Immobilisation of lipase on the surface of magnetic nanoparticles and non-porous glass beads for regioselective acetylation of prednisolone.

Magnetic nanoparticles (MNPs) were prepared by co-precipitation of Fe+3/Fe+2 (2:1; molar ratio) with a mean size of 40 ± 5 nm. Scanning electron microscopy, dynamic light scattering, X-ray diffraction, vibrating sample magnetometer and thermogravimetric analysis were used to characterise the MNPs. The MNPs and also hydroxylated nonporous glass beads were similarly functionalised with the aid of glutaraldehyde to be cross-linked to lipase originated from Thermomyces lanuginosus (TLL). The yield and efficiency for immobilised lipase on the MNPs were calculated as 72 ± 2.4 and 63 ± 3.5%, whereas

Comparison between magnetic and non magnetic multi-walled carbon nanotubes-dispersive solid-phase extraction combined with ultra-high performance liquid chromatography for the determination of sulfonamide antibiotics in water samples.

In this manuscript, a new method based on the use of off-line dispersive solid-phase extraction (dSPE) combined with ultra-high performance liquid chromatography with diode-array detection was developed to determine 11 sulfonamide antibiotics (sulfanilamide, sulfacetamide, sulfadiazine, sulfathiazole, sulfamerazine, sulfadimidin, sulfamethoxypyridazine, sulfadoxine, sulfamethoxazole, sulfisoxazole and sulfadimethoxine) in mineral waters with different mineral content. For this purpose, pristine multi-walled carbon nanotubes (MWCNTs) and magnetic-MWCNTs (m-MWCNTs) were used as sorbents. Magneti

Bisphosphonate-anchored PEGylation and radiolabeling of superparamagnetic iron oxide: long-circulating nanoparticles for in vivo multimodal (T1 MRI-SPECT) imaging.

The efficient delivery of nanomaterials to specific targets for in vivo biomedical imaging is hindered by rapid sequestration by the reticuloendothelial system (RES) and consequent short circulation times. To overcome these two problems, we have prepared a new stealth PEG polymer conjugate containing a terminal 1,1-bisphosphonate (BP) group for strong and stable binding to the surface of ultrasmall-superparamagnetic oxide nanomaterials (USPIOs). This polymer, PEG(5)-BP, can be used to exchange the hydrophobic surfactants commonly used in the synthesis of USPIOs very efficiently and at room tem

Epidermal growth factor receptor-targeted ultra-small superparamagnetic iron oxide particles for magnetic resonance molecular imaging of lung cancer cells in vitro.

BACKGROUND: Magnetic resonance (MR) molecular imaging can detect abnormalities associated with disease at the level of cell and molecule. The epidermal growth factor receptor (EGFR) plays an important role in the development of lung cancer. This study aimed to explore new MR molecular imaging targeting of the EGFR on lung cancer cells. METHODS: We attached ultra-small superparamagnetic iron oxide (USPIO) particles to cetuximab (C225) anti-human IgG using the carbodiimide method. We made the molecular MR contrast agents C225-USPIO and IgG-USPIO, the latter as a control reagent, and determined

Magnetic graphene nanosheets based electrochemiluminescence immunoassay of cancer biomarker using CdTe quantum dots coated silica nanospheres as labels.

A highly sensitive electrochemiluminescence (ECL) immunosensor for the detection of prostate specific antigen (PSA) was designed using biofunctionalized magnetic graphene nanosheets (G@Fe(3)O(4)) as immunosensing probes and CdTe quantum dots coated silica nanospheres (Si/QDs) as signal amplification labels. In this work, a sandwich-type immunosensor was fabricated, which was assembled on the surface of indium tin oxide glass (ITO). The analyte was detected in a home-made flow injection ECL (FI-ECL) cell through the immunosensor. Owing to the signal amplification of G@Fe(3)O(4) composite and Si

Functional MRI using intravascular contrast agents: detrending of the relative cerebrovascular (rCBV) time course.

In pharmacological fMRI experiments in animal models, blood pool contrast agents may be used to map cerebral blood volume change as a surrogate for neural activation. When the background signal drift due to contrast agent washout is non-negligible over the duration of the signal changes of interest, time-course detrending is essential for accurate interpretation of the experiment. Detrending approaches based on estimation of the background signal from a baseline period of the time course prior to pharmacological (or functional) challenge were evaluated with the aim of identifying a robust meth

Quantitative analysis of cyclosporine A in whole blood samples of liver transplant recipients using a stir bar-assisted magnetic dispersive solid phase extraction combined with HPLC-MS/MS.

Cyclosporine A is an immunosuppressive drug which is used in the organ transplantation and treatment of autoimmune diseases. Also, it has been approved to be used as a hepatoprotective agent in the liver transplant recipients. Nephrotoxicity is the major toxic side effect of cyclosporine and due to its narrow therapeutic index, the doses must be adjusted individually. In the current research, a stir bar-assisted magnetic dispersive solid phase extraction method followed by high performance liquid chromatography-tandem mass spectrometry has been introduced and validated for the quantitative ana

Magnesium-Enhanced Magnetic Hyperthermia for Triggering Vaccine-Like Antitumor Immunotherapy.

Cancer immunotherapies, such as programmed cell death protein 1 (PD-1) inhibitors, cancer vaccines, and chimeric antigen receptor T cells (CAR-T cells), have become first-line treatments for multiple tumors by eliciting antitumor immunity. However, their limited efficacy in immunosuppressive tumors, particularly Head and Neck Squamous Cell Carcinoma (HNSCC), underscores the urgent need for versatile therapeutics. Hereinto, we develop an injectable hydrogel incorporating Mg microparticles and Fe3O4 magnetic nanoparticles (Mg/Fe3O4@Gel) to enhance antitumor immunotherapy, achieving potent vaccin

Magnetic nanoparticle-based method for microorganism concentration in sterile body fluids: Validation and clinical applications.

Rapid and accurate detection of microorganisms in sterile body fluids, particularly cerebrospinal fluid (CSF), is crucial for effective diagnosis and treatment. Conventional methods, such as centrifugation, may result in low microbial recovery and false negatives, limiting diagnostic accuracy. An alternative, efficient, and accessible microbial concentration method is needed. This study evaluates a nanoparticle-based microbial concentration method to enhance pathogen recovery from CSF. The method was optimized for interaction time (1 min) and nanoparticle dosage (0.01 g/mL) using standard micr

CIMNE-CRISPR: A novel amplification-free diagnostic for rapid early detection of African Swine Fever Virus.

African Swine Fever Virus (ASFV) is a highly contagious pathogen with nearly 100% mortality in swine, causing severe global economic loss. Current detection methods rely on nucleic acid amplification, which requires specialized equipment and skilled operators, limiting accessibility in resource-constrained settings. To address these challenges, we developed the Covalently Immobilized Magnetic Nanoparticles Enhanced CRISPR (CIMNE-CRISPR) system. This amplification-free diagnostic system seamlessly combines target recognition, sequence-specific enrichment, and signal generation. This approach us

In situ supported Pd NPs on biodegradable chitosan/agarose modified magnetic nanoparticles as an effective catalyst for the ultrasound assisted oxidation of alcohols and activities against human breast cancer.

In this content, a green approach for the ultrasound promoted in situ immobilization of Pd NPs over biodegradable chitosan/agarose modified ferrite NP (Fe3O4@CS-Agarose/Pd) is developed. The structural and physicochemical features of the material were estimated using advanced analytical techniques like FT-IR, ICP-OES, FESEM, EDS, XRD, TEM and VSM. The magnetic material was catalytically explored in the oxidation of alcohols under ultrasonic waves. Sonication had a significant role in enhancing the catalytic performance in the alcohol's oxidation as compared to conventional heating. The heterog

Enzymatic degradation of ginkgolic acids by laccase immobilized on core/shell Fe3O4/nylon composite nanoparticles using novel coaxial electrospraying process.

Enzyme immobilization can increase enzyme reusability to reduce cost of industrial production. Ginkgo biloba leaf extract is commonly used for medical purposes, but it contains ginkgolic acid, which has negative effects on human health. Here, we report a novel approach to solve the problem by degrading the ginkgolic acid with immobilized-laccase, where core/shell composite nanoparticles prepared by coaxial electrospraying might be first applied to enzyme immobilization. The core/shell Fe3O4/nylon 6,6 composite nanoparticles (FNCNs) were prepared using one-step coaxial electrospraying and can b

GE11-PDA-Pt@USPIOs nano-formulation for relief of tumor hypoxia and MRI/PAI-guided tumor radio-chemotherapy.

Radio-chemo combination therapy has synergetic therapeutic effects on tumors. However, the tumor microenvironment, e.g. hypoxia and elevated H2S levels, limits its treatment efficacy. In this study, we developed a cisplatin-loaded, poly dopamine-coated and GE11 peptide-conjugated multi-functional theranostic system (GE11-PDA-Pt@USPIOs) based on poly acrylic acid-coated ultra-small superparamagnetic iron oxide nanoparticles (PAA@USPIOs) for modulation of the tumor hypoxic microenvironment and magnetic resonance imaging/photoacoustic imaging (MRI/PAI) guided radio-chemotherapy of tumors. Monitoring of impaired phagocytic function of Kupffer cells in an obstructive cholangitis rat model using superparamagnetic iron oxide MRI and contrast-enhanced ultrasound.

BACKGROUND: Kupffer cells (KC) have an important role in the host defense in obstructive cholangitis. Non-invasive monitoring of phagocytic function of KC is pivotal. Several studies showed the possibility of non-invasive monitoring of phagocytic function of KC using superparamagnetic iron oxide-enhanced magnetic resonance imaging (SPIO-MRI) or contrast-enhanced ultrasound (CEUS). PURPOSE: To investigate the serial change of KC function using SPIO-MRI and CEUS and whether the SPIO-MRI parameter correlates with the CEUS parameter in obstructive cholangitis rat models. MRI/optical dual-modality imaging of vulnerable atherosclerotic plaque with an osteopontin-targeted probe based on Fe3O4 nanoparticles.

Rupture of vulnerable atherosclerotic plaque is the major pathological cause of luminal thrombosis in acute coronary syndromes. Since foamy macrophages have been identified as a prominent component in vulnerable atherosclerotic lesions and osteopontin (OPN) is reported to be highly expressed in foamy macrophages, OPN could be a potential target for vulnerable atherosclerotic plaque imaging. The current study designed an OPN-specific MRI/optical dual-modality probe to detect vulnerable plaques. Fluorescence imaging revealed that 24 h after injection of the Cy5.5-OPN-DMSA-MNPs (COD-MNPs), the at
