Genome-Wide Analysis of BnaRLCK VII Gene Family in Brassica napus and Investigation of Its Function in Resistance to Sclerotinia sclerotiorum.

UNLABELLED: Receptor-like cytoplasmic kinases (RLCKs) constitute a core family of signaling proteins that modulate diverse cellular activities and participate in multiple physiological and biochemical processes in plants. As a unique subclade of RLCKs, the receptor-like cytoplasmic kinases VII (RLCK VII) subfamily features a conserved kinase domain but lacks extracellular and transmembrane domains, and has been proven to exert crucial functions in plant immunity, growth and development, and yield formation. BACKGROUND/OBJECTIVES: This study systematically identified and characterized RLCK VII…

A structurally defined acidic Angelica sinensis polysaccharide synergizes with αGalCer as adjuvants to enhance humoral and cellular immunity.

The development of effective adjuvants is important for enhancing subunit vaccine efficacy. A novel acidic polysaccharide (ASP-3b) was isolated and purified from Angelica sinensis roots. We characterized its structure and evaluated its potential as a vaccine adjuvant, particularly in synergy with the glycolipid α-galactosylceramide (αGalCer or αGC). ASP-3b is a pectin polysaccharide with a molecular weight of 346,628 Da. It was composed of rhamnose, glucuronic acid, galacturonic acid, glucose, galactose, and arabinose in a molar ratio of 2.16: 1.84:78.21:1.24:11.23:5.32. Structural analyses re

TaWRKY24 Positively Regulates Wheat Resistance to Puccinia striiformis f. sp. Tritici via Modulation of Defense Signaling and Reactive Oxygen Species Accumulation.

Wheat stripe rust, caused by Puccinia striiformis f. sp. tritici (Pst), remains a major threat to global wheat production, which necessitates a better understanding of the molecular mechanisms underlying resistance. WRKY transcription factors are pivotal regulators of plant immunity; however, their specific functions in the wheat-Pst pathosystem remain poorly characterized. In this study, we identified and functionally characterized a WRKY transcription factor gene, TaWRKY24, in wheat. Expression profiling revealed that TaWRKY24 was continuously induced upon Pst inoculation. Subcellular locali…

Coronatine promotes Pseudomonas syringae virulence in plants by activating a signaling cascade that inhibits salicylic acid accumulation.

Phytopathogens can manipulate plant hormone signaling to access nutrients and counteract defense responses. Pseudomonas syringae produces coronatine, a toxin that mimics the plant hormone jasmonic acid isoleucine and promotes opening of stomata for bacterial entry, bacterial growth in the apoplast, systemic susceptibility, and disease symptoms. We examined the mechanisms underlying coronatine-mediated virulence and show that coronatine activates three homologous NAC transcription factor (TF) genes, ANAC019, ANAC055, and ANAC072, through direct activity of the TF, MYC2. Genetic characterization

Comparative Wound Healing Processes in Plants and Animals: Bioinspired Strategies for Advancing Regenerative Medicine.

Wound healing is a fundamental biological process essential to maintaining structural integrity and survival across both plant and animal life. Despite the profound evolutionary distance separating these kingdoms, wound healing provides one of those momentous occasions when these biological universes collide, revealing significant evolutionary parallels in the core mechanisms of healing, despite clear molecular and physiological differences. However, two challenges have hindered systematic cross-kingdom comparisons. First, unlike animal wound healing, the major phases of plant wound healing ha

Elicitation of Innate Immunity by a Bacterial Volatile 2-Nonanone at Levels below Detection Limit in Tomato Rhizosphere.

Bacterial volatile compounds (BVCs) exert beneficial effects on plant protection both directly and indirectly. Although BVCs have been detected in vitro, their detection in situ remains challenging. The purpose of this study was to investigate the possibility of BVCs detection under in situ condition and estimate the potentials of in situ BVC to plants at below detection limit. We developed a method for detecting BVCs released by the soil bacteria Bacillus velezensis strain GB03 and Streptomyces griseus strain S4-7 in situ using solid-phase microextraction coupled with gas chromatography-mass

Arabidopsis NLR Immune Receptors: Structural Diversification, Signaling Networks, and Emerging Principles of Immune Activation.

Plant intracellular nucleotide-binding leucine-rich repeat receptors (NLRs) are central to effector-triggered immunity (ETI), but the rapid expansion of structural data has outpaced an integrated explanation of how receptor architecture, oligomerization, and network context determine immune output. Recent structural and biochemical studies, particularly in Arabidopsis, have revealed that NLRs operate as dynamic signaling assemblies rather than simple pathogen-recognition switches. Here, we synthesize recent evidence across coiled-coil NLRs (CNLs), Toll/interleukin-1 receptor NLRs (TNLs), and R

Ultrasound-driven plant lipid vesicles amplify oncolytic virotherapy in colorectal cancer.

Colorectal cancer (CRC) remains a highly aggressive malignancy, and its treatment with oncolytic virotherapy is hindered by multiple delivery barriers and delayed therapeutic kinetics. To address these challenges, we developed a plant lipid-based vesicular system (P-M@Ce6-OVs) co-encapsulating oncolytic viruses (OVs) and sonosensitizer chlorin e6 (Ce6), enabling ultrasound (US)-guided, spatiotemporally controlled delivery to deep colorectal tumors. Under US exposure, the system enhanced mucus infiltration, tumor penetration, and lysosomal escape, while activating sonodynamic therapy to rapidly

The Janus Face of Aspergillus in Fish Aquaculture: From Pathogenic Threat to Functional Feed Additive.

The dramatic rise in global aquaculture has brought renewed focus on filamentous fungi impacting fish health and aquafeeds. The fungal genus Aspergillus has a Janus-like role in aquaculture. On the pathogenic side, opportunistic pathogens like A. flavus, A. niger and A. fumigatus cause outbreaks of aspergillosis, with granulomatous lesions and significant mortality, especially in fish with impaired immunity and/or poor environmental conditions. Toxigenic strains of A. flavus and A. parasiticus also contaminate aquafeeds with aflatoxins, some of the most toxic natural hepatotoxins and carcinoge

Novel but stable endosymbionts have contrasting effects on aphid dispersal and plant feeding damage in the cereal pest Diuraphis noxia.

Endosymbiotic bacteria can affect many ecological attributes of their insect hosts, including (in herbivorous insects) how they interact with plants where they feed. This raises the issue of whether deliberate endosymbiont introductions could be used to decrease crop damage caused by insect pests. Here, we investigate how transinfecting Rickettsiella viridis and Regiella insecticola endosymbionts into a novel pest aphid host, the Russian wheat aphid (Diuraphis noxia), influences population growth, alate production, dispersal ability, and crop damage. Both the Rickettsiella (originating from pe

Efficacy and Safety of Add-On Plant-Based Drugs for COVID-19 Patients: A Review of the Randomized Control Trials.

COVID-19 caused by the infection of SARS-CoV-2 is still a global concern. WHO reported that from 13 March to 9 April 2023, there were 3 million new cases and approximately 23,000 deaths, mostly occurring in the South-East Asia and Eastern Mediterranean regions, which is predicted due to the new Omicron variant, Arcturus XBB.1.16. Many studies have reported the potency of medicinal plants in enhancing the function of the immune system to combat virus infection. The literature review aimed to describe the efficacy and safety of add-on plant-based drugs for COVID-19 patients. The articles were ex

Comparative analysis of salicylic acid levels and gene expression in resistant, tolerant, and susceptible cassava varieties following whitefly-mediated SLCMV infection.

Sri Lankan cassava mosaic virus (SLCMV), the primary pathogen responsible for cassava mosaic disease in cassava plantations, is transmitted via infected cutting stems and the whitefly vector, Bemisia tabaci. To obtain better insights into the defense mechanism of cassava against SLCMV, whiteflies were used to induce SLCMV infection for activating the salicylic acid (SA) signaling pathway, which triggers the innate immune system. The study aimed to investigate the specific interactions between viruliferous whiteflies and SA accumulation in resistant (C33), tolerant (Kasetsart 50; KU50), and sus

Involvement of potato (Solanum tuberosum L.) MKK6 in response to potato virus Y.

Mitogen-activated protein kinase (MAPK) cascades have crucial roles in the regulation of plant development and in plant responses to stress. Plant recognition of pathogen-associated molecular patterns or pathogen-derived effector proteins has been shown to trigger activation of several MAPKs. This then controls defence responses, including synthesis and/or signalling of defence hormones and activation of defence related genes. The MAPK cascade genes are highly complex and interconnected, and thus the precise signalling mechanisms in specific plant-pathogen interactions are still not known. Her

Antagonism of Transcription Factor MYC2 by EDS1/PAD4 Complexes Bolsters Salicylic Acid Defense in Arabidopsis Effector-Triggered Immunity.

In plant immunity, pathogen-activated intracellular nucleotide binding/leucine rich repeat (NLR) receptors mobilize disease resistance pathways, but the downstream signaling mechanisms remain obscure. Enhanced disease susceptibility 1 (EDS1) controls transcriptional reprogramming in resistance triggered by Toll-Interleukin1-Receptor domain (TIR)-family NLRs (TNLs). Transcriptional induction of the salicylic acid (SA) hormone defense sector provides one crucial barrier against biotrophic pathogens. Here, we present genetic and molecular evidence that in Arabidopsis an EDS1 complex with its part…

Does chromatin remodeling mark systemic acquired resistance?

The recognition of plant pathogens activates local defense responses and triggers a long-lasting systemic acquired resistance (SAR) response. Activation of SAR requires the hormone salicylic acid (SA), which induces SA-responsive gene expression. Recent data link changes in gene expression to chromatin remodeling, such as histone modifications and histone replacement. Here, we propose a model in which recruitment of chromatin-modifying complexes to SA-responsive loci controls their basal and SA-induced expression. Basal repression of these loci requires the post-translational modifier SUMO (SM
