The ferroptosis-neuroinflammation feedback loop in traumatic brain injury: mechanisms and therapeutic implications.

Traumatic brain injury (TBI) is a major cause of death and disability worldwide, with secondary injury processes such as ferroptosis and neuroinflammation adding to its detrimental effects. Recent studies reveal a profound bidirectional interaction. On one hand, inflammation drives ferroptosis by disrupting iron homeostasis through the IL-6/Janus kinase 2 (JAK2)/signal transducer and activator of transcription 3 (STAT3)-hepcidin axis, impairing the system Xc-/glutathione (GSH)/GPX4 antioxidant system and promoting immune cell infiltration. Conversely, ferroptotic cells release damage-associate

Immune evasive DNA donors and recombinases license kilobase-scale writing.

Genome-editing technologies that use recombinases to insert kilobase-scale DNA sequences into mammalian genomes canonically require large double-stranded DNA (dsDNA) donors1,2. However, dsDNA molecules evoke problematic and toxic innate immune responses, limiting integration efficiencies and generally constraining applicability to ex vivo or immune-deficient contexts. By harnessing mechanisms of integrative prokaryotic viruses and mobile genetic elements, here we demonstrate that recombinases are compatible with immune evasive circular single-stranded DNA molecules optimally bearing a partial-

Effects of Prunella vulgaris polysaccharide-based O/W emulsions on water retention, protein conformation, and antioxidant properties of ultrasonically treated reduced-fat pork batter.

This study examined how Prunella vulgaris polysaccharides (PVP, 0, 2% and 4% w/w in the O/W emulsion) and ultrasonic treatment (0 and 30 min) affect key quality attributes of reduced-fat pork batter. Combined PVP and ultrasound significantly improved (P < 0.05) emulsifying stability and water-holding capacity, cutting cooking loss from 11.75% to 5.88%. PVP alone enhanced water-protein-polysaccharide interactions: it shortened T21 and T22 relaxation times, raised immobilized water from 86.18% to 93.95%, and lowered overall water mobility. It also altered myofibrillar protein structure, reducing

Dibutyl phthalate exposure-induced AhR activation drives ferroptosis and HMGB1/TLR4-mediated inflammatory liver injury via NRF2-HO-1 signaling.

The widespread presence of plastic pollutants, particularly phthalate esters such as dibutyl phthalate (DBP), poses a significant threat to both ecosystems and human health. Despite the accumulating evidence of DBP toxicity, the mechanisms driving DBP-induced hepatotoxicity remain poorly understood. In this study, an integrated approach combining network toxicology, molecular docking, and multidimensional experimental validation was applied to elucidate the molecular pathways through which DBP causes liver damage. Ferroptosis was identified as a central player in DBP-induced hepatotoxicity. Th

Volumetric variations in the buccal fat pad, parapharyngeal adipose corpus, and orbital fat according to sex, age, and body mass index: an MRI study with segmentation.

The buccal fat pad (BFP), pharyngeal adipose corpus (PAC), and orbital fat are deep fatty compartments of the face that are often considered as non-mobilized fat and are excluded from lipid metabolism. There is considerable clinical inter-individual variability in BFP volume. The PAC volume, which is influenced by weight gain, may be involved in obstructive sleep apnoea syndrome. The aim of this study was to (1) establish well-defined segmentation protocols for the deep fat compartments of the midface; (2) study the volumetric variation in the deep fat compartments of the face according to bod

Unravelling the Signature Follicular Fluid Metabolites in Dairy Cattle Follicles Growing Under Negative Energy Balance: An In Vitro Approach.

The astringent selection criteria for milk-oriented traits in dairy cattle have rendered these animals prone to various metabolic disorders. Postpartum lactational peak and reduced feed intake lead to negative energy balance in cattle. As a compensatory mechanism, cattle start mobilizing fat reserves to meet the energy demand for vital body functions. Consequently, diminished glucose concentrations and elevated ketone body levels lead to poor ovarian function. The impaired follicular development and subpar oocyte quality diminish the conception rates, which poses significant economic repercuss

Predicting Collision Cross-Section Values for Small Molecules through Chemical Class-Based Multimodal Graph Attention Network.

Libraries of collision cross-section (CCS) values have the potential to facilitate compound identification in metabolomics. Although computational methods provide an opportunity to increase library size rapidly, accurate prediction of CCS values remains challenging due to the structural diversity of small molecules. Here, we developed a machine learning (ML) model that integrates graph attention networks and multimodal molecular representations to predict CCS values on the basis of chemical class. Our approach, referred to as MGAT-CCS, had superior performance in comparison to other ML models

Emerging Mechanisms of Skeletal Muscle Homeostasis and Cachexia: The SUMO Perspective.

Mobility is an intrinsic feature of the animal kingdom that stimulates evolutionary processes and determines the biological success of animals. Skeletal muscle is the primary driver of voluntary movements. Besides, skeletal muscles have an immense impact on regulating glucose, amino acid, and lipid homeostasis. Muscle atrophy/wasting conditions are accompanied by a drastic effect on muscle function and disrupt steady-state muscle physiology. Cachexia is a complex multifactorial muscle wasting syndrome characterized by extreme loss of skeletal muscle mass, resulting in a dramatic decrease in li

Effects of Palmitoylethanolamide on Neurodegenerative Diseases: A Review from Rodents to Humans.

Palmitoylethanolamide (PEA) stands out among endogenous lipid mediators for its neuroprotective, anti-inflammatory, and analgesic functions. PEA belonging to the N-acetylanolamine class of phospholipids was first isolated from soy lecithin, egg yolk, and peanut flour. It is currently used for the treatment of different types of neuropathic pain, such as fibromyalgia, osteoarthritis, carpal tunnel syndrome, and many other conditions. The properties of PEA, especially of its micronized or ultra-micronized forms maximizing bioavailability and efficacy, have sparked a series of innovative research

Differential expression of perilipin 2 and 5 in human skeletal muscle during aging and their association with atrophy-related genes.

Sarcopenia, the progressive loss of muscle mass and strength, is a phenomenon characterizing human aging whose etiology is still not clear. While there is increasing evidence for the influence of inter-muscular adipose tissue infiltration in the development of sarcopenia, much less is known about a possible role for intra-muscular triglycerides (IMTG). IMTG accumulate in form of lipid droplets decorated by proteins such as Perilipins (Plins). In skeletal muscle the most abundant are Plin2 and Plin5. In this study we compared the expression of these two Plins in Vastus lateralis muscle samples

High-mobility group box 1, oxidative stress, and disease.

Oxidative stress and associated reactive oxygen species can modify lipids, proteins, carbohydrates, and nucleic acids, and induce the mitochondrial permeability transition, providing a signal leading to the induction of autophagy, apoptosis, and necrosis. High-mobility group box 1 (HMGB1) protein, a chromatin-binding nuclear protein and damage-associated molecular pattern molecule, is integral to oxidative stress and downstream apoptosis or survival. Accumulation of HMGB1 at sites of oxidative DNA damage can lead to repair of the DNA. As a redox-sensitive protein, HMGB1 contains three cysteine

Peroxisomal multifunctional protein-2 deficiency causes motor deficits and glial lesions in the adult central nervous system.

In humans, mutations inactivating multifunctional protein-2 (MFP-2), and thus peroxisomal beta-oxidation, cause neuronal heterotopia and demyelination, which is clinically reflected by hypotonia, seizures, and death within the first year of life. In contrast, our recently generated MFP-2-deficient mice did not show neurodevelopmental abnormalities but exhibited aberrations in bile acid metabolism and one of three of them died early postnatally. In the postweaning period, all survivors developed progressive motor deficits, including abnormal cramping reflexes of the limbs and loss of mobility,

Assignment of Tangier disease to chromosome 9q31 by a graphical linkage exclusion strategy.

A low level of high density lipoprotein (HDL) cholesterol is a strong predictor of ischaemic heart disease (IHD) and myocardial infarction. One cause of low HDL-cholesterol is Tangier disease (TD), an autosomal codominant inherited condition first described in 1961 in two siblings on Tangier Island in the United States of America. Apart from low HDL-cholesterol levels and an increased incidence of atherosclerosis, TD is characterized by reduced total cholesterol, raised triglycerides, peripheral neuropathy and accumulation of cholesteryl esters in macrophages, which causes enlargement of the l

Exploration of Colonic Mucus as A Dissolution Compartment for Orally Administered Drugs.

Colonic mucus has gained interest as a dissolution compartment in the large intestine due to the large fraction of bound water. Notably, the mucus layer contains more water than what is freely mobile within the colonic lumen. Here, we studied solubility of a series of structurally and physicochemically diverse orally administered drugs. In total, 15 compounds were measured for their solubility in an artificial colonic mucus solution. Equilibrium solubility was measured using the traditional shake-flask method at 37°C in several important media for dissolution studies in the colon: the mucus so

Tryptanthrin Nanoliposomal Lotion as a Corticosteroid-Free Strategy for Safe and Effective Treatment of Atopic Dermatitis.

Consuming a high-fat diet (HFD) is widely recognized to cause obesity and result in chronic brain inflammation that impairs cognitive function. Repetitive transcranial magnetic stimulation (rTMS) has shown effectiveness in both weight loss and cognitive improvement, although the exact mechanism is still unknown. Our study examined the effects of rTMS on the brain and intestinal microecological dysfunction. rTMS successfully reduced cognitive decline caused by an HFD in behavioral assessments involving the Y maze and novel object recognition. This was accompanied by an increase in the number of

Metabolic footprint of microplastics and nanoplastics: From environmental exposure to metabolic diseases.

Microplastics and Nanoplastics (MPs/NPs), as emerging environmental pollutants, are characterized by their resistance to degradation, high mobility, and strong adsorption capacity. They are widely distributed across global environments and enter the human body through multiple pathways, where they interfere with metabolic health. This review introduces the concept of "metabolic footprint" to systematically analyze the environmental behavior of MPs/NPs, human exposure routes, and the associations between MPs/NPs, metabolic pathways, and metabolic diseases. MPs/NPs can induce energy metabolism d

Viral Capsid-Membrane Interactions Propel Non-Brownian Movements of Non-enveloped Reoviruses during Entry.

Understanding how non-enveloped viruses breach host cell membranes is critical for developing strategies to block viral entry, a key step in infection. Despite extensive study, how viral capsids and host lipid membranes dynamically cooperate during membrane penetration remains poorly defined. Here, using reovirus as a model non-enveloped virus and planar model membranes, we identify previously unrecognized non-Brownian membrane motions of infectious subvirion particles (ISVPs) by single-virus tracking. We then integrate experiments with computational modeling to dissect the stepwise, processiv

Aspects of plant intelligence (ii).

Plant intelligence was earlier described as adaptively variable behavior during the lifetime of the individual plant (Trewavas 2003). This general definition for animals and plants (biological intelligence) is now justified from generic terms and emphasis on adaptive behavior. Plant intelligence can also be defined from dictionary definitions of intelligence. Artificial intelligence has been indicated as adaptive behavior. Prominent psychological opinion now indicates that human intelligence is adaptive: IQ does not measure human intelligence. The uniformity of intelligence definitions negates

High-resolution untargeted metabolomics reveals altered lysophosphatidylcholine, palmitoleic acid, triglycerides, and isopulegone caffeate associated with ovarian injury in BTBR ob/ob mice.

Diabetes mellitus adversely affects female reproductive health, leading to ovarian dysfunction and infertility. In this study, we investigated metabolic alterations in the ovaries of BTBR ob/ob mice, a well-established type 2 diabetes mellitus model, using high-resolution untargeted metabolomics combined with ion mobility spectrometry. Our analysis identified significant changes in key lipid metabolites, including polyunsaturated lysophosphatidylcholine, palmitoleic acid, triglyceride, and isopulegone caffeate, which are closely linked to inflammatory and oxidative stress pathways. Notably, th

Research progress on the mechanism of chondrocyte ferroptosis in osteoarthritis.

Osteoarthritis (OA) is a chronic, progressive joint disease characterized by degenerative changes in articular cartilage, subchondral bone sclerosis, accompanied by synovitis and chondrocyte apoptosis. With the aging of society, it has become one of the major factors endangering the mobility of middle-aged and elderly people worldwide. The pathogenesis of OA remains unclear, and current treatments can only control symptoms without effectively repairing damaged cartilage. Ferroptosis, a novel form of programmed cell death proposed and confirmed in 2012, is characterized by iron-dependent accumu
