A snapshot of reproductive and sexual health and rights of women living with HIV in Asia.

OBJECTIVE: A brief review of status of sexual and reproductive health and rights of women living with human immunodeficiency virus (HIV) in Asia with a focus on their access to related services. SUBJECT(S) AND METHODS: The content has primarily emerged from several meetings, workshops and research studies that regional networks of women living with HIV have conducted in the region. We used published and unpublished literature in English after the year 2000. RESULTS: We found many hindrances to realization of sexual and reproductive rights by HIV-positive women and their access to related hea

A novel central nervous system-penetrating protease inhibitor overcomes human immunodeficiency virus 1 resistance with unprecedented aM to pM potency.

Antiretroviral therapy for HIV-1 infection/AIDS has significantly extended the life expectancy of HIV-1-infected individuals and reduced HIV-1 transmission at very high rates. However, certain individuals who initially achieve viral suppression to undetectable levels may eventually suffer treatment failure mainly due to adverse effects and the emergence of drug-resistant HIV-1 variants. Here, we report GRL-142, a novel HIV-1 protease inhibitor containing an unprecedented 6-5-5-ring-fused crown-like tetrahydropyranofuran, which has extremely potent activity against all HIV-1 strains examined wi

Correlation between human immunodeficiency virus type 1 RNA levels in the female genital tract and immune activation associated with ulceration of the cervix.

To address the hypothesis that local immune activation resulting from genital ulceration enhances human immunodeficiency virus type 1 (HIV-1) replication and shedding into the genital tract, paired plasma and cervicovaginal lavage (CVL) samples were obtained from 12 HIV-infected women before and after treatment of cervical intraepithelial lesions. Two weeks after treatment, inflammation and ulceration of the cervix were accompanied by major increases in mean concentrations of HIV-1 RNA (200-fold), tumor necrosis factor-alpha, interleukin 6, and soluble markers shed by activated lymphocytes and

A Case of Toxoplasmic Encephalitis in an HIV-Negative Patient With Multiple Cerebral Infarction-Like Findings in the Early Stages.

Toxoplasmic encephalitis (TE) is an opportunistic infection that typically manifests in immunocompromised individuals, such as those with HIV infection or AIDS. The characteristic imaging finding is a mass-like lesion with ring enhancement. We report a rare case of TE in a patient who was not infected with HIV with atypical imaging features. An 81-year-old woman developed progressive impaired consciousness over several weeks. Initially, the brain MRI revealed multiple lesions resembling cerebral infarctions, which gradually enlarged and increased in number. Subsequently, gadolinium-enhanced MR

Targeting the pDC/IFN-I axis in HIV-1 immunotherapy.

PURPOSE OF REVIEW: Recent findings on the critical pathogenic role of inflammatory pDC and type 1 interferons (IFN-I) in HIV-1 pathogenesis in humanized mice and its correlation with inflammatory diseases in people living with HIV-1 (PLWH) suggest that targeting the pDC/IFN-I signaling pathway will reverse HIV-induced inflammation to treat HIV-associated end-organ diseases and rescue anti-HIV immunity to reduce or control HIV-1 reservoirs. RECENT FINDINGS: In both humanized mice and in PBMC from people living with HIV-1 (PLWH), depletion of pDC or inhibition of IFN-I signaling resolves IFN-as

Interleukin-2 enhances production in 24 hours of infectious human immunodeficiency virus type 1 in vitro by naturally infected mononuclear cells from seropositive donors.

When peripheral blood mononuclear cells from HIV-1 seropositive patients were treated with rIL-2 in vitro a rapid increase of 2-12 fold in production of extracellular infectious HIV-1 occurred, in 6 of 9 experiments. Overall, the increase in the 9 experiments was significant (p less than 0.01) and provides more direct evidence for an enhancing role of IL-2 in naturally infected cells.

Roles of fragment crystallizable-mediated effector functions in broadly neutralizing antibody activity against HIV.

PURPOSE OF REVIEW: 'Broadly neutralizing antibodies' (bNAbs), are rare HIV-specific antibodies which exhibit the atypical ability to potently neutralize diverse viral isolates. While efforts to elicit bNAbs through vaccination have yet to succeed, recent years have seen remarkable preclinical and clinical advancements of passive immunization approaches targeting both HIV prevention and cure. We focus here on the potential to build upon this success by moving beyond neutralization to additionally harness the diverse effector functionalities available to antibodies via fragment crystallizable-ef

Improved oral bioavailability and brain transport of Saquinavir upon administration in novel nanoemulsion formulations.

The aim of this investigation was to develop novel oil-in-water (o/w) nanoemulsions containing Saquinavir (SQV), an anti-HIV protease inhibitor, for enhanced oral bioavailability and brain disposition. SQV was dissolved in different types of edible oils rich in essential polyunsaturated fatty acids (PUFA) to constitute the internal oil phase of the nanoemulsions. The external phase consisted of surfactants Lipoid-80 and deoxycholic acid dissolved in water. The nanoemulsions with an average oil droplet size of 100-200 nm, containing tritiated [(3)H]-SQV, were administered orally and intravenous

Distress tolerance and decreased cognitive fusion tendencies explain the connection between mindfulness, posttraumatic growth and health-related quality of life among HIV patients.

BACKGROUND: Dispositional mindfulness is one psychological resource that consistently promotes posttraumatic growth (PTG) and health-related quality of life (HRQoL). However, the precise mechanisms through which mindfulness performs this function are not clear. The current study aimed to examine the mediating role of distress tolerance and cognitive fusion in the relationship between mindfulness, PTG and HRQoL among HIV patients. METHOD: Using a cross-sectional design, 491 HIV patients (M age = 31.41; SD = 10.37; gender = 211 (47%) males; and 280 (53%)) females were randomly selected from a te

A daisy chain of inferences: the role of single-cell and single-genome proviral sequencing in characterizing HIV-1 reservoirs.

PURPOSE OF REVIEW: Understanding and targeting the HIV reservoir requires navigating a hierarchy of inferential assays. Quantitative viral outgrowth assays, FLIP-seq, and intact proviral DNA assays (IPDA) - though methodologically distinct - are all fundamentally single-cell technologies. Each relies on limiting dilution to isolate and interrogate individual proviral genomes derived from single infected cells, offering high-resolution proxies for the outcome of greatest interest: replication competence and the risk of viral rebound. Rather than providing direct measurements, these assays infer

Peptide Bispecifics Inhibiting HIV-1 Infection by an Orthogonal Chemical and Supramolecular Strategy.

Viral infections pose a significant threat to human health, and effective antiviral strategies are urgently needed. Antiviral peptides have emerged as a promising class of therapeutic agents due to their unique properties and mechanisms of action. While effective on their own, combining antiviral peptides may allow us to enhance their potency and to prevent viral resistance. Here, we developed an orthogonal chemical strategy to prepare a heterodimeric peptide conjugate assembled on a protein-based nanoplatform. Specifically, we combined the optimized version of two peptides inhibiting HIV-1 by

Antiretroviral action of Rosemary oil-based atazanavir formulation and the role of self-nanoemulsifying drug delivery system in the management of HIV-1 infection.

Atazanavir or ATV is an FDA-approved, HIV-1 protease inhibitor that belongs to the azapeptide group. Over time, it has been observed that ATV can cause multiple adverse side effects in the form of liver diseases including elevations in serum aminotransferase, indirect hyper-bilirubinemia, and idiosyncratic acute liver injury aggravating the underlying chronic viral hepatitis. Hence, there is an incessant need to explore the safe and efficacious method of delivering ATV in a controlled manner that may reduce the proportion of its idiosyncratic reactions in patients who are on antiretroviral the

Simultaneous efficacy of IL-15 and the HSP70 fragments in increasing immunostimulatory effects of a HIV-1 DNA vaccine candidate against SCR HIV-1.

Interleukin (IL)-15 serves as a potent adjuvant that can enhance T cell-directed vaccine responses. Moreover, heat shock proteins (HSPs) have been used as potent adjuvants in immunotherapy of tumors and infectious diseases, as well as in vaccine development. In this study, we evaluated the ability of IL-15 and HSP70 fragments (C-terminal (CT)-Hsp70 and N-terminal (NT)-Hsp70) to act as adjuvants for enhancing the immunostimulatory effects of a HIV-1 DNA vaccine candidate against single-cycle replicable (SCR) HIV-1. These adjuvants were administered individually and in combination with the HIV-1

Dimethyl Fumarate Prevents HIV-Induced Lysosomal Dysfunction and Cathepsin B Release from Macrophages.

HIV-associated neurocognitive disorders (HAND) are prevalent despite combined antiretroviral therapy, affecting nearly half of HIV-infected patients worldwide. During HIV infection of macrophages secretion of the lysosomal protein, cathepsin B, is increased. Secreted cathepsin B has been shown to induce neurotoxicity. Oxidative stress is increased in HIV-infected patients, while antioxidants are decreased in monocytes from patients with HIV-associated dementia (HAD). Dimethyl fumarate (DMF), an antioxidant, has been reported to decrease HIV replication and neurotoxicity mediated by HIV-infecte

Dynamics of adrenal steroids are related to variations in Th1 and Treg populations during Mycobacterium tuberculosis infection in HIV positive persons.

Tuberculosis (TB) remains the most frequent cause of illness and death from an infectious agent, and its interaction with HIV has devastating effects. We determined plasma levels of dehydroepiandrosterone (DHEA), its circulating form DHEA-suphate (DHEA-s) and cortisol in different stages of M. tuberculosis infection, and explored their role on the Th1 and Treg populations during different scenarios of HIV-TB coinfection, including the immune reconstitution inflammatory syndrome (IRIS), a condition related to antiretroviral treatment. DHEA levels were diminished in HIV-TB and HIV-TB IRIS patien

Educational impact on women's HIV knowledge and stigma beliefs: evidence from universal primary education in Malawi.

This paper examines the impact of education on women's comprehensive knowledge of HIV and stigma in Malawi, utilizing data from three waves of the nationally representative Malawi Demographic Health Survey. Education plays a critical role in improving health knowledge and countering misinformation, as underscored by SDG3. Our study employs the universal primary education policy implemented in 1994 as an instrumental variable, revealing that women affected by the reform experienced improvements in their education. We find that each additional year of schooling increases the likelihood of women
