Expression of the familial cardiac valvular dystrophy gene, filamin-A, during heart morphogenesis.

Myxoid degeneration of the cardiac valves is a common feature in a heterogeneous group of disorders that includes Marfan syndrome and isolated valvular diseases. Mitral valve prolapse is the most common outcome of these and remains one of the most common indications for valvular surgery. While the etiology of the disease is unknown, recent genetic studies have demonstrated that an X-linked form of familial cardiac valvular dystrophy can be attributed to mutations in the Filamin-A gene. Since these inheritable mutations are present from conception, we hypothesize that filamin-A mutations presen

Palatal fusion - where do the midline cells go? A review on cleft palate, a major human birth defect.

Formation of the palate, the organ that separates the oral cavity from the nasal cavity, is a developmental process characteristic to embryos of higher vertebrates. Failure in this process results in palatal cleft. During the final steps of palatogenesis, two palatal shelves outgrowing from the sides of the embryonic oronasal cavity elevate above the tongue, meet in the midline, and rapidly fuse together. Over the decades, multiple mechanisms have been proposed to explain how the superficial mucous membranes disappear from the contact line, thus allowing for normal midline mesenchymal confluen

Epithelial-mesenchymal interaction in gastric mucosal restoration.

In this review article we discuss the role of growth factors in gastric ulcer healing using an in vitro wound repair model with gastric epithelial and mesenchymal cells. Several growth factors accelerate gastric epithelial and mesenchymal wound healing in vitro with acceleration of cell migration and proliferation. Epidermal growth factor, transforming growth factor-alpha (TGFalpha), hepatocyte growth factor, and insulin accelerate predominantly gastric epithelial wound healing; and TGFbeta and basic fibroblast growth factor predominantly accelerate gastric mesenchymal wound healing. Platelet-

Non-nuchal-type fibroma associated with Gardner's syndrome. A hitherto-unreported mesenchymal tumor different from fibromatosis and nuchal-type fibroma.

We describe a unique benign mesenchymal tumor in paraspinal location in a 13-year-old patient with Gardner's syndrome. The Gardner's syndrome in this patient consisted of multiple (more than 100) polyps throughout the entire colon with most in the cecum and rectum, three osteomas in the frontal area of the skull and one in the third right rib, and multiple superficial skin tumors. One of these cutaneous tumors was excised and histologically diagnosed as an epidermal cyst. Both father and uncle of this patient suffered from Gardner's syndrome as well. Microscopically the mesenchymal tumor was h

Development of the submandibular gland and its closer neighboring structures in human embryos and fetuses of 19-67 mm CRL.

The fetal development and arrangement of the human submandibular gland was studied by means of serial sections of human embryos and fetuses ranging from 19 mm to 67 mm CRL. Computer assisted 3-dimensional reconstruction leads to the following observations: 1. The orifice of the submandibular duct is located in the medial paralingual sulcus. 2. There is evidence that the extension and location of WHARTON's duct is influenced by the surrounding structures. 3. The surface of the submandibular gland primordium shows impressions of the neighboring structures. 4. The glandular tissue is encapsulated

The effects of colchicine on the ultrastructure of odontogenic cells in the common skate, Raja erinacae.

Ultrastructural alterations induced by colchicine were investigated to determine the secretory activities of odontogenic cells during formation of tooth enameloid matrix in skates. Treated skate inner dental epithelial (IDE) cells did not display dilated cisternae of the granular endoplasmic reticulum (GER) nor accumulate Golgi-associated secretory granules at any dose level or time interval examined. This response was markedly different from that observed in teleost IDE cells synthesizing the enameloid collagen matrix. Treated skate IDE cells did show increased accumulations of glycogen-conta

Role of actin filaments in shape formation of mesenteric mesothelial cells of the bullfrog.

The role of actin filaments in the development of cellular shape in the mesenteric mesothelium of the bullfrog was studied by using a simple, new technique for making en face preparations of mesothelial sheets. By using these mesothelial cell preparations, the distribution of actin was determined by means of fluorescence microscopy with 7-nitrobenz-2-oxa-1,3-diazole (NBD)-phallacidin and that of myosin by means of immunofluorescence microscopy. Although fluorescence produced by both NBD-phallacidin and antimyosin staining was found exclusively along the margins of the cells, its intensity was

The spatial phenotype of genotypically distinct meningiomas demonstrate potential implications of the embryology of the meninges.

Meningiomas are the most common primary brain tumor and their incidence and prevalence is increasing. This review summarizes current evidence regarding the embryogenesis of the human meninges in the context of meningioma pathogenesis and anatomical distribution. Though not mutually exclusive, chromosomal instability and pathogenic variants affecting the long arm of chromosome 22 (22q) result in meningiomas in neural-crest cell-derived meninges, while variants affecting Hedgehog signaling, PI3K signaling, TRAF7, KLF4, and POLR2A result in meningiomas in the mesodermal-derived meninges of the mi…

Novel Mode of Defective Neural Tube Closure in the Non-Obese Diabetic (NOD) Mouse Strain.

Failure to close the neural tube results in birth defects, with severity ranging from spina bifida to lethal anencephaly. Few genetic risk factors for neural tube defects are known in humans, highlighting the critical role of environmental risk factors, such as maternal diabetes. Yet, it is not well understood how altered maternal metabolism interferes with embryonic development, and with neurulation in particular. We present evidence from two independent mouse models of diabetic pregnancy that identifies impaired migration of nascent mesodermal cells in the primitive streak as the morphogenet…

Cowden syndrome with Lhermitte- Duclos disease presenting as ataxia.

Cowden syndrome or multiple hamartoma syndrome is a rare genodermatosis of autosomal dominant inheritance characterized by multiple hamartomatous lesions of ectodermal, mesodermal and endodermal origin. A 45-year-old man presented to us with a history of dural arteriovenous fistula and intracerebral bleed in the past with gradually progressive difficulty in walking. Magnetic resonance imaging (MRI) of the brain showed a heterogeneous lesion in the cerebellum which was diagnosed as adult Lhermitte-Duclos disease which is considered a component of Cowden syndrome. On examination we found florid …

Development of the carapacial ridge: implications for the evolution of genetic networks in turtle shell development.

Paleontologists and neontologists have long looked to development to understand the homologies of the dermal bones that form the "armor" of … genetic networks in the development of the turtle shell. This work establishes a new role for this mesodermal compartment and highlights the importance of changes in genetic regulation in the evolution of morphology.

Endoscopic resection of primary rectal teratoma.

A 76-year-old female was noted to have a rectal mass on evaluation for postmenopausal bleeding. There was no history of abdominal pain, constipation, or rectal bleeding. Flexible sigmoidoscopy revealed a 3-cm pedunculated rectal mass at 10 cm. Hair fibers were visible through the smooth glistening surface. At colonoscopy, the polyp was removed by snare polypectomy. Histology showed epidermal, mesodermal, and endodermal components diagnostic of benign cystic teratoma. Subsequent total abdominal hysterectomy and bilateral salpingo-oophorectomy revealed endometrial carcinoma and postmenopausal ov…

Biomechanical considerations of high myopia: Part I--Physiological characteristics.

Prior to a discussion of biomechanical considerations in high myopia, a review of anatomical and physiological patterns in high myopia is presented. A scleral pathogenesis or mesodermal hypothesis of the anomaly is presented with the inclusion of supporting evidence. Retinal, choroidal, and vitreous malformations in high myopia also are highlighted. Notable is a pattern consistent with the concept of gradual stretching of the vitreous chamber. The association of high myopia with other diseases is shown, with special attention being given to the concomitant features of high myopia and glaucoma.…

Bilateral choanal atresia associated with malformation of the anterior skull base: embryogenesis and clinical implications.

A number of craniofacial and systemic malformations have been described in association with choanal atresia. We report a case of bilateral choanal atresia associated with congenital absence of the cribriform plate, crista galli, and perpendicular plate of the ethmoid bone. The anterior skull base defect was detected by using high-resolution computed tomography with three-dimensional reconstructions. The findings support the mesodermal flow theory of choanal atresia, in which there is excess migration of neural crest cells into the developing nasal septum and posterior choanae. This occurs at t…

[Tumors of the anterior mediastinum--surgical treatment and prognosis].

This paper reports on 96 mediastinal tumors situated exclusively or mainly in the anterior mediastinum. Early surgical intervention is alway … trosternal goiters, none died of a cause connected with the goiter and nine out of 11 patients with mesodermal tumors survived. The indication for surgery should be established extensively, since one third of the tumors is malignant and the surgical lethality (1.8%) is very low.

TGF-β Signaling in Cranial Neural Crest Affects Late-Stage Mandibular Bone Resorption and Length.

Malocclusions are common craniofacial malformations which cause quality of life and health problems if left untreated. Unfortunately, the current treatment for severe skeletal malocclusion is invasive surgery. Developing improved therapeutic options requires a deeper understanding of the cellular mechanisms responsible for determining jaw bone length. We have recently shown that neural crest mesenchyme (NCM) can alter jaw length by controlling recruitment and function of mesoderm-derived osteoclasts. Transforming growth factor beta (TGF-β) signaling is critical to craniofacial development by d…
