Analysis of the nasal vestibule mycobiome in patients with allergic rhinitis.

Advances in culture-independent sequencing methods have been utilised in recent studies to understand the phylogenetic composition of the human microbiome of healthy and diseased skin. Allergic rhinitis (AR) is an inflammatory condition of the nasal cavity caused by environmental allergens. Although nasal microbial communities have been considered important contributors in human health, no studies to date have comprehensively compared fungal communities (mycobiome) of the nasal vestibule using the culture-independent pyrosequencing method. This study aimed to investigate how fungal communities

Microbiomes of the dust particles collected from the International Space Station and Spacecraft Assembly Facilities.

BACKGROUND: The International Space Station (ISS) is a unique built environment due to the effects of microgravity, space radiation, elevated carbon dioxide levels, and especially continuous human habitation. Understanding the composition of the ISS microbial community will facilitate further development of safety and maintenance practices. The primary goal of this study was to characterize the viable microbiome of the ISS-built environment. A second objective was to determine if the built environments of Earth-based cleanrooms associated with space exploration are an appropriate model of the

PSMε controls skin commensal CD8 + T cell activation.

Upon skin colonization, the prevalent human skin commensal S. epidermidis can elicit a CD8 + T cell response that protects against pathogens or clears tumors. The microbial features that drive this response are undefined, limiting our ability to understand and predict commensal-immune crosstalk and to engineer potent commensal-derived immunotherapies. To uncover these microbial features, we harnessed both the natural variation in CD8 + T cell induction across primary human isolates of Staphylococcus and our ability to genetically manipulate these strains. Stimulatory strains exhibit increased

High Humidity Exacerbates Psoriasiform Skin Disease Relapse by Increasing Tissue-Resident Memory T Cells via Altering Skin Microbiota.

Psoriasis recurrence remains a common and difficult medical problem, while environmental humidity appears to have an impact on psoriasis morbidity. However, it remains unknown whether and how high humidity impacts psoriasis relapse. Using unique mouse models of psoriasis relapse, we found that high humidity exposure exacerbated psoriasis recurrence by increasing skin-resident memory CD8+ T (TRM) cells through a mechanism depending on its upregulation of IL-15Rα on keratinocytes. Keratinocyte-specific knockout of IL-15Rα or administrating soluble sIL-15Rα to block IL-15 abrogated these effects.

Pou2f3 Modulates Mosquito Host-Seeking Behavior by Regulating Skin Microbiota and Their Production of Volatiles: Implications for Eco-Friendly Pest Control Strategies.

Skin microbiota-derived volatile metabolites are critical for mosquito host-seeking and disease transmission. Similar volatile organic compounds (VOCs)-mediated mechanisms may apply to agricultural pests (e.g., aphids, whiteflies). We demonstrate that Pou2f3, a skin-enriched transcription factor, regulates β-defensin1 expression, shaping microbiota composition and volatile production. Pou2f3-deficient mice exhibited increased Staphylococcus abundance and elevated 1-methoxy-2-propanol levels, enhancing attraction in Culex and Aedes mosquitoes. This VOC, alongside other microbiota-derived compou

Microbiota and Alzheimer's disease: mechanistic insights from a multi-organ perspective.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder driven by multifactorial mechanisms. Increasing evidence suggests that dysbiosis, a term denoting an imbalance in the composition of the microbiota, may play a pivotal role in the pathogenesis of AD across multiple bodily sites, including the gut, oral cavity, nasal passages, lungs, and skin. Microbial imbalances may promote neuroinflammation, immune dysfunction, and metabolic disturbances through complex host-microbiota networks. This review synthesizes current advances in the understanding of microbiota-driven modulation of

Topical maltotetraose alleviates chronic pruritus by restoring skin barrier function and cutaneous homeostasis in C57BL/6J mice.

Food-grade ingredients are attracting increasing attention in cosmetic applications owing to their safety, biocompatibility, sustainability, and multifunctionality. Although maltotetraose (G4) has been reported to possess moisturizing and potential anti-pruritic properties, systematic in vivo evidence supporting its efficacy against chronic pruritus remains limited. Therefore, this study evaluated the anti-pruritic effects of topical G4 in an acetone-ether-water (AEW)-induced chronic pruritus mouse model and characterized the associated changes in skin barrier function, oxidative stress, infla

A skin colonizer disrupts inflammatory and humoral immune defenses in hidradenitis suppurativa.

Hidradenitis suppurativa (HS) is a chronic inflammatory skin disease associated with a polybacterial dysbiosis devoid of a known pathogen. Here, we report that HS patients mount IgA and IgG responses against skin colonizers, notably Porphyromonas uenonis (Pu), a rare species selectively enriched in severe disease. In these patients, Pu foci are detected in the epidermis, surrounded by IgA deposits, and anti-Pu IgGs cross-react with self-antigens expressed by healthy keratinocytes. Using healthy human skin explants, we demonstrate that patient-derived Pu can cross an intact epidermal barrier, i

The role of skin microbiota in lichen planus from a Mendelian randomization perspective.

Lichen planus is a chronic skin lesion characterized by pruritic violaceous papules, which has a high risk of morbidity. Skin microbiota plays an important role in the maintenance of cutaneous mucosal barrier and human health and immune homeostasis. Studies have shown that skin microbiota may play a role in the pathogenesis of lichen planus, but it is not yet clear. MR studies have been performed to determine causal associations. Lichen planus samples from Finn database were extracted from published GWAS data, including 6,411 cases and 405,770 controls; skin microbiota samples were gathered fr
