BACKGROUND: Radiogenomic modelling faces a significant challenge due to the high-dimensional nature of quantitative radiomic features and limited sample sizes. Feature selection is therefore essential to eliminate irrelevant features and mitigate overfitting. Particle swarm optimisation (PSO) has shown promise for effectively navigating large feature spaces, yet its effectiveness in radiogenomics remains unexplored. This study investigates the value of PSO-based methods, both independently and in combination with other advanced techniques, for MRI-based prediction of human papillomavirus (HPV)

Chromosomal rearrangement (CR) events have been implicated in many tumor and non-tumor human diseases. CR events lead to their associated diseases by disrupting gene and protein structures. Also, they can lead to diseases through changes in chromosomal 3D structure and gene expression. In this study, we search for CR-associated diseases potentially caused by chromosomal 3D structure alteration by integrating Hi-C and ChIP-seq data. Our algorithm rediscovers experimentally verified disease-associated CRs (polydactyly diseases) that alter gene expression by disrupting chromosome 3D structure. In

Nonhuman primates have been a common animal model to evaluate experimentally induced malformations. Reports on spontaneous malformations are important in determining the background incidence of congenital anomalies in specific species and in evaluating experimental results. Here we report on a stillborn cynomolgus monkey (Macaca fascicularis) with multiple congenital anomalies from the colony maintained at the Southwest National Primate Research Center at the Texas Biomedical Research Institute, San Antonio, Texas. Physical findings included low birth weight, craniorachischisis, facial abnorma

PURPOSE: To describe a challenging case of acute retinal necrosis.
METHODS: Clinical data, including medical history, clinical findings and imaging features were retrospectively collected in a 28-year-old female with unilateral active retinitis.
RESULTS: The yellowish area of retinitis surrounding a pigmented chorioretinal scar and the full thickness retinal hyper-reflectivity associated with diffuse increase in choroidal demonstrated a challenging case of necrotising retinitis, where baseline clinical and tomographic features were atypical and misleading towards a toxoplasmic aetiology. The

Atypical choroid plexus papilloma (aCPP) is very rarely seen in adults. Here, we present the case of a 47-year-old male with several months of headache, nausea, dizziness, and imbalance who was found to have an enhancing mass of the fourth ventricle with imaging findings suggestive of likely ependymoma. The patient underwent suboccipital craniotomy with C1 laminectomy and telovelar approach for gross-total resection of the lesion, with final pathology demonstrating WHO grade II aCPP. Subsequent genomic analysis showed a biologically relevant TERT mutation, as well as several variants of unknow

We present a case of sigmoid volvulus in a young male patient with culture-proven Salmonella Typhi in the blood which was sensitive to Meropenem and Azithromycin only, presented with fever, vomiting, loose stools, hematochezia, abdominal distention and tenderness with no signs of perforation on erect chest x-ray. Further, radiological imaging showed signs of sigmoid volvulus. An urgent colonic decompression with untwisting of the mesentery was performed. In our case, it can be said that sigmoid volvulus was developed as a complication of multiple drug-resistant strains of Salmonella Typhi. The

Breast cancer is biologically heterogeneous, yet conventional clinical decisions rely on static, single-marker proxies (ER, PR, HER2, and Ki-67) that capture only a narrow slice of tumor biology. This mini-review argues that AI-driven multi-omics biomarkers offer a valuable shift toward systems-level, model-derived probabilities integrating genomic, transcriptomic, proteomic, metabolomic, and imaging information to explain and predict disease behavior more faithfully. We synthesize recent advances across three domains: (i) computational strategies for multi-omics integration, from classical pe

Predicting spatial transcriptomics from histology images offers cost-effective insights but faces privacy barriers preventing cross-institutional data sharing. To address this, we present privateST, a homomorphic-encryption-optimized framework that substantiates the feasibility of secure spatial transcriptomics prediction. To accommodate the computational constraints of homomorphic encryption, we downsampled the input images using bilinear interpolation. To improve prediction accuracy for the 100 target genes, we incorporated predictions for an auxiliary set of 150 highly expressed genes. This

Centrosome abnormalities (CA) are a hallmark of epithelial cancers, yet their spatial complexity and phenotypic heterogeneity remain poorly understood due to limitations in conventional image analysis. Here we present CenSegNet (Centrosome Segmentation Network), a modular deep learning framework for high-throughput segmentation of centrosomes and epithelial architecture, enabling accurate and generalisable centrosome phenotyping at spatial and single-cell resolution across imaging modalities and tissue contexts. Applied to tissue microarrays comprising 911 breast cancer cores from 127 patients

Radiomics allows for quantitative utilisation of radiological data and carries great potential for improving the diagnosis and management of hepatopancreatobiliary (HPB) cancers. Radiomic features represent changes at the mesoscopic scale and serve as noninvasive markers for tumour heterogeneity. Using deep learning and machine learning approaches, retrospective studies have demonstrated that radiomic signatures have the capability to improve the diagnosis of hepatocellular carcinoma (HCC), pancreatic cancer, and cholangiocarcinoma, in conjunction with radiological evaluation. Radiomic models

Pancreatic cystic lesions (PCLs) represent a highly heterogeneous category of pancreatic abnormalities with an increasing prevalence due to advances in imaging modalities and an aging population. While the majority of PCLs are benign, a subset harbors a variable risk of malignant transformation. Current guidelines rely primarily on imaging features for risk stratification, but limitations in diagnostic accuracy have led to many unnecessary surgical procedures and conversely, missed opportunities for timely resection in cases that progress to malignancy.Recent advances in biomarker discovery ho

BACKGROUND: Patients with mitochondrial encephalomyopathy, lactic acidosis, and stroke-like episodes (MELAS) usually present with multisystemic dysfunction with a wide range of clinical manifestations. When the tests for common mitochondrial DNA (mtDNA) point mutations are negative and the mtDNA defects hypothesis remains, urine epithelial cells can be used to screen the mitochondrial genome for unknown mutations to confirm the diagnosis.
CASE PRESENTATION: A 66-year-old Chinese woman presented with symptoms of MELAS and was initially misdiagnosed with acute encephalitis at another institutio

Follicular lymphoma (FL) is generally considered an indolent disorder. With modern day treatments, long remissions are often achieved both in the front-line and relapsed setting. However, a subset of patients has a more aggressive course and a worse outcome. Their identification is the main purpose of modern day prognostic tools. In this review, we attempt to summarize the evidence concerning prognostic and predictive factors in FL, including (1) pre-treatment factors, from baseline clinical characteristics and imaging tests to histological grade, the microenvironment and genomic abnormalities

The combined use of androgen deprivation therapy (ADT) and image-guided radiotherapy (IGRT) can improve overall survival in aggressive, localized prostate cancer. However, owing to the adverse effects of prolonged ADT, it is imperative to identify the patients who would benefit from this combined-modality therapy relative to the use of IGRT alone. Opportunities exist for more personalized approaches in treating aggressive, locally advanced prostate cancer. Biomarkers--such as disseminated tumour cells, circulating tumour cells, genomic signatures and molecular imaging techniques--could identif

Cell-to-cell heterogeneity in infection outcome is a general feature of most viruses, but the underlying mechanisms are poorly understood. Here, we developed a live-cell single-molecule imaging technology to visualize infection by unmodified influenza A viruses (IAVs) with unprecedented resolution. Using this approach, we generated a detailed kinetic map of IAV infection, which identified viral ribonucleoprotein (vRNP) replication, nuclear export, and virion budding as important sources of heterogeneity. Mechanistically, we show that infection heterogeneity is caused by differential viral gene

BACKGROUND: Pro-neurotensin (pro-NT) is the stable circulating precursor of neurotensin (NT), a neuropeptide expressed mainly in the central nervous system and small intestine that regulates key physiological processes like fatty acid absorption in the gut and suppresses appetite via central mechanisms. Studies in NT-deficient mice and humans implicate NT in obesity and insulin resistance, highlighting its role in metabolic regulation.
METHODS: To explore the genetic determinants of circulating pro-NT and its causal relationships with obesity and brain phenotypes, we conducted a genome-wide m

BACKGROUND: Truly understanding the cancer biology of heterogeneous tumors in precision medicine requires capturing the complexities of multiple omics levels and the spatial heterogeneity of cancer tissue. Techniques like mass spectrometry imaging (MSI) and spatial transcriptomics (ST) achieve this by spatially detecting metabolites and RNA but are often applied to serial sections. To fully leverage the advantage of such multi-omics data, the individual measurements need to be integrated into 1 dataset.
RESULTS: We present the Multi-Omics Imaging Integration Toolset (MIIT), a Python framework

Researchers have conducted extensive research on the correlation between brain resting-state functional activities (RSFA) and migraine. However, we still do not fully understand the exact nature of the causal relationship between these RSFA and migraine. We conducted a bidirectional two-sample Mendelian randomization (MR) study to investigate the causal association between migraine and RSFA. We gathered summary statistics from genome-wide association studies for 191 resting-state functional magnetic resonance imaging phenotypes. We employed various analytical methods for bidirectional two-samp

Radiogenomics relationships (RRs) aims to identify statistically significant correlations between medical image features and molecular characteristics from analysing tissue samples. Previous radiogenomics studies mainly relied on a single category of image feature extraction techniques (ETs); these are (i) handcrafted ETs that encompass visual imaging characteristics, curated from knowledge of human experts and, (ii) deep ETs that quantify abstract-level imaging characteristics from large data. Prior studies therefore failed to leverage the complementary information that are accessible from fu
