In the muscle biopsy of a female patient with chronic progressive external ophthalmoplegia (CPEO), myopathy, and exercise intolerance, the heteroplasmic deletion of a single nucleotide (DeltaT5885) in the mitochondrial tRNA tyrosine gene (tRNA(Tyr)) was found. The mutation was associated with the mitochondrial phenotype of individual muscle fibers, suggesting a causal association of DeltaT5885 with the mitochondrial disease phenotype. The microdeletion was absent from the patient's and her relatives' blood, indicating a spontaneous somatic origin.

Nitric oxide is an important bioregulatory molecule with a range of physiological functions. Nitric oxide can also react with oxygen species to produce a range of reactive nitrogen oxides that can damage DNA and lead to mutations of the DNA base sequence. The mutagenicity of a variety of reactive nitrogen oxide species and related DNA damaging agents in the supF assay are reviewed here, in the context of recent reports that relate to the nature of the DNA lesions responsible for the induced mutations. Mutations induced by nitric oxide in the supF assay are compared to those induced by N(2)O(3)

The clinical trials of immunotherapy of renal carcinoma have been based on the theory that manipulation of the host-immune response can influence the growth of malignant cells in the host. The results cited above do not demonstrate a clear therapeutic role of immunotherapy but they do provide evidence to encourage further studies. This is especially pertinent in view of the current absence of effective systemic agents. The stimulation of the host response appears to be finite and cannot be expected to control massive tumor burden. The clinical trials analyzed above must be interpreted with thi

Control of classical swine fever (CSF) in endemic regions relies heavily on vaccination of breeding herds. Optimizing vaccination strategies is essential for sustainable disease control covering pigs belonging to different categories. Data describing direct comparisons between the classical C-strain and recombinant DIVA-compatible vaccines in the context of gestational timing, maternal antibody transfer, and early postnatal virological outcomes remain very limited. This study evaluated how vaccination in the middle to late pregnancy periods (at 72 days of gestation) with either the live attenu

OBJECTIVES: Dilated cardiomyopathy (DCM) is a complex cardiovascular disease with unknown etiology. Although nuclear genes play active roles in DCM, mitochondrial dysfunction was believed to be involved in the pathogenesis of DCM. The objective of this study was to analyze the association between mitochondrial tRNA (mt-tRNA) mutations and DCM.
MATERIALS AND METHODS: We performed a mutational analysis of mt-tRNA genes in a cohort of 318 patients with DCM and 200 age- and gender-matched control subjects. To further assess their pathogenicity, phylogenetic analysis and mitochondrial functions in

We describe a young woman with a progressive mitochondrial myopathy that started with muscle weakness and went on to include deafness, dementia and ataxia. Skeletal muscle showed the histological and biochemical features of mitochondrial respiratory chain dysfunction. Genetic analysis identified a novel, heteroplasmic, A to G transition in tRNA(Ser(UCN)) at position 7480 affecting a highly conserved base in the anticodon loop. Single-fibre PCR showed highest levels of mutation in cytochrome c-oxidase-deficient fibres and quantification in two biopsies taken 5 years apart showed no change in pe

The influence of small Alu-like RNA, isolated from specific RNP complexes (alpha-RNP), on the activity of RNA polymerase III in cell-free system has been studied. The RNAs transcribed in vitro from Alu-DNA template (BLUR, 8) were isolated and subjected to polyacrylamide gel electrophoresis. A specific stimulation of RNA polymerase III activity by alpha-RNA was demonstrated.

Cytoplasts from patients with myoclonus epilepsy with ragged-red fibers harboring a pathogenic point mutation at either nucleotide 8344 or 8356 in the human mitochondrial tRNA(Lys) gene were fused with human cells lacking endogenous mitochondrial DNA (mtDNA). For each mutation, cytoplasmic hybrid (cybrid) cell lines containing 0 or 100% mutated mtDNAs were isolated and their genetic, biochemical, and morphological characteristics were examined. Both mutations resulted in the same biochemical and molecular genetic phenotypes. Specifically, cybrids containing 100% mutated mtDNAs, but not those c

The activity of cytochrome oxidase (CO), the terminal enzyme of the mitochondrial electron transport chain, has been reported to be lower in the brains of Alzheimer disease (AD) patients. This suggests that a modification of mitochondrial DNA (mtDNA) may be responsible for this decrease of CO activity. Many mtDNA variants were found by different studies at a higher frequency in AD patients, suggesting that mtDNA variants could confer a genetic susceptibility to AD. In this study, we sequenced the entire mitochondrial genome region that encompasses the three CO genes and the 22 mitochondrial tR

Integrative and conjugative elements (ICEs) are mobile genetic elements that widely disseminate adaptive traits among bacterial populations. Despite their prevalence, the regulatory mechanisms orchestrating ICE integration and excision remain poorly understood. Here, we investigate the transcriptional regulation of the integrase gene (int) in ICEKp1 of Klebsiella pneumoniae. Our results demonstrate that the integrated ICEKp1 exploits the adjacent host transfer RNA-asn promoter to drive int transcription, whereas the excised ICEKp1 switches to a 3'-end promoter. Remarkably, the messenger RNA st

Mutations in mitochondrial tRNA (mt-tRNA) genes are associated with non-syndromic hearing loss (NSHL), though their detailed molecular mechanisms and pathogenic potential remain unclear. In this study, we reported three Han Chinese pedigrees exhibiting maternally transmitted NSHL. Mitochondrial genomes from probands were sequenced and compared to revised Cambridge reference sequence (rCRS), followed by phylogenetic analysis to assess pathogenicity. We further established cybrid cell lines to measure mtDNA copy number, ATP levels, mitochondrial membrane potential (MMP), reactive oxygen species

Utilization of endogenous RNA interference (RNAi) mechanisms via delivery of exogeneous small interfering RNA (siRNA) molecules offers a transformative approach to treatment of disease by enabling sequence specific silencing of mutated gene expression. Nanotechnology-based platforms have enabled delivery of siRNA and have already been clinically validated for intravenous (IV) infusion administration (e.g patisiran). Oral administration of siRNA remains an unmet challenge due to formidable biological barriers in the gastrointestinal (GI) tract. Nanotechnology-enabled strategies for oral siRNA d

Paternal exposure to environmental stressors elicits distinct changes to the sperm sncRNA profile, modifications that have significant post-fertilization consequences. Despite this knowledge, there remains limited mechanistic understanding of how paternal exposures modify the sperm sncRNA landscape. Here, we report the acute sensitivity of the sperm sncRNA profile to the reproductive toxicant acrylamide. Furthermore, we trace the differential accumulation of acrylamide-responsive sncRNAs to coincide with sperm transit of the proximal (caput) segment of the epididymis, wherein acrylamide exposu

The gene transfer agent (GTA) of the a-proteobacterium Rhodobacter capsulatus is a cell-controlled genetic exchange vector. Genes that encode the GTA structure are clustered in a 15-kb region of the R. capsulatus chromosome, and some of these genes show sequence similarity to known bacteriophage head and tail genes. However, the production of GTA is controlled at the level of transcription by a cellular two-component signal transduction system. This paper describes homologues of both the GTA structural gene cluster and the GTA regulatory genes in the a-proteobacteria Rhodopseudomonas palustris

1. 3H-, 125I-, and 32P-labeling methods were developed for base composition and sequence analysis of minute amounts of nonradioactive nucleic acids containing modified constituents. 2. Base composition analysis showed tRNA from two "liver-like" minimal deviation hepatomas, Morris hepatomas 5123D and 7777, to exhibit typical alterations when compared with liver tRNA. Our observations, which were made for different transplant generations of the tumors, indicated a trend toward undermethylation and undermodification of tRNA. 3. In order to extend the usefulness of the quantitation of urinary nucleoside markers, studies were undertaken to explore the adaptability of such determinations for early detection in cancer-prone populations such as asbestos workers. Another study was aimed at exploring the usefulness of therapy in individual patients. During these studies, two heretofore unknown phenomena serendipitously emerged which expand the versatility of the marker determinations: (a) radiation damage in animals and humans causes an excretion of urinary BAIB which from preliminary studies appears to be proportional to t

In vitro incubation of hyperimmune rabbit lymphoid cells with the hallucinogenic indole alkaloid, d-lysergic acid diethylamide (LSD), results in biosynthesis of modified, secretable immunoglobulin peptides. Modification involves covalent attachment of the lysergyl moiety to COOH-terminal portions of the peptides. Aalogous effects occur when cells are incubated in vitro in the presence of the non-hallucinogenic LSD analogue, d-lysergic acid, and N-[3H]-carboxymethyl-d-lysergamide. The phenomenon is reversed by tryptophan and is inhibited by puromycin and cycloheximide. In vitro attachment of th

Several point mutations in mitochondrial tRNA genes have been linked to distinct clinical subgroups of mitochondrial diseases. A particularly large number of different mutations is found in the tRNA(Leu)(UUR) gene. We show that base substitutions at nucleotide position 3256, 3260, and 3271 of the mitochondrial genome, located in the D and anticodon stem of this tRNA, and mutation 3243 changing a base involved in a tertiary interaction, significantly impair the processing of the tRNA precursor in vitro. In correlation with other studies, our results suggest that inefficient processing of certai

The rational design of polymer-nucleic acid nanoparticles requires precise control over colloidal stability, interfacial structure, and assembly under biologically relevant conditions. Here, we report a hierarchically assembled, antibody-functionalized polyplex nanoplatform for targeted messenger RNA (mRNA) delivery. A biodegradable cationic poly(β-amino ester) (PβAE-447) enables efficient condensation of in vitro-transcribed mRNA into nanoscale polyplexes. To enhance stability and introduce modular functionality, polyplexes were coated with a poly(γ-glutamic acid)-avidin (PGA-avidin) corona,
