HLA-DQB1-mediated B cell-epithelial crosstalk drives EBV-associated inflammation in Hunner-type interstitial cystitis.

Hunner-type interstitial cystitis (HIC) is a chronic condition marked by persistent pain and inflammation. To elucidate its immunogenetic drivers, we integrated bulk RNA sequencing and single-cell RNA sequencing datasets with targeted major histocompatibility complex (MHC) sequencing. Transcriptomic analysis revealed selective expansion of B cells and epithelial cells, with strong enrichment of Epstein-Barr virus (EBV) response signatures. CellChat and NicheNet modeling uncovered bidirectional communication wherein B cells secrete IL-1β, FGF2, LIF, and TNFSF9, activating prostaglandin synthesi

Epigenetic-epitranscriptomic crosstalk through TaHAG1-TaNSUN2 coordinates thermotolerance in wheat.

High temperature is a primary abiotic stress that severely constrains crop productivity. Deciphering the regulatory pathways underlying heat responses is essential for breeding heat-tolerant crops with stable yields. Although both epigenetic and epitranscriptomic regulations are involved in plant heat adaptation, their mechanistic interplay remains unclear. Here, integrated epigenomic (H3K9Ac/H3K14Ac) and transcriptomic profiling under heat stress identifies the mRNA m⁵C methyltransferase TaNSUN2 as a key regulator of thermotolerance in wheat. We demonstrate that TaNSUN2 is transcriptionally a

Integrated Metabolomic and Proteomic Analyses Reveal Altered Iron Homeostasis and Immunometabolic Remodeling in the Spleen of Solea senegalensis during Vibriosis.

Vibrio anguillarum is a major pathogen impacting Solea senegalensis aquaculture. While host-pathogen competition for iron drives disease progression, the in vivo molecular evidence linking iron homeostasis and immune responses in fish remains limited. Here, we integrated metabolomic and proteomic analyses to characterize the splenic response of S. senegalensis during experimental vibriosis. Multiomics integration revealed disruption of iron homeostasis alongside coordinated immunometabolic reprogramming. We detected the bacterial siderophore piscibactin directly in spleen tissue of sole exhibi

Glycine max (L.) Merr. (Soybean) metabolome responses to potassium availability.

Potassium (K+) has vital physiological and metabolic functions in plants and its availability can impact tolerance to biotic and abiotic stress conditions. Limited studies have investigated the effect of K+ fertilization on soybean metabolism. Using integrated omics, ionomics and metabolomics, we investigated the field-grown Glycine max (soybean) response, after four K+ soil fertilization rates. Soybean leaf and pod tissue (valves and immature seeds) extracts were analysed by ultra-performance liquid chromatography coupled to high-resolution mass spectrometry (UPLC-HRMS) and inductively couple

Dose-response-defined physiological framework enables comparative analysis of transcriptional response dynamics across multiple herbicides in Arabidopsis thaliana.

Herbicides elicit diverse physiological responses in plants largely due to distinct modes of action of individual compounds. However, it remains unclear how plants differentially respond to herbicides with distinct modes of action when applied at herbicide-specific treatment levels corresponding to comparable degrees of growth inhibition. In this study, we integrated physiological and transcriptomic analyses to characterize the responses of Arabidopsis thaliana exposed to seven herbicides (2,4-dichlorophenoxyacetic acid, acifluorfen, atrazine, glyphosate, imazethapyr, mesotrione, and paraquat)

Endoplasmic reticulum stress preconditioning modifies intracellular mercury content by upregulating membrane transporters.

Endoplasmic reticulum (ER) stress preconditioning protects cells against methylmercury (MeHg) cytotoxicity by inducing integrated stress responses such as eIF2α phosphorylation, ATF4 accumulation, and nonsense-mediated mRNA decay (NMD) suppression. Here we demonstrated that ER stress preconditioning results in the upregulation of membrane transporters, leading to a decrease in intracellular mercury content. Our analyses showed that ER stress preconditioning upregulated the expression of methionine transporters that affect the cellular influx of MeHg, LAT1, LAT3, and SNAT2; and a membrane trans…

Hormonal and transcriptomic reprogramming underlie responses to recurrent drought in the halophyte Atriplex halimus L.

This study provides the first integrative assessment of drought stress memory in a halophyte, highlighting the role of recurrent drought in shaping physiological and molecular responses in plants adapted to unpredictable and stressful environments. Here, we investigated the physiological, biochemical and transcriptomic responses of the xerohalophyte Atriplex halimus L. exposed to one, two or three consecutive drought cycles (D1-D3). Each cycle consisted of severe water deficit (<5% field capacity) followed by a rehydration phase, evaluating water relations, gas exchange, phytohormone profiles,

Resistance to pyrethroids in Aedes aegypti: Insights into transcriptomic response to different insecticide concentrations.

Insecticide spraying is a common strategy for controlling dengue outbreaks, but its effectiveness is compromised by the development of resistance in mosquito populations. In this study, we subjected a strain of Aedes aegypti known for its exceptional ability to develop resistance to controlled permethrin and lambda-cyhalothrin insecticides pressure using two different concentrations. We analyzed resistance mechanisms that are enhanced at each concentration and used RNA sequencing to identify transcripts specifically associated with these exposure levels. Our objective was to uncover the molecu

Cellular clarity: a logistic regression approach to identify root epidermal regulators of iron deficiency response.

BACKGROUND: Plants respond to stress through highly tuned regulatory networks. While prior works identified master regulators of iron deficiency responses in A. thaliana from whole-root data, identifying regulators that act at the cellular level is critical to a more comprehensive understanding of iron homeostasis. Within the root epidermis complex molecular mechanisms that facilitate iron reduction and uptake from the rhizosphere are known to be regulated by bHLH transcriptional regulators. However, many questions remain about the regulatory mechanisms that control these responses, and how th

Using New Approaches in Neurobiology to Rethink Stress-Induced Amnesia.

PURPOSE OF REVIEW: Psychological stress can impact memory systems in several different ways. In individuals with healthy defense and coping systems, stress results in the formation of negatively valenced memories whose ability to induce emotional and somatic distress subsides with time. Vulnerable individuals, however, go on to develop stress-related disorders such as post-traumatic stress disorder (PTSD) and suffer from significant memory abnormalities. Whether expressed as intrusive trauma memories, partial amnesia, or dissociative amnesia, such abnormalities are thought to be the core sourc

Architecting Co-continuous PVDF Composites via Multi-material 3D Parallel Printing for Enhanced Piezoelectricity and Wearable Sensing.

Flexible piezoelectric composites have attracted considerable attention for self-powered sensing; however, achieving efficient electromechanical response while maintaining structural integrity under repeated deformation remains challenging. Herein, a structure-driven multi-material 3D parallel printing strategy is developed to construct polyvinylidene fluoride (PVDF)-based piezoelectric composites with a co-continuous dual-phase architecture. In this design, ionic liquid (IL)-modified PVDF serves as the piezoelectric phase, where the IL promotes β-phase formation and dipole orientation, while

Heat-shock proteins can promote as well as regulate autoimmunity.

Heat-shock proteins (Hsps) are among the most highly conserved and immunogenic proteins shared by microbial agents and mammals. Under physiological conditions, the ubiquitously distributed Hsps maintain the integrity and function of other cellular proteins when cells are exposed to stressful stimuli. However, owing to their conserved nature and stress inducibility, Hsps may become targets of immune response. The T cells and/or antibodies induced by a microbial Hsp may crossreact with the corresponding mammalian Hsp (molecular mimicry) and trigger an autoimmune response, which if unchecked can

Mobile small RNAs as mediators of stress-induced epigenomic remodeling and systemic stress adaptation.

Plants rely on sophisticated long-distance communication networks to coordinate adaptive responses to changing environmental conditions. Among these, mobile small RNAs (sRNAs), including microRNAs (miRNAs) and small interfering RNAs (siRNAs), function as critical systemic signaling molecules that move between tissues through plasmodesmata and the phloem. While their roles in post-transcriptional gene regulation are well established, emerging evidence indicates that mobile sRNAs also mediate stress-induced epigenomic remodeling in recipient tissues. Specifically, 24-nt siRNAs can direct RNA-dep

Stress levels of glucocorticoids inhibit LHβ-subunit gene expression in gonadotrope cells.

Increased glucocorticoid secretion is a common response to stress and has been implicated as a mediator of reproductive suppression upon the pituitary gland. We utilized complementary in vitro and in vivo approaches in the mouse to investigate the role of glucocorticoids as a stress-induced intermediate capable of gonadotrope suppression. Repeated daily restraint stress lengthened the ovulatory cycle of female mice and acutely reduced GnRH-induced LH secretion and synthesis of LH β-subunit (LHβ) mRNA, coincident with increased circulating glucocorticoids. Administration of a stress level of gl

Stress and personality.

Stress is an adaptation reaction of living organisms in response to internal or external threats to homeostasis. It is considered as a complex defence mechanism representing the final endpoint of numerous dynamic and interconnected factors of biological, psychological and social nature. Stress is not a simple, stimulus-response reaction, but the interaction between an individual and the environment, involving subjective perception and assessment of stressors, thus constituting a highly personalized process. Specific inherited characteristics, early experience in life, and particular, learned c

Integrating genomics, multi-omics, CRISPR and speed breeding for stress-resilient vegetable legume improvement.

Vegetable legumes are nutritionally and ecologically important crops. However, their genetic improvement has not kept pace with the increasing challenges posed by climate change due to the polygenic nature of stress tolerance, narrow genetic diversity, and the persistent gap between molecular discoveries and field-level cultivar development. Although recent reviews have examined individual genomic tools or specific stress responses, a comprehensive synthesis integrating genomics-assisted breeding, multi-omics technologies, genome editing, and speed breeding within a unified crop improvement fr

Role of starvation survival response mechanisms on ribosome integrity, antibiotic tolerances, and virulence of Pseudomonas aeruginosa biofilms.

Bacterial biofilms contain physiologically diverse subpopulations of cells, including cells that are nutrient stressed or dormant. We determined how two dormancy pathways, ribosome hibernation and the stringent response, contribute to the survival and antibiotic tolerance of Pseudomonas aeruginosa biofilms. Analyses of whole biofilms and single cells showed that these pathways have differing effects on biofilm cell physiology. Ribosome hibernation, mediated by hibernation promoting factor (HPF), is essential for optimal survival and resuscitation of starved biofilm cells. In the absence of HPF
