Co-occurrence of triple ESBL-associated genes (blaCTX-M, blaTEM and blaSHV) in Gram-negative bacteria from clinical specimens at a provincial hospital in Far-Western Nepal.

BACKGROUND: Antimicrobial resistance (AMR) among Gram-negative bacteria, particularly those producing extended-spectrum β-lactamase (ESBLs), represents a significant global healthcare challenge. CTX-M enzymes have achieved global dominance, often disseminated through plasmid-mediated horizontal gene transfer, complicating treatment in resource-limited settings. This study aimed to determine prevalence, antimicrobial resistance patterns and ESBL-associated gene profiles in clinical Gram-negative isolates. METHODS: A hospital-based cross-sectional study was conducted at Mahakali Provincial Hosp

Characterization of Phylogroups, Virulence Factors and Antimicrobial Resistance in Canine Pyometra-associated Escherichia coli.

The presence of multidrug-resistant extraintestinal pathogenic Escherichia coli (ExPEC) in pets poses a significant threat to animal health and represents a potential source of antimicrobial resistance with zoonotic transmission to humans. A total of 56 pyometra swab samples from dogs were analysed for the presence of E. coli using standard microbiological procedures. Phylogrouping of the E. coli isolates was performed using the quadruplex, and 12 virulence genes were detected by single polymerase chain reaction (PCR) method. Antimicrobial susceptibility testing was conducted against 12 antimi

Tuberous sclerosis associated with MDR1 gene expression and drug-resistant epilepsy.

Intractable seizures are the most common manifestation in severe cases of tuberous sclerosis. Multidrug resistance type 1 (MDR1) gene expression is directly linked to the resistance of tumor cells to chemotherapy as the major cause of treatment failure, but it has not been reported in tuberous sclerosis cells nor has the relationship between the MDR1 gene and antiepileptic drugs been described. A 4-month-old female is described with poorly controlled seizures secondary to tuberous sclerosis. The patient was treated with antiepileptic drugs, including phenytoin, phenobarbital, and lorazepam, wi

Potentiation of pirarubicin activity in multidrug resistant cells by rifampicin.

The effect of the anti-tuberculosis drug rifampicin on pirarubicin activity was investigated in multidrug-resistant cells overexpressing P-glycoprotein. Rifampicin increased the sensitivity of pirarubicin to anthracycline-resistant mouse leukemic P388 cells and significantly enhanced the cytotoxicity and intracellular accumulation of pirarubicin in resistant cells, but had no effect in parent cells. By contrast, two other rifamycins, rifamycin B and SV, had no effect on pirarubicin accumulation in resistant cells. Rifampicin also enhanced pirarubicin-induced apoptosis and G2/M blockade on the

Antibiotic Treatment Administered to Pigs and Antibiotic Resistance of Escherichia coli Isolated from Their Feces and Carcasses.

Antimicrobial resistance (AMR) in bacteria is a frequent and widespread phenomenon. The European Food Safety Authority (EFSA) reports that multidrug resistant (MDR) Escherichia coli is considered an important hazard to public health. The lack of data on the correlation between the administration of antibiotics to pigs and the diffusion of MDR E. coli necessitates an in-depth study. The aims of our study were first of all to determine the presence of MDR and/or extended spectrum β-lactamase (ESβL) E. coli isolated from feces and carcasses of pigs; and second, to evaluate the correlation between

Two new Salmonella genomic islands 1 from Proteus mirabilis and description of blaCTX-M-15 on a variant (SGI1-K7).

OBJECTIVES: To characterize the structure of Salmonella genomic islands 1 (SGI1s) from two clinical Proteus mirabilis isolates: one producing an ESBL and the other a penicillinase. METHODS: WGS completed by PCR and Sanger sequencing was performed to determine sequences of SGI1s from Pm2CHAMA and Pm37THOMI strains. RESULTS: Two new variants of SGI1 named SGI1-Pm2CHAMA (53.6 kb) and SGI1-K7 (55.1 kb) were identified. The backbone of SGI1-Pm2CHAMA shared 99.9% identity with that of SGI1. Its MDR region (26.3 kb) harboured two class 1 integrons (an In2-type integron and an In4-type integron) con

Resistance to the antiproliferative effect induced by a short-chain ceramide is associated with an increase of glucosylceramide synthase, P-glycoprotein, and multidrug-resistance gene-1 in cervical cancer cells.

PURPOSE: Ceramide is glycosylated to glucosylceramide or lactosylceramide, and this glycosylation is a novel multidrug-resistance (MDR) mechanism. In this work, a short-chain ceramide (C6), lactosylceramide (LacCer), and an inhibitor of ceramide glycosylation (D-threo-1-phenyl-2-decanoylamino-3-1-propanol, PDMP) were evaluated on the proliferation of cervical cancer cells. The participation of glucosylceramide synthase (GCS), P-glycoprotein (P-gp), and multidrug-resistance gene-1 (MDR-1) in the resistance to the antiproliferative effect induced by C6 was also evaluated. METHODS: Cell prolifer

Detection of polymyxin-resistant Enterobacteriaceae from poultry farms in Brazil: continued mcr gene dissemination.

The emergence and persistence of plasmid-mediated polymyxin resistance in Brazilian poultry production pose a significant One Health challenge. Here, cloacal swabs from 202 broilers across four farms in the State of Rio de Janeiro yielded 125 Enterobacteriaceae isolates growing on polymyxin-EMB agar. Escherichia coli accounted for 99% of resistant isolates, with one Klebsiella pneumoniae. Multidrug resistance (MDR) was observed in 75% of polymyxin-resistant strains. PCR screening revealed mcr-1 and mcr-5 genes. Conjugation assays demonstrated horizontal transfer of mcr-1 plasmids (48.5–194 kb)

Antibiogram of isolated bacteria and overview of resistome and virulome of multi-drug resistant Escherichia coli from ready-to-eat foods vended in Lagos, Nigeria.

BACKGROUND: Ready-to-eat (RTE) foods are popular fast-foods due to their convenience and affordability. However, they pose huge public health risks due to the presence of antibiotic-resistant and virulent pathogens. Nevertheless, such data is sparse in Nigeria where RTE food consumption is widespread. This study determined antibiotic susceptibility patterns, and identified antibiotic resistance and virulence genes in bacteria from 199 RTE food samples (aadun, biscuits, bread, eko, kokoro and shawarma) vended within Lagos State, Nigeria. Bacteria were recovered on nutrient agar and MacConkey ag

Fluoroquinolone resistant Escherichia coli in community: molecular analyses of resistance genes, virulence factors, and antimicrobial susceptibility test.

INTRODUCTION: Most urinary tract infections in all populations are caused by gram -negative, facultative anaerobic, uropathogenic E. coli (UPEC). Multidrug resistance (MDR) to first line antibiotics recommended for treatment of UTI is a global health problem. The misuse of antimicrobial agents for UTI treatment can lead to selection of bacterial MDR, able to transfer mechanism of resistance through mobile genetic elements (MGEs), such as transposons, integrons and conjugative plasmids. This study aims to characterize MLST, virulence and resistance genes to quinolones and β-lactams of UPEC samp

Antimicrobial resistance profiles of salmonella spp. and escherichia coli isolated from fresh nile tilapia (oreochromis niloticus) fish marketed for human consumption.

BACKGROUND: Salmonella spp. and pathogenic strains of Escherichia coli are among the major foodborne zoonotic pathogens. These bacterial pathogens cause human illnesses characterized by hemorrhagic colitis, vomiting, nausea, and other agent-related symptoms. The increasing occurrence of antimicrobial resistance in these pathogens is also a serious public health concern globally. Regular surveillance of phenotypes and genotypes of Salmonella spp. and Escherichia coli from animal-derived foods is necessary for effective reduction and control of these foodborne pathogens. This study was conducted

First report of MDR virulent Pseudomonas aeruginosa in apparently healthy Japanese quail (Coturnix japonica) in Bangladesh.

Pseudomonas aeruginosa (P. aeruginosa) is a major pathogen associated conditions like septicaemia, respiratory disorders, and diarrhoea in poultry, particularly in Japanese quail (Coturnix japonica). The infection causes huge economical losses due to its high transmissibility, mortality and zoonotic potential. This study aimed to isolate, identify, detect virulence genes, and profile multidrug resistance (MDR) of P. aeruginosa from Japanese quail. Oral and rectal swabs were collected from 110 apparently healthy quail birds across various districts in Bangladesh. Bacterial isolation and identif

Prevalence and antimicrobial resistance characterization of multidrug-resistant Staphylococcus epidermidis isolated from raw milk of dairy cattle and ewes.

Milk is a vital and widely consumed food, but contamination by biological, chemical, and physical factors can lead to milk-borne diseases. Mastitis, particularly subclinical mastitis (SCM), is a significant biological factor that deteriorates milk quality. Among the 135 agents causing SCM, Staphylococcus epidermidis, a Gram-positive coagulase-negative staphylococcus, plays a notable role. The excessive and indiscriminate use of antibiotics in treating mastitis has led to the emergence of multidrug-resistant (MDR) strains, posing threats to both animal and human health. This study aimed to asse

Houseflies (Musca domestica) as vectors of multidrug-resistant, ESBL-producing Escherichia coli in broiler poultry farms of North India: implications for antibiotic resistance transmission.

The transmission of antibiotic resistance (AR) from farm animals to healthy human communities, beyond the food chain, is often facilitated by biological vectors, notably houseflies (Musca domestica). This study aimed to evaluate the role of M. domestica collected from commercial broiler chicken farms as a carrier of multidrug-resistant (MDR), extended-spectrum β-lactamase (ESBL)-producing Escherichia coli. E. coli were isolated separately from the housefly's external surface (ES) and internal homogenate (IH) to determine the primary AR transmission route within houseflies. Remarkably, 68.6% ho

Genome-Based Molecular Diversity of Extended-Spectrum β-Lactamase-Producing Escherichia coli From Pigeons in China.

Extended-spectrum β-lactamase-producing Escherichia coli (ESBL-EC) strains present a significant menace to the well-being of both animals and humans. However, limited information is available regarding their profiles in pigeons. Using a combination of whole genome sequencing, drug susceptibility testing, and bioinformatics analysis, we examined the genomic features and epidemiology of 95 ESBL-EC strains (41 racing and 54 meat pigeons) that were isolated from 11 Chinese cities. These strains belonged to seven phylogenetic groups (A, B1, B2, C, D, E, and F). Moreover, these isolates have 51 sero

Antimicrobial Resistance and Extended-Spectrum Beta-Lactamase (ESBL) Genes in E. coli Isolated from Equine Fecal Samples in Turkey.

The extensive use of antibacterial agents used for treatment in human and veterinary clinics to increase yield in livestock and aquaculture in developing countries causes the detection and spread of multidrug-resistant (MDR) strains in different sources. The presence of Escherichia coli strains is very common in racehorses and it's a serious problem on horse farms. Extended-spectrum beta-lactamase (ESBL) production is an important source of MDR development in Gram-negative strains such as E.coli. This study aimed to detect the presence of ESBL genes in E. coli strains isolated from horse farms
