Interindividual variability in static apnoea performance is partly explained by genetic factors.

A large interindividual variability exists in maximal apnoea duration and the physiological adjustments that occur during apnoea. Although a few genes have been found to be associated with some of the cardiovascular responses during apnoea, the extent to which genetic and environmental factors contribute to individual differences in apnoea performance has not been studied. Accordingly, 12 monozygotic and 10 dizygotic twin pairs (n = 44) performed a series of five maximal apnoeas with the face immersed in cold water to determine the heritability estimates for apnoea duration and the accompanyin

An update on adult forms of hereditary pheochromocytomas and paragangliomas.

PURPOSE OF REVIEW: Pheochromocytomas and paragangliomas (PPGL) display a strong genetic determinism with 40% of inherited forms. The purpose of this review is to provide an update on current knowledge on adult forms of hereditary PPGL and their management. RECENT FINDINGS: PPGL are genetically-driven in 70% of cases, with germline and/or somatic mutations identified in more than 20 genes. Although eight new susceptibility genes have recently emerged, mutations on SDHx genes remain the most frequent. In addition to SDHB, mutations in SLC25A11, FH and MDH2 may predispose to a metastatic disease…

Corpse dismemberment: A case series. Solving the puzzle through an integrated multidisciplinary approach.

The finding of corpse parts poses several challenges for the forensic pathologist presenting implications for identification, diagnosis of death and determination of wounds vitality. Further interpretative difficulties in cases of cadaveric dismemberment derive from the scarcity of tanatochronological parameters useful to estimate the post-mortem interval (PMI) and the absence of uniform investigative protocols in the different centres of forensic pathology. The present study proposes an investigation protocol for the cadaveric dismemberment through the discussion of a case series. The study g

Causative Mutations and Genetic Risk Factors in Sporadic Early Onset Alzheimer's Disease Before 51 Years.

BACKGROUND: Pathogenic variants in the autosomal dominant genes PSEN1, PSEN2, or APP, APOE4 alleles, and rare variants within TREM2, SORL1, and ABCA7 contribute to early-onset Alzheimer's disease (EOAD). However, sporadic EOAD patients have been insufficiently studied to define the probability of being a carrier of one of these variants. OBJECTIVE: To describe the proportion of each genetic variation among patients with very young-onset sporadic AD. METHODS: We first screened PSEN1, PSEN2, and APP in 154 EOAD patients with an onset before 51 years and a negative family history. Among 99 pati

Vienna-Chicago: the cultural transformation of the model system of the un-opposed molar.

The discussion over the roles of genes and environment on the phenotypical specification of organisms has held a central role in science philosophy since the late 19(th) century and has re-emerged in today's debate over genetic determinism and developmental plasticity. In fin-de-siecle Vienna, this debate coincided with a philosophical debate over empiricism/materialism versus idealism/vitalism. Turn-of-the-century Vienna's highly interdisciplinary environment was also the birthplace for the model system of the un-opposed molar. The un-opposed molar system features new tissue formation at the …

[Williams syndrome (microdeletion 7q11.23), model of behavioral phenotype].

A specific behavioral phenotype is recognized in some genetic entities, particularly in microdeletion syndromes secondary to cytogenetically undetectable chromosomal deletions. Williams syndrome (WS) is a developmental disorder displaying dysmorphic signs, heart malformations and behavioral phenotype associated, in most cases, with a deletion of chromosome 7q11.23. We described physical and neuro-psychological assessment of nine cases of SW. Molecular studies were performed using Southern blot analyses or FISH, and identified a 7q11.23 deletion in all cases. Behavioral phenotype of WS is chara

Genome-wide association study reveals key genomic regions underlying colostrability in Italian Holstein cattle.

BACKGROUND: Colostrum yield (CY, L) and concentration of immunoglobulin G (IgG, g/L) are important phenotypes to monitor in dairy farms because of their association with the risk of failure of the passive transfer of immunity in the newborn calf. This can occur when the CY of the parturient cow is insufficient or when the IgG concentration is low. Given that both of these traits are heritable, the present study aimed to investigate their genetic determinism by identifying significant genomic regions. A genome-wide association study coupled with an exploratory functional enrichment analysis was…

Deciphering obesity: a conceptual overview and synthesis.

Obesity represents a multifactorial disorder rooted in genetic, developmental, and environmental influences. Its persistence and heterogeneity warrant deeper understanding of pathology beyond genetic determinism. This Perspective synthesizes a conceptual model that draws from fundamental concepts, establishes cellular and organ levels of pathology, and provides a functional hierarchy of molecular signals in energy regulation. At the cellular level, DNA, RNA, and epigenetic regulators act as scripts, playwrights, and directors shaping metabolic behavior, establishing a "metabolic memory" that i…

Transcription from a gene desert in a melanoma porcine model.

The genetic mechanisms underlying cutaneous melanoma onset and progression need to be further understood to improve patients' care. Several studies have focused on the genetic determinism of melanoma development in the MeLiM pig, a biomedical model of cutaneous melanoma. The objective of this study was to better describe the influence of a particular genomic region on melanoma progression in the MeliM model. Indeed, a large region of the Sus scrofa chromosome 1 has been identified by linkage and association analyses, but the causal mechanisms have remained elusive. To deepen the analysis of th…

Physician Liability for Genetic Risk-Based Prescription Decisions.

The use of AvertD, a genetic test to assess an individual's risk of developing opioid use disorder, will expose physicians to liability. Critics of the test argue that the test itself is unreliable, often resulting in false positives, which will lead to potential undertreatment of pain, and false negatives, which will lead to inappropriate opioid prescriptions. But if the test is available, physicians will use it. And often, due to a combination of genetic determinism and genetic illiteracy, physicians will rely on the test to make opioid prescription decisions without also looking to the envi…

OBJECTIVE: Huntington's disease (HD) is a genetic neurodegenerative disease characterized by cognitive, motor, and psychiatric dysfunction. … lop psychobehavioral symptoms. CONCLUSIONS: In the past, the HD gene was considered the epitome of genetic determinism. Our results are consistent with recent evidence that other factors might modulate its impact. Genetic background, gender, age, body temperature, and arterial blood pH have a major impact on myocardial infarct size in the mouse and need to be carefully measured and/or taken into account: results of a comprehensive analysis of determinants of infarct size in 1,074 mice.

Evidence of phenotypic and genetic relationships between sociality, emotional reactivity and production traits in Japanese quail.

The social behavior of animals, which is partially controlled by genetics, is one of the factors involved in their adaptation to large breeding groups. To understand better the relationships between different social behaviors, fear behaviors and production traits, we analyzed the phenotypic and genetic correlations of these traits in Japanese quail by a second generation crossing of two lines divergently selected for their social reinstatement behavior. Analyses of results for 900 individuals showed that the phenotypic correlations between behavioral traits were low with the exception of signi

Detection of selection signatures within candidate regions underlying trypanotolerance in outbred cattle populations.

Breeding indigenous African taurine cattle tolerant to trypanosomosis is a straightforward approach to control costs generated by this disease. A recent study identified quantitative trait loci (QTL) underlying trypanotolerance traits in experimental crosses between tolerant N'Dama and susceptible Boran zebu cattle. As trypanotolerance is thought to result from local adaptation of indigenous cattle breeds, we propose an alternative and complementary approach to study the genetic architecture of this trait, based on the identification of selection signatures within QTL or candidate genes.
