The Pathophysiology of Small Intestinal Foreign Body Obstruction and Intraoperative Assessment of Tissue Viability in Dogs: A Review.

Small intestinal foreign body obstructions occur commonly in dogs, accounting for 80% of all canine intestinal obstructions. Such obstructions induce local aberrations in secretion, absorption, and intestinal motility that can precipitate devastating systemic consequences, including a systemic inflammatory response, sepsis, and multiorgan dysfunction. Radiographic diagnosis is poorly sensitive relative to ultrasonography for diagnosing the presence of obstructive foreign material. Emergent surgical intervention is indicated for dogs with obstructive foreign material due to an inability to asse

Mucosal mast cells. I. Isolation and functional characteristics of rat intestinal mast cells.

We have developed a procedure for the dispersion of mast cells from the intestinal lamina propria (LP) and epithelium of rats infected with the intestinal nematode, Nippostrongylus brasiliensis. The dispersed cells are morphologically and histochemically similar to intestinal mucosal mast cells (MMC) in situ and are distinguishable from peritoneal mast cells (PMC). MMC derived from the LP or epithelium of parasitized animals secrete histamine in response to the specific parasite antigens as well as anti-IgE. Unlike PMC, these cells are unresponsive to the basic secretagogues 48/80 and bee veno

Expand, push, break, release, repair: A unique way to secrete macromolecules.

Regulated exocytosis is a fundamental process in specialized secretory organs. In this issue of Science Signaling, Dolan et al. describe a distinctive modality of secretion in the goblet cells of the small intestine crypts driven by intracellular cargo expansion.

Tracking Vibrio cholerae Cell-Cell Interactions during Infection Reveals Bacterial Population Dynamics within Intestinal Microenvironments.

Vibrio cholerae is the causative agent of the diarrheal disease cholera. Although many V. cholerae virulence factors have been studied, the role of interbacterial interactions within the host gut and their influence on colonization are poorly understood. Here, we utilized the conjugative properties of a Vibrio-specific plasmid to serve as a quantifiable genetic marker for direct contact among V. cholerae cells in the infant rabbit model for cholera. In conjunction, we also quantified contact-dependent type 6 secretion system (T6SS)-mediated killing of co-infecting V. cholerae strains. Tracking

Clinical significance of changes of flora in the upper digestive tract.

Dysbiosis does not necessarily mean disease. Typical clinical picture of disease caused by metabolic activity of dysbiotic microflora is the microbial over-growth syndrome. We studied case histories of 434 patients without consideration of their basic illnesses and correlated them with types of dysbioses of duodenal juice. 55% of 314 patients with gastrointestinal disorders revealed a dysbiosis. A dysbiosis could also be found in up to 64% of pancreatic diseases, whereas intestinal dysfunctions and chronic inflammatory diseases usually show normal colonization. These cases express alteration o

Duodenal microflora in infants with acute and persistent diarrhoea.

The duodenal bacterial population was determined qualitatively and quantitatively in 3 groups of infants with diarrhoeal disease of varying duration. Infants with acute self limiting diarrhoea (group 1) had an abnormal overgrowth of organisms in the upper small bowel which was similar to that found in infants whose diarrhoea persisted after 4 days in hospital (group 2). The total duodenal organism count in infants with diarrhoea persisting after 7 days in hospital (group 3) was almost 100 times greater than either of the other groups. The findings may have important implications in preventing

Pancreatic function tests in tropical sprue is pancreas involved?

Pancreatic functions were studied in 15 patients with tropical sprue. The diagnosis of tropical sprue was made on accepted criteria of demonstrating malabsorption of at least two unrelated food substances in the absence of any other cause for malabsorption in patients residing in a tropical zone. Exogenous secretion (Boots) in a dose of one clinical unit per kg was employed for direct stimulation of exocrine pancreatic secretion. Of the 15 patients, 12 showed normal pancreatic functions. In two patients, volume output was marginally low i.e. 1.6 and 1.6 ml/kg/hr respectively (Normal value grea

Analysis of the mechanism of action of calcium-induced exocrine pancreatic secretory changes in the dog.

In conscious dogs, provided with gastric and duodenal fistulas (Thomas cannula), the effect of an acute i.v. calcium infusion (10 mg./kg./hr.) on the plateau levels of secretin-induced (GIH, 1 CU./kg./hr.) pancreatic juice parameters were analyzed without or with previous cholingergic blockade by a continuous atropine perfusion (1.5 mg./hr.). In the former condition, i.v. calcium raised all pancreatic juice parameters, mainly protein concentration (148%) and output (173%). During atropine blockade, i.v. calcium changed neither flow rate nor bicarbonate output of secretin-induced pancreatic sec

Mpi-driven N-glycosylation orchestrates mucin O-glycosylation and intestinal homeostasis.

The intestinal mucus barrier physically separates the epithelium from the dense microbial community of the gut and is essential for intestinal homeostasis. The principal component, the gel-forming mucin MUC2, is extensively glycosylated, yet how different classes of glycans regulate mucin function remains unclear. Here we show that N-glycosylation is required for proper MUC2 maturation and mucus barrier integrity. Using mouse models with hypomorphic or intestinal epithelial-specific loss of the mannose-generating enzyme MPI, which is required for N-glycan synthesis, we find that impaired N-gly

Immune Regulation of Goblet Cell and Mucus Functions in Health and Disease.

The mucosal surfaces of the body are the most vulnerable points for infection because they are lined by single or multiple layers of very active epithelial cells. The main protector of these cells is the mucus system generated by the specialized goblet cell secreting its main components, the gel-forming mucins. The organization of the mucus varies from an attached mucus that is impenetrable to bacteria in the large intestine to a nonattached, more penetrable mucus in the small intestine. The respiratory tract mucus system clears particles and microorganisms from healthy lungs but causes diseas

[The proteoglycan content of the duodenal juice in peptic ulcer patients].

Examined were 21 patients with duodenal ulcer during exacerbation, 18 patients at the stage of clinical remission and 37 patients with exacerbation of chronic cholecystitis with the purpose of evaluating the content of glycosaminoglycans in the duodenal juice. In was established that patients with duodenal ulcer during exacerbation showed a marked reduction of the release of glycosaminoglycans while the content of these substances was increased at the stage of clinical remission and patients with exacerbation of chronic cholecystitis. The authors discuss the role of glycosaminoglycans in the p

[Pseudopancreas insufficiency in 3 young infants with extrahepatic bile duct atresia].

As was to be expected, the duodenal juice in 3 infants with extra-hepatic biliary atresia was different from that in 43 healthy infants similarly investigated. Following the injection of Secretin and Pancreozymin, the amylase and bicarbonate values were normal, while the trypsin activity in the 3 patients was significantly reduced. Trypsinogen could not be found in the duodenal juice but was presesnt as Trypsin which was mainly inactive. It is concluded that the dissociated pancreatic insufficiency is caused by a lack of bile secretion.

Expansion of Intestinal Secretory Cell Population Induced by Listeria monocytogenes Infection: Accompanied With the Inhibition of NOTCH Pathway.

Listeria monocytogenes, as a model organism, is a causative agent of enteric pathogen that causes systemic infection. However, the interaction of L. monocytogenes and small intestinal epithelium has not been fully elucidated yet. In this study, mice and intestinal organoids were chosen as the models to investigate the influence of L. monocytogenes infection on the intestinal secretory cells and its differentiation-related pathways. Results confirmed the phenomenon of intestinal damage that L. monocytogenes infection could lead to villi damage in mice, which was accompanied by the increase of T
