Interactions between zinc, essential fatty acids and prostaglandins: relevance to acrodermatitis enteropathica, total parenteral nutrition, the glucagonoma syndrome, diabetes, anorexia nervosa and sickle cell anaemia.

Many of the features of zinc deficiency and of essential fatty acid (EFA) deficiency are similar in both animals and humans. The two agents interact in various ways. EFAs are important in zinc absorption, probably after conversion to prostaglandins (PGs). Zinc seems necessary for at least two stages in EFA metabolism, the conversion of linoleic acid to gamma-linolenic acid, and the mobilisation of dihomogammalinolenic acid (DGLA) for the synthesis of 1 series PGs. Zinc may also be important in the conversion of DGLA to arachidonic acid and in arachidonic acid mobilisation for 2 series PG forma

An approach using lecithin treatment for olivopontocerebellar atrophies.

A therapeutic trial with two different lecithins, with 32 and 7% phosphatidylcholine respectively, was performed for 3 months on 11 patients with clinical diagnosis of dominant, recessive and sporadic olivopontocerebellar atrophies. The correlation between plasma choline levels and the clinical picture shows a clinical worsening with very high choline levels and a slight improvement with smaller increases of plasma choline levels. The possible role in these disorders of phosphatidylcholine and linoleic acid - both present in lecithin - is discussed, with relationship to these findings.

Untargeted metabolomics analysis reveals spatial metabolic heterogeneity in different intestinal segments of type 1 diabetic mice.

Type 1 diabetes (T1D) has been reported to cause systematic metabolic disorders, but metabolic changes in different intestinal segments of T1D remain unclear. In this study, we analyzed metabolic profiles in the jejunum, ileum, cecum and colon of streptozocin-induced T1D and age-matched control (CON) mice by an LC-MS-based metabolomics method. The results show that segment-specific metabolic disorders occurred in the gut of T1D mice. In the jejunum, we found that T1D mainly led to disordered amino acid metabolism and most amino acids were significantly lower relative to CON mice. Moreover, fat

9- and 13-Hydroxy-octadecadienoic acids (9+13 HODE) are inversely related to granulocyte colony stimulating factor and IL-6 in runners after 2h running.

This study utilized a pro-inflammatory exercise mode to explore potential linkages between increases in 9- and 13-hydroxy-octadecadienoic acid (9+13 HODE) and biomarkers for inflammation, oxidative stress, and muscle damage. Male (N=10) and female (N=10) runners ran at ∼70% VO2max for 1.5h followed by 30min of downhill running (-10%). Blood samples were taken pre-run and immediately-, 1-h-, and 24-h post-run, and analyzed for 9+13 HODE, F2-isoprostanes, six cytokines, C-reactive protein (CRP), creatine kinase (CK), and myoglobin (MYO). Gender groups performed at comparable relative heart rate

Differential effects of hypothalamic long-chain fatty acid infusions on suppression of hepatic glucose production.

Our objective was to investigate whether the direct bilateral infusion of the monounsaturated fatty acid (MUFA) oleic acid (OA) within the mediobasal hypothalamus (MBH) is sufficient to reproduce the effect of administration of OA (30 nmol) in the third cerebral ventricle, which inhibits glucose production (GP) in rats. We used the pancreatic basal insulin clamp technique (plasma insulin ∼20 mU/ml) in combination with tracer dilution methodology to compare the effect of MBH OA on GP to that of a saturated fatty acid (SFA), palmitic acid (PA), and a polyunsaturated fatty acid (PUFA), linoleic a

[Changes in the level of free fatty acids in patients with acute alcoholic hepatitis].

The study covered 25 patients with acute alcoholic hepatitis mean age of 48 years 25 patients with alcoholic liver steatosis (mean age of 49 years), and 41 clinically healthy persons - controls (mean age of 42 years). Plasma levels of individual free fatty acids (FFA) were determined in any case. There was a statistically significant decrease of palmitinc, stearinic, oleinic, linolic and arachidonic acid and of total FFA in the patients with acute alcoholic hepatitis (AAH) as compared with the controls (p less than 0.001). Similar changes were found in patients with liver steatosis but they we

[Effect of acute overheating of animals on the processes of lipid peroxidation].

In liver tissue, brain, lungs and skeletal muscles of rats, subjected to overheating, content of conjugated diens and lipid peroxides was distinctly increased. The decrease in the period of malon dialdehyde induction and in the antioxidative activity of lipids correlated with the alteration in content of peroxidation products. The overheating of animals was accompanied by a decrease in unsaturation of lipids in liver tissue, brain and skeletal muscles due to a decrease in amounts of arachidonic acid and to an increase in content of palmitinic and stearinic acids. In lipids of lungs the unsatur

Alteration of Fatty Acid Profile in Fragile X Syndrome.

Fragile X Syndrome (FXS) is the most prevalent monogenic cause of Autism Spectrum Disorders (ASDs). Despite a common genetic etiology, the affected individuals display heterogenous metabolic abnormalities including hypocholesterolemia. Although changes in the metabolism of fatty acids (FAs) have been reported in various neuropsychiatric disorders, it has not been explored in humans with FXS. In this study, we investigated the FA profiles of two different groups: (1) an Argentinian group, including FXS individuals and age- and sex-matched controls, and (2) a French-Canadian group, including FXS

Fatty acid abnormalities in cystic fibrosis-the missing link for a cure?

The care for cystic fibrosis (CF) has dramatically changed with the development of modulators, correctors, and potentiators of the CFTR molecule, which lead to improved clinical status of most people with CF (pwCF). The modulators influence phospholipids and ceramides, but not linoleic acid (LA) deficiency, associated with more severe phenotypes of CF. The LA deficiency is associated with upregulation of its transfer to arachidonic acid (AA). The AA release from membranes is increased and associated with increase of pro-inflammatory prostanoids and the characteristic inflammation is present be

Myo-inositol deficiency in gerbils: comparative study of the intestinal lipodystrophy in Meriones unguiculatus and Meriones libycus.

Like Mongolian gerbils (Meriones unguiculatus), Meriones libycus develop an intestinal lipodystrophy due to myo-inositol deficiency. Fat accumulation was observed in the intestine of both species when a myo-inositol-deficient diet containing coconut oil was fed to female gerbils. It began in the duodenum and gradually extended to the entire small intestine. Starvation partially removed the accumulated fats. The efficiency of fat absorption was not affected. Most of the accumulated fats were lauric acid-rich triglyceride with the fatty acid composition reflecting the pattern of the intestinal l

Dihomo-γ-Linolenic Acid Elevation with Desaturase Imbalance in Metabolic Dysfunction-Associated Steatotic Liver Disease in a Japanese Health Checkups Cohort: HOZUGAWA Study, a Multi-Omic, Diet Adjusted Analysis.

Background/Objectives: Metabolic dysfunction-associated steatotic liver disease (MASLD) has been linked to dietary fat quality and polyunsaturated fatty-acid metabolism. We evaluated whether dietary n-6 fatty-acid intake, serum dihomo-γ-linolenic acid (DGLA), and desaturase-based indices for Δ5-desaturase (D5D) and Δ6-desaturase (D6D) are associated with MASLD. Methods: We conducted a cross-sectional analysis within the HOZUGAWA health checkup cohort in Japan (n = 289; 100 MASLD, 189 non-MASLD). Participants underwent hepatic ultrasonography, dietary assessment using the Brief Self-Administere

Effects of protein-energy malnutrition and human immunodeficiency virus-1 infection on essential fatty acid metabolism in children.

This report summarizes data on the availability of essential fatty acids (EFAs) and their long-chain polyunsaturated fatty acid (LCPUFA) metabolites in protein-energy malnutrition (PEM), in human immunodeficiency virus-1 (HIV-1) infection for which less information is available, and the combination of both PEM and HIV-1. The contribution of different EFAs and LCPUFAs to the fatty-acid composition of plasma and erythrocyte membrane lipids was found to be reduced in children with PEM in comparison with well-nourished children. In addition to limited dietary EFA supply, reduced bioconversion of E

Severity-Dependent Long-Term Post-Traumatic Changes in the Circulating Oxylipin Profile.

Trauma causes the breakdown of membrane phospholipids and the subsequent degradation of the released polyunsaturated fatty acids (PUFAs) to partially bioactive oxylipins. Here, we screened for circulating PUFAs and oxylipins in patients (n = 34) differing from those of uninjured controls (n = 25) and analyzed their diagnostic potential. Patients were followed up for 1 to 240 h after minor/moderate, severe, and very severe injuries. Of the targeted oxylipins, 13 out of 80 (13/80) were detected in almost all patients and controls. Injury caused a long-term decrease in 9- and 13-hydroxyoctadecadi

The Influence of Fatty Acid Expression Methods on Associations with Cardiometabolic Biomarkers: Evidence from NHANES 2011-2012.

Background/Objectives: Fatty acids (FAs) are critical mediators of human physiology, yet associations between dietary intake, circulating FA levels, and health outcomes remain debated. Circulating FAs are typically reported as a percentage of total FA (% total), whereas absolute concentrations are measured infrequently. Although mathematically related, these metrics provide distinct information. NHANES 2011-2012 uniquely provided serum FA concentrations, enabling direct comparison of concentration and % total within the same individuals. Methods: Serum FAs measured by electron capture negative

Antioxidant and antiradical activities of resorcinarene tetranitroxides.

Resorcinarene oxazines bearing four TEMPO fragments at the wide rim of the macrocycle were prepared through the aminomethylation of resorcinarene octols with 4-amino-TEMPO and formaldehyde. Tetra-TEMPO resorcinarenes are efficient scavengers of 1,1-diphenyl-2-picrylhydrazyl radicals. The model studies revealed that macrocyclic structure and intramolecular hydrogen bonding make considerable contribution to antiradical activity of these compounds. Tetra-TEMPO resorcinarenes show also superoxide dismutase-like activity and efficiently inhibit ABAP-induced peroxidation of linoleic acid.
