Stress, the HPA axis, and nonhuman primate well-being: A review.

Numerous stressors are routinely encountered by wild-living primates (e.g., food scarcity, predation, aggressive interactions, and parasitism). Although many of these stressors are eliminated in laboratory environments, other stressors may be present in that access to space and social partners is often restricted. Stress affects many physiological systems including the hypothalamic-pituitary-adrenocortical (HPA) axis, which is the focus of this review. The glucocorticoid, cortisol, is the ultimate output of this system in nonhuman primates, and levels of this hormone are used as an index of st…

Designing primate models to assess the prodromal phase of Huntington's disease.

Huntington's disease (HD) is an inherited autosomal-dominant neurodegenerative disorder characterized by frontal-type cognitive deficits, involuntary choreiform movements and progressive neuronal degeneration, primarily affecting the caudate-putamen complex. There is currently no effective therapy for this disorder. Numerous efforts are directed towards the search and validation of new therapies to prevent or slow down disease progression. To this end, different animal models, including in nonhuman primates, have been developed to mimic the early phase of neuronal dysfunction that precedes deg…

Cytokine expression in malaria-infected non-human primate placentas.

Malaria parasites are known to mediate the induction of inflammatory immune responses at the maternal-foetal interface during placental malaria (PM) leading to adverse consequences like pre-term deliveries and abortions. Immunological events that take place within the malaria-infected placental micro-environment leading to retarded foetal growth and disruption of pregnancies are among the critical parameters that are still in need of further elucidation. The establishment of more animal models for studying placental malaria can provide novel ways of circumventing problems experienced during pl

Accumulation of intrahepatic islet amyloid in a nonhuman primate transplant model.

Islet amyloid is hypothesized to play a role in nonimmunologic transplanted islet graft loss. We performed a quantitative histologic analysis of liver biopsies from intrahepatic islet grafts transplanted in streptozotocin-induced diabetic cynomolgus macaques. Seven animals treated with antithymocyte globulin (ATG) and rapamycin or ATG and rituximab experienced islet graft rejection with lymphocytic infiltrates present on islet graft biopsies. Except for one case involving the oldest and largest donor where amyloid was present on initial biopsy 1 month after transplant, none of the six other ca

Experimental video analysis of eye blink reflex in a primate model.

BACKGROUND: The establishment of reliable methods for functional assessment in experimental models of peripheral nerve regeneration is crucial. METHODS: We present a straightforward method for video analysis of the eye blink reflex in a model of facial nerve damage in a nonhuman primate (Callithrix sp.). RESULTS: Our 6-level dynamic analysis demonstrated good reproducibility between independent observers, as measured by Cohen's kappa index. Our static analysis, which was based on 4 semiautomated metric parameters, showed low correlation during the early stage of facial movement recovery (the

Hour of birth and birth assistance: from a primate to a medicalized pattern?

OBJECTIVES: Previous studies generally agree that in Homo sapiens births without medical intervention occur mostly at night, although with a less accentuated pattern than in other primate species. The present study has three main objectives: (a) to establish the hourly pattern of births in a modern medicalized population, (b) to explore the association between the hour of birth and maternal and fetal variables and mode of delivery, and (c) to evaluate the risk for medical intervention at different hours of the day. METHODS: The hourly distribution of 25,779 deliveries at the "La Paz" Madrid U

Development of a novel nonhuman primate model for Rift Valley fever.

Rift Valley fever (RVF) virus (RVFV) can cause severe human disease characterized by either acute-onset hepatitis, delayed-onset encephalitis, retinitis and blindness, or a hemorrhagic syndrome. The existing nonhuman primate (NHP) model for RVF utilizes an intravenous (i.v.) exposure route in rhesus macaques (Macaca mulatta). Severe disease in these animals is infrequent, and large cohorts are needed to observe significant morbidity and mortality. To overcome these drawbacks, we evaluated the infectivity and pathogenicity of RVFV in the common marmoset (Callithrix jacchus) by i.v., subcutaneou

Evolution of an X-linked primate-specific micro RNA cluster.

Micro RNAs (miRNAs) are a class of small regulatory RNAs, which posttranscriptionally repress protein production of the targeted messenger RNAs (mRNAs). Accumulating evidence has suggested lineage-specific miRNAs have contributed to lineage-specific characteristics. However, the birth and death of these miRNAs, particularly in primates, largely remain unexplored. We herein characterized the evolutionary history of a newly discovered miRNA cluster on primate X-chromosome, spanning a approximately 33-kb region in human Xq27.3. The cluster consists of six distinct miRNAs, four of which are compac…

[Cultivated corneal endothelial cell sheet transplantation in a primate model].

Our new surgical treatment for corneal endothelial dysfunction involves replacing damaged the corneal endothelium with healthy corneal endot … ed a model of cultivated corneal endothelial transplantation for corneal endothelial dysfunction in primates whose corneal endothelial cells have less proliferative capacity in vivo. Our results suggest that this is a useful model for long-term observation in advance of future clinical application of cultivated corneal endothelial transplantation.

Localization of arm representation in the cerebral peduncle of the non-human primate.

Motor deficit severity and the potential for recovery in patients with brain injury depend on the integrity of descending corticofugal projections. Clinical assessment of these conditions following subtotal brain trauma requires a comprehensive understanding of the anatomical structures involved in the lesion as well as those structures that are spared. To assist in this endeavor, we investigated motor fiber organization in the crus cerebri of the cerebral peduncle (ccCP) in the rhesus monkey. Fibers originating from the arm representations of the primary (M1), supplementary (M2), rostral cing

Developmental changes in cellular prion protein in primate visual cortex.

Cellular prion protein (PrP(c)) is a cell surface glycoprotein highly expressed in neurons, and a protease-resistant conformer of the protein accumulates in the brain parenchyma in prion diseases. In human prion diseases, visual cortex and visual function can be affected. We examined both the levels and the localization of PrP(c) in developing visual cortex of the common marmoset. Western blot analysis showed that PrP(c) increased from the day of birth through adulthood, and this increase correlated with the progression of synapse formation. Immunohistochemistry showed that PrP(c) was present

Integration of cognitive and motivational information in the primate lateral prefrontal cortex.

The prefrontal cortex (PFC), particularly the lateral prefrontal cortex (LPFC), has an important role in cognitive information processing. The area receives projections from sensory association cortices and sends outputs to motor-related areas. Neurons in LPFC code the behavioral significance of stimuli, which can be abstract precursors for complex motor commands and are structured hierarchically. Loss of these neurons leads to a lack of flexibility in decision making, such as seen in stereotyped behaviors. However, to make more appropriate decisions the code for behavioral significance has to

Multifocal visual evoked potentials in the anesthetized non-human primate.

PURPOSE: To evaluate monkey multifocal visual evoked cortical potentials (mfVEPs) recorded from central and peripheral fields for reliabilit … ere statistically distinct from noise. CONCLUSIONS: mfVEPs can be reliably recorded from non-human primates anesthetized with pentobarbital. Under the recording conditions described, mfVEPs are not contaminated by ERG activity. mfVEPs may be useful in animal models of diseases that differentially affect macular and peripheral visual field responsiveness.

The significance of platelet-activating factor and fertility in the male primate: a review.

Since its discovery nearly 30 years ago platelet-activating factor (PAF) has emerged as one of the more important lipid mediators known. PAF (1-O-alkyl-2-O-acetyl-sn-glycero-3-phosphorylcholine) exists endogenously as a mixture of molecular species with structural variants of the alkyl moiety. PAF is a novel potent signaling phospholipid that has unique pleiotropic biological properties in addition to platelet activation. PAF also plays a significant role in reproduction and is present in the sperm of a number of primate species. PAF content in squirrel monkey sperm is significantly higher dur

We found no major differences in the distribution and shape of labelled cells in the zona incerta of MPTP-treated monkeys when compared to control. In conclusion, we report that the primate zona incerta shows considerable cyto and chemoarchitectoni …

Detection of Fruit and the Selection of Primate Visual Pigments for Color Vision.

Primates have X chromosome genes for cone photopigments with sensitivity maxima from 535 to 562 nm. Old World monkeys and apes (catarrhines) and the New World (platyrrhine) genus Alouatta have separate genes for 535-nm (medium wavelength; M) and 562-nm (long wavelength; L) pigments. These pigments, together with a 425-nm (short wavelength) pigment, permit trichromatic color vision. Other platyrrhines and prosimians have a single X chromosome gene but often with alleles for two or three M/L photopigments. Consequently, heterozygote females are trichromats, but males and homozygote females are d…

Spinal cord involvement in the nonhuman primate model of Lyme disease.

Lyme borreliosis is a multisystemic disease caused by infection with various genospecies of the spirochete Borrelia burgdorferi. The organs most often affected are the skin, joints, the heart, and the central and peripheral nervous systems. Multiple neurological complications can occur, including aseptic meningitis, encephalopathy, facial nerve palsy, radiculitis, myelitis, and peripheral neuropathy. To investigate spinal cord involvement in the nonhuman primate (NHP) model of Lyme borreliosis, we inoculated 25 adult Macaca mulatta with B. burgdorferi sensu strictu strains N40 by needle (N=9)

Non-human primate models of childhood psychopathology: the promise and the limitations.

Although non-human primate models have been used previously to investigate the neurobiology of several sensory and cognitive developmental pathologies, they have been employed only sparingly to study the etiology of childhood psychopathologies for which deficits in social behavior and emotion regulation are major symptoms. Previous investigations of both adult human and non-human primates have indicated that primate social behavior and emotion are regulated by a complex neural network, in which the amygdala and orbital frontal cortex play major roles. Therefore, this review will provide inform…
