Plasmodium falciparum transcriptome analysis reveals pregnancy malaria associated gene expression.

BACKGROUND: Pregnancy-associated malaria (PAM) causing maternal anemia and low birth weight is among the multiple manifestations of Plasmodium falciparum malaria. Infected erythrocytes (iEs) can acquire various adhesive properties that mediate the clinical severity of malaria. Recent advances on the molecular basis of virulence and immune evasion have helped identify var2csa as a PAM-specific var gene. METHODOLOGY/PRINCIPAL FINDINGS: The present study presents a genome-wide microarray transcript analysis of 18 P. falciparum parasite isolates freshly collected from the placenta. The proportion

Impact of placental Plasmodium falciparum malaria on pregnancy and perinatal outcome in sub-Saharan Africa: II: effects of placental malaria on perinatal outcome; malaria and HIV.

Placental malaria is recognized as a common complication of malaria in pregnancy in areas of stable transmission, and, as a consequence, serious health problems arise for the mother and especially her baby [1]. Although malaria in pregnancy is a major factor associated with adverse perinatal outcome, the link between malaria and perinatal morbidity/mortality is less clear in areas with stable endemic malaria where pregnant women have acquired immunity [2]. Histological examination of the placenta is a predictor of fetal morbidity, as well as being the most sensitive detector of maternal infect

Thymic involution with loss of Hassall's corpuscles mimicking thymic dysplasia in a child with transfusion-associated graft-versus-host disease.

A 7-year-old leukemic girl developed pancytopenia following chemotherapy and was given several transfusions of nonirradiated blood. Within 2 weeks she developed a maculopapular rash, fever, abnormal liver function, diarrhea, and wasting. She became septic and died 6 weeks later. Transfusion-associated graft-versus-host disease (GVHD) was suspected clinically. At autopsy, changes diagnostic of GVHD were present in the skin and liver. The remarkable feature of the case was the histopathology of the thymus, which was morphologically "dysplastic," i.e., minute, lymphoid depleted, devoid of a corti

Challenges in the rabbit haemorrhagic disease 2 (RHDV2) molecular diagnosis of vaccinated rabbits.

Molecular methods are fundamental tools for the diagnosis of viral infections. While interpretation of results is straightforward for unvaccinated animals, where positivity represents ongoing or past infections, the presence of vaccine virus in the tissues of recently vaccinated animals may mislead diagnosis. In this study, we investigated the interference of RHDV2 vaccination in the results of a RT-qPCR for RHDV2 detection, and possible associations between mean Cq values of five animal groups differing in age, vaccination status and origin (domestic/wild). Viral sequences from vaccinated rab

A Case of Neonatal Lupus Presenting with Myocardial Dysfunction in the Absence of Congenital Heart Block (CHB): Clinical Management and Brief Literature Review of Neonatal Cardiac Lupus.

Neonatal lupus (NLE) is a rare acquired autoimmune disorder caused by transplacental passage of maternal autoantibodies to Sjogren's Syndrome A or B (SSA-SSB) autoantigens (Vanoni et al. in Clin Rev Allerg Immunol 53:469-476, 2017) which target fetal and neonatal tissues for immune destruction. The cardiac trademark of NLE is autoimmune heart block, which accounts for more than 80% of cases of complete atrioventricular heart block (AVB) in newborns with a structurally normal heart (Martin in Cardiol Young 24: 41-46, 2014). NLE presenting with cardiac alterations not involving rhythm disturbanc

Material awareness on natural feeding.

INTRODUCTION: Natural breastfeeding is the only proper way to feed newborns and infants because it ensures their proper development. Breastfeeding enhances health and protects against the development of many diseases in childhood and adulthood. The primary benefits of breastfeeding include reduced incidences of infection in the respiratory system as well as a reduction in gastrointestinal and systemic infections. The benefits of breastfeeding also include decreased inflammation and improved immunity to disease in the infant. Further benefits of breastfeeding are reduced incidences of type 1 di

Seroprevalence of Anti-Cytomegalovirus Antibodies in Pregnant Women from South-West Romania.

Cytomegalovirus (CMV), in addition to other agents, is part of the TORCH complex (Toxoplasma gondii, Rubella virus, Cytomegalovirus, Herpes simplex viruses, and other agents). CMV infection is the most frequent cause of congenital malformations. This study aimed to establish the variation of prevalence of anti-CMV antibodies in pregnant women from the South-West region of Romania, according to demographic factors, such as age and area of residence, in two separate time periods (2013-2016 and 2019-2022). We collected from the hospital records the age, place of residence, and anti-CMV antibody t

The nanoemulsion adjuvant provides antigen dose-sparing effects and enhances maternal passive immune protection for the cell-cultured quadrivalent influenza virus subunit vaccine.

To elicit a robust immune response, an adjuvant can be combined with the antigen in influenza vaccine formulations. In this study, we evaluated the dose-sparing effect and safety of a squalene-based oil-in-water nanoemulsion (NE) adjuvant formulated with a cell culture-derived quadrivalent influenza vaccine. Immune responses-including anti-HA IgG antibody levels and hemagglutination inhibition (HAI) titers-were assessed, along with protection against a homologous challenge with influenza B virus (strain B/Maryland/15/2016 B/Victoria). We also investigated the influence of antigen dose on vacci

Randomized, placebo-controlled phase 3 trial evaluating safety, immunogenicity, and reactogenicity of RSVPreF3-Mat in high-risk pregnant women and their infants.

Respiratory syncytial virus (RSV) causes respiratory illness among infants; maternal vaccination confers passive protection through placental antibody transfer. This phase 3, randomized, placebo-controlled trial evaluated safety, reactogenicity, and immunogenicity of RSV prefusion protein F3 maternal vaccine (RSVPreF3-Mat), administered at 240/7 to 360/7 weeks of gestation to high-risk pregnant women (women with obstetric complications and/or human immunodeficiency virus infection or in pregnant adolescents), and their infants up to 1 year post-birth. Enrollment was stopped and the study unbli

Gestational age-dependent dynamics of transplacental CMV and VZV IgG transfer: Weekly comparative analysis in preterm and full-term neonates (24-41 weeks).

BACKGROUND: Transplacental transfer of maternal IgG antibodies promotes foetal immunity against cytomegalovirus (CMV) and varicella-zoster virus (VZV) infections. Comprehensive data on antibody transfer across gestational ages, particularly before 28 weeks, remain limited. METHODS: This prospective cohort study analysed paired maternal and newborn blood samples from n = 564 mother-child pairs (gestational weeks 24-41). Anti-CMV and anti-VZV IgG concentrations were measured using ELISA-tests and CMV neutralising capacity was assessed using an in-house cell culture-based assay. RESULTS: Newbor

[Immunologic platelet tests].

Platelet immunological assays have been used for several decades. The antiglobulin consummation test was the first to be introduced followed by the immunoradiometric technique, immunofluorescence and enzyme linked immunological specific assay, as well as the different monoclonal antibody specific immobilisation of platelet antigens (MAIPA) assays. Using these different tests, it is possible to detect, quantify and define the specificity of platelet antibodies in neonatal alloimmune thrombocytopenia, in idiopathic thrombocytopenic purpura and post transfusion purpura.

Impact of maternal antibodies and microbiota development on the immunogenicity of oral rotavirus vaccine in African, Indian, and European infants.

Identifying risk factors for impaired oral rotavirus vaccine (ORV) efficacy in low-income countries may lead to improvements in vaccine design and delivery. In this prospective cohort study, we measure maternal rotavirus antibodies, environmental enteric dysfunction (EED), and bacterial gut microbiota development among infants receiving two doses of Rotarix in India (n = 307), Malawi (n = 119), and the UK (n = 60), using standardised methods across cohorts. We observe ORV shedding and seroconversion rates to be significantly lower in Malawi and India than the UK. Maternal rotavirus-specific an

Maternal- and Fetal-Encoded Perforin-2 Limits Placental Infection by a Bloodborne Pathogen.

Placental immune responses are highly regulated to strike a balance between protection and tolerance. For relatively mild infections, protection encompasses both the mother and fetus; however, during worsening conditions, protection becomes exclusively reserved for the mother. Previously, we and others have shown that the host factor perforin-2 plays a central role in protecting mice and cells against infection. In this study, we analyzed perforin-2 activity in the mouse placenta to determine whether perforin-2 plays a similarly protective role. We show that perforin-2 is critical for inhibiti

[Virological diagnosis of HIV infection in children].

The laboratory diagnosis of HIV-1 infection in newborns should be carried out using virological methods, viral isolation or molecular methods for the detection of proviral DNA by PCR. Serological methods have poor sensitivity in the first months of life because the IgG of the seropositive mother crosses the placenta. Once HIV-1 infection is diagnosed, other assays are available which indicate the clinical progression of the infection, the genetic variability of the virus, its biological behavior and sensitivity to different antiretroviral medications. These studies can be used independently of

Shigella-specific antibodies in the first year of life among Zambian infants: A longitudinal cohort study.

INTRODUCTION: Shigellosis, is a leading cause of moderate-to-severe diarrhoea and related mortality in young children in low and middle income countries (LMICs). Knowledge on naturally acquired immunity can support the development of Shigella candidate vaccines mostly needed in LMICs. We aimed to quantify Shigella-specific antibodies of maternal origin and those naturally acquired in Zambian infants. METHODS: Plasma samples collected from infants at age 6, 14 and 52-weeks were tested for Shigella (S. sonnei and S. flexneri 2a) lipopolysaccharide (LPS) antigen specific immunoglobulin G (IgG) a
