The C. elegans homolog of the murine cystic kidney disease gene Tg737 functions in a ciliogenic pathway and is disrupted in osm-5 mutant worms.

Cilia and flagella are important organelles involved in diverse functions such as fluid and cell movement, sensory perception and embryonic patterning. They are devoid of protein synthesis, thus their formation and maintenance requires the movement of protein complexes from the cytoplasm into the cilium and flagellum axoneme by intraflagellar transport (IFT), a conserved process common to all ciliated or flagellated eukaryotic cells. We report that mutations in the Caenorhabditis elegans gene Y41g9a.1 are responsible for the ciliary defects in osm-5 mutant worms. This was confirmed by transgen

Molecular characterization of the prevalent soil-transmitted helminths in Narathiwat Province, southern Thailand.

Soil-transmitted helminths (STHs) are prevalent in tropical regions, including southern Thailand. However, limited data on their abundance in this area and molecular characteristics exist. This study evaluated 210 fecal specimens from the Narathiwat Province to determine the molecular profiles of STHs. Microscopic examination revealed that the predominant STHs were Trichuris spp. and Ascaris spp., with prevalences of 14.76% and 6.67%, respectively. Based on ITS1 sequencing, two haplotypes of Ascaris spp. were identified, classified as A. lumbricoides and A. suum. ITS1-based phylogeny revealed

Host-parasite cross-species regulation by Schistosoma japonicum EV-miRNA sja-miR-2a-3p limits hepatic fibrosis via BCL2 suppression in hepatic stellate cells.

Schistosomiasis remains an important zoonotic parasitic disease, and hepatic fibrosis is a major cause of chronic pathology following Schistosoma japonicum infection. In schistosomiasis, hepatic fibrosis is the major cause of severe pathological damage. However, during the long-term co-evolution and coexistence between Schistosoma parasites and their hosts, the parasite can modulate the host hepatic fibrotic process through extracellular vesicle (EV)-mediated cross-species communication. In this study, we identified an EV-derived microRNA from Schistosoma japonicum, sja-miR-2a-3p, that suppres

Eucalyptus oil: A promising anticoccidial agent with multifaceted protective effects.

In-feed preventive coccidiostat additives are regarded as the primary choice of coccidiosis control, whereas the lasting appearance of drug resistance seriously hampered its application. Eucalyptus oil (EUC) has been evidenced to possess anti-malaria and anti-helminth efficacy. Confronting the urgent requirements for novel anticoccidial remedies, EUC was picked to scrutinize its anticoccidial efficacy with the in vivo coccidiosis model. Birds were orally administrated with 8 × 104 sporulated oocysts and treated with 20 mg/kg EUC in feed during the whole experimental period, diclazuril (DIC) wa

Helminths infection of Schizothorax niger in Kashmir, India: morphological and molecular characterization.

BACKGROUND: The identification of helminth parasites in Schizothorax spp. from Kashmir, including Schyzocotyle acheilognathi, Pomphorhynchus kashmirensis, and Adenoscolex oreini, is hindered by morphological limitations and high intraspecific variation. While previous studies have relied on morphological diagnosis, a comprehensive molecular characterization is lacking. To address these limitations and achieve more accurate species identification, this study employs molecular techniques such as DNA sequencing and phylogenetic analysis. METHOD: Genomic DNA was extracted using standardized proto

Brugia malayi miRNAs and potential targets within the feline host (Felis catus).

Host specificity is a critical feature of the survival and proliferation of parasites. In the context of interactions with the host, numerous mechanisms have been identified, particularly in parasitic helminths, that enable manipulation of the host immune system to enhance their own survival. The evolutionary history of these interactions often results in hosts becoming disease-tolerant or asymptomatic, even when burdened with a high number of worms, until a disruption in the host's immune system can trigger a disease state. However, the molecular mechanisms underlying host specificity and the

Morphological and molecular identification of cardiopulmonary nematodes in the European wildcat (Felis silvestris) and nine other carnivores in Spain.

Cardiopulmonary nematodes (CPN) are among the most pathogenic helminths, with wild carnivores serving as hosts for a variety of species. Knowledge of their presence in the Iberian Peninsula is limited, and for certain hosts, such as the European wildcat (Felis silvestris), no data are currently available. Lung and heart samples from 112 road-killed individuals belonging to 10 different species, primarily wildcats (n = 33), collected in Navarra (northern Spain) were collected. Morphological identification of nematodes was done and confirmed, when possible, by sequencing fragments of the cytochr

Neglected zoonotic helminthiases in wild canids: new insights from South America.

The global threat of neglected tropical diseases (NTDs) constitutes a public health issue in underdeveloped countries. Zoonotic helminthiases are the most common human NTD agents in developing countries in sub-Saharan Africa, Asia, and the Americas, causing a global burden of disease that exceeds that of more recognized infectious diseases such as malaria and tuberculosis. Wild canids are well-known mammals that act as natural reservoirs of zoonotic-relevant helminthiasis worldwide, thus playing a pivotal role in their epidemiology and transmission to humans. Here we evaluate the occurrence of

Molecular characterization of a novel GSTO2 of Fasciola hepatica and its roles in modulating murine macrophages.

Fascioliasis is an important zoonotic helminthic disease caused by Fasciola hepatica and poses a serious threat to global public health. To evade the immune response of its host (humans or animals), F. hepatica secretes various antioxidant enzymes such as glutathione transferase (GST) to facilitate its invasion, migration and parasitism in vivo. To investigate the biological functions of a novel omega-class GST (GSTO), the molecular features of GSTO2 of F. hepatica were analyzed by online software, and the biochemical properties in vitro of recombinant GSTO2 (rGSTO2) were dissected. Then, the

Dynamics of the bacterial gut microbiota during controlled human infection with Necator americanus larvae.

Hookworms are soil-transmitted helminths that use immune-evasive strategies to persist in the human duodenum where they are responsible for anemia and protein loss. Given their location and immune regulatory effects, hookworms likely impact the bacterial microbiota. However, microbiota studies struggle to deconvolute the effect of hookworms from confounders such as coinfections and malnutrition. We thus used an experimental human hookworm infection model to explore temporal changes in the gut microbiota before and during hookworm infection. Volunteers were dermally exposed to cumulative dosage

TRP channels as potential targets for antischistosomals.

Ion channels are membrane protein complexes that underlie electrical excitability in cells, allowing ions to diffuse through cell membranes in a regulated fashion. They are essential for normal functioning of the neuromusculature and other tissues. Ion channels are also validated targets for many current anthelmintics, yet the properties of only a small subset of ion channels in parasitic helminths have been explored in any detail. Transient receptor potential (TRP) channels comprise a widely diverse superfamily of ion channels with important roles in sensory signaling, regulation of ion homeo

Fat-associated lymphoid cluster in Cyprinus carpio: Characterisation and its relation with peritoneal haemangiosarcoma.

FALC cells are natural helper cells producing Th2-type cytokines, which express c-kit, Sca-1, IL7R and CD45 in mouse and human. These cells are involved in allergic responses and contribute to the inflammatory reactions of adipose tissue; however, a lack of information prevails about the presence of these cells in other species. The aim of the study was to identify and characterise FALC cells in the common carp (Cyprinus carpio) using immunohistochemistry and molecular biology techniques as well as to explore their relationships with their microenvironment. Histological description of the FALC

Critical role of Trib1 in differentiation of tissue-resident M2-like macrophages.

Macrophages consist of at least two subgroups, M1 and M2 (refs 1-3). Whereas M1 macrophages are proinflammatory and have a central role in host defence against bacterial and viral infections, M2 macrophages are associated with responses to anti-inflammatory reactions, helminth infection, tissue remodelling, fibrosis and tumour progression. Trib1 is an adaptor protein involved in protein degradation by interacting with COP1 ubiquitin ligase. Genome-wide association studies in humans have implicated TRIB1 in lipid metabolism. Here we show that Trib1 is critical for the differentiation of F4/80(+

Congenital optic disc cyst concomitant with persistent hyaloid artery--a case report.

The aim is to present a rare case of solitary malformation in the form of a congenital optic disc cyst concomitant with the persistent hyaloid artery. The intrabulbar congenital cyst of the optic disc partially covering the medial part of the disc was found in a 3-month old infant. B-San ultrasound confirmed the presence of the intrabulbar heterogeneous mass (7.0 x 2.5 x 5.4 mm) within the vitreous cavity and the concomitant persistent hyaloid artery was shown in Colour Doppler Imaging. The axial length of the involved eye was shorter than of the healthy one (16.68 mm vs. 18.42 mm). The magnet

Alterations in the porcine colon microbiota induced by the gastrointestinal nematode Trichuris suis.

Helminth parasites ensure their survival by regulating host immunity through mechanisms that dampen inflammation. These properties have recently been exploited therapeutically to treat human diseases. The biocomplexity of the intestinal lumen suggests that interactions between the parasite and the intestinal microbiota would also influence inflammation. In this study, we characterized the microbiota in the porcine proximal colon in response to Trichuris suis (whipworm) infection using 16S rRNA gene-based and whole-genome shotgun (WGS) sequencing. A 21-day T. suis infection in four pigs induced

Human helminth co-infection: no evidence of common genetic control of hookworm and Schistosoma mansoni infection intensity in a Brazilian community.

Strong statistical associations between soil-transmitted helminths and schistosomes are frequently observed in co-endemic human populations, although the underlying explanations remain poorly understood. This study investigates the contribution of host genetics and domestic environment to hookworm and Schistosoma mansoni infection intensity and evaluates the role of genetic and non-genetic factors in co-variation of infection intensity. Detailed genealogical information allowed assignment of 1303 individuals living in the Brazilian community of Americaninhas, Minas Gerais state, to 25 pedigree

Double-stranded RNAs from the helminth parasite Schistosoma activate TLR3 in dendritic cells.

Stimulation of dendritic cells (DCs) by the egg stage of the helminth parasite Schistosoma mansoni activates a signaling pathway resulting in type I interferon (IFN) and IFN-stimulated gene (ISG) expression. Here, we demonstrate that S. mansoni eggs disjointedly activate myeloid differentiation factor 88 (MyD88)-dependent and MyD88-independent pathways in DCs. Inflammatory cytokine expression and NF-kappa B activation in DCs from MyD88-deficient mice were impaired, whereas signaling transducer activator of transcription (STAT) 1(Tyr701) phosphorylation and ISG expression were intact in MyD88 o

CD28-deficient mice generate an impaired Th2 response to Schistosoma mansoni infection.

Engagement of CD28 on T cells provides a co-stimulatory signal necessary for T cell activation and differentiation. Recent findings suggest that priming of T helper (Th)2 cells is more dependent on CD28 activation that Th1 cells. The present study examines whether mice that lack expression of CD28 as a result of gene targeting are capable of generating a Th2 response characteristic during infection with the intravascular trematode parasite Schistosoma mansoni. Mutant and control mice were either inoculated in the footpad with S. mansoni eggs (a potent inducer of a Th2 response) or infected per

Haematological and biochemical profiling combined with HRAMS-based metabolomics reveals the potential biomarkers of Wuchereria bancrofti asymptomatic infections: towards early interventions and diagnostics.

Lymphatic filariasis (LF) encompasses grotesque and irreversible chronic conditions such as lymphedema, hydrocele, and elephantiasis. Asymptomatic or subclinical infections remain largely unnoticed as microscopy, serological tests, and antigen detection typically lack sufficient sensitivity to detect circulating microfilariae. Herein, we utilised high-resolution accurate mass spectrometry (HRAMS)-based metabolomics to thoroughly examine host metabolic responses in 25 patients with active microfilaremic infections compared with 30 healthy controls. Metabolites were annotated using the Compound
