Non-coding RNAs-regulated SLC7A11 modulates ferroptosis: a new strategy for cancer therapy.

Ferroptosis is an iron-dependent form of regulated cell death that plays a dual role in cancer progression and suppression. Solute carrier family 7 member 11 (SLC7A11/xCT) is a key regulator of tumor cell ferroptosis that promotes cystine uptake and glutathione synthesis. However, the regulatory mechanisms of ferroptosis remain unclear, which limits its application in cancer therapy. Recent studies have found that non-coding RNAs (ncRNAs), including lncRNAs, miRNAs, and circRNAs, participate in the process of ferroptosis by regulating SLC7A11. In this review, we summarize the mechanisms of ncR

Exosomal ncRNAs in seminal fluid: Unraveling their regulatory roles in male infertility.

By transmitting bioactive substances, including proteins, lipids, and a varied spectrum of non-coding RNAs, seminal exosomes-including epididymosomes and prostasomes-are shown to be crucial in sperm maturation, motility, and protection. The paper emphasizes how miRNAs, lncRNAs, and circRNAs within these exosomes affect gene expression during spermatogenesis and are starting to be interesting non-invasive biomarkers for several infertility abnormalities, including azoospermia and asthenozoospermia. It also addresses the therapeutic possibilities of exosome-based approaches in reducing inflammat

Decoding immune tolerance in infertility: Exploring immune pathways and non-coding RNAs as pioneering biomarkers and therapeutic targets.

Infertility, impacting a significant number of couples, is characterized by the failure to conceive after one year of consistent, unprotected sexual intercourse. It is multifactorial, with etiological contributors including ovulatory dysfunction, male reproductive anomalies, and tubal patency issues. Approximately 15% of infertility cases are classified as "unexplained," highlighting the complexity of this condition. Lifestyle determinants such as obesity and smoking further complicate reproductive outcomes, while infertility can also indicate underlying chronic health conditions. A specialize

An updated review on abnormal epigenetic modifications in the pathogenesis of systemic lupus erythematosus.

Systemic lupus erythematosus (SLE) is a chronic autoimmune disease. The inconsistent prevalence of SLE between monozygotic twins suggests that environmental factors affect the occurrence of this disease. Abnormal epigenetic regulation is strongly associated with the pathogenesis of SLE. Epigenetic mechanisms may be involved in the development of lupus through DNA methylation, histone modification, noncoding RNAs, and other modifications. This review aims to show numerous studies as a treasure map to better understand the effects of aberrant epigenetic modification in the onset and development

Mechanistic and therapeutic insights into the function of different cell death modalities in rheumatoid arthritis: emphasis on the crosstalk with non-coding RNAs.

Rheumatoid arthritis (RA), a prevalent autoimmune disorder, imposes a substantial burden on global health due to its progressive disability and compromised patient well-being. Although the precise etiology of this condition is still not fully understood, current research implicates intricate interactions between dysregulated immune cells and pro-inflammatory mediators. Recent scientific advancements have highlighted the pathogenic significance of programmed cell death (PCD) mechanisms (including spanning apoptosis, autophagy, ferroptosis, necroptosis, senescence, and pyroptosis) in RA pathophy

Non-coding RNAs in Wilson's Disease: Plausible drivers of hepatic symptom heterogeneity.

Wilson's disease (WD) is an autosomal recessive disorder caused by mutations in the ATP7B gene, which impair cellular copper excretion and lead to toxic copper accumulation in the liver, brain, and other organs. Clinically, WD presents with a broad spectrum of hepatic and neurological manifestations. The pronounced phenotypic variability among patients harboring identical ATP7B mutations, including affected siblings, suggests the influence of additional genetic and/or epigenetic factors such as non-coding RNAs (ncRNAs), in modulating disease presentation. This review explores the potential inv

Hypermethylation and small RNA expression are associated with increased age in almond (Prunus dulcis [Mill.] D.A. Webb) accessions.

The focus of this study is to profile changes in DNA methylation and small RNA expression occurring with increased age in almond breeding germplasm to identify possible biomarkers of age that can be used to assess the potential of individuals to develop aging-related disorders. To profile DNA methylation in almond germplasm, 70 methylomes were generated from almond individuals representing three age cohorts (11, 7, and 2 years old) using an enzymatic methyl-seq approach followed by analysis to call differentially methylated regions (DMRs) within these cohorts. Small RNA (sRNA) expression was p

Modulation of immunosuppressive cells and noncoding RNAs as immunotherapy in osteosarcoma.

The most common bone cancer is osteosarcoma (OS), which mostly affects children and teenagers. Early surgical resection combined with chemotherapy significantly improves the prognosis of patients with OS. Existing chemotherapies have poor efficacy in individuals with distant metastases or inoperable resection, and these patients may respond better to novel immunotherapies. Immune escape, which is mediated by immunosuppressive cells in the tumour microenvironment (TME), is a major cause of poor OS prognosis and a primary target of immunotherapy. Myeloid-derived suppressor cells, regulatory T ce

Interaction between N6-methyladenosine (m6A) modification and noncoding RNAs in cancer.

As a critical internal RNA modification in higher eukaryotes, N6-methyladenosine (m6A) has become the hotspot of epigenetics research in recent years. Extensive studies on messenger RNAs have revealed that m6A affects RNA fate and cell functions in various bioprocesses, such as RNA splicing, export, translation, and stability, some of which seem to be directly or indirectly regulated by noncoding RNAs. Intriguingly, abundant noncoding RNAs such as microRNAs, long noncoding RNAs, circular RNAs, small nuclear RNAs, and ribosomal RNAs are also highly modified with m6A and require m6A modification

Alterations in mRNA 3' UTR Isoform Abundance Accompany Gene Expression Changes in Human Huntington's Disease Brains.

The huntingtin gene has two mRNA isoforms that differ in their 3' UTR length. The relationship of these isoforms with Huntington's disease is not established. We provide evidence that the abundance of huntingtin 3' UTR isoforms differs between patient and control neural stem cells, fibroblasts, motor cortex, and cerebellum. Huntingtin 3' UTR isoforms, including a mid-3' UTR isoform, have different localizations, half-lives, polyA tail lengths, microRNA sites, and RNA-binding protein sites. Isoform shifts in Huntington's disease motor cortex are not limited to huntingtin; 11% of alternatively p

Successful disabling of the 5' UTR of HCV using adeno-associated viral vectors to deliver modular multimeric primary microRNA mimics.

Chronic hepatitis C virus (HCV) infection is a major health concern and is strongly associated with cirrhosis, hepatocellular carcinoma and liver-related mortality. The HCV genome is the template for both protein translation and viral replication and, being RNA, is amenable to direct genetic silencing by RNA interference (RNAi). HCV is a highly mutable virus and is capable of escaping RNAi-mediated silencing. This has highlighted the importance of developing RNAi-based therapy that simultaneously targets multiple regions of the HCV genome. To develop a multi-targeting RNAi activator, a novel a

Regulation of non-coding RNA networks in the nervous system--what's the REST of the story?

Recent advances are now providing novel insights into the mechanisms that underlie how cellular complexity, diversity, and connectivity are encoded within the genome. The repressor element-1 silencing transcription factor/neuron-restrictive silencing factor (REST/NRSF) and non-coding RNAs (ncRNAs) are emerging as key regulators that seem to orchestrate almost every aspect of nervous system development, homeostasis, and plasticity. REST and its primary cofactor, CoREST, dynamically recruit highly malleable macromolecular complexes to widely distributed genomic regulatory sequences, including th

CircNFIB attenuates hypoxia/reoxygenation-induced ferroptosis in AC16 cardiomyocytes with involvement of the miR-338-3p/GPX4 pathway.

Cardiomyocyte ferroptosis contributes to myocardial ischemia/reperfusion injury, but the upstream noncoding RNA mechanisms remain incompletely defined. This study examined whether nuclear factor I B-derived circular RNA (circNFIB) regulates hypoxia/reoxygenation (H/R)-induced ferroptosis in human ventricular cardiomyocyte-derived AC16 cells and whether the miR-338-3p/GPX4 pathway contributes to this effect. The circNFIB back-splice junction was verified by Sanger sequencing. Cell viability, malondialdehyde, intracellular Fe2+, boron-dipyrromethene (BODIPY) 581/591 C11 oxidation, and glutathion

METTL3-mediated m6A modification exacerbates vascular endothelial inflammation in coronary artery disease by regulating DNMT1 mRNA stability.

Chronic vascular endothelial inflammation plays a pivotal role in the pathogenesis of coronary artery disease (CAD). N6-methyladenosine (m6A), the predominant epitranscriptomic modification in messenger RNA (mRNA), regulates pro-atherogenic inflammatory responses in coronary endothelial cells. This study explores the role of the m6A methyltransferase METTL3 in CAD-associated endothelial inflammation through its regulation of DNA methyltransferase 1 (DNMT1) mRNA stability. Using cultured human coronary artery endothelial cells (HCAECs), an apolipoprotein E-deficient (ApoE-/-) mouse model of ath

RNA-binding protein SORBS2 increases human trophoblast cell migration via stabilizing HK2 mRNA in preeclampsia.

IN BRIEF: SORBS2, an RNA-binding protein, is identified as a regulator of aerobic glycolysis, which is essential for trophoblast migration and placental development. Reduced SORBS2 expression in preeclampsia may impair trophoblast migration by affecting mRNA stability and glycolysis, suggesting its role in the disease's pathogenesis. ABSTRACT: Insufficient trophoblast migration and impaired uterine spiral artery remodeling are implicated in the pathogenesis of preeclampsia, contributing to inadequate placentation. However, the molecular mechanism underlying this process remains unclear. Aerob
