Development and validation of TaqMan-based real-time PCR assays for translucent post larvae disease (TPD) detection in Penaeus vannamei.

Translucent Post larvae Disease (TPD), caused by Vibrio parahaemolyticus, has emerged as a critical disease impacting shrimp hatcheries across several Asian countries. This disease severely affects the early larval stages and can lead up to 100 % mortality. Thus, controlling TPD is essential to reduce losses in shrimp hatcheries. Although PCR-based assays have recently been reported for the detection of TPD-causing V. parahaemolyticus (VpTPD), these methods have not yet undergone comprehensive validation in accordance with the Aquatic Animal Health Code of the World Organization for Animal Hea

Development and application of a novel TaqMan qPCR assay targeting the gN gene for genotyping pseudorabies virus.

Pseudorabies virus (PRV), known as porcine herpesvirus type I, belongs to the α-herpesvirus subfamily of the herpesviridae family. Its virions are oval or round, with a genome of approximately 143 000 bp and a high guanine-cytosine (GC) content of 73% (Pomeranz et al., 2005). While swine are the natural host and the virus has caused significant economic loss to the pig industry, PRV can infect other mammals, causing acute infectious diseases characterized by neurological symptoms (He et al., 2019; Cheng et al., 2020). Young pigs are more susceptible to PRV and the infection in piglets is nearl

Establishment of a 23S rRNA assay for Brucella and its application in evaluating bacterial growth status.

Brucellosis presents notable diagnostic challenges, particularly during the chronic phase of infection, due to the typically low bacterial load in tissues, which may evade detection by conventional Polymerase Chain Reaction (PCR) methods. Traditional PCR techniques are susceptible to cross-reactions and can produce false-positive or false-negative results. Furthermore, bacteriological and serological tests have recognized limitations in sensitivity and specificity, potentially complicating accurate diagnosis. We developed a novel reverse transcription quantitative PCR (RT-qPCR) assay targeting

Application of real-time PCR for the identification of the endangered species Galemys pyrenaicus through faecal samples.

BACKGROUND: Currently, many micromammals are important targets for study. The endangered Galemys pyrenaicus is an outstanding example. Globally, their populations have suffered a substantial decline in last 20 years. In the surveyed area, the capture of desman is legally forbidden due to the high conservation concerns. Reason by non-invasive sampling through faeces is proposed for its monitoring. Furthermore, the confusion between faeces from desman and Mediterranean water shrews must be considered. Thus, the aim of this study was focused on developing RT-PCR assays to determine the presence o

Non-lethal detection of Eubothrium crassum (Cestoda) in farmed Atlantic salmon, Salmo salar, using anal swabs and real-time PCR.

Detection of intestinal parasites in fish typically requires autopsy, resulting in the sacrifice of the fish. Here, we describe a non-lethal method for detecting the tapeworm Eubothrium crassum in fish using anal swabs and real-time PCR detection. Two assays were developed to detect cytochrome oxidase I (COI) mitochondrial DNA and 18S ribosomal DNA sequences of E. crassum, respectively. The assays were tested on swab samples from confirmed pathogen free Atlantic salmon (Salmo salar L.) and on samples from farmed Atlantic salmon, where the presence and intensity of parasites had been establishe

Validation of real-time PCR assays for detecting Plasmodium and Babesia DNA species in blood samples.

Malaria and babesiosis are global health threats affecting humans, wildlife, and domestic animals, particularly in Africa, the Americas, and Europe. Malaria can lead to severe outcomes, while babesiosis usually resembles a mild illness but can be severe and fatal in individuals with weakened immune systems. Swift, accurate detection of these parasites is crucial for treatment and control. We evaluated a real-time PCR assay for diagnosing five Plasmodium and three Babesia species from blood samples, assessing its sensitivity, specificity, and analytical performance by analyzing 46 malaria-posit

Verification of a Rapid Analytical Method for the Qualitative Detection of Listeria spp. and Listeria monocytogenes by a Real-Time PCR Assay according to EN UNI ISO 16140-3:2021.

Microbial contamination and foodborne infections are a significant global public health concern. For this reason, the detection, monitoring, and characterization of pathogens represent a significant challenge in quality control settings. Standard approaches, such as culture methods and biochemical tests, are known to be very time-consuming and intensive. Conversely, molecular technologies based on the genomic identification of bacteria are quick and low-cost. Listeria monocytogenes is an opportunistic pathogen and a major concern especially in food industries. It is important to understand and

Ultrasensitive and rapid diagnostic tool for detection of Acanthamoeba castellanii.

Acanthamoeba keratitis is a devastating infectious disease of the cornea caused by an opportunistic amoeba, Acanthamoeba castellanii. It is poorly recognized, and diagnostic delays can lead to irreversible damage to the vision. The gold standard for diagnosis has been a sample culture that lasts approximately 2 weeks. Nevertheless, the essence of time has led to the need for an accurate and fast technique to detect A. castellanii from a sample. We developed both traditional and quantitative real-time-PCR-based methods to detect A. castellanii in less than 3 hours and with the sensitivity of on

Evaluation of a rapid, chip-based, micro-PCR assay for detection of rabies virus in human and canine specimens.

Rabies, a lethal zoonotic encephalitis, remains a significant global health concern, causing an estimated 60 000 annual fatalities worldwide. Dogs serve as the primary reservoirs and vectors for transmitting this infection to humans. Definitive diagnosis of rabies in both human and animal cases necessitates laboratory testing involving various clinical specimens. However, the complexity of laboratory infrastructure and the need for skilled personnel, along with the challenge of maintaining cold-chain integrity during sample referral, hinder the decentralization of diagnostic facilities. This s

A Pilot Study to Detect Viable Salmonella spp. in Diarrheal Stool Using Viability Real-Time PCR as a Culture-Independent Diagnostic Tool in a Clinical Setting.

Frontline laboratories are adopting culture-independent diagnostic testing (CIDT) such as nucleic acid amplification tests (NAATs) due to numerous advantages over culture-based testing methods. Paradoxically, the viability of pathogens, a crucial factor determining active infections, cannot be confirmed with current NAATs alone. A recent development of viability PCR (vPCR) was introduced to mitigate this limitation associated with real-time PCR (qPCR) by using a DNA-intercalating dye to remove residual and dead cell DNA. This study assessed the applicability of the vPCR assay on diarrheal stoo

Decreased expression of miR-23b is associated with poor survival of endometrial cancer patients.

Endometrial cancer (EC) is one of the most common types of cancer of the female reproductive system. EC is classified into two types (EC1 and EC2). MiRNAs are single-stranded RNA molecules that regulate gene expression posttranscriptionally. They have aberrant expression profiles in cancer, including EC. This study aimed to assess the level of expression of a panel of 16 miRNAs in both types of EC and healthy endometrium (HE). A total of 45 patients were enrolled into the study, 18 patients diagnosed with EC1, 12 diagnosed with EC2, and 15 HE controls. Tumor tissues or healthy endometrial tiss
