Toxoplasma Effectors Targeting Host Signaling and Transcription.

Early electron microscopy studies revealed the elaborate cellular features that define the unique adaptations of apicomplexan parasites. Among these were bulbous rhoptry (ROP) organelles and small, dense granules (GRAs), both of which are secreted during invasion of host cells. These early morphological studies were followed by the exploration of the cellular contents of these secretory organelles, revealing them to be comprised of highly divergent protein families with few conserved domains or predicted functions. In parallel, studies on host-pathogen interactions identified many host signali

Current standing and frontiers of gene therapy for meningiomas.

Meningiomas are among the most common intracranial tumors. The treatment of choice for these lesions is complete resection, but in 50% of cases it is not achieved due to tumor location and/or surgical morbidities. Moreover, benign meningiomas have high recurrence rates of up to 32% in long-term follow-up. Molecular analyses have begun to uncover the genetics behind meningiomas, giving rise to potential genetics-based treatments, including gene therapy. The authors performed a literature review on the most relevant genes associated with meningiomas and both current and potential gene therapy st

Abnormalities of T cell signaling in systemic lupus erythematosus.

Systemic lupus erythematosus (SLE) is an autoimmune disease resulting from a loss of tolerance to multiple self antigens, and characterized by autoantibody production and inflammatory cell infiltration in target organs, such as the kidneys and brain. T cells are critical players in SLE pathophysiology as they regulate B cell responses and also infiltrate target tissues, leading to tissue damage. Abnormal signaling events link to defective gene transcription and altered cytokine production, contributing to the aberrant phenotype of T cells in SLE. Study of signaling and gene transcription abnor

Protective effect of RNA interference targeting Ikkβ on hepatic fibrosis in rats.

Nuclear factor-κB (NF-κB) is an important regulatory factor in cells. NF-κB has a wide range of biological activities. After activation, it participates in the transcription and regulation of many genes and plays a role in infection, inflammatory response, oxidative stress, cell multiplication, and apoptosis. The activation of the NF-κB signal pathway is dependent on the degradation of the IκB kinase β (IKKβ) complex. IKK β is the key kinase in the NF-κB activation pathway. After inhibition, it can block the activation of NF-κB. IKKβ is a key regulator of NF-κB activation, also an early regula

Correction of hyperbilirubinemia in gunn rats by surgical delivery of low doses of helper-dependent adenoviral vectors.

Helper-dependent adenoviral (HDAd) vectors are attractive for liver-directed gene therapy because they can drive sustained high levels of transgene expression without chronic toxicity. However, high vector doses are required to achieve efficient hepatic transduction by systemic delivery because of a nonlinear dose response. Unfortunately, such high doses result in systemic vector dissemination and dose-dependent acute toxicity with potential lethal consequences. We have previously shown in nonhuman primates that delivery of HDAd in surgically isolated livers resulted in a significantly higher

MECP2 isoform-specific vectors with regulated expression for Rett syndrome gene therapy.

BACKGROUND: Rett Syndrome (RTT) is an Autism Spectrum Disorder and the leading cause of mental retardation in females. RTT is caused by mutations in the Methyl CpG-Binding Protein-2 (MECP2) gene and has no treatment. Our objective is to develop viral vectors for MECP2 gene transfer into Neural Stem Cells (NSC) and neurons suitable for gene therapy of Rett Syndrome. METHODOLOGY/PRINCIPAL FINDINGS: We generated self-inactivating (SIN) retroviral vectors with the ubiquitous EF1alpha promoter avoiding known silencer elements to escape stem-cell-specific viral silencing. High efficiency NSC infect

Phosphoinositide 3-kinase (p110α) gene delivery limits diabetes-induced cardiac NADPH oxidase and cardiomyopathy in a mouse model with established diastolic dysfunction.

Phosphoinositide 3-kinase [PI3K (p110α)] is able to negatively regulate the diabetes-induced increase in NADPH oxidase in the heart. Patients affected by diabetes exhibit significant cardiovascular morbidity and mortality, at least in part due to a cardiomyopathy characterized by oxidative stress and left ventricular (LV) dysfunction. Thus, PI3K (p110α) may represent a novel approach to protect the heart from diabetes-induced cardiac oxidative stress and dysfunction. In the present study, we investigated the therapeutic potential of a delayed intervention with cardiac-targeted PI3K gene therap

Methodological Evaluation of a P2C-Based ReMOT CRISPR/Cas9 System in Aedes aegypti.

Mosquito-borne infectious diseases remain a major challenge to public health, highlighting the need for efficient and accessible gene editing approaches. Receptor-mediated ovary transduction of cargo (ReMOT) offers an alternative to embryonic microinjection, in which P2C, an ovary-targeting peptide, enables ovarian delivery of the editing components. However, key design parameters and operational boundaries of the P2C-based ReMOT system have not been clearly defined. Here, we performed a methodological evaluation of the P2C-mediated ReMOT CRISPR/Cas9 system in Aedes aegypti. Cas9-P2C fusion pr

Optimized AAV capsids robustly transduce airway epithelial cells.

Gene therapies have demonstrated transformative potential for a range of genetic disorders, including immunodeficiencies, hematopoietic conditions, and neuromuscular diseases. However, the application of these approaches to cystic fibrosis (CF) and other airway diseases remains constrained by the challenge of efficient gene delivery to target epithelial cells. Adeno-associated virus (AAV) vectors are widely used for in vivo gene delivery due to their favorable safety profile and capacity for long-term transgene expression in non-dividing cells. Nonetheless, current AAV capsids require high dos

AtZIK4, a MAPK kinase kinase, positively regulates phosphate uptake gene expression to modulate phosphate starvation tolerance in Arabidopsis.

The mitogen-activated protein kinase (MAPK) cascade is a highly conserved signal transduction module in eukaryotes. We investigated the function of ZIK4, a MAPK kinase, in the phosphate starvation response of Arabidopsis. Phenotypic analysis revealed that the zik4 mutant had stronger tolerance to phosphate starvation than that to the wild type. However, the kinase domain was still expressed in zik4. The CRISPR/Cas9 system was used to construct two independent knockout mutants zik4-c3 and zik4-c15 by targeting two different sites in the AtZIK4 kinase domain. Physiological measurements showed th

Genetic and epigenetic landscape of non-familial sudden cardiac death.

Sudden cardiac death (SCD) remains a major global health challenge, characterized by high incidence, low survival rates, and often presenting as the first manifestation of underlying disease. This review provides a comprehensive overview of the genetic and epigenetic landscape of non-familial SCD, emphasizing the multifactorial nature of this complex clinical endpoint. We summarize common single nucleotide polymorphisms and insertion/deletion variants associated with SCD, identified through candidate-gene-based studies and genome-wide association studies, highlighting risk variants clustered i

Rice cold tolerance: Physiological responses, molecular regulatory networks, and improvement strategies.

Low-temperature stress is a critical abiotic constraint that severely impairs rice yield and quality, posing a substantial threat to global food security-especially as rice, a staple food for more than half of the world's population, is inherently susceptible to cold stress. This review systematically summarizes the typical phenotypic and physiological impairments induced by cold stress across key rice growth stages, and elaborates on the multi-layered molecular regulatory networks underpinning rice cold tolerance, which integrate the entire process of cold signal perception, intracellular tra

TrkB Overexpression via Gene Therapy: Enhanced Optic Nerve Uptake With Associated Increases in Mitochondria and Axoplasmic Transport.

INTRODUCTION: Glaucoma is a progressive neurodegenerative disease that affects retinal ganglion cells (RGCs), ultimately leading to vision loss. In this study, we investigated gene therapy-mediated transduction of RGCs and examined axonal transport changes in the optic nerve using a viral vector designed to upregulate tropomyosin receptor kinase B (TrkB) expression. METHOD: TrkB expression was evaluated in retinae and optic nerves of rats following genetic intravitreal delivery of AAV2-TrkB. Axonal transport and preliminary mitochondrial changes were assessed in optic nerves by immunohistoche

Transient receptor potential channels as emerging therapeutic targets: mechanisms and therapeutic insights in inflammatory bowel disease.

Transient Receptor Potential channels (TRPs) are a family of non-selective cation channels widely distributed across diverse tissues and cells in mammals. They participate in numerous physiological processes, including environmental stimulus perception, signal transduction, neurotransmitter release, and insulin secretion. The involvement of TRPs in pain syndromes, neurodegenerative diseases, metabolic disorders, and tumorigenesis has established them as prominent targets in translational medicine. Inflammatory Bowel Disease (IBD), encompassing Crohn's disease (CD) and ulcerative colitis (UC),

Transcriptional changes in non-human primate tissues after intrathecal delivery of serotype 9 adeno-associated viral vector: Insights into organ toxicities.

Adeno-associated virus (AAV)-based gene transfer has brought transformative therapeutic benefits to patients with otherwise untreatable genetic diseases. However, treatment-related organ toxicities, particularly for high doses, remain a safety concern in the clinic with some translatability in preclinical species. In the present study, we conducted an RNA sequencing (RNA-seq) analysis in non-human primates administered intrathecally with scAAV9-CBA-GFP, empty viral capsid particles, or a "Promoterless" vector. This analysis revealed a broad and long-lasting (4 weeks after dosing) transcription

Two-Component MprAB System Regulates the Expression of Genes Involved in Cell Envelope Biosynthesis in Corynebacterium glutamicum.

To accurately detect internal and environmental cues, bacteria have evolved signal transduction pathways such as two-component systems (TCSs) to reprogram appropriate genetic and physiological functions for adaptation and survival. The MprAB TCS is commonly found in actinobacteria and has been associated with important processes such as mycobacterial virulence, nutrient starvation, and environmental stress, particularly cell envelope stress. However, a comprehensive investigation of the function and response network of the MprAB TCS in corynebacteria remains to be carried out. In this study, w

Transmembrane Protein 43: Molecular and Pathogenetic Implications in Arrhythmogenic Cardiomyopathy and Various Other Diseases.

Transmembrane protein 43 (TMEM43 or LUMA) encodes a highly conserved protein found in the nuclear and endoplasmic reticulum membranes of many cell types and the intercalated discs and adherens junctions of cardiac myocytes. TMEM43 is involved in facilitating intra/extracellular signal transduction to the nucleus via the linker of the nucleoskeleton and cytoskeleton complex. Genetic mutations may result in reduced TMEM43 expression and altered TMEM43 protein cellular localization, resulting in impaired cell polarization, intracellular force transmission, and cell-cell connections.
