses of 0.5, 5, and 50 mg/kg resulted in hepatocyte damage and elevated serum levels of total bilirubin. Albumin, alanine transaminase, and aspartate aminotransferas

The biological effects of bisphenol AF in reproduction and development: What do we know so far?

Due to the established endocrine-disrupting effects of Bisphenol A (BPA), alternative bisphenols entered the market. Bisphenol AF (BPAF) is now commonly used in the industrial manufacturing of polycarbonate plastics and epoxy resins. However, BPAF's effects on reproduction and development have not been thoroughly reviewed. We investigated the relationship between BPAF exposure and reproduction and early development. We performed a literature review of studies on BPAF and reproductive physiology. Using keywords, we searched PubMed, Medline, Cochrane Library Database, Embase, and ClinicalTrials.

High-performance formaldehyde electrochemical sensor utilizing Nafion as solid electrolyte for human exhaled breath detection.

Formaldehyde (HCHO), as a representative biomarker associated with lung cancer screening, plays a key role in distinguishing healthy individuals from those with diseases through its ppb-level concentration differences. Therefore, achieving high sensitivity and low limit of detection (LOD) for specific detection of HCHO is critically important. In this work, a HCHO electrochemical sensor was prepared using a two-step drop-coating method on a flexible planar screen-printed electrode, with 60 % Pt/C-Nafion catalyst as the sensing layer and Nafion as the solid-state electrolyte. Associations between per- and polyfluoroalkyl substances and reproductive outcomes among women undergoing in vitro fertilization/intracytoplasmic sperm injection treatment.

Exposure to per- and poly-fluoroalkyl substances (PFAS) has been reported to impair fertility in experimental models. However, studies on female reproductive outcomes are limited to a few legacy PFAS. This study linked exposure to various PFAS with reproductive outcomes in women undergoing IVF/ICSI. A total of 275 women were recruited, and plasma concentrations of 17 PFAS were quantified. Based on analysis using the adjusted generalized linear models, an increase in the quartile of 6:2Cl-PFESA was associated with a decrease in the number of 2PN zygotes [9.33, 95 %CI: (3.63, 15.03)] and 2PN cle

The association between per- and polyfluoroalkyl substances and biological aging among middle and older age adults: The moderating effect of caffeine.

Perfluoroalkyl and polyfluoroalkyl substances (PFAS) are pervasive environmental pollutants associated with aging-related diseases and mortality risks, yet their relationship with biological aging remains underexplored. Notably, dietary antioxidants like caffeine have been shown to alleviate oxidative stress and inflammation, and these pathways are closely related to PFAS-induced toxicity and aging processes. However, whether dietary caffeine intake could mitigate the adverse effect of PFAS on biological aging is still unknown. This study aims to investigate the association between PFAS exposu

A simple and low-cost electrode based on Nafion-stabilized silver nanoparticles supported on FTO for the electrochemical determination of Pb (II) and Cu (II).

Increasing awareness of the environmental risks posed by heavy metal accumulation in the environment-due to their toxicity and persistence in biological systems-has driven the development of more efficient and accessible detection methods. Conventional techniques, despite their accuracy, are often expensive, time-consuming, and reliant on non-portable specialized equipment. This study presents a novel, low-cost electrochemical sensor using a fluorine-doped tin oxide (FTO) electrode modified with Nafion-stabilized silver nanoparticles (AgNPs) for the rapid and accurate detection of Pb (II) and

Birth weight in relation to maternal and neonatal biomarker concentration of perfluorooctane sulfonic acid: a meta-analysis and meta-regression from a systematic review.

BACKGROUND: Perfluorooctane sulfonic acid (PFOS) is a legacy chemical, that while banned in some countries, is still found in various environmental media and in nearly all humans given its long half-life. OBJECTIVE: We examined mean birth weight (BW) differences in relation to PFOS exposure biomarkers using systematic review methods. METHODS: We fit a random effects model to obtain the overall pooled effect and for stratified analyses examining biomarker sample type and timing, study confidence, scaling factors, and country of study origin. We also conducted a meta-regression to assess the i

Perfluorooctanoic acid at environmentally relevant concentrations interfere with nitrogen fixation and amino acid synthesis in soil-soybean system.

Perfluorooctanoic acid (PFOA) contamination in soils is of concern, stemming from its detrimental impact on ecosystem functions and human health at high levels. However, considering its actual pollution levels in cultivated soil, it is necessary to comprehensively evaluate its biological effects under environmental concentrations. Results on the effect of PFOA at levels of 10 and 20 μg/kg on the entire life cycle of soybeans (Glycine max L.) showed increased accumulation in the order of root > leaf > stem > pod > bean, ranging from 0.5 to 155.4 μg/kg. PFOA accumulated in beans (0.5-1.1 μg/kg)

Perfluorodecanoic acid (PFDA) increases oxidative stress through inhibition of mitochondrial β-oxidation.

Per- and polyfluoroalkyl substances (PFAS) are a large group of synthetic organic chemicals that are ubiquitous environmental pollutants. Among PFAS, perfluorodecanoic acid (PFDA) is one of the most toxic compounds, but the molecular basis behind its toxicity is not fully understood. In an interspecies comparison with placental cells (HTR-8/SVneo) and zebrafish embryos, we demonstrate that PFDA induces mitochondrial dysfunction and impairs fatty acid β-oxidation. Reduced β-oxidation leads to less TCA cycle activity, resulting in less NADH and consequently NADPH production. Thereby NADPH-depend
