Role of Wnts in prostate cancer bone metastases.

Prostate cancer (CaP) is unique among all cancers in that when it metastasizes to bone, it typically forms osteoblastic lesions (characteriz … ression decreases allowing for unmasking of Wnt's osteoblastic activity and ultimately resulting in osteosclerosis at the metastatic site. We believe that DKK-1 is one of the switches that transitions the CaP bone metastasis activity from osteolytic to osteoblastic. Wnt/DKK-1 activity fits a model of CaP-induced bone remodeling occurring in a continuum composed …

[Zonography in solitary bone cysts].

The zonography method is gaining more and more users today and arouses a definite interest of specialists in medicine. This is due to the peculiarities of the method and the tremendous clinical possibilities which make it possible to improve and accelerate the diagnosis not only of diseases of the lungs and kidneys, but also of some segments of the bone system. The availability of zonography allows it to be used successfully in the diagnosis of various, pathological among others, types of bone fractures, tumors of the skeleton and, therefore, makes it possible to watch the reparative manifesta

Anterior chest wall involvement in patients with pustulosis palmoplantaris.

With the aim of determining the frequency and radiographic features of anterior chest wall involvement in patients with pustulosis palmoplantaris, a questionnaire was sent to 107 patients. Ninety-three patients returned the questionnaire, five of whom were excluded from further analysis due to the appearance of psoriatic lesions. Twenty-five (28%) of the remaining 88 patients reported pain and/or swelling of joints or bones in the anterior chest wall. All were examined radiographically, using tomography, and a group of 20 patients without anterior chest wall complaints were examined similarly.

Osteonecrosis of bone. Current concepts as to etiology and pathogenesis.

Experimental and some clinical evidence suggest that one or more mechanisms, alone or in combination, induce bone necrosis. These include marrow cell hypertrophy, microembolic phenomena, and lipid-induced osteocyte necrosis. Such events may result in local inflammatory exudates and/or vascular impedence. Either may effect an increase in the hydrostatic pressures in bone with the potential to limit blood flow and magnify the initial insult. Further experimental investigations are necessary to gain knowledge of etiologic mechanisms and a rational means of prophylaxis or treatment.

Terminal phalangeal sclerosis in sickle cell disease.

Hand films of 100 patients with sickle cell disease were analyzed in order to establish the type, distribution, and incidence of terminal phalangeal sclerosis. Fifty hand films from patients with no known sickle cell disease matched for age and race were also studied. Twenty-four of 100 patients with sickle cell disease, with 14 less than 20 years old, showed clear evidence of terminal phalangeal sclerosis, while only 10% of the controls were positive. The incidence was significantly different from the control group (chi 2 = 4.1949, d.f. = 1, p less than 0.05) suggesting that terminal phalange

Arthroprostheses without cement in osteoarthrosis of the hip (preliminary report) (16 cases of Sbarbaro arthroprosthesis).

The authors report the results obtained with arthroprostheses without cement. In this preliminary report they examine sixteen cases treated with Sbarbaro's arthroprosthesis from 1972 to 1976, at the Orthopaedic Division of the Specialized Regional Hospital, Udine. They stress the efficacy of this kind of arthroprosthesis, which can be used in subjects under the age of sixty years who have severe bilateral lesions, especially of the femoral head. The operation is contraindicated when there is severe dysplasia of osteoporosis of the acetabulum. Weight bearing must be avoided for six months, the

[Is gonarthrosis with varus deformity a clinical entity].

Studies were conducted in a group of patients with varose deformity. The present paper gives the results of indicidual bone metabolism studies conducted on 663 healthy controls and patients with articular disease without genu varum or with varose deformity. In subjects with genu varum intravenous administration of calcium causes the urinary elimination of hydroxyproline and nitrogen to fall. Among the groups under investigation this group of patients excretes the least amount of urine, has a relatively low calciuria and shows the highest retention rate of intravenously administered calcium. Generation and Characterization of an Osteoclastic Cell Model of Gaucher's Bone Disease.

Gaucher disease (GD) is caused by recessive mutations in the GBA1 gene (Glucosylceramidase beta 1) generating lysosomal accumulation of glucosylceramides. Bone phenotypes related to GD include Erlenmeyer flask deformity, osteosclerosis, osteonecrosis, osteopenia, and bone pain. Lysosomes modulate osteoclast differentiation, playing a fundamental role in osteoclastogenesis and bone resorption. However, the role of GBA1 mutations in the osteoclast function remains poorly understood. We aimed to generate and characterize an in vitro osteoclast cell model of GD. The Gba1 gene was knocked out with …

Reported bone abnormalities in CGL1 and CGL2 included osteolytic lesions, bone cysts, osteosclerosis, and pseudo-osteopoikilosis, predominantly in long bones. Bone mineral density (BMD) was generally normal to high at trabecular sites. In contrast, FPLD2 was characterized by reduced BMD at cortical sites and lower trabecular bone score, suggesting …

Effect of dense bone islands on orthodontic tooth movement and root resorption during space closure with fixed orthodontic appliances: A longitudinal study on panoramic radiography.

INTRODUCTION: Idiopathic osteosclerosis (IO), also known as "dense bone island," is a localized and well-defined radiopaque lesion often close to the radicular area of teeth. The present study investigated whether IO affected orthodontic tooth movement and root resorption during the closure of extraction spaces. METHODS: A total of 1624 pretreatment and posttreatment panoramic radiographs were retrospectively screened. Forty adolescents and young adults who received orthodontic treatment with symmetrical mandibular premolar extraction and presented with unilateral IO were selected. Extraction…

Notably, pacritinib prevented the splenomegaly, reticulin fibrosis and osteosclerosis observed in untreated KO mice. Herein, we report a cas … ISCUSSION: The case is unusual as the fracture occurred despite normal bone density and significant osteosclerosis, which was previously considered protective against fractures. Additionally, rather than the spine, the fracture occurred in the hip, which is an uncommon site for mastocytosis-induced fractures. CONCLUSION: Mastocytosis is a rare cause of osteopor …

Skeletal Fluorosis: A Case of Inhalant Abuse Leading to a Diagnosis of Colon Cancer.

Skeletal fluorosis is a long-term bone disease that develops when prolonged fluoride toxicity leads to osteosclerosis and bone deformities that result in crippling pain and debility. The disease is endemic to many countries due to environmental or industrial exposures. However, rare cases in the United States have been reported from various causes including heavy toothpaste ingestion, excessive tea consumption, voriconazole use, and inhalant abuse. Here, we present a case of a 41-year-old man who presented for weight loss and severe joint pains due to bony sclerotic lesions found on X-rays. So…

A case report of pycnodysostosis with atypical femur fracture diagnosed by next-generation sequencing of candidate genes.

RATIONALE: Pycnodysostosis is a rare autosomal recessive skeletal dysplasia characterized by short stature, craniofacial dysmorphism, acro-osteolysis, osteosclerosis, and brittle bone with poor healing. Pycnodysostosis results from the deficient activity of cathepsin K, a lysosomal cysteine protease that is encoded by CTSK. PATIENT CONCERNS: We report a Korean adult patient with pycnodysostosis and atypical femur fracture whose diagnosis was confirmed by next-generation sequencing (NGS) of candidate genes. A 41-year-old female patient was presented with a left femur fracture after falling dow…
