Ubiquitin E3 ligase activity in TRIpartite Motif (TRIM) family proteins.

TRIpartite Motif (TRIM) family proteins are required for healthy development and homeostasis, while their dysregulation is observed in a wide range of diseases. Although diverse cellular roles are emerging for TRIMs, the majority function as ubiquitin E3 ligases by means of their RING (really interesting new gene) domain. Here, we delve into what is known about TRIM ubiquitin E3 ligase mechanisms, their ubiquitin chain specificities, and how they can be regulated through homo- and hetero-multimerisation and post-translational modifications.

Alternative control: what's WASp doing in the nucleus?

Wiskott-Aldrich syndrome (WAS) is a rare X-linked recessive immunodeficiency disorder of childhood that is caused by mutations in the WAS gene. WAS encodes WASp, a protein that is known to function in the cytoplasm of hematopoietic cells and is required for the induced differentiation of CD4+ T helper type 1 (TH1) lymphocytes. Now, a paper in Science Translational Medicine describes another mechanism for impaired immunity in WAS by showing that WASp localizes in the nucleus and regulates histone modifications and chromatin structure, thereby modulating expression of the TH1 master gene TBX21 (

Lysinoalanine crosslinking in the extracellular flagellar hook of Synergistota.

UNLABELLED: Bacterial motility is driven by the flagellar motor, a conserved macromolecular complex that enables cells to navigate chemical gradients. In spirochetes, motility depends on periplasmic flagella whose function is supported by a unique post-translational modification (PTM) in the hook protein FlgE: a covalent lysinoalanine (Lal) crosslink between adjacent subunits. This modification has been shown to be conserved across the phylum and is required for motility. In spirochetes, Lal is thought to provide additional hook stability needed to meet the demands of flagellar rotation in the

In Vitro Analysis of Profilin 1 Binding to Recombinant Actin.

Profilin 1 (PFN1) is a key actin-binding protein (ABP) involved in cytoskeletal dynamics, regulating actin polymerization and filament remodeling. While native actin has been extensively studied, recombinant actin produced through biotechnological approaches remains less explored due to solubility and folding challenges. Previously, we assessed the effects of S-nitrosylation on actin binding, providing insights into post-translational modifications that regulate actin function. In the present study, we provide a comprehensive protocol to produce recombinant β-actin using an in vitro transcript

S-acylation of TDP43 regulates its condensation in amyotrophic lateral sclerosis.

TDP43 inclusion bodies are widely present in the majority of patients with familial and sporadic amyotrophic lateral sclerosis (ALS). The mechanisms regulating TDP43 solubility remain incompletely understood. Here, we report that TDP43 undergoes S-acylation primarily at the Cys244 residue by the S-acyltransferase zDHHC23. This S-acylation maintains the liquid-like properties of TDP43 by reducing the aberrant interaction with poly(ADP-ribose) polymerase 1 (PARP1) and PARylated proteins, thereby countering the pathological condensation of TDP43. S-acylation-deficient TDP43 inclusions sequester t

Lactate as a metabolic-epigenetic hub in neurological disorders.

Lactate, once considered merely a metabolic waste product of glycolysis, has emerged as a pivotal signaling molecule that bridges cellular metabolism and epigenetic regulation in the central nervous system. The discovery of protein lactylation in 2019 revealed a novel post-translational modification that directly linked lactate accumulation to changes in gene expression, fundamentally reshaping our understanding of how metabolic states influenced neural function and dysfunction. This comprehensive review synthesizes recent advances in lactate biology, encompassing the expanding enzymatic lands

Hypoxia-mediated regulation of mRNA metabolism: from transcription to stability.

BACKGROUND: Hypoxia plays a critical role in both physiological and pathological processes. Cells adapt to hypoxia through the extensive reprogramming of gene expression. While the transcriptional regulations of hypoxia-inducible factors (HIFs) are well-studied, recent advances have emphasized the importance of post-transcriptional regulation, including mRNA translation, modification, stability, and degradation. Summarizing the regulation of mRNA homeostasis under hypoxia will improve our understanding of how hypoxia contributes to diseases such as cancer and ischemic injury. MAIN BODY: This r

ERK-METTL3 axis acts as a novel regulator of antiviral innate immunity combating pseudorabies virus infection.

The ERK-mediated phosphorylation of the core m6A methyltransferase METTL3 has been linked to the regulation of embryonic stem cell differentiation and tumorigenesis. However, its role in the antiviral innate immune response remains unclear. In this study, we found that during infection with the prototypical alpha-herpesvirus Pseudorabies virus (PRV), ERK2 protein expression increased significantly, while METTL3 expression decreased both in vitro and in vivo. Overexpressing ERK2 and METTL3 effectively reduced PRV replication, while their knockdown promoted viral replication. The C-terminal doma

SARS-CoV-2 disrupts host epigenetic regulation via histone mimicry.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) emerged at the end of 2019 and caused the devastating global pandemic of coronavirus disease 2019 (COVID-19), in part because of its ability to effectively suppress host cell responses1-3. In rare cases, viral proteins dampen antiviral responses by mimicking critical regions of human histone proteins4-8, particularly those containing post-translational modifications required for transcriptional regulation9-11. Recent work has demonstrated that SARS-CoV-2 markedly disrupts host cell epigenetic regulation12-14. However, how SARS-CoV-2

What makes a prognostic biomarker in CNS diseases: strategies for targeted biomarker discovery? Part 2: chronic progressive and relapsing disease.

INTRODUCTION: Compared with acute neurological conditions, biomarker discovery in chronic progressive and relapsing neurodegenerative diseases may be more difficult. First, the degenerative process may be so slow that the biomarker discovered is released only in small quantities making it a fishing exercise with very little to catch. Second, it may be difficult to differentiate acute new damage, relevant to prognostic estimates, from already existing background damage. A potential advantage in chronic conditions is that there may be sufficient time for the disease to leave a specific signature

High-resolution NMR spectroscopy of proteins in intact mitochondria.

Understanding protein structure and function within mitochondria is essential for unraveling the molecular mechanisms underlying cellular energy production, stress response and disease. Here we present an approach for NMR observation of proteins within intact mitochondria by delivering proteins directly into isolated mitochondria via electroporation. Using this method, we investigate the interaction of α-synuclein with the mitochondrial membrane and examine how post-translational modifications regulate this interaction. In addition, we assessed the stability of GB1 and the dimerization of its

Methyl-CpG binding proteins: specialized transcriptional repressors or structural components of chromatin?

DNA methylation is an epigenetic modification that is implicated in transcriptional silencing. It is becoming increasingly clear that both correct levels and proper interpretation of DNA methylation are important for normal development and function of many organisms, including humans. In this review we focus on recent advances in understanding how proteins that bind to methylated DNA recognize their binding sites and translate the DNA methylation signal into functional states of chromatin. Although the function of methyl-CpG binding proteins in transcriptional repression has been attributed to

S-Nitrosylation of GPCR Regulatory Machinery as a Mechanism to Bias Signaling.

Upon stimulation by endogenous ligands, G protein-coupled receptors (GPCRs) activate downstream signaling pathways through multiple mechanisms, including G protein subtypes, β-arrestins, and receptor-specific partners. Synthetic ligands may activate only a subset of these pathways, resulting in functional selectivity or signaling bias. Since not all signaling outputs are therapeutically desirable, there is pharmaceutical interest in exploiting biased signaling. Although much effort is focused on designing ligands to induce receptor conformations that result in signal bias, it is also true that

Targeting Multidrug Resistance in Cancer: Impact of Retinoids, Rexinoids, and Carotenoids on ABC Transporters.

The active efflux of drugs by adenosine triphosphate (ATP)-binding cassette (ABC) trans-porters, such as multidrug resistance protein 1 (MDR1/ABCB1), multidrug resistance-associated protein 1 and 2 (MRP1/ABCC1; MRP2/ABCC2), and breast cancer resistance protein (BCRP/ABCG2), is a well-established mechanism contributing to multidrug resistance (MDR). Interestingly, various vitamin A-based molecules have been found to influence the expression or function of these transporters. This work investigated the current evidence on the effects of retinoids, rexinoids, and carotenoids on ABC transporters a

Could cyanogenic glycoside rich diet cause increased risk for carbamylation-induced protein damage in individuals with chronic inflammatory diseases?

Cyanogenic glycosides are found in a diverse group of plants and are metabolized into thiocyanate by the intestines and liver. Conversion of plant derived thiocyanates into cyanide and isocyanic acid occurs by the activity of neutrophil-derived enzyme myeloperoxidase. Therefore, increased intake of cyanogenic glycoside rich plant based diet may lead to increased isocyanic acid induced protein carbamylation in chronic inflammatory states (increased myeloperoxidase activity). As there is a close relationship between non-enzymatic post-translational modification and protein function, carbamylatio

[Peptidylarginine deiminase type4 (PADI4) role in immune system].

Rheumatoid arthritis (RA) is one of the representative auto-immune diseases, characterized by systemic inflammatory synovitis. Various genetic and environmental factors which contribute to RA pathogenesis, has been suggested, however, detailed mechanism remained unknown. While around 100 genetic risk factors for RA have been reported, Peptidylarginine deiminase type4 (PADI4) was firstly identified as a non-MHC RA genetic risk factor. Furthermore, PADI4 risk allele possessed the association with bone damage regardless of anti citrullinated peptide antibody (ACPA) positivity in Asian RA patients

Bottlenecks in Proteomics: An Addendum.

Mass spectrometry (MS) is the core for advanced methods in proteomic experiments. When effectively used, proteomics may provide extensive information about proteins and their post-translational modifications, as well as their interaction partners. However, there are also many problems that one can encounter during a proteomic experiment, including, but not limited to, sample preparation, sample fractionation, sample analysis, data analysis and interpretation, and biological significance. Here, we discuss some of the problems that researchers should be aware of when performing a proteomic exper

Context-dependent protective and pathogenic functions of YTHDF1 in MASLD and hepatocellular carcinoma: Mechanisms and design of therapeutics.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is the most common chronic liver disease and can progressively develop into inflammatory metabolic dysfunction-associated steatohepatitis (MASH) and liver fibrosis. During this progression, activation of hepatic stellate cells (HSCs) leads to excessive accumulation of extracellular matrix. There is a growing trend to develop into hepatocellular carcinoma (HCC). The limited availability of effective therapeutics highlights the need to better understand the underlying pathogenic mechanisms. N6-methyladenosine (m6A) is a key post-tr

RNA m6A modification-regulated ferroptosis in cancer: mechanism and therapeutic potential.

Ferroptosis, an iron-dependent form of regulated cell death, is increasingly recognized as a context-dependent therapeutic vulnerability in cancer, particularly as malignant cells adapt to oxidative, metabolic, and therapy-induced stresses. As a prevalent and reversible epitranscriptomic modification, RNA N6-methyladenosine (m6A) modification orchestrates RNA stability, translation, splicing and decay; consequently, its dysregulation contributes to cancer progression and therapeutic resistance. The intersection of m6A regulation and ferroptosis is therefore biologically important because many
