Mechanism of the toxic effects of tetracycline on blood meal digestion in Haemaphysalis longicornis.

The extensive utilization of antibiotics in the field of animal husbandry gives rise to various concerns pertaining to the environment and human health. Here, we demonstrate that the administration of tetracycline impedes blood meal digestion in the tick Haemaphysalis longicornis. Tissue sectioning, 16S rRNA high-throughput sequencing, and transcriptome sequencing of the midgut were employed to elucidate the mechanism underlying tetracycline toxicity. The treatment group consisted of engorged female ticks that were subjected to tetracycline microinjections (75 µg per tick), whereas the control

[Epilepsy genetics and genetic epilepsies].

During the last two decades, the molecular revolution in medicine has had a strong impact in the field of epilepsies. The quest for epilepsy-genes has been focused mainly on large mendelian pedigrees. This approach has allowed the identification of new causative genes and has provided new information about the pathogenesis of many epilepsy syndromes. Neverthless, the clinical implication of this information still has a relatively little impact on the genetic counselling for most of the syndromes.

[Ser49gly polymorphism as predictor of development of hereditary sick sinus node syndrome].

We demonstrated for the first time on clinico-genetic material an association of hereditary sick sinus node syndrome (SSNS) with polymorphism of beta-adrenorecetor gene. We found that heterozygous variant of Ser49gly of beta-adrenoreceptor gene was significantly more often met in patients with SSNS and their healthy relatives than in subjects of control group. In the group of patients with SSNS contrary to control group we noted statistically significant preponderance of carriers of mutant Gly49 allele of.

[Hereditary hypermobility of the joints].

The authors applied clinico-genetic methods of analysis to work out the data concerning familial spread of hypermobility of the joints (567 inhabitants of Tbilisi, 302 children with congenital dislocation of the hip joint). A common genetic character of this pathology and congenital dislocation of the hip was established (88% of common genes). Approximate evaluations of relative contribution of the genetic component into the general dispersion (93%) and of the penetrance of the gene were obtained. A monogenous dominant type of inheritance was revealed.

Incidence of small-intestinal mucosal abnormalities and of clinical coeliac disease in the relatives of children with coeliac disease.

Evidence is presented of a higher than normal incidence both of clinical coeliac disease and of small-intestinal mucosal abnormalities in relatives of children with coeliac disease. In such relatives the incidence of mucosal abnormality may differ from the incidence of clinical coeliac disease. The data show an absence of any simple Mendelian pattern of inheritance: in place of the hypothesis that inheritance is through a dominant gene of reduced penetrance, it is argued that the pathogenesis of coeliac disease is multifactorial, the genetic basis of susceptibility being polygenic and interact

Dysregulation of myddosome complex genes its related to alendronate treatment failure in osteoporosis postmenopausal patients.

Some patients with osteoporosis (OP) do not respond to treatment with bisphosphonates; pathways that stimulate osteoclatogenesis may be involved in this failure, such as the myddosome pathway. A total of 40 OP patients and 20 controls were included in the group study. Patients treated with sodium alendronate (SA) for two years were classified according to bone mineral density (BMD) variations of the lumbar spine, femoral neck, and total hip, measured by the method of dual-energy x-ray absorptiometry (DXA) as responsive patients (OP-R) (n = 20) and non-responders (OP-NR) (n = 20), to evaluate t

Spinocerebellar Ataxia Type 23 (SCA23): A Rare Cause of SCA in the Americas.

Spinocerebellar ataxia type 23 (SCA23) is a rare autosomal dominant hereditary ataxia caused by a pathogenic variant in the PDYN gene. It usually presents in adulthood, with a mean age of onset around 43 ± 15 years (reported range: 10–73 years), and progresses slowly with cerebellar symptoms. We report a case of a Brazilian 25-year-old female patient whose symptoms began at 19 years of age, characterized by progressive dysarthria, tremor, dysphagia, and gait disturbance. She had no relatives with similar symptoms. The initial genetic ataxia panel, which included the most prevalent hereditary a

Case Report: diffuse entire gastrointestinal tract involvement of ALK-positive anaplastic large cell lymphoma harboring JAK-STAT pathway mutations in an adolescent with leukemoid reaction.

Anaplastic lymphoma kinase-positive anaplastic large cell lymphoma (ALK+ ALCL) is an aggressive mature T-cell non-Hodgkin lymphoma. Its typical characteristics include positive CD30 and ALK expression detected by immunohistochemistry, and it is often accompanied by ALK gene translocation, among which the chromosomal translocation t (2;5) is the most common. This disease predominantly affects young individuals, with extranodal involvement being relatively common. However, cases involving the digestive tract are relatively rare. We report a rare case of a 14-year-old female with ALK+ALCL present

Effects of composting on the fate of doxycycline, microbial community, and antibiotic resistance genes in swine manure and broiler manure.

Aerobic composting is an economical and effective technology that is widely used to treat animal manure. To study the fate of doxycycline (DOX), the microbial community, and antibiotic resistance genes (ARGs) during composting, aerobic composting of broiler manure and swine manure was carried out under natural environmental conditions. Aerobic composting effectively removed DOX (with a removal rate > 97%) and most ARGs from animal manure. The microbial diversity and the numbers of ARGs were higher in composted swine manure compared with composted broiler manure. The microbial community structu

Increased gene expression and copy number of mutated blaKPC lead to high-level ceftazidime/avibactam resistance in Klebsiella pneumoniae.

BACKGROUND: Resistance to ceftazidime-avibactam was reported, and it is important to investigate the mechanisms of ceftazidime/avibactam resistance in K. pneumoniae with mutations in blaKPC. RESULTS: We report the mutated blaKPC is not the only mechanism related to CZA resistance, and investigate the role of outer porin defects, efflux pump, and relative gene expression and copy number of blaKPC and ompk35/36. Four ceftazidime/avibactam-sensitive isolates detected wild type blaKPC-2, while 4 ceftazidime/avibactam-resistant isolates detected mutated blaKPC (blaKPC-51, blaKPC-52, and blaKPC-33)

Hydranencephaly in a newborn with a FLVCR2 mutation and prenatal exposure to cocaine.

BACKGROUND: Hydranencephaly is a relatively rare but severe structural brain abnormality that often results in perinatal death. Although several factors including infection and multiple births have been reported to be associated with this birth defect, the underlying etiology is not well understood. Recently, FLVCR2 gene mutations have been implicated in a subset of hydranencephaly cases, following an autosomal recessive pattern of inheritance. CASE: We report a male infant with hydranencephaly found to have a previously unreported six amino acid deletion in one copy of the FLVCR2 gene follow

Clinical and genetic investigation of a Japanese family with cardiac fabry disease. Identification of a novel α-galactosidase A missense mutation (G195V).

Fabry disease is an X-linked lysosomal storage disorder caused by mutations of the α-galactosidase A gene (GLA), and the disease is a relatively prevalent cause of left ventricular hypertrophy mimicking idiopathic hypertrophic cardiomyopathy. We assessed clinically 5 patients of a three-generation family and also searched for GLA mutations in 10 family members. The proband had left ventricular hypertrophy with localized thinning in the basal posterior wall and late gadolinium enhancement (LGE) in the near-circumferential wall in cardiovascular magnetic resonance images and her sister had vasos

Neurofibromatosis type 2 attributable to gonosomal mosaicism in a clinically normal mother, and identification of seven novel mutations in the NF2 gene.

Neurofibromatosis type 2 (NF2) is an autosomal dominant cancer syndrome that predisposes to the development of bilateral vestibular schwannomas sometimes associated with schwannomas at other locations, meningiomas, ependymomas and juvenile posterior subcapsular lenticular opacities. This disease is caused by inactivating mutations in the NF2 tumour-suppressor gene, located in 22q12. Recently, somatic mosaicism has been demonstrated in some "de novo" NF2 patients. We here report the genetic study of 33 NF2 patients from 33 unrelated Italian families. Twelve mutations were characterised, includi

[Dysplastic nevi and the risk of melanoma: a guideline for patient care. Nederlandse Melanoom Werkgroep van de Vereniging voor Integrale Kankercentra].

Consensus was recently reached in the Netherlands regarding the clinical management of dysplastic naevi and the definitions in clinical and pathological diagnostics. The term 'dysplastic' is reserved for histological diagnostics; the term preferred for clinical use is 'clinically atypical naevus'. A naevus is defined as clinically atypical if it meets three of the following five criteria: > or = 5 mm in diameter, vaguely bordered, asymmetrically shaped, irregularly pigmented and a red hue (erythema). Presence of clinically atypical naevi is a main risk factor for melanoma. Dysplastic naevus sy

Expression and interactions of the two closely related homeobox genes Phox2a and Phox2b during neurogenesis.

Recent evidence suggests that specific families of homeodomain transcription factors control the generation and survival of distinct neuronal types. We had previously characterized the homeobox gene Phox2a, which is expressed in differentiating neurons of the central and peripheral autonomic nervous system as well as in motor nuclei of the hindbrain. Targeted deletion of the Phox2a gene affects part of the structures in which it is expressed: the locus coeruleus, visceral sensory and parasympathetic ganglia and, as we show here, the nuclei of the IIIrd and IVth cranial nerves. We now report on

Comprehensive 16s rRNA sequencing and metabolomics to investigate the effect of anticancer bioactive peptides combined with oxaliplatin on gastric cancer.

The occurrence and development of gastric cancer are closely related to the gut microbiota. Previous studies have found that the combination of anticancer bioactive peptides (ACBP) and oxaliplatin (OXA) has a significant therapeutic effect on gastric cancer. However, the impact of ACBP-OXA on the gut microbiota remains unclear. In this study, we established a nude mouse model of ACBP-OXA for gastric cancer. Here, we set out to investigate the diversity of gut microbiota and fecal metabolomics and the correlation between gut microbiota and metabolites. We demonstrated that ACBP-OXA has a signif

Subtyping of primary aldosteronism with adrenal vein sampling: Hormone- and side-specific effects of cosyntropin and metoclopramide.

BACKGROUND: Cosyntropin and metoclopramide can affect the subtyping of primary aldosteronism when used with adrenal vein sampling by exerting hormone- and side-specific effects on cortisol and aldosterone secretion. We investigated how these stimuli affect the selectivity index, the relative aldosterone secretion index, and the lateralization index in consecutive primary aldosteronism patients submitted to adrenal vein sampling. METHODS: We recruited 171 patients; of these, 149 underwent adrenal vein sampling before and after stimulation with cosyntropin (250 µg intravenous bolus, n= 53, 73%
