Perioperative tislelizumab plus lenvatinib treatment for resectable hepatocellular carcinoma at high risk of recurrence: single-arm phase II trial.

Postoperative recurrence significantly impacts long-term survival in patients with resectable hepatocellular carcinoma (HCC). In this single-arm, phase II trial (NCT04834986), 27 patients with resectable HCC at high recurrence risk received perioperative tislelizumab (anti-PD-1) plus lenvatinib (multi-kinase inhibitor). Co-primary endpoints were safety and feasibility. Secondary endpoints included objective response rate (ORR), 1-year and 2-year disease-free survival (DFS), median DFS, and pathologic complete response (pCR). Post-hoc exploratory machine learning analyses identified potential p

USP33 contributes to EMT and Lenvatinib resistance by enhancing eEF1A1-mediated ILEI protein synthesis in hepatocellular carcinoma.

Lenvatinib resistance represents a primary cause of treatment failure in patients with advanced hepatocellular carcinoma (HCC), while its underlying molecular mechanisms remain incompletely understood. Using a CRISPR knockout library targeting human ubiquitination-related proteins, we identified ubiquitin-specific protease 33 (USP33) as a key factor of lenvatinib resistance in HCC. Transcriptome sequencing further demonstrated that USP33 induces epithelial-mesenchymal transition (EMT), and its downregulation suppresses EMT and enhances lenvatinib sensitivity in HCC cells. Through proteomic ana

Quizartinib-induced resistance drives clonal emergence of MV4-11 cells with molecular alterations enabling multidrug antileukemic escape.

FLT3 inhibitors have become a cornerstone in the treatment of FLT3-mutated acute myeloid leukemia (AML), however, durable clinical responses are frequently limited by the emergence of acquired resistance. In this study, we established and comprehensively characterized a quizartinib-resistant FLT3-ITD AML model to elucidate the molecular and functional mechanisms underlying therapeutic failure. Prolonged exposure of MV4-11 cells to escalating concentrations of quizartinib resulted in the selection of quizartinib resistant clones (MV4-11QR), displaying an increase in IC50 and a shift from cytoto

Intraocular delivery of crystalline sorafenib provides sustained, potent inhibition of wet age-related macular degeneration.

Neovascular age-related macular degeneration (nAMD) is a leading cause of blindness worldwide. Anti-VEGF agents are very beneficial for nAMD, with intravitreal (IVT) delivery becoming the gold standard for treatment of nAMD and other retinal conditions. Incomplete response remains a major challenge, as does the requirement for high-frequency, repeat IVT injections, which pose a significant burden on patients. Many potential drug candidates have attractive pharmacodynamic profiles but are limited by their insolubility, precluding IVT administration. These include tyrosine kinase inhibitors (TKI

Hepatic stellate cells promote structural and functional remodeling of three-dimensional hepatocellular carcinoma coculture spheroids.

BACKGROUND: Hepatic stellate cells (HSCs) are major regulators of the hepatocellular carcinoma (HCC) tumor microenvironment, contributing to extracellular matrix remodeling, tumor progression, and therapeutic resistance. Although three-dimensional (3D) coculture spheroid models better represent tumor-stroma interactions than conventional monocultures, the impact of HSCs on spheroid architecture and function remains incompletely characterized. This study investigated how HSCs influence the structural and functional properties of a 3D HCC model. METHODS AND RESULTS: Spheroids were produced usin

Targeting FAM111B attenuates mitophagy and increases the sensitivity to lenvatinib treatment by increasing MFN2 stability in hepatocellular carcinoma.

Lenvatinib resistance significantly limits its clinical efficacy and application in the treatment of hepatocellular carcinoma (HCC). Mitofusin 2 (MFN2) is an important GTPase involved in mitochondrial fusion, energy balance and mitophagy. The role and regulatory mechanism of MFN2 in HCC progression and lenvatinib resistance remain unclear. Herein, we demonstrated that the family with sequence similarity 111 member B (FAM111B) regulated the stability of MFN2 and the sensitivity to lenvatinib in HCC. Mechanistically, FAM111B promoted MFN2 ubiquitination by recruiting RAN-binding protein 9 (RANBP

The Metazoan SpoT Homolog 1 promotes ferroptosis by regulating the intracellular redox cycle and iron levels in hepatocellular carcinoma.

BACKGROUND: Ferroptosis, a form of programmed cell death, is a potential target for cancer therapy. Metazoan SpoT Homolog 1 (MESH1) possesses intracellular NADPH phosphatase activity, which has been linked to ferroptosis. However, the molecular effects on the ferroptosis in hepatocellular carcinoma (HCC) remain unclear. This study aimed to investigate the relationship between MESH1 expression and the prognosis of patients with HCC, as well as its impact on ferroptosis in HCC cells. METHODS: We used resected specimens from patients to assess the relationship between MESH1 expression and progno

CXCR2 immunomodulatory therapy protects against microstructural white matter injury and gait abnormalities but does not mitigate deficits of cognition in a preclinical model of cerebral palsy.

Minimizing central nervous system (CNS) injury from preterm birth depends upon understanding the critical pathways that underlie essential neurodevelopmental and CNS pathophysiology. Signaling by chemokine (C-X-C motif) ligand 1 (CXCL1) through its cognate receptor, CXCR2 [(C-X-C motif) receptor 2] is essential for neurodevelopment. Increased CXCR2 signaling, however, is implicated in a variety of uterine and neuropathologies, and their role in the CNS injury associated with perinatal brain injury is poorly defined. To evaluate the long-term efficacy of CXCR2 blockade in functional repair of b

Lenvatinib after progression on pemigatinib and futibatinib in FGFR2 fusion-positive biliary tract cancer with an acquired kinase point mutation.

Biliary tract cancers (BTC) represent a heterogeneous group of malignancies with a poor prognosis and rising incidence. Oncogenic FGFR2 fusions are one of several actionable molecular alterations. In this context, selective FGFR tyrosine kinase inhibitors have demonstrated promising and durable response rates and are now approved and included in clinical guidelines. However, secondary kinase mutations frequently arise over time, leading to resistance against these drugs. We present the case of a 41-year-old male patient with metastatic BTC who underwent molecular analysis after disease progres

Inhibition of levodopa-induced abnormal involuntary movements (AIMs) using a selective α7 nicotinic positive allosteric modulator.

Chronic administration of nicotine and nicotinic ligands have been shown to reduce levodopa-induced dyskinesia (LID) in rodents and primates. Due to its unique extra-striatal localisation and biochemical signalling properties, the α7 subtype of nicotinic acetylcholine receptors (nAChRs) may represent an important and unique target for drug development for the treatment of dyskinesia, particularly since positive allosteric modulator (PAM) at the α7 nAChRs subtype may provide an opportunity to reduce dyskinesia without side effects. In this study, we report on the anti-dyskinetic actions of a se
