The Potential of Systemic Control of Sand Flies Using Fipronil in the Novel Leishmania major (Kinetoplastida: Trypanosomatidae) Reservoirs Meriones tristrami (Rodentia: Muridea) and Meriones crassus (Rodentia: Muridea).

Leishmania major (Yakimoff & Schokhor, 1914), an important causative agent of Old World Cutaneous Leishmaniasis (CL), is transmitted by sand flies among a limited number of gerbilline reservoir-species. We can take advantage of this strong dependency to break the pathogen transmission cycle by using systemic insecticides that render the host toxic to the blood-feeding vector. We evaluated the potential of this approach with two novel reservoir species, incriminated for CL expansion in several sites in the Middle East. Specifically, we evaluated: 1) the residuality of the systemic insecticide f…

Transport and release of electron donors and alkalinity during reductive dechlorination by combined emulsified vegetable oil and colloidal Mg(OH)2: Laboratory sand column and microcosm tests.

Emulsified vegetable oil combined with colloidal Mg(OH)2 (EVO-CM) can slowly release electron donors and OH- into groundwater and is therefore regarded as a promising amendment for enhanced in situ treatment of chlorinated solvents, such as tetrachloroethene (PCE) and trichloroethene (TCE). However, its migration ability in different porous media and the simultaneous release of electron donor and pH buffer during enhanced reductive dechlorination (ERD) have never been evaluated in detail. In this study, EVO-CM with uniform drop size and desirable stability was prepared. Laboratory-scale column

Taxonomic, molecular and ecological approach reveals high diversity of vector sand flies, varied blood source supply and a high detection rate of Leishmania DNA in Colombian Amazon region.

BACKGROUND: The Amazon region is home to more than 30% of the sand flies species in Colombia, including vectors of Leishmania mainly in the genus Lutzomyia and Psychodopygus. Advances in morphological and molecular taxonomy of sand flies facilitate the development of updated and robust species inventories in understudied areas, such as the departments of Amazonas and Caquetá. Currently, integrating the detection of blood meal sources and Leishmania DNA represents a key approach under the "One Health" concept by providing insights into human and animal health and the dynamics of different ecosy…

Species diversity of phlebotomine sand flies and sympatric occurrence of Leishmania (Mundinia) martiniquensis, Leishmania (Leishmania) donovani complex, and Trypanosoma spp. in the visceral leishmaniasis focus of southern Thailand.

Autochthonous leishmaniasis in Thailand has recently been a public health concern due to an increasing number of new clinical cases. Most indigenous cases were diagnosed with Leishmania (Mundinia) martiniquensis, and Leishmania (Mundinia) orientalis. However, some doubts regarding vector misidentification have arisen and need to be elucidated. Accordingly, we aimed to assess the species composition of sand flies and determine the molecular prevalence of trypanosomatids in the transmission area of leishmaniasis in southern Thailand. In the present study, a total of 569 sand flies were caught fr…

Development of a next generation DNA sequencing-based multi detection assay for detecting and identifying Leishmania parasites, blood sources, plant meals and intestinal microbiome in phlebotomine sand flies.

Leishmaniasis is a disease caused by Leishmania parasites transmitted by phlebotomine sand flies (Diptera: Psychodidae). Human infections with different Leishmania species cause characteristic clinical manifestations; cutaneous or visceral leishmaniasis. Here we describe the development and application of a Miseq Next GenerationSequencing (NGS)-based Multi Detection Assay (MDA) designed to characterize metagenomics parameters pertinent to the sand fly vectors which may affect their vectorial capacity for Leishmania. For this purpose, we developed a MDA by which, DNA fragments were amplified th…

Molecular Detection of Leishmania in Wild Caught Sand Flies of Larroussius Subgenus in Iran: Combined Use of Internal and External Morphological Characters as a Mean to Differentiate Morphologically Similar Females.

OBJECTIVE: Phlebotomine sand flies are the primary vectors of leishmaniasis; visceral form of disease is transmitted mainly by species belonging to Larroussius and Adlerius subgenus. Species identification of some females in Larroussius subgenus is not easy due to the high similarity. Accurate species identification enables control operations to target only main vectors and increase understanding of ecological needs, bionomic characteristics, and behavioral traits. The purpose of current study was to use two approaches based on internal and external morphological characters to identify the wil…

Comparative analysis of the microbiota of sand fly vectors of Leishmania major and L. tropica in a mixed focus of cutaneous leishmaniasis in southeast Tunisia; ecotype shapes the bacterial community structure.

Phlebotomine sand flies are vectors of the protozoan parasite Leishmania spp. Although the intestinal microbiota is involved in a wide range of biological and physiological processes and has the potential to alter vector competence, little is known about the impact of host species and environment on the gut microbiome. To address this issue, a comparative analysis of the microbiota of sand fly vector populations of Leishmania major and L. tropica in a mixed focus of cutaneous leishmaniasis in Tunisia was performed. Bacterial 16S rRNA gene amplification and Illumina MiSeq sequencing were used t…

Understanding a wood-derived biochar's impact on stormwater quality, plant growth, and survivability in bioretention soil mixtures.

Bioretention systems are planted media filters used in stormwater infrastructure. Maintaining plant growth and survival is challenging because most designs require significant sand. Conventional bioretention soil media (BSM) might be augmented with biochar to make the BSM more favorable to plants, to improve nutrient removal efficiency, and enhance plant survivability during drought while replacing compost/mulch components that have been linked to excess nutrient export. Pots with BSMs representing high and moderate sand content were amended with wood biochar, planted with switchgrass, and sub…

Bimetallic Fe/Al-MOF for the adsorptive removal of multiple dyes: optimization and modeling of batch and hybrid adsorbent-river sand column study and its application in textile industry wastewater.

Bimetallic metal organic framework (MOF) has garnered interest over the years with its applications in industrial wastewater treatment. In this work, Fe-Al-1,4-benzene-dicarboxylic acid (FeAl(BDC)) MOF was synthesized, and adsorptive removal of Rhodamine B dye in batch and unique hybrid FeAl (BDC)-river sand fixed-bed column was studied. The experimental data from the batch studies corroborated well with the pseudo-second-order (PSO) (R2: 0.97) and Freundlich adsorption isotherm models (R2: 0.98) and achieved a maximum adsorption capacity of 48.59 mg/g in 90 min. Furthermore, a fixed-bed colum…

Community deployment of a synthetic pheromone of the sand fly Lutzomyia longipalpis co-located with insecticide reduces vector abundance in treated and neighbouring untreated houses: Implications for control of Leishmania infantum.

BACKGROUND: The rising incidence of visceral leishmaniasis due to Leishmania infantum requires novel methods to control transmission by the sand fly vector. Indoor residual spraying of insecticide (IRS) against these largely exophilic / exophagic vectors may not be the most effective method. A synthetic copy of the male sex-aggregation pheromone of the key vector species Lutzomyia longipalpis in the Americas, was co-located with residual pyrethroid insecticide, and tested for its effects on vector abundance, hence potential transmission, in a Brazilian community study. METHODS: Houses within …

The Gut Microbial Community Structure of the North American River Otter (Lontra canadensis) in the Alberta Oil Sands Region in Canada: Relationship with Local Environmental Variables and Metal Body Burden.

The Alberta Oil Sands Region in Canada is home to one of the largest oil bitumen deposits in the world. The North American river otter (Lontra canadensis) is a top predator with a small home range and is sensitive to disturbances; it has been designated as a sentinel species for the potential impacts of the natural resource exploitation on freshwater ecosystems in the Alberta Oil Sands Region. With an increasing interest in noninvasive biomarkers, recent studies suggest that gut microbiota can be used as a potential biomarker of early biological effects on aquatic wildlife. The goal of the pre…

Field evaluation of a 0.005% fipronil bait, orally administered to Rhombomys opimus, for control of fleas (Siphonaptera: Pulicidae) and phlebotomine sand flies (Diptera: Psychodidae) in the Central Asian Republic of Kazakhstan.

Plague (Yersinia pestis) and zoonotic cutaneous leishmaniasis (Leishmania major) are two rodent-associated diseases which are vectored by fleas and phlebotomine sand flies, respectively. In Central Asia, the great gerbil (Rhombomys opimus) serves as the primary reservoir for both diseases in most natural foci. The systemic insecticide fipronil has been previously shown to be highly effective in controlling fleas and sand flies. However, the impact of a fipronil-based rodent bait, on flea and sand fly abundance, has never been reported in Central Asia. A field trial was conducted in southeaster…

Emerging risk of visceral leishmaniasis in a non-endemic area of Southern Brazil: Sand fly survey and identification of a dog positive for Leishmania infantum using a kDNA-based PCR assay.

In the state of Santa Catarina (SC), Brazil, the first autochthonous case of canine visceral leishmaniasis (CVL) was reported in Florianópolis in 2010, which remains the only municipality with confirmed human and canine cases. The primary vector of Leishmania infantum, Lutzomyia longipalpis, has not yet been detected in SC, and data on sand fly fauna and CVL are scarce. This study employed a three-pronged approach in the non-endemic municipality of Biguaçu, adjacent to Florianópolis: (i) development of a PCR assay targeting the kDNA minicircle of the Leishmania donovani complex for detection o…

Sand fly fauna (Diptera: Psychodidae) from Serra da Mocidade Nacional Park: report of vectors and putative vectors of American cutaneous leishmaniasis and new records for the state of Roraima, Brazil.

Phlebotomine sand flies are insects of medical importance, as they are vectors of viruses, bacteria and trypanosomatids of the genus Leishmania Ross, the causative agent of leishmaniasis (Forattini 1973). During the past 20 years approximately 500,000 cases of leishmaniasis have been reported in Brazil. Most of these cases were reported in the North and Northeast regions. In the State of Roraima, almost 2,000 cases, including both cutaneous leishmaniasis (CL) and visceral leishmaniasis (VL), have been reported during the past three years (SINAN 2017). Usually the transmission by infected femal…
